황색포도상구균 분리배지 비교 (Comparison of Four Different Isolation Media for Staphylococcus aureus)

  • 황색포도상구균 분리에 사용되고 있는 난황첨가 만니톨 식염한천배지, Baird-Parker 한천평판배지, Baird-Parker 한천평판배지(RPF) 및 Petrifilm에 대하여 성능 비교실험을 실시하였다. 민감도는 30종의 황색포도상구균에 대하여 분리배지의 특징적인 콜로니를 형성하는 비율로 측정하였는데 Baird-Parker 한천평판배지(RPF)> Petrifilm> Baird-Parker 한천평판배지> 난황첨가 만니톨 식염한천배지의 순으로 나타났다. 황색포도상구균이 아닌 균주를 이용하여 측정한 선택성은 4종 분리배지 모두 위양성을 나타내지 않았다. 인위적으로 접종한 식품에서의 회수실험은 4종 배지 모두 회수 효율이 비슷한 것으로 측정되었다. 수산건제품에서 황색포도상구균으로 추정되는 콜로니를 분리하여 4종 배지에서 위양성, 위음성 발현여부를 측정하였다. 그 결과, 황색포도상구균 이외의 대부분 분리균주에서는 위양성이 나타나지 않았지만 S. carnosus ssp carnosus의 경우 난황첨가 만니톨 식염한천배지, Baird-Parker 한천평판배지와 Petrifilm에서 위양성을 나타내었다. 그러나 Baird-Parker 한천평판배지(RPF)에서는 위양성 반응이 나타나지 않아 실험에 공시된 배지 중에서는 선택성이 상대적으로 높은 것으로 판단되었다.

천일염 생산공정별 미생물 분포 조사 및 호염미생물 동정 (Distribution and Identification of Halophilic Bacteria in Solar Salts Produced during Entire Manufacturing Process)

  • 우리나라 전남지역에서 생산되는 천일염의 생산공정별 미생물 분포 조사 및 배양 분리법을 이용하여 호염미생물을 분리하여 동정하는 것을 이번 연구의 목표로 삼았다. 천일염 시료는 생산지를 고려하여 육지 염전인 영광군 한 지역과 해안 염전 지역인 신안군 두 군데에서 생산공정별로 세분화하여 총 28개 분석시료를 수집하여 사용하였으며, 이들 시료를 십진 희석하여 4종류의 미생물 분리용 배지에 도말, 배양한 후 나타난 집락(colony)을 계수하였다. 그 결과 천일염 생산 공정 중 대장균 등의 식품 오염지표 미생물은 검출되지 않았지만, 저장수에서 증발지 단계로 진행되면서 $1.1{\times}10^3{\sim}1.8{\times}10^5$ CFU/g의 호염성 미생물들이 검출되었다. 그러나 이후 단계인 함수저장고, 결정지에서는 미생물 수가 점차적으로 감소되었으며, 소금저장소에 보관된 천일염 시료에서는 미생물이 전혀 검출되지 않았다. 이들 분리균들은 형태학적 특성에 따라 무작위로 62개 집락을 선정하여 분리하였다. 순수 분리된 미생물들은 PCR 기법을 통해 16S rRNA 유전자의 염기서열을 분석한 후, 기존에 보고된 미생물 유전자 database와 비교함으로써 12속의 천일염 유래 호염균들의 존재를 확인하였다. 이번 연구 결과 천일염 생산 단계에서 분리된 halophilic bacteria은 Halobacillus, Halomonas, Bacillus, Idiomarina, Marinobacter, Pseudoalteromonas, Vibrio, Salinivibrio, Virgibacillus, Alteromonas, Staphylococcus 및 un-known 등이다.

