Kinetic Properties of Manganese Peroxidase from the Mushroom Stereum ostrea and its Ability to Decolorize Dyes

  • Manganese peroxidase (MnP) was isolated from the culture filtrate of the wood log mushroom Stereum ostrea (S. ostrea), grown on Koroljova medium, and then purified by ammonium sulfate [70% (w/v)] fractionation, DEAE-cellulose anion exchange chromatography, and Sephadex G-100 column chromatography, with an attainment of 88.6-fold purification and the recovery of 22.8% of initial activity. According to SDS-PAGE the molecular mass of the MnP was 40 kDa. The optimal pH and temperature were found to be 4.5 and $35^{\circ}C$, respectively. The enzyme was stable even after exposure to a pH range of 4.5 to 6.0, and at temperatures of up to $35^{\circ}C$ at a pH of 4.5 for 1h. The $K_m$ and $V_{max}$ values for the substrate phenol red were found to be $8{\mu}m$ and 111.14 U/mg of protein, respectively. The MnP also oxidized other substrates such as guaiacol, DMP, and veratryl alcohol. Sodium azide, EDTA, SDS, $Cu^{2+}$, and $Fe^{2+}$, at 1-5 mM, strongly inhibited enzyme activity, whereas $Ca^{2+}$ and $Zn^{2+}$ increased enzyme activity. The participation of the purified enzyme in the decolorization of dyes suggests that S. ostrea manganese peroxidase could be effectively employed in textile industries.

Exposure Assessment of Hazardous Chemical Agents for Dental Technicians in Ulsan City (울산지역 치과기공사들의 화학적 유해요인 노출 평가)

  • Objectives: This study was conducted to evaluate the exposure level of hazardous chemical agents for dental technicians in Ulsan. Methods: We measured airborne total dusts and metals such as Nickel, Manganese, Cobalt, and Chromium in 10 dental laboratories by the NIOSH Methods 0500 and 7300, respectively. Methyl methacrylate (MMA), a key ingredient in acrylic resin, was also monitored using passive samplers for long-term sampling and Tenax tubes for short-term sampling. Results: Measured levels of all items were below 10% of the Korean exposure limit except for Nickel. The geometric mean concentration and geometric standard deviation of total dust, Nickel, and MMA were $0.14mg/m^3$ (2.16), $165.3{\mu}g/m^3$ (3.31), and 0.2 ppm (2.5) respectively. Airborne Nickel concentration of two dental laboratories exceeded the exposure limit ($1000{\mu}g/m^3$). The major emission sources of Nickel were metal trimming and casting processes. Conclusions: We found that Nickel, a carcinogen, should be controled most urgently to protect dental technicians.

  • Seaweeds produce antioxidants to counteract environmental stresses, and these antioxidant genes are regarded as important defense strategies for marine algae. In this study, the expression of Pyropia yezoensis (Bangiales, Rhodophyta) ascorbate peroxidase (PyAPX) and manganese-superoxide dismutase (PyMnSOD) was examined by qRT-PCR in P. yezoensis blades under abiotic stress conditions. Furthermore, the functional relevance of these genes was explored by overexpressing them in Chlamydomonas. A comparison of the different expression levels of PyAPX and PyMnSOD after exposure to each stress revealed that both genes were induced by high salt and UVB exposure, being increased approximately 3-fold after 12 h. The expression of the PyAPX and PyMnSOD genes also increased following exposure to $H_2O_2$. When these two genes were overexpressed in Chlamydomonas, the cells had a higher growth rate than control cells under conditions of hydrogen peroxide-induced oxidative stress, increased salinity, and UV exposure. These data suggest that Chlamydomonas is a suitable model for studying the function of stress genes, and that PyAPX and PyMnSOD genes are involved in the adaptation and defense against stresses that alter metabolism.

  • Mancozeb (MCZ), a polymeric complex of zinr and manganese salts of ethylene bisthiocarbamate, is widely used in agriculture as fungicidel, insecticides, and herbicides. MCZ can be occupationally and environmentally exposed to human and has been reported to have detrimental effects on the reproductive system, but the toxicity of MCZ on immune responses has not been systematically investigated. We investigated the effects of MCZ exposure on the activities of murine splenocytes through evaluation of splenocytes cellularity and INF-$\gamma$ synthesis. Splenocytes were examned ex vivo from mice orally treated with various doEes of MCZ for 1 day (acute exposure, 2,100, 5,000, 10,000 mg/kg) or ior 5 consecutive days per week for 4 weeks (subacute exposure,250, 1,000, 1,500 mg/kg/day) fellowed by culture for 2 days in the presence of Con A or PHA plus IL-2. Splenocytes Iron naive mice were cultured with various concentration of MCZ (50, 500, 1,000 ng/ml) in the presence of Con A or PHA plus IL-2 for 2 days in vitro. IFN-$\gamma$ production was decreased with the in vitro exposure to all concentration of MCZ. The spleen cellularity and IFN -$\gamma$ production by splenocytes from MCZ -acutely and - subacutely exposured mice were decreased in comparision with that oi control group.

