Aspergillus awamori 가 생산하는 konjak mannan 분해효소에 관한 연구 (A Study on the Konjak Mannan-hydrolyzing Enzymes from Aspergillus awamori)

  • Aspergillus awamori가 생산하는 konjak mannan 분해 효소군(酵素郡)에 관한 연구로서 효소생산조건, 효소의 정제 및 성질, 효소분해에 미치는 감마선 조사(照射)의 영향을 조사하여 다음과 같은 결과를 얻었다. 1) 보존균주(保存菌株) 80여개중 konjak mannan 분해효소를 다량 생산하는 것으로 A. awamori를 선정하였다. 2) A. awamori를 밀기울 고체배양하는 경우 최적 배양일수는 3일, 살수(撒水)의 pH는 4, 살수량(撒水量)은 100%가 가장 좋았다. 3) A. awamori를 액체 진탕 배양하는 경우 최적 배양일수는 6일, 기본배지(基本培地)에 대한 추가탄소원(追加炭素源)으로는 xylose 0.1%, 질소원(窒素源)으로는 peptone 0.04%가 가장 좋았다. 4) A. awamori의 고체배양물에서 konjak mannan분해효소를 유안염석(硫安鹽析) 및 DEAE-Sephadex column chromatography에 의하여 정제(精製)하고 fraction I과 fraction II를 분리하였다. 5) fraction I과 fraction II의 작용최적 pH는 모두 4이었고 pH 안정(安定)범위는 각각 $3.5{\sim}5$$3{\sim}6$이었으며 konjak mannan에 대한 가수분해도는 각각 9%, 50%, guar gum에 대한 것은 각각 14%, 11%이 었 다. 6) 조효소액(粗酵素液)에 의한 konjak mannan의 가수분해도는 0.5 Mrad이상의 감마선 조사(照射)에 의하여 약간 촉진되었으며 특히 수화(水和)상태에서의 조사(照射)에 의하여 그 효과가 현저하였다. 7) konfak mannan의 감마선 조사(照射)는 점도(粘度)강하와 환원력(還元力)증가를 초래하였으며 건조상태보다는 수화(水和) 상태에서 그 효과가 현저하였다.

  • 적색효모 세포벽 용해효소의 기질특이성을 밝히기 위해 기질로 사용가능한 다당을 screening하여 Rh.glutinis IFO 0695가 생산하는 mannan을 얻었다. 이 mannan의 구조는 $\beta$-1,3과 $\beta$-1,4 결합이 교대로 연결된 직쇄상의, 중합도 약 650인 수용성의 다당으로 밝혀졌다. 이 다당을 기질로하여 양 효소의 기질특이성을 조사한 결과, Bacillus생산효소는 $\beta$-1. 3, $\beta$-1. 4 mannan을 4당 또는 6당 단위로 가수분해하는 효소로서 그 절단점은 $\beta$-1, 4결합 이었다. 이 $\beta$-1,4 결합을 가수분해하기 위해서는 인접 결합이 $\beta$-1,3결합이어야만 하는 새로운 type의 $\beta$-1,4 mannauase이었다. 한편, Penicillium생산효소는 $\beta$-1, 3, $\beta$-1, 4 mannan의 $\beta$-1, 3결합을 2당 또는 4당의 단위로 가수분해하여 최종적으로는 2당을 생성하는 $\beta$-1, 3 mannanase의 일종이었다.

Mannan-binding lectin of the sea cucumbers Stichopus japonicus has common antigenic determinants with human serum mannan-binding lectin

  • The host defense system or immune system of all modern animals has their roots in very ancient organisms. After analyzing literature data concerning properties of invertebrates and vertebrates lectins we suggest that mechanism of mannans recognition may exist in marine invertebrates, as a universal mechanism for homeostasis maintenance and host defense, and mannan-binding lectins family of vertebrates has ancient precursor, as was shown for another S-type lectins family. We carried out the screening of mannan-binding type lectin among different species of echinoderms inhabiting in Piter the Grate Bay, the sea of Japan. As a result, the C-type lectins (SJL-32) specific for high mannose glycans was isolated from the coelomic plasma of the sea cucumbers Stichopus japonicus by ion-exchange chromatography on a DEAE-Toyopearl 650M, affinity chromatography on a mannan-Sepharose 6B and gel filtration on a Sephacryl S-200. SJL-32 is homodimer with molecular mass about 32 kDa on SDS-PAGE under non-reducing conditions. Protein part of the lectin has high conteins Asn, Glu, Ser. Hemagglutination of trypsin-treated O blood group human erythrocytes by SJL-32 was competitively inhibited by high-branched -D-mannan composed of -1,2 and -1,6 linked D-mannopyranose residues. In contrast, a variety of mono-, oligo-, and polysaccharides composed of residues of galactose and fucose showed absence or little inhibitory activities. The lectin activity strong depends on Ca2+ concentration, temperature and pH. Monospecific polyclonal antibodies were obtained to the lectin. As was shown by ELISA assay, antibodies to SJL-32 cross-reacted with human serum mannan-binding lectin. This data allows making conclusion about common antigenic determinants and structural homology of both lectins. In our opinion, SJL-32 belongs to evolutionary high conservative mannan-binding lectins (MBLs) family and takes part in the host defense against pathogenic microorganisms.

