Caponization Effects on Growth Performance and Lipid Metabolism in Taiwan Country Chicken Cockerels

  • This trial was designed to study the caponization effects on the appearance, carcass characteristics, blood constituents and lipid metabolism of Taiwan country chicken cockerels. Cockerels were caponized at 8 weeks of age. Sixteen-week-old chickens, including 10 capons, 5 slips (incomplete caponized male chickens) and 20 normal chickens of equal sexes were selected for a 10 week ad libitum feeding trial. Results showed that the testosterone concentrations in the capons and females were lower (p<0.05) than that of intact males. The comb length, height and weights were also lower (p<0.05). The weight of the slips was between that of the capons and intact males, but was heavier (p<0.05) than that of the capon. The live-weight, carcass weight and shank perimeter in the capons were higher than those of the other groups (p<0.05). Hepatic lipogenic enzyme activity analyses showed that NADP-malic dehydrogenase (MDH) activity in the capons and female chickens was higher than that in intact male chickens (p<0.05). The MDH activity in the slips was between that for the capon and intact male chickens (p>0.05). The abdominal fat weight and relative abdominal fat weight of the capons and females were heavier than that for intact males (p<0.05); the slips were between the capons and intact males. The blood lipid content results showed that the triacylglycerol and cholesterol in the capons were higher than that for intact males (p<0.05). However, the percentage of low-density lipoprotein (LDL) was lower than that in intact male chickens (p<0.05). It appears that the increase in lipid accumulation in caponized male chickens is attributed mainly to an increase in MDH activity and the changes in lipid transportation in the capons.

  • This study was undertaken to elucidate the effect of DHA-rich fish oil (DHA-rich oil) added to different dietary fats on lipid metabolism. Rats were fed perilla oil, sesame oil and beef tallow with or without DHA-rich oil for 12 weeks. The weight gain was higher in groups with DHA-rich oil than that of groups without DHA-rich oil, with DHA-rich oil, while weight of epididymal fat pad was lower in perilla oil and beef tallow groups with DHA-rich oil. The contents of total lipid and triglyceride in plasma were not affected by dietary fat types, but that of total and HDL cholesterol in plasma were higher in sesame oil group than perilla oil and beef tallow groups without DHA-rich oil. The contents of total lipid, triglyceride, total cholesterol, HDL cholestrol and LDL cholesterol in plasma were decreased by DHL-rich oil addition. The contents of total lipid, total cholesterol and triglyceride in the liver were not affected by dietary fat type. The contents of total cholesterol and triglyceride in the liver were not affected by dietary fat type. The contents of total lipid and TG in liver were not affected by DHA-rich oil addition while hepatic cholesterol increased by DHA-rich oil addition. The activities of glucose 6-phosphate dehydrogenase and malic enzyme were highest in beef tallow group without DHA-rich oil and decreased by DHA-rich oil addition. Peroxisomal ${\beta}$-oxidation had an inverse relationship against the activities of lipogenic enzymes. In conclusion, dietary DHA-rich oil decreased fat accumulation and had hypolipidemic effect, especially in beef tallow group. Also groups with DHA-rich oil showed more hypolipidemic effect than perilla oil group. And DHA-rich oil addition to diets resulted in increasing dietary n-3/n-6 ratio. Therefore increase in n-3/n-6 ratio as well as dietary DHA were considered to be responsible for the hypolipidemic effect resulted from DHA-rich oil addition.

갈색 느티만가닥버섯의 영양특성 및 생리기능성 분석 (Analysis of Nutritional Characteristics and Physiological Functionality of Hypsizygus marmoreus (Brown cultivar))

  • 갈색 느티만가닥버섯으로부터 새로운 생리기능성 물질을 개발하기 위하여 갈색 느티만가닥버섯 자실체의 영양 특성과 생리기능성을 조사하였다. 갈색 느티만가닥 버섯의 자실체에는 조단백질 27.3%, 조지방 2.7%, 탄수화물 55.8%을 함유하고 있었고 유리당 중에서는 전화당이 건조 중량 100 g 당 8.6 mg을, 유기산 중에서는 11,109.3 mg의 사과산을 가장 많이 함유하고 있었다. 갈색 느티만가닥버섯 자실체의 물 추출물과 에탄올 추출물을 제조하여 이들의 생리기능성을 측정한 결과 물 추출물이 66.7%의 항고혈압성 안지오텐신 전환효소 저해 활성(ACE)과 37.3%의 항통풍성 잔틴옥시데이즈 저해활성을 보였으나 에탄올 추출물은 대체로 낮은 생리기능성을 보였다. 또한 느티만가닥버섯의 자실체를 증류수에 넣고 $70^{\circ}C$에서 12시간 추출하였을때 가장 많이(저해활성: 84.4%) 추출되었으나 $50^{\circ}C$에서 12시간 동안 추출하여 얻은 추출물에서도 높은 ACE 저해활성(81.4%)을 보여 경제적 생산면에서 더 유리 할 것으로 사료된다.

