개 자연발생 유선종양 2종의 세포주 확립 및 특성분석 (Establishment and Characterization of Canine Mammary Gland Cancer Cell Lines)

  • 개에서 자연발생한 유선종양으로부터 채취, 배양된 세포주 2개를 확립하고 특성을 분석하였다. 9세 령의 퍼그 종 개와 동일연령의 토이푸들 종의 개에 발생한 종양을 무균 상태를 유지하여 수술적으로 채취한 후 primary culture를 실시하였다. 두 종양의 조직병리검사에서는 각각 선암종과 혼합암종이 진단되었으며, 이후 두 마리 모두 전이로 인해 폐사하였다. 배양된 종양세포는 1년 이상의 기간 동안 60회 이상 계대를 반복하면서 형태학적으로 일관성을 유지하였고, 특성분석을 위해 광학현미경검사, 성장곡선 산출, 배가 시간 계산, 누드 마우스에 이종이식, 면역조직 화학검사를 실시하였다. 각 세포주는 다각형의 긴 세포형태를 보였으며, 세포질 연결을 형성하였으며, 배가 시간은 각각 47.1 시간과 18.6 시간이었다. 암컷 누드마우스의 등 부위에 피하이식 후 4주 이내에 10마리 중 9마리에서 촉진이 가능한 종괴의 형성이 확인되었으며, 면역조직화학검사 시 한 세포주에서는 keratin과 cytokeratin 8에서 다른 세포주에서는 smooth muscle actin과 cytokeratin 8에서 강한 염색성이 확인되었다. 두 세포주는 개의 유선종양의 시험관내, 체내 연구에 있어 모두 유용하게 사용될 수 있을 것으로 생각된다.

애완견 유방암의 수술적 치료 후에 사용한 Phytolacca D$_{2}$의 종양조직의 재발 방지 효과 (The use of Phytolacca D$_{2}$ for the treatment of Mammary Tumors in Bitches)

  • The effectiveness of two different methods for the treatment of mammary tumors in bitches was examined in 30 dogs to determine the use of homepathy induction by Phytolacca $D_{2}$ preparations. The animals in group 1 received only Phytolacca $D_{2}$. In group 2the tumors were removed surgically and were treated with Phytolacca $D_{2}$ post operation. For the clinical sigh post treatment the dogs were examined regularly to detect neoplasia, reoccurrences and metastases. Treatment with Phytolacca $D_{2}$ resulted in following effects; In group 1 was not efficient. In group 2 was an inhibition of tumor growth and treated animals appeared to be revitalized.

Clomipramine treatment of acral lick dermatitis in a dog

  • An 11-year-old, toy poodle dog was presented with dermatologic lesions and mammary gland tumor (MGT) evaluation. A solitary, lobulated MGT (size $2.5{\times}3.5$ cm) was affecting the 5th left mammary gland. Firm, oval plaque skin lesions were present on the left dorsal carpal area. The skin lesions were alopecic and salivary staining. The dog had historical separation anxiety and excessive licking of skin lesions were observed. Based on the clinical and histopathologic examinations, MGT was diagnosed with mammary complex adenoma and the skin lesions were diagnosed with acral lick dermatitis. Behavior modification treatment using oral clomipramine was effective.

Effect of Low Doses of Genistein and Equol on Protein Expression Profile in MCF-7 Cells

  • Although action modes of equol and genistein have been extensively studied, their precise roles in tumor cells remain elusive. To address possible effects of these compounds on protein expression in mammary tumor cells, proteins modulated in MCF-7 mammary tumor cells when incubated in absence and presence of 10 uM equol or genistein were identified through 2-dimensional gel electrophoresis, MALDI-TOF MS/MS, and NCBInr database search using Mascot software. Most proteins differentially expressed in MCF-7 cells after treatment with 10 uM genistein or equol were identified as being the same. Exposure to both compounds caused decreased cellular expression of RNA-binding protein regulatory subunit and oncogene DJ1 tubulin beta-1 chain, and increased expression of heterogeneous ribonucleoproteins F and L, KH-type splicing regulatory protein, and translation elongation factor EF-Tu precursor. Genistein and equol at dose used in this study showed common action mechanism.

개의 유선에 발생한 골격외 골육종 1예 (Extraskeletal Osteosarcoma of the Mammary Gland in a Dog)

  • A 16-year-old female Poodle with multiple mammary masses was referred to the Veterinary Medical Teaching Hospital of Seoul National University. Radiographic finding indicated a round mass of soft tissue density at the ventral portion of the 4th-7th sternum, which was 7 cm in diameter and presented multiple mineralization within the mass. Cytologically, aspirate of the mammary mass was moderately cellular and revealed well-differentiated osteoblasts with moderately basophilic cytoplasm, various sized and eccentrically placed nuclei and distinct nucleoli. Osteoblasts interspersed with osteoid-suspected pink-staining intercellular matrix. Some cells displayed malignant features such as macronuclei, macronucleoli, binucleation, abnormal mitotic figures, anisocytosis and anisokaryosis. On which the mass was diagnosed as osteosarcoma. Histopathologic examinations of the mass were compatible with a diagnosis of osteosarcoma. The patient was treated with surgery alone. The patient died 45 days later from surgery.