Association with Combined Occupational Hazards Exposure and Risk of Metabolic Syndrome: A Workers' Health Examination Cohort 2012-2021

  • Background: This study aimed to evaluate the association between exposure to occupational hazards and the metabolic syndrome. A secondary objective was to analyze the additive and multiplicative effects of exposure to risk factors. Methods: This retrospective cohort was based on 31,615 health examinees at the Pusan National University Yangsan Hospital in Republic of Korea from 2012-2021. Demographic and behavior-related risk factors were treated as confounding factors, whereas three physical factors, 19 organic solvents and aerosols, and 13 metals and dust were considered occupational risk factors. Time-dependent Cox regression analysis was used to calculate hazard ratios. Results: The risk of metabolic syndrome was significantly higher in night shift workers (hazard ratio = 1.45: 95% confidence interval = 1.36-1.54) and workers who were exposed to noise (1.15:1.07-1.24). Exposure to some other risk factors was also significantly associated with a higher risk of metabolic syndrome. They were dimethylformamide, acetonitrile, trichloroethylene, xylene, styrene, toluene, dichloromethane, copper, antimony, lead, copper, iron, welding fume, and manganese. Among the 28 significant pairs, 19 exhibited both positive additive and multiplicative effects. Conclusions: Exposure to single or combined occupational risk factors may increase the risk of developing metabolic syndrome. Working conditions should be monitored and improved to reduce exposure to occupational hazards and prevent the development of the metabolic syndrome.

  • Effects of exposure to a neurotoxicant, fenitrothion on antioxidant enzyme activities in Chironomus riparius Mg. (Diptera, Chironomidae) larvae were evaluated under laboratory conditions. Exposure to this chemical led to an increase of cupper, zinc type superoxide dismutase and manganese type superoxide dismutase activities and to a decrease of glutathion peroxidase activity. An activation of catalase was observed in the larvae exposed to high fenitrothion concentration. The response of superoxide dismutase was rapid and sensitive to low chemical concentrations, but changes in catalase, total peroxidase and glutathion peroxidase were less sensitive. In this study, antioxidant enzyme activities in Chironomus riparius larvae were identified as pertinent biomarkers for environmental monitoring.

It is reported to induce teratogenic and carcinogenic effect in laboratory animals. But the immunomodulating effects of Mancozeb exposure have not been systemically evaluated. The purpose of this study was to investigate the effects of Mancozeb on cell-mediated immunity in mice. For ex vivo assessment, mice were orally exposed to Mancozeb dissolved in distilled water as concentrations of 2,500, 5,000, 10,000 mg/kg for single occasion (acute exposure) or 250, 1,000, 1,500 mg/kg/day 5 days a week for 30days(subacute). Exposure Evaluation to Total Welding Fume and Manganese at Technical High Schools in Choong-Nam Area (충청지역 일부 공업고등학교 실습생의 용접흄 및 망간에 대한 노출 평가)

  • Geometric mean of airborne welding fume concentration at technical high schools was 4.80mg/㎥)N.D~35.39 mg/ ㎥ and the percentage of samples exceeded TLV of the Korean ministry of labor was 43.6%, Geometric mean of airborne Mn concentration was 0.06 mg/㎥(N.D~0.42mg/㎥) and the percentage of samples exceeded TLV of ACGIH was 15.4 % In case of airborne Me concentration, there is a significant difference among schools (p<0.05) Mn concentrations in blood of the exposed and control groups were 1.84$\mu\textrm{g}$/dl and 1.91 mg/dl respectively. Mn concentrations in urine of the exposed and control groups were 1.36$\mu\textrm{g}$/ιand 0.57$\mu\textrm{g}$/ι respectively. In case of Mn concentrations in urine there is a significant difference between both groups(P<0.001) and among schools(p<0.05) Mn concentrations in blood and urine of exposed group were not over BEIs of the Korean ministry of labor. Mn levels in blood and urine were not significantly affected by smoking, drinking and residence, There was no correlation between Mn concentration in air and blood but there was a statistically significant correlation between Mn concentration in air urine(r=0.323). There was no a statistically significant correlation between Mn concentration in blood and urine.

  • In this study, a corrosion failure analysis of a heat transport pipe was conducted, as the result of a pinhole leak. Interestingly, the corrosion damage occurred externally in the pipeline, resulting in severe thickness reduction near the seam line. Also, while a stable magnetite protective film formed on the inner surface, the manganese oxide formation occurred only on the outer surface. The interior and exterior of the pipe were composed of ferrite and pearlite. The large manganese sulfide and alumina inclusions were found near the seam line. In addition, the manganese sulfide inclusions resulted in grooving corrosion, which progressed in the seam line leading to the reduction in the thickness, followed by the exposure of the alumina in the matrix to the outer surface. To note, the corrosion was accelerated by pits generated from the boundaries separating the inclusions from the matrix, which resulted in pinhole leaks and water loss.