High-Molecular-Weight Poly-Gamma-Glutamate Protects Against Hypertriglyceridemic Effects of a High-Fructose Diet in Rat

  • We studied the effects of 2 different dosages of high-molecular-weight poly-${\gamma}$-glutamic acid (hm ${\gamma}$-PGA) derived from Bacillus subtilis chungkookjang on lipid metabolism in a high-fructose diet-induced hypertriglyceridemic animal model. For 4 weeks, rats were fed either AIN-93 diet (normal control, NC; n = 10) or modified AIN-93 diet in which cornstarch was substituted with 63% fructose (n = 30) to induce hypertriglyceridemia. After 4 weeks, the hypertriglyceridemic rats were treated with daily oral doses of 0 mg (hypertriglyceridemic control, HC), 2.5 mg (hypertriglyceridemic, low hm ${\gamma}$-PGA, HL), or 5 $mg{\cdot}kg{\cdot}bw^{-1}{\cdot}d^{-1}$ (hypertriglyceridemic, high hm ${\gamma}$-PGA, HH) hm ${\gamma}$-PGA for 4 weeks. The HL and HH groups exhibited significantly lower levels of serum triglyceride, total cholesterol, LDL cholesterol, and free fatty acids than the HC group. The administration of hm ${\gamma}$-PGA reduced serum ALT and AST levels. The activities of lipogenic enzymes such as hepatic malic enzyme and glucose-6-phosphate dehydrogenase as well as glucose-6-phosphate dehydrogenase mRNA expression were significantly decreased by hm ${\gamma}$-PGA administration (p < 0.05). These results indicate that hm ${\gamma}$-PGA has an anti-hypertriglyceridemic effect in high-fructose diet-induced hypertriglyceridemic rats.

Pathogenicity of Korean isolates of Acanthamoeba by observing the experimental infection and zymodemes of five isoenzymes

  • To determine the pathogenicity of Acanthamoeba spp. isolated in Korea and to develop a isoenzymatic maker, the mortality rate of infected mice, in vitro cytotoxicity against target cells and isoenzyme band pattern were observed. Five isolates of Acanthamoeba spp. (YM-2, YM-3, YM-4, YM-5 and YM-7) were used in this study as well as three reference Acanthamoeba spp. (A. culbertsoni, A. hatchetti, and A. royreba). According to the mortality rate of infected mice, Korean isolated could be categorized into three groups: high virulent (YM-4), low virulent (YM-2, YM-5, YM-7) and the nonpathogenic group (YM-4), In addition, the virulence of Acanthamoeba spp. was enhanced by brain passage in mice. In the cytotoxicity assay against chinese hamster ovary cells, especially, the cytotoxicity of brain-passaged amoebae was relatively higher than the long-term cultivated ones. The zymodeme patterns of glucose-6-phosphate dehydrogenase (G6PD), malate dehydrogenase (MDH), hexokinase (HK), glutamate oxaloacetate transaminase (GOT) and malic enzyme (ME)of Acanthamoeba spp. were different among each isolate, and also between long-term cultrued amoebae and brain passaged ones. In spites of the polymorphic zymodemes, a slow band of G6PD and K, and an intermediate band of MDH were only observed in pathogenic Acanthamoeba spp., which should be used as isoenzymatic makers.

골쇄보 열수추출물의 급여가 갱년기 유도 흰쥐의 대사개선 효능에 미치는 영향 (Effects of Drynariae Rhizoma Hot Water Extract on Metabolic Improvement in the Ovariectomized Rat Model of Menopause)

  • Background: Many menopausal women suffer from health problems including metabolic diseases such as dyslipidemia and osteoporosis. Thus they need natural products and functional foods particularly highly nutritional food products, that can help alleviate these diseases. This study was carried out to determine the effect of Drynariae Rhizoma water extract on the lipid and bone metabolism of ovariectomized Sprague-Dawley rats. Methods and Results: The animals were randomly divided into six dietary groups comprising SHAM-operated rats, OVX rats (normal diet), and OVX-DR rats (Drynariae Rhizoma extract). After 8 weeks, plasma, liver, and fat samples were collected to analyze the lipid metabolism, plasma Ca, alkaline phosphatase (ALP), osteocalcin and C-terminal telopeptide (CTx) concentrations, which are biochemical makers of bone metabolism. The left femurs of rats were also collected for histological analyses. OVX counteracted menopause induced body weight gain, as well as increases in triglycerides, total cholesterol, and free fatty acids. The Drynariae Rhizoma group showed low levels of triglycerides, high HDL-cholesterol, and decreased lipogenesis based on activity of the lipid-regulating enzymes (fatty acid synthase and malic enzyme). Decreased serum levels of ALP and osteocalcin were observed in Drynariae Rhizoma group. Conclusions: The results of this study show that Drynariae Rhizoma extract may effectively regulate hyperlipidemia and improve bone density.

