High frequency somatic embryogenesis and plant regeneration of interspecific ginseng hybrid between Panax ginseng and Panax quinquefolius

  • Background: Interspecific ginseng hybrid, Panax ginseng ${\times}$ Panax quenquifolius (Pgq) has vigorous growth and produces larger roots than its parents. However, F1 progenies are complete male sterile. Plant tissue culture technology can circumvent the issue and propagate the hybrid. Methods: Murashige and Skoog (MS) medium with different concentrations (0, 2, 4, and 6 mg/L) of 2,4-dichlorophenoxyacetic acid (2,4-D) was used for callus induction and somatic embryogenesis (SE). The embryos, after culturing on $GA_3$ supplemented medium, were transferred to hormone free 1/2 Schenk and Hildebrandt (SH) medium. The developed taproots with dormant buds were treated with $GA_3$ to break the bud dormancy, and transferred to soil. Hybrid Pgq plants were verified by random amplified polymorphic DNA (RAPD) and inter simple sequence repeat (ISSR) analyses and by LC-IT-TOF-MS. Results: We conducted a comparative study of somatic embryogenesis (SE) in Pgq and its parents, and attempted to establish the soil transfer of in vitro propagated Pgq tap roots. The Pgq explants showed higher rate of embryogenesis (~56% at 2 mg/L 2,4-D concentration) as well as higher number of embryos per explants (~7 at the same 2,4-D concentration) compared to its either parents. The germinated embryos, after culturing on $GA_3$ supplemented medium, were transferred to hormone free 1/2 SH medium to support the continued growth and kept until nutrient depletion induced senescence (NuDIS) of leaf defoliation occurred (4 months). By that time, thickened tap roots with well-developed lateral roots and dormant buds were obtained. All Pgq tap roots pretreated with 20 mg/L $GA_3$ for at least a week produced new shoots after soil transfer. We selected the discriminatory RAPD and ISSR markers to find the interspecific ginseng hybrid among its parents. The $F_1$ hybrid (Pgq) contained species specific 2 ginsenosides (ginsenoside Rf in P. ginseng and pseudoginsenosides $F_{11}$ in P. quinquefolius), and higher amount of other ginsenosides than its parents. Conclusion: Micropropagation of interspecific hybrid ginseng can give an opportunity for continuous production of plants.

Overcoming sterility by Caffeine and Temperature in Oriental-Asiatic Interspecific Lily Hybrid (백합 Oriental-Asiatic 종간잡종의 임성 회복을 위한 카페인과 변온처리)

  • In order to recover fertility from sterile interspecific OA-1 $F_1$ hybrid (Oriental hybrid 'Mero Star' ${\times}$ Asiatic hybrid 'Connecticut King'), various concentrations (0.1, 0.3 and 0.5%) of caffeine were injected directly into flower buds and then confirmed the viability of OA-1 $F_1$ hybrid at the flowering time. After the caffeine treatment, fertilized $F_1$ hybrids were crossed as female with Asiatic hybrid 'Lanzarote' as male. Five plantlets were obtained from seven embryos of 16 pollinated flowers at 0.3% treatment of caffeine while 0.5% treatment of caffeine obtained one plant let and 0.1 % treatment of caffeine plantlet did not produce at all. Thus 0.3% of caffeine treatment was considered as optimum concentration to produce subsequent progenies and the OA-l $F_1$ hybrid treated with caffeine produced 51% of putative 2n gametes. Pollen germination of OA-2 ('Romero Star' ${\times}$ 'Lady Rosa') and OA-3 ('Expression' ${\times}$ 'Lady Rosa') was not differ between temperature treatment alone and in combination with caffeine and temperature treatment. In the reciprocal crosses of OA-1 and Asiatic hybrid 'Lanzarote' or Oriental hybrid 'Sorbonne', A ('Lanzarote') ${\times}$ OA-1 or OA-1 ${\times}$ A crosses showed better results than O ('Sorbonne') ${\times}$ OA-1 or OA-1 ${\times}$ a crosses in plant obtaining. All progenies obtained from A ${\times}$ OA-1 or OA-1 ${\times}$ A crosses were confirmed as triploids by GISH analysis.