Analysis of Different Activation Statuses of Human Mammary Epithelial Cells from Young and Old Groups

  • Human mammary epithelial cells have different proliferative statuses and demonstrate a close relationship with age and cell proliferation. Research on this topic could help understand the occurrence, progression and prognosis of breast cancer. In this article, using significance analysis of a microarray algorithm, we analyzed gene expression profiles of human mammary epithelial cells of different proliferative statuses and different age groups. The results showed there were significant differences in gene expression in the same proliferation status between elderly and young groups. Three common differentially expressed genes were found to dynamically change with the proliferation status and to be closely related to tumorigenesis. We also found elderly group had less status-related differential genes from actively proliferating status to intermediate status and more statusrelated differential genes from intermediate status than the young group. Finally, functional enrichment analyses allowed evaluation of the detailed roles of these differentially-expressed genes in tumor progression.

Interleukin-10 UP-regulates TRAIL Gene Expression in the Mammary Epithelial Cell at the Involution Stage

  • Interleukin-10 (IL-10) is known as a regulator of inflammation and pathogenesis in mammalian organs, but its precise role is little known in the mammary gland. Our initial experiment showed that IL-10 expression levels in mice decreased at the lactation stage otherwise increased at the involution stage. To reveal the effects of IL-10 on the involution of mammary gland, expression profiles of the apoptosis-related genes were examined in transgenic mice expressing human IL-10 as well as in knock-out mice (IL-10-/-). Mild inflammatory legions by lymphocytes were observed in the mammary glands of transgenic lines at the lactation stage. The expression of TRAIL (Tumor necrosis factor-Related Apoptosis-Inducing Ligand) among the apoptosis-related genes was highly elevated in the transgenic mice while others were not significantly changed. Furthermore, TRAIL was down regulated by four fold in the IL-10-/- mice at the involution stage. The expression of DR4 was elevated at the involution stage of normal mice. DR4 was detected in the milk of transgenic mice but absent in that of normal mice. Our results proposed that the elevated IL-10 at the involution stage recruit lymphocytes and induce TRAIL and DR4 genes, therefore, lead to enter involution stage of mammary glands.

Vascular Morphometric Changes During Tumor Growth and Chemotherapy in a Murine Mammary Tumor Model Using OCT Angiography: a Preliminary Study

  • To develop a biomarker predicting tumor treatment efficacy is helpful to reduce time, medical expenditure, and efforts in oncology therapy. In clinics, microvessel density using immunohistochemistry has been proposed as an indicator that correlates with both tumor size and metastasis of cancer. In the preclinical study, we hypothesized that vascular morphometrics using optical coherence tomography angiography (OCTA) could be potential indicators to estimate the treatment efficacy of breast cancer. To verify this hypothesis, a 13762-MAT-B-III rat breast tumor was grown in a dorsal skinfold window chamber which was applied to a nude mouse, and the change in vascular morphology was longitudinally monitored during tumor growth and metronomic cyclophosphamide treatment. Based on the daily OCTA maximum intensity projection map, multiple vessel parameters (vessel skeleton density, vessel diameter index, fractal dimension, and lacunarity) were compared with the tumor size in no tumor, treated tumor, and untreated tumor cases. Although each case has only one animal, we found that the vessel skeleton density (VSD), vessel diameter index and fractal dimension (FD) tended to be positively correlated with tumor size while lacunarity showed a partially negative correlation. Moreover, we observed that the changes in the VSD and FD are prior to the morphological change of the tumor. This feasibility study would be helpful in evaluating the tumor vascular response to treatment in preclinical settings.

개에 있어서 악성 림프부종 일례 (A Case of Malignant Lymphedema in a Dog)

  • A five-year-old, female Great Dane dog with edema, localized trauma, mild pain, and lameness of the right hind limb was referred to the Veterinary Medical Teaching Hospital of Chungbuk National University. This dog had a history of mammary tumor excisions 6 months ago. Abnormal changes were not seen in the values of complete blood count and serum biochemical tests. But pedal direct lymphangiography using aqueous-based radiographic agent showed the obstructed lymph flow in right popliteal lymph node. Based on these observations, the dog was suspected as lymphedema resulted from lymph drainage flilure without any other possibilities of inflammation or other causes. Although recommended chemotherapy and physiotherapy had been applied for resolvinr presented problems for one month, there was no improvement on edema of damaged region and any other clinical signs. Therefore, the necropsy was performed after euthanasia under agreement of the owner of patient. In histopatholofical examination, the most characteristic lesions in the mass of femoral region were diffuse edema fibrosis and neoplastic cells in the lymphatics. Also, the neoplastic cells were very similar to those found in the tumor mass of mammary gland, which had diagnosed as fibrosing carcinoma. These facts suggested that the cause of obstructed lymph flow was the neoplasia in lymphatics of the right hind limb. With these results, a diagnosis of malignant lymphedema was made in this dog.