제주 재래마아 쓰시마 재래마의 혈액내 단백질의 다형 (Polymorphisms of Blood Proteins In Cheju Native Horses and Tsushima Native Horses)

  • 제주 재래마와 쓰시마 재래마 간의 유전적 유연관계를 16개 혈액 단백질 좌위의 유전적 다형현상을 분석하여 연구하였다. 두 지역의 재래마 집단에서 5개의 단백질 좌위를 제외한 11개 좌위에서는 유전적 다형현상을 보였다. 다형현상을 나타내는 좌위에서 분석된 유전자 빈도를 이용하여 평균 이형접합자 빈도를 분석한 결과 제주 재래마에서는 0.375로 쓰시마 재래마의 0.304 보다 다소 높게 나타났다. Nei방법에 의해 계산된 Da distance와 유전자 동일성은 각각 0.108과 0.868이었다. 본 연구결과와 이미 보고된 아메리카말 집단들에서의 결과를 이용하여 phylogenetic tree를 구성하여 본 결과 크게 세 개의 cluster를 이루었다. 즉 아메리카말 집단들이 하나의 cluster를 이루었고 제주 재래마 집단이 하나의 cluster를 이루었으며, 이 두 cluster는 현존 말의 기원으로 보는 몽고 야생마 cluster에서 분지됨을 알 수 있었다.

한국산 연어류에서 Genetic Marker 개발을 위한 생화학적 연구 (A Biochemical Study for the Development of Genetic Marker on Salmonids in Korea)

  • 연어, 산천어, 무지개송어 등 우리나라의 연어과 어류에 있어서 종의 식별 및 3배체 어류의 판정에 동위효소를 genetic marker로 활용할 수 있는지의 가능성을 타진하고자 LDH, MDH, IDH, a-GPDH, ME, 6-PGD, PGI 및 PGM 등 8개 동위효소에 대하여 골격근 조직을 중심으로 분석을 실시하였다. 이중 골격근 조직의 MDH-B와 IDH loci에서 종간에 뚜렷한 차이를 나타내었으며 특히 MDH-B loci의 b 유전자의 출현빈도는 연어나 산천어에서는 거의 나타나지 않았으나 무지개송어에서는 매우 높게 나타났다. 또한 IDH도 무지개송어와 산천어간의 genetic marker로 유용할 것으로 보인다. 한편, PGI는 3배체 어류 생산시 친어를 상호 대립유전자(allele)의 동형접합(homozygote)인 개체를 사용할 경우 이의 효율적인 판정을 위한 새로운 marker로 활용할 수 있는 가능성을 가지고 있는 것으로 나타났으며 다른 loci에서는 별다른 차이를 발견할 수가 없었다.

A Strategy to Increase Microbial Hydrogen Production, Facilitating Intracellular Energy Reserves

  • Overexpression of the genes encoding phosphoeneolpyruvate carboxykinase (pckA) and NAD-dependent malic enzyme (maeA) facilitates higher intracellular ATP and NAD(P)H concentrations, respectively, under aerobic conditions in Escherichia coli. To verify a hypothesis that higher intracellular energy reserves might contribute to H2 fermentation, wild-type E. coli strains overexpressing pckA and maeA were cultured under anaerobic conditions in a glucose minimal medium. Overexpression of pckA and maeA enabled E. coli to produce 3-times and 4-times greater H2 (193 and 284 nmol, respectively) than the wild type (66 nmol H2). The pckA and maeA genes were further overexpressed in a hydrogenase-3-enhanced E. coli strain. The hydrogenase-3-enhanced strain (W3110+fhlA) produced 322 nmol H2, whereas the ATP-enhanced strain (W3110+fhlA+pckA) produced 50% increased H2 (443 nmol). Total H2 in the NAD(P)H-enhanced strain (W3110+fhlA+maeA) was similar to that in the control strain at 319 nmol H2. Possible explanations for the contribution of the increased cellular energy reserves to the enhanced hydrogen fermentation observed are discussed based on the viewpoint of metabolic engineering strategy.

Porphyrin Derivatives from a Recombinant Escherichia coli Grown on Chemically Defined Medium

  • We have reported previously that a recombinant Escherichia coli co-expresses aminolevulinic acid (ALA) synthase, an NADP-dependent malic enzyme, and a dicarboxylate transporter-produced heme, an iron-chelated porphyrin, in a succinate-containing complex medium. To develop an industrially plausible process, a chemically defined medium was formulated based on M9 minimal medium. Heme synthesis was enhanced by adding sodium bicarbonate, which strengthened the C4 metabolism required for the precursor metabolite, although a pH change discouraged cell growth. Increasing the medium pH buffering capacity (100mM phosphate buffer) and adding sodium bicarbonate enabled the recombinant E. coli to produce heme at rates 60% greater than those in M9 minimal medium. Adding growth factors (1 mg/l thiamin, 0.01 mg/l biotin, 5 mg/l nicotinic acid, 1 mg/l pantothenic acid, and 1.4 mg/l cobalamin) also induced positive heme production effects at levels twice of heme production in M9-based medium. Porphyrin derivatives and heme were found in the chemically defined medium, and their presence was confirmed by liquid chromatography/mass spectroscopy (LC/MS). The formulated medium allowed for the production of $0.6{\mu}M$ heme, $29{\mu}M$ ALA, $0.07{\mu}M$ coproporphyrin I, $0.21{\mu}M$ coproporphyrin III, and $0.23{\mu}M$ uroporphyrin in a 3 L pH-controlled culture.