항암 화학요법중인 급성 골수성 백혈병 환자의 구강내 세균변화에 관한 연구 (Changes in the Oral Microflora in Patients with Acute Myeloid Leukemia during the Period of Induction Therapy)

  • To investigate the changes in aerobic and facultative anaerobic oral microflora during remission-induction chemotherapy in patients with acute myeloid leukemia, 10 consecutive patients were studied during a period of 28 days. One day before, during and after the induction therapy, patients were given 10% Betadine solution for mouthrinses after breakfast and kept from eating and drinking. After 3 hours, paraffin-stimulated whole saliva was obtained for 2 minutes and transported to the laboratory. The samples were dispersed and homogenized by use of vortex mixer for 20 seconds. From these samples 10-fold serial dilutions (from 10-1 through 10-3) were prepared. Each dilution of 0.1 ml was plated on duplicate set of one nonselective medium (Blood agar) and four selective media (Sabourauds dextrose agar, Mannitol salt agar, Mac-Conkey agar, SF medium ) using applicator woods. All agar plate were incubated at 37$^{\circ}C$ for 48 hours. The total number of microorganisms was calculated and the percentage distribution of the various microorganisms from each specimen was drawn. 1. The salivary flow rate decreased by 66%, going from 5.38 ml/2min to 1.81 ml/2min over two days during the chemotherapy. 2. The total number of microorganisms in saliva increased by 22%, going from 4.88$\times$105/ml to 6.00$\times$105/ml over two days during the chemotherapy. 3. The salivary flow rate and the total number of microorganisms in saliva were recovered within 28 days after the chemotherapy. 4. The quantitative alteration in oral Enterobacteria, Enterococci, Staphylococci, Cndida during the chemotherapy had no statistical significance. 5. In saliva of the patients with acute myeloid leukemia who ahd intraoral ulcer, Enterobacteria was quantitatively predominent. Our study suggests that chemotherapy-induced transient xerostomia may induce acute oral infection. Consequently, the use of saliva substitute, the removal of intraoral infection source and the consistent oral hygiene care seem to be required to avoid the transmission of potential pathogenes in this group of patients.

Isolation and Identification of Staphylococcus sp. from Korean Fermented Fish Products

  • In order to find out if staphylococci occur in significant numbers in Korean fermented fish products, a total of 40 different fermented fish products were collected from different markets in Korea and analyzed for their physico-chemical and microbiological states. The pH, salt concentration and water activity of the products were measured and the total viable cell count and the number of Staphylococcus grown on mannitol salt agar were determined. The identification of the strains of Staphylococcus were made by API Staph Strip and MIS identification kits, and the physiological properties of the identified strains were further characterized by different conventional methods. The pH, salt content and water activity of fermented fish samples varied widely from 4.8 to 7.1, 7.4-28.7$%$ and 0.77-0.84, repectively, depending on the type of product. The total viable cell count varied from $10^4-10^9$ cfu/ml, and most of the samples had $10^5-10^6$ cfu/ml No correlation was found between the viable cell count and the pH, NaCl concentration and water activity of the samples. Among the 35 colonies identified as Staphylococcus strains by the identification kits, S. xylosus was the most frequently occurring strain marking 17, and S. warneri was 8, S. epidermidis 4 and S. cohnii 2. S. hominis, S. saprophyticus, S. haemolyticus and S. aureus were also identified once each. In some samples (K-3, P-6, K-8, G-5 and G-10), 2-3 different species of Staphylococcus were found. Considering the region of sampling, among the 10 samples from Kunsan 5 were identified as S. warneri, while in the other regions S. xylosus was predominant. Although the physiological characteristics of the identified strains were generally consistent with those in Bergey's Manual, some discrepances were also observed. All the strains were highly salt tolerant, growing in the media containing over 18$%$ NaCl. All the strains except S. aureus (G-11) showed negative in hemolysis activity, plasma coagulation and DNase tests. All the strains including S. aureus (G-11) showed negative in enterotoxin test.

농산물 중 Staphylococcus aureus의 분리를 위한 선택배지 평가 (Evaluation of Selective Media for Isolation of Staphylococcus aureus from Agricultural Products)

