Polymorphism Of A Deep Marine Benthic Bacterium From The Gulf Of Mexico

  • An interesting bacterium (#271) was isolated from the abyssal plain of the Sigsbee Deep of the Gulf of Mexico. The organism exhibits marked polymorphism (baciloid, coryneform and myceloid morphologies) in response to certain cultural conditions. the organism has been observed undergoing reproduction by transverse fission, fragmentation and arthrospore production. The presence of arthrospores indicates the bacterium is a member of the genus Arthrobacter; however, computed affinity coefficients do not confirm this genus. Until further studies have been completed on this isolate the authors are reluctant to place it in a generic group.

Inhibitory Effect of Transition Metal Gallium [Ga(NO3)3] on Biofilm Formation by Fish Pathogens (전이금속 갈륨(Ga(NO3)3)을 이용한 biofilm을 형성하는 어류질병세균의 억제)

  • The prevalence of pathogenic bacteria such as Streptococcus parauberis (Sp), Streptococcus iniae (Si), and Edwardsiella tarda (Et) in flounder fish farms in Jeju Island and their management by gallium treatment was studied. Sp, Si, and Et were found to exhibit a low rate of cell growth and high biofilm formation. Hence, in the present study, cell growth and biofilm formation were measured spectrophotometrically 72 h after the addition of different concentrations of gallium (2, 4, or 8 mg/ml). In addition, cell death was measured by resazurin and propidium iodide staining assays. The results showed that bacterial cell death increased and biofilm formation decreased with an increasing concentration of gallium. Hence, the present study signifies that the use of gallium against bacterial pathogens could be useful for disease management in flounder farms.

Variations in Species Composition, Biomass, and Density in Shrimp Trawl Bycatch Across Seasons and Tidal Phases in Southern Korean Waters: Developing a Fisheries Risk Management Approach

  • We quantitatively investigated compositions of fish and invertebrate bycatch in Korean shrimp fisheries. We sampled shrimp trawl bycatch from 48 commercial trawls over 1 year. These samples contained 108 taxa from 50 families, with 60 fish taxa contributing 51.2% of the total biomass and 48 invertebrate taxa 48.8%. However, 86.32% of the total individual organism density comprised invertebrates, while individual fishes made up the remainder. Within the bycatch, two fish species varied in population size structure by season, suggesting recruitment is seasonal in these taxa. Overall general catch characteristics (total bycatch biomass and organism density) varied significantly by season and tide, and a significant interaction effect was observed (season$\times$tide) on total density, but not on total biomass. The data collected will be used in designing a program of long-term bycatch monitoring.

Cyst formation of the marine ciliate, Vorticella sp. using MCCF medium (MCCF 배지를 이용한 해산 섬모충 Vorticella sp.의 cyst형성)

  • The ciliate, Vorticella was often observed in the rotifer mass culture tanks as common co-existing organism. This Vorticella performed as a predator for aquatic bacteria population in the rotifer mass culture tanks. This study was carried out to investigate a cyst formation medium of Vorticella in the laboratory for keeping Vorticella seed. The test organism Vorticella sp. was isolated from culture water of rotifer mass culture tanks. The cyst of Vorticella was formed by dried-method for the formation and maintainance of cyst. MCCF (Marine Ciliate Cyst Formation) medium was used for cyst formation (incystment), preservation and return to moving cell (excystment) of the marine ciliate, Vorticella sp. The cyst shape and size were ellipical type and $30.51 \pm1.98\;\mu$m (Avg. $\pm$ SD) of minor axis and $28.89 \pm2.12\;\mu$m (Avg. $\pm$ SD) of minor axis (n=10), The Vorticella cyst was kept in the room temperature ($10\~35^{\circ}C$) and total dark condition (24D:0L) during 1 year. The preserved cyst was transferred to moving cell state (excystment) only by the addition of fresh sea water in the MCCF medium. The five Vorticella sp. moving cells of excysted from cysts showed the growth up to 912$\pm$64 cells/10 ml in MCCF medium during the culture period of 16 days. This MCCF medium was very useful tool for cyst formation and species preservation of marine ciliate Vorticella.

Melon Growth Enhancement by Fucoidan and Fucoidan Decomposing Bacteria (후코이단과 후코이단 이용 박테리아의 멜론 성장 촉진 효과 검증)

  • BACKGROUND: Marine algae is a productive organism that is consumed as a nutritious food. However, large amounts of unused portions of the algae are incinerated as trash or dumped in the sea, causing pollution. Recycling algae is important for saving resources and conserving the environment. In this study, the fucoidan which is a major carbohydrate of marine algae was tested as a source of fertilizer for farming. METHODS AND RESULTS: The growth rate of the melon was examined after treating fucoidan and the melon growth factors, weight and length of stem were measured. To discover the mechanism of melon growth promotion of fucoidan, bacteria that decomposed fucoidan were isolated from soil and abalone. Bacillus wiedmannii and Stenotrophomonas pavanii were isolated from terrestrial soil and Pseudomonas sp. was isolated from abalone. Among these three bacteria, Pseudomonas sp. had the highest and most specific fucoidan-decomposing activity. When Pseudomonas sp. was treated with fucoidan on melon-growing soil, the growth of melon was relatively improved compared to the treatment with fucoidan alone. CONCLUSION: We found that fucoidan, the main carbohydrate of marine algae, promoted melon growth. Fucoidan-decomposing microorganisms were isolated from terrestrial soil and marine organism, and we found that these bacteria stimulated the effect of melon growth promotion of marine algae. This is the first report that confirms the fertilizer effect of marine algae and shows the use of bacteria with marine algae.

