Ocular transport of hydrophilic drugs: Enhancement of the paracellular penetration across cornea and conjunctiva in the rabbit (수용성약물의 안점막 투과기전에 관한 연구: 토끼의 각막 및 결막 세포간극경로의 투과촉진)

  • The objective of this study was to determine whether 4-phenylazobezyloxycarbonyl-Pro-Leu-Gly-Pro-D-Arg (Pz-peptide), an enhancer of hydrophilic solute permeability in the intestine, could elevate the paracellular permeability of hydrophilic drugs across cornea and conjunctiva in the rabbit. The in-vitro penetration of hydrophilic drugs (mannitol, atenolol) and lipophilic drug (propranolol) across the rabbit cornea and conjunctiva was studied either in the presence or absence of 3 mM Pz-peptide. Drug penetration was evaluated using the modified Ussing chamber. The conjunctiva was more permeable than the cornea to all drugs. Pz-peptide showed enhanced effects on the drug transport across cornea and conjunctiva in a concentration dependent manner. Effects or ion transport inhibitor on the mannitol penetration were then investigated. Mannitol penetration was not changed by serosal addition of $100\;{\mu}M$ ouabain, suggesting that $Na^+/K^+$ ion tranporter was not involved in the Pz-peptide induced elevation of paracellular drug permeability. Furthermore, effects of Pz-peptide and EDTA on the transport of atenolol and propranolol into the ocular tissues or blood circulation after its administration into both eyes were investigated. EDTA showed enhanced effect on propranolol transport into the ocular tissues, but Pz-peptide did not show significant difference. Systemic absorption of propranolol by the addition of EDTA or Pz-peptide was not changed. On the other hand, EDTA and Pz-peptide elavated the atenolol transport into the ocular tissues. The transport of atenolol into the blood circulation was also enhanced by the addition of EDTA, but no effect was observed by the addition of Pz-peptide. The above findings suggest that Pz-peptide would be used as an paracellular pathway enahncer of hydrophilic drugs into the eye, without affecting the systemic absortion of topically applied opthalmic drugs.

Standardization of Chemically Defined Medium for the Production of Mycelium and Basidiocarps in Flammulina velutipes (팽나무버섯 균사체 및 자실체 생산을 위한 화학합성배지의 최적화)

  • Nutritional requirements for the growth of Flammulina velutipes were studied. Mannitol, glutamic acid and ammonium nitrate were chosen for the maximum mycelial growth when various carbon and nitrogen sources tested. Optimum C : N ratio for the mycelial growth was 20 : 1. Potassium dihydrogen phosphate was selected among the phosphate sources. Magnesium sulphate and thiamine HCl stimulate mycelial growth. Final compositions of optimized chemically defined medium were 1.5% mannitol, 0.082% $NH_4NO_3$, 0.312% glutamic acid, 0.25% $KH_2PO_4$, 0.06% $MgSO_4{\cdot}7H_2O\;and\;0.3\;{\mu}g/l$ thiamin HCl. This medium not only support mycelial growth but also induce fruit body formation.

  • Granulocyte-macrophage colony-stimulating factor (GM-CSF) is a cytokine that stimulates the production of granulocytes, macrophages, and white blood cells. The effects of osmotic pressure on secretion of human GM-CSF into the culture medium were investigated in suspension cultures of transgenic tobacco cells. An increase in osmotic pressure caused by the addition of mannitol decreased the cell size index, with the effect being more pronounced when cells were measured wet rather than dry. Increased osmotic pressure enhanced the secretion of hGM-CSF. At 90 g/L mannitol, the maximum concentration tested, hGM-CSF was present in the culture medium at 980 ug/L. As the concentration of mannitol increased, the total amount of protein secreted also increased, but was disproportionately enriched in GM-CSF NaCl, another osmoticum, had very similar effects on cell growth and hGM-CSF production, but did not cause enrichment for hGM-CSF Additionally, protein-stabilizing polymer was added to culture broth to enhance stability of secreted recombinant protein. Finally, above two method were applied together to maximize the productivity.

  • The ripe fruit of Hass avocado contains one of the highest elvels of cytochrome P-450 protein found in the plant kingdom. To determine whether wounded roots of avocado contain P-450 protein, the roots of avocado were wounded by slicing, and then allowed to incubate in sealed plastic bags, in 0.4M mannitol, and in the solution to make protoplast preparation containing cellulysin and macerase during the specified times. The microsomal proteins were extracted from the samples, separated by SDS-PAGE, and then subjected to Western blot analysis using polyclonal antibodies which are generated against the CYP71A1 protein. wounded roots in sealed bags produced CYP71A1 within 6 hours after cutting, and those in 0.4M mannitol did not produce CYP71A1 even after 72 hours, but those in the protoplast preparation by cellulysin and macerase induced and produced CYP71A was induced in only 24 hours. These results indicate that CYP 71A1 plays a role for wound healing for root tissue o avocado, and would-inducible P-450 protein was not detected in the mannitol solution by preventing a synthesis of ethylene in a liquid state, but the softening of tissues by cellulysin and macerase to make protoplast preparation was involved in an activation of CYP 71A1 even in the liquid state.