  • 본 연구는 농산물로부터 S. aureus분리에 적합한 배지를 선발하고자 난황첨가 mannitol salt agar (MSA), 난황과 tellurite과 첨가된 Barid-Parker agar (BPA), 토끼혈장이 첨가된 Barid-Parker agar (BPA+RPF), 3M petrifilm Staph Express count plates (Petriflm) 및 CHROMagar Staphylococcus aureus (CSA)의 민감도, 특이도, 손상된 세포에 대한 회복능, 농산물로부터 회수율에 대하여 비교 실험을 하였다. 민감도와 특이도를 조사하기 위하여 S. aureus 21주, Stapylococcus spp. 20주, 그 외 식중독균 33주를 각 선택배지에 접종하였으며 손상된 세포에 대한 회복능은 조사를 위하여 열($60^{\circ}C$, 90초), 산(1% lactic acid, 10분), 저온($-20^{\circ}C$, 1시간) 처리하였다. 또한 양상추, 토마토, 고추에 S. aureus를 4.0 log CFU/g농도로 접종하고 각 선택배지에 도말하였다. 민감도는 BPA+RPF(100%) = CSA(100%)=petrifilm(100%) > MSA(90.5%) > BPA(90.5%), 특이도는 BPA+RPF(100%) = CSA(100%) > MSA(84.6%) > BPA(75.0%) > petrifilm(67.3%) 순으로 나타났다. 한편, 손상된 세포의 회복능과 농산물에서 회수율은 5종의 배지에서 차이가 없었다. 따라서 농산물 중 S. aureus를 분리할 시 BPA+RPF와 CSA를 사용하는 것이 가장 효과적일 것으로 판단된다.

Isolation Rate of Methicillin-Resistant Staphylococcus aureus (MRSA) from Nasal cavity inferior regions and Cellular phones

  • Nosocomial infection and community-acquired infection with Staphylococcus aureus, especially methicillin-resistant S. aureus (MRSA), has become a strong concern in human body sites and related effects. The aim of this study is investigate the isolation rate of MRSA from nasal cavity inferior regions and cellular phones to assess the risk factor of nosocomial infection and community-acquired infection. 34.7% and 37.2% isolates were MRSA from the nasal cavity inferior regions and cellular phones according to a Mannitol salt agar (added oxacillin $6{\mu}g/mL$) culture and PCR according to S. aureus specific 16S rRNA and mecA primers. Thus, the distribution of S. aureus and the isolation rate of MRSA represent a very high risk factor regards nosocomial infection and community-acquired infection.

Mathematical Models to Predict Staphylococcus aureus Growth on Processed Cheeses

  • 본 연구는 가공치즈에서 Staphylococcus aureus의 생장을 예측하기 위한 수학적 모델을 개발하였다. 모짜렐라 슬라이스 치즈와 체다 슬라이스 치즈에 S. aureus 혼합균액(ATCC13565, ATCC14458, ATCC23235, ATCC27664, NCCP10826) 0.1 ml (log CFU/g)을 접종한 후 $4^{\circ}C$ (1440 h), $15^{\circ}C$ (288 h), $25^{\circ}C$ (72 h), and $30^{\circ}C$ (48 h)에 저장하면서 총 세균수와 S. aureus 세균수를 tryptic soy agar와 mannitol salt agar를 이용해 각각 확인하였다. S. aureus의 세균 수를 Baranyi model로 분석하여 생장률(${\mu}_{max}$; ${\log}CFU{\cdot}g^{-1}{\cdot}h^{-1}$), 유도기(LPD; h), 초기 세균 수(log CFU/g), 최대 생장 세균수(log CFU/g)를 계산함으로써 1차 모델을 개발하였다. 또한 저장온도와 S. aureus의 ${\mu}_{max}$, LPD의 관계를 분석하기 위해 square root model과 exponential decay model을 이용하였고 이를 통해 2차모델을 개발하였으며 개발된 모델의 평균제곱근 편차(RMSE)를 계산하여 적합성을 검증하였다. $4^{\circ}C$에서는 모든 가공치즈에서 황색포도상구균의 생장이 관찰되지 않았으나 $15^{\circ}C$, $25^{\circ}C$, $30^{\circ}C$에서는 모짜렐라 슬라이스와 체다 슬라이스 치즈에서 황색포도상구균이 생장하였으며($R^2=0.785-0.996$) 저장온도가 높아짐에 따라 생장률은 증가한 반면 유도기는 감소하였다($R^2=0.879-0.999$). 또한 개발된 모델의 RMSE 값은 0.3500-0.5344로 적합하였다. 따라서 본 연구결과는 가공치즈에서 황색포도상구균의 생장 예측에 유용하게 사용될 것이다.