Microbiological Pollution Investigation of Spring water in Seogwipo city of Jeju Island (제주도내 서귀포시 해안가 저지대용천수의 미생물학적 오염도 조사)

  • The bacteriological examination of spring water in Seogwipo-city was conducted. A total 11 spring water samples were performed from January to April, 2010. During the study period, the range of temperature was from 0.7 to $15.4^{\circ}C$, and result of the analyses showed that hydrogen ion concentrations (pH) for spring water was 0.33 to 7.8. salinity levels for sample average of 0.5 to 1.04‰, as the result of measuring dissolved oxygen (DO) for spring water showed that water dissolved oxygen were 1.02 to 7.14 mg/${\ell}$. The range of total coliform of spring water sample at 11 stations located in the designated spring water were <1.8~>1,600 MPN/100mL. And the range of geometric mean of total coliform were 1.9~117.1 MPN/100mL, The range of fecal coliform of spring water sample at 11 stations located in the designated spring water were <1.8~>1,600 MPN/100mL. And the range of geometric mean of fecal coliform were 1.8~68.1 MPN/100mL, respectively. Level of microbial contamination was examined in 11 samples for indication of bacterial contamination such as heterophic bacteria, EscherichiacoliO157;H7, salmonella spp., Vibrio parahaemolyticus, Staphylococcus aureus, Shigella spp. Were frequently detected from the spring water. Salmonella spp., S.aureus were detected in the range of $0{\sim}1.0{\times}10^1$, $0{\sim}3.0{\times}10^1$ CFU/ml, respectively. And the Escherichia coli O157;H7, Vibrio parahaemolyticus, Shigella spp. Were not detected from the examined spring water samples.

  • The purpose of this study is to research on founding of the department of marine leisure industry in fisheries high school. Research methods were used a survey district, students and teachers in fisheries high school and industry human are 353 people in the target. The results of this research are as follows. First, the ratio in favor of making the marine leisure industrial department had more than 81%. The second, teachers and industry people selected core factors include oceanography, water leisure, leisure diving, commercial diving, lifeguard, rafting. Finally, subject for the department of marine leisure industry in fisheries high school are fisheries shipping industry basis, understanding of the ocean, marine leisure tourism, underwater technology, navigation, seamanship, maritime laws regulations, heat engines and aquatic organism.

  • As methods reducing damages by jellyfish which enter the coastal areas of Korea, attaching cutting devices to towing nets of otter trawls or pair trawls and/or using a canvas type of cutting nets of small fishing boats have been widely utilized. In order to reduce shut-down damages of power plants in coastal areas due to the mass influx of marine organisms including jellyfish, a possible improvement of the traveling water screen system and various jellyfish influx blocking devices were suggested in this study. The results could be utilized as an important index for reducing damages by jellyfish bloom which cause on a massive scale in summer in Korea.

  • Copepod is useful live food organism in the larval rearing. However of fin fish, culcures of copepod species (Specially calanoida and cyclopoida) were very difficult under artificial management conditions. In this study, we report successful culture of a free swimming cyclopodia copepod species (Apocyclops sp.) by easily baker's yeast as food. The maximum culture density of Apocyclops sp. was observed 11,200±2,660 ind/ℓ during the 30 days culture periods in the 1.5ℓ culture tanks. The nauplii were growth up maximum density 6,400±432 ind/ℓ on day 16th. The number of egg carring female was observed maximum density (666.7 ind./ℓ) of two times after 6days and 28 days of initial culture.

  • Upon gene inactivation in animal models, the zebrafish (Danio rerio) has become a useful model organism for many reasons, including the fact that it is amenable to various forms of genetic manipulation. Genome editing is a type of genetic engineering in which DNA is inserted, deleted, modified, or replaced in the genome of a living organism. Mainly, CRISPR (clustered regularly interspaced short palindromic repeats) Cas9 (CRISPR-associated protein 9) is a technology that enables geneticists to edit parts of the genome. In this study, we utilized this technology to generate an mmp15b mutant by using zebrafish as an animal model. MMP15 is the membrane-type MMP (MT-MMP) which is a recently identified matrix metalloproteinase (MMP) capable of degrading all kinds of extracellular matrix proteins as well as numerous bioactive molecules. Although the newly-established mmp15b zebrafish mutant didn't exhibit morphological phenotypes in the developing embryos, it might be further utilized to understand the role of MMP15 in liver-related diseases, such as liver fibrosis, and associated pathogeneses in humans.