Optimized Production of Biosurfactant by the Indigenous Bacterium, Pseudoalteromonas sp. HK-3 Originating from Oil-Spilled Areas (유류누출 지역에서 유래한 토착세균, Pseudoalteromonas sp. HK-3 배양에서 생물계면활성제의 최적 생산)

  • The principal objective of this study was to determine the optimal conditions for the production of biosurfactant by the indigenous bacterium, Pseudoalteromonas sp. HK-3, originating from oil-spilled areas. The relationship between total biosurfactant production and the factors affecting biosurfactant production were evaluated by statistical analysis using SPSS software. The effects of various supplemental carbon sources (e.g., glucose, dextrose, mannitol, citrate, acetate) on the maximal production of biosurfactant by the test culture of Pseudoalteromonas sp. HK-3 was then evaluated. As a result, mannitol was found in this study to be the best supplemental carbon source for the production of biosurfactant. A spot inoculation of crude cultural liquid containing the HK-3 cells generated the largest clear zone, whereas only small clear zones appeared around the spots inoculated with either supernatant only or cell pellets following centrifugation. Our results demonstrated that the HK-3 test culture supplemented with 2% mannitol at an initial pH of 6 generated the maximal amount of biosurfactant within 72 h of incubation.

  • LPC analogues including natural and unnatural LPC, 3-L-2-PC, acetylated LPC and ethylene glycol derivative are prepared from D-mannitol using in convenient procedures by only changing the synthetic sequences, and their protective activities against cecal ligation and puncture (CLP)-induced severe sepsis are compared. The chirality at C2 position in LPC is found to be required as (S)-configuration for sepsis inhibition, comparing from the protection activity between LPC 6 and unnatural LPC 8. The hydroxyl functionality is also very important and required at C2 or C3 position as shown in the protection activities of ethylene glycol analogue 11 and 3-L-2-PC 9.

Fundamental study on development of latent heat storage material for waste heat recovery of biomass gasification

  • Recently, latent heat thermal energy storage system (LHTES) has gained attention in order to utilize middle temperature (373~573 K) waste heat from biomass gasification. This paper has investigated thermo-physical properties of erythritol [$CH_2OHCHOH$ $CHOHCH_2OH$], mannitol [$CH_2OH$ $(CHOH)_4CH_2OH$] and their compounds as phase change materials (PCMs). The differential scanning calorimetry (DSC) was applied to measure the melting point and latent heat of these PCMs. Also the melting and solidification characteristics of these PCMs were observed in a glass tube with a digital camera. In the DSC measurement, when the amount of mannitol content was more than 40 mass%, the melting point of these compounds show two melting points. The experimental results showed that the velocity of melting and solidification were different for every mixture ratio of compounds. These compounds had the super-cooling phenomenon during the solidification process.

Wort Fermentation by Leuconostoc citreum Originated from Kimchi and Sensory Properties of Fermented Wort

  • Fermentation of wort was investigated with an ultimate goal to develop a fermented beverage rich in prebiotics and functional ingredients as well as desirable in flavors. Wort was fermented with Leuconostoc citreum HJ-P4 originated from kimchi and subjected to sensory descriptive analysis. L. citreum HJ-P4 produced various organic acids (e.g., lactic acid, acetic acid) as well as functional sugars (e.g., mannitol, panose) during wort fermentation. The concentration and ratio of lactic acid and acetic acid were significantly influenced by roasting conditions of malts used for wort preparation and aeration conditions during fermentation. The concentration of mannitol and panose varied depending on the sucrose content of wort and aeration conditions. Sensory characteristics of the fermented worts were clearly differentiated according to the roasting conditions of malts used for wort preparation and aeration conditions during fermentation. These results indicate that metabolite concentration of fermented wort and its sensory properties can be manipulated with roasting conditions of malts and fermentation conditions.

  • The optically active glycerol acetonides are often used as important chiral intermediates for many syntheses. In connection with the development of inhibitors of phospholipases, we have compared the synthetic routes to (S)-and (R)-glycerol acetonide from D-mannitol and D-isoascorbic acid, and L-serine and L-ascorbic acid, respectively. In our hands, the conversions of L-serine to (R)-glycerol acetonide and of D-mannitol to (S)-glycerol acetonide were found to be most effective.

A Study on the Physical and Chemical Propeties of Hydrous Aluminum Oxide (합성알루마나수화물의 물리화학적 성질에 관한 연구)

  • Physical and chemical properties on the aging inhibition mechanism of hydrous aluminum oxide were studied by means of dehydration velocity, activation energy, DTA, TGA, IR spectra, X-ray diffraction and TMA. During aging, changes may occur in the hydrous aluminum oxide structure which results in a loss of acid reactivity and in crystal formation to the hydrated hydrous alumina. The results obtained from the X-ray diffraction pattern and DTA, TGA thermogram studies showed that the aging product stabilized with either sorbitol or mannitol was hydrous aluminum oxide ($Al_{2}O_{3}{\cdot}xH_{2}O$) but the aging product not stabilized with either sorbitol or mannitol product not stabilized was hydrated hydrous aluminum oxide $Al_{2}O_{3}{\cdot}xH_{2}O{\cdot}yH_{2}O$. The activation energy of dehydration of the hydrous almina was about 17 Kcal. mol$^{1}$ deg$^{-1}$ which was observed a little less than that of 22 kcal.mol.$^{-1}$ deg.$^{-1}$ of or mannitol, the inhibition mechanism in the aging process from oxide is assumed to prevent the formation of the hydrated hydrous aluminum oxide and the aging process is thought of as analogous to the polymorphic transformations which occur as a system converts to its most stable state.