Clinical Study on Mannan Peptide Combined with TP Regimen in Treating Patients with Non-small Cell Lung Cancer

  • Purpose: To investigate short-term response rate, quality of life and toxicities of mannan peptide combined with TP regimen in treating patients with non-small cell lung cancer (NSCLC). Patients and Methods: Forty one patients with NSCLC were divided into an experimental group treated with TP regimen combined with mannan peptide (21 patients) and a control group treated with TP alone (20 patients). Results: Response rates were 61.9% (13/21) for the experimental and 60% (12/20) for the control group (p>0.05). Regarding toxicity, white blood cell decreased more frequently in the control group (65%, 13/20) than in the experimental group (33.3%, 7/21) (p<0.05); nausea and vomiting also occurred more frequently in the control group (55%, 11/20 vs 23.8%, 5/21) (p<0.05). In terms of quality of life, this index was improved by 57.1% (12/21) and 25% (5/20) in experimental and control groups, respectively (p<0.05). Conclusions: Response rate of TP after combined with mannan peptide is mildly increased, while this combination alleviates bone marrow suppression as well as nausea and vomiting of TP, and improves quality of life when treating patients with NSCLC. However, this conclusion should be confirmed by randomized clinical trails.

Catabolic Repression 및 Derepression에 의한 효모 세포의 다당류 함량 변화와 무기 폴리 인산(제 5 보) (Changes in Amounts of Polysaccharides and Polyphosphates under Catabolic Repression and Derepression in Yeast (V))

  • 본 연구는, catabolic repression시킨 효모세포를 완전배지와 최소배지에서 derepression시켜, 배양시기 및 인산 첨가농도(free, limited, sufficient)에 따른 5종의 다당류 합성변화를 조사하였다. 그리고 다당류 합성과 무기폴리인산 축적량 및 인지질 합성 사이의 상관지수를 구하여 합성시 관련되는 유의한 정도를 검정하였다. 그 결과, 최소배지에서 catabolic derepression시킨 효모세포가, 완전배지에서 derepression시킨 세포에 비하여, glycogen의 합성이 발리 그리고 많이 일어났고, acid soluble glycogen type이 주된 함량을 나타내었으며, alkali soluble glycogen은 당이 많이 소모된 24시간 배양 후에 소량 나타났다. 무기인산 첨가정도에 따라 total glycogen합성이 일정한 비율로 빨리 그리고 높게 일어났다. Glucan의 합성에는 ALPase 중 ALPase "C"가 관련할 것으로 추정되었다. Mannan은 ezponential phase초기와 정체기때, acid soluble 분획은 정체기때 최대함량을 나타내었다. Mannan 합성과 poly-P "C"축적량 사이의 상관지수는 0.866, mannan합성과 인지질 사이의 상관지수는 0.726으로 나타나 매우 유의하였다.

Inhibition of Yeast Growth by Broadly Cross-Reactive Antisera Elicited by Heterologous Mannan-Protein Conjugate

  • A new approach to obtain broadly cross-reactive antisera against important yeast pathogens by intensive hyperimmunization with polysaccharide-protein conjugates is described here. Surface mannan of Candida albicans and capsular galactoglucoxylomannan of Cryptococcus laurentii were isolated and chemically linked to human serum albumin. Antisera elicited by a 7-week vigorous immunization of rabbits with the conjugates showed effective cross-reactive growth inhibition of different representatives of Candida spp. as well as Cryptococcus spp. IgG antibodies are evidenced as the effective component of the antisera.

  • The effects of dietary mannan oligosaccharides, Lactobacillus plantarum, Bacillus subtilis, and Bacillus licheniformis supplementation on hybrid grouper Epinephelus akaara ♀×Epinephelus lanceolatus ♂ were evaluated. The fish were fed a basal diet and five other diets consisting of 0.6% mannan oligosaccharides, L. plantarum, B. subtilis, and B. licheniformis and mixture of each 0.15% prebiotic and all the probiotics (designated as MOS, LP, BS, BL, and SYN) for 56 days. Growth performance and feed utilization showed no significant differences among all experimental groups. Lipid level of whole-body was significantly high in MOS and BL groups. Plasma aspartate aminotransferase was significantly low in BL and SYN groups. Nitro-blue tetrazolium, lysozyme and anti-protease, and glutathione peroxidase in BS, SYN, and all probiotic groups, respectively, were significantly high. Intestinal Vibrio bacteria was significantly low in all probiotic and SYN groups. Gene expression of interleukin-1β and interleukin-10 in SYN group; transforming growth factor β2 in MOS and BS groups, toll-like receptor 2-2 in BS and BL groups; and C-type lectin in MOS, LP and SYN groups were significantly upregulated. Our findings indicate that mannan oligosaccharides, L. plantarum, B. subtilis, and B. licheniformis could improve innate immunity, antioxidant capacity, anti-inflammation, and intestinal microbiota of hybrid grouper.

The Effect of Feeding Mannan-Oligosaccharides (Bio-MOS) on the Performance of Meat Chickens under Two Different Vaccination Programs

  • The effects of feeding a mannan oligosaccharide (Bio-Mos) from 0 to 3 g/kg diet and vaccination program on 1- to 35-day performance (growth and feed efficiency), metabolizable energy, nitrogen utilization and carcass composition of meat chickens were investigated. A general vaccination program was used against IB, IBD and ND with half of the birds per diet receiving a booster dose of IB and ND vaccines at 12 days of age. Dietary supplementation of Bio-Mos (BM) did not influence body weight gain, feed efficiency and nutrient utilization. The highest dietary BM (3 versus 1.5 or 0 g/kg) increased carcass abdominal fat and reduced the proportion of drumstick in the carcass of meat chickens. The booster dose reduced the performance of birds. It was concluded that the addition of BM to the diet of chickens did not significantly influence the performance and nutrient utilization of meat chickens.