배양검사를 통해 확인한 중환자실 직원의 손씻기 효과 (Hand Hygiene Effects Measured by Hand Culture in Intensive Care Unit)

  • The effect of hand hygiene was measured by hand culture before and after hand hygiene for 86 nurses, doctors, and nurses aide/housekeepers in Surgical Intensive Care Unit. The subjects were asked to press their dominant hand in hand-shaped Mannitol salt agar immediately after patient contact and then washed their hand by preferred hand hygiene agents [soap and water, waterless alcohol gel, or 4% chlorhexidine gluconate detergent (CHG)], and cultured one hand again Amount of isolated microorganism was calculated by counting the number of divided areas ($1{\times}1cm$) which is culture positive in hand culture plate. The amount of microorganisms were significantly reduced from 58.1(${\pm}38.59$) to 27.4(${\pm}30.4$) cells after hand hygiene. The staff nurse's hand hygiene was more effective compared to medical doctors and nurses aide/housekeepers. Methicillin-resistant Staphylococcus aureus(MRSA) was isolated in 41(47.1%) subjects ; but only removed 100% in 28(32.2%) subjects. When the amount of hand microorganisms was compared by subject's preferred hand hygiene agents, it was decreased in order of 4% CHG, waterless alcohol solution, soap and water, and water. The hand hygiene practice was inadequate to reduce hand microorganisms and significantly different by occupations. Further research and development of hand hygiene improvement program which emphasize the quality of hand hygiene is recommended.

Prevalence and Molecular Characterization of Methicillin-Resistant Staphylococcus aureus from Nasal Specimens: Overcoming MRSA with Silver Nanoparticles and Their Applications

  • Staphylococcus aureus is a cause of high mortality in humans and therefore it is necessary to prevent its transmission and reduce infections. Our goals in this research were to investigate the frequency of methicillin-resistant S. aureus (MRSA) in Taif, Saudi Arabia, and assess the relationship between the phenotypic antimicrobial sensitivity patterns and the genes responsible for resistance. In addition, we examined the antimicrobial efficiency and application of silver nanoparticles (AgNPs) against MRSA isolates. Seventy-two nasal swabs were taken from patients; MRSA was cultivated on Mannitol Salt Agar supplemented with methicillin, and 16S rRNA sequencing was conducted in addition to morphological and biochemical identification. Specific resistance genes such as ermAC, aacA-aphD, tetKM, vatABC and mecA were PCR-amplified and resistance plasmids were also investigated. The MRSA incidence was ~49 % among the 72 S. aureus isolates and all MRSA strains were resistant to oxacillin, penicillin, and cefoxitin. However, vancomycin, linezolid, teicoplanin, mupirocin, and rifampicin were effective against 100% of MRSA strains. About 61% of MRSA strains exhibited multidrug resistance and were resistant to 3-12 antimicrobial medications (MDR). Methicillin resistance gene mecA was presented in all MDR-MRSA strains. Most MDR-MRSA contained a plasmid of > 10 kb. To overcome bacterial resistance, AgNPs were applied and displayed high antimicrobial activity and synergistic effect with penicillin. Our findings may help establish programs to control bacterial spread in communities as AgNPs appeared to exert a synergistic effect with penicillin to control bacterial resistance.

콜라겐 함유식품의 미생물 성장특성 연구 (Microbial Growth Characteristics on Foods Containing Collagen)

  • 콜라겐 함유량이 높은 한식메뉴에 대한 미생물학적 위해 평가를 위해 S. aureus와 Salmonella를 삼계탕, 어글탕, 오돌뼈 볶음, 전약, 족발에 접종하여 다양한 저장온도에서 세균수를 관찰하였다. $4^{\circ}C$의 저장온도에서는 5가지 콜라겐 함유식품에서 세균의 성장이 관찰되지 않았고, $10^{\circ}C$의 경우 삼계탕과 족발이 다른 식품에 비해 세균이 생장하는 패턴을 나타냈다. 저장온도 $20^{\circ}C$에서는 전약을 제외한 나머지 식품에서 세균이 생장하였고, $30^{\circ}C$에서도 전약만 세균수가 감소하는 것으로 관찰되었다. 5가지 콜라겐 함유식품 중 전약만 고온 저장온도에서 위해세균의 활성이 관찰되지 않고 감소하는 경향을 나타냈기 때문에 전약에 첨가된 정향, 대추, 계피, 생강, 후추에 대한 항균활성을 실험하였다. 그 결과 정향, 계피, 후추, 생강이 S. aureus에 대해 항균성을 보였고, Salmonella에 대해서는 정향과 계피만 항균력이 나타났으며, 대추는 항균능력이 없는 것으로 나타났다.