Breeding of F1 Hybrid for Oriental Mustard(Brassica juncea L. Czern) Using the Cytoplasmic Male Sterile Line (웅성불임 인자를 이용한 갓(Brassica juncea L. Czern)의 F1 육종)

  • Recently, the demand for new cultivar of oriental mustard(Brassica juncea) is increased as the consumption of oriental mustard has increased dramatically in the market due to Kimchi is attracting the world's attention. However, absence of seed supply system can cause many problems including inbreeding depression due to self seed production, deterioration of the seed purity and heterogeneity of commercial seed. To establish the $F_1$ variety breeding system of oriental mustard, pure line and inbred lines were screened from inbreeding genetic resources. Male-sterile line was also selected from breeding combinations for the quality improvement of mustard. The combining ability from(Indojasai ${\times}$ Goheungdamyang) combinations and isolation line of(MS910 ${\times}$ Japan red mustard 8 ${\times}$ Ganghwa mustard 9) was highest, thus these lines were selected as parental lines. PCR(Polymerase Chain Reaction) and gene sequence analysis revealed that the genes related to CMS(orf263, orf220, and orf288) were distributed in mitochondria. The isolated lines from this study also showed good performance in yield test and farmhouse prove test.

Development of new broccoli varieties from elite lines obtained by microspore cultivation method (소포자 배양 유래 계통을 활용한 브로콜리 신품종 조기 육성)

  • Since the year 2000, the production and consumption of broccoli have rapidly increased in Korea. And, the average production area and amount were about 1,700 ha and 29,000 ton for the past 5 years. Even with the increase of these cultivation and consumption, more than 95% of the broccoli seeds are currently imported from foreign countries such as Japan and Netherlands. Therefore, development of domestic broccoli varieties is needed to relieve Korean farmers' production cost for broccoli. In this situation, National Institute of Horticultural and Herbal Science (NIHHS) of Korea has tried to develop F1 hybrid varieties from elite lines that were obtained by microspore cultivation method from 2008. As the results, about 850 lines of broccoli were obtained and self-pollinated. Then their ploidy levels of the genome were confirmed to select double haploid (D.H.) lines. And the D.H. lines' horticultural traits were evaluated in open field. After the selection of 17 elite D.H. lines, they were cross-pollinated with a male sterile (MS) line to produce F1 hybrid seeds. After 2 to 3 years field trials of these F1 hybrid varieties at the area of Suwon, Gangneung, and Jeju respectively, two hybrid varieties such as 'Wongyo8011' and 'Wongyo8012' are selected for the application of variety protection. With these 4 years of research, we found that the microspore cultivation method is a powerful tool for the conventional breeding program, especially for the development of various inbred lines and even F1 hybrid varieties in short time.

New Mid-late Maturing F1 Hybrid Onion Cultivar, "Yeongpunghwang" (양파 추파형 중·만생 다수성 신품종 "영풍황")

  • New onion hybrid cultivar, "Yeongpunghwang" was developed from cross between "MOS8", male sterile line and "Mokpo 11", pollen line by Mokpo Sub-Station, National Institute of Horticultural & Herbal Science, RDA in 2008. The crossing line was performed yield test in 2003. IT was evaluated as "Mokpo 11" in the regional yield trial test(RYT) from 2006 to 2008 during three years. This line is semi-erect plant growth type and has green leaf colour. It has broad ovate bulb shape and yellow skin colour in bulb characteristics. Plant height, leaf number, and bolting rate was 78cm, 7.9, and 3.9%, respectively. Lodging date as harvesting indicator was on 26 May. The yield potential of "Yeongpunghwang" was out-yielding 6% compare to check variety ("Sunpower") by with 8,344 kg/10a in RYT. It showed moderated cold hardness in all regional location which was conducting RYT, including Muan, Goheung, Changyeong and Andong. So this variety was recommend as a leading variety at southern area of korea.

Development of PVY resistant flue-cured $F_1$ hybrid variety 'KF120'

  • Potato Virus Y (PVY), PVY-vein necrosis strain, causes severe damage at growth, yield and leaf quality on flue-cured tobacco in Korea. The development of PVY resistant flue-cured varieties without quality deterioration is therefore urgently desired. The flue-cured tobacco, KF120 (Korea Flue-cured 120), was a male-sterile (ms) $F_1$ hybrid derived from the cross between msKF117 and KF0007-7. msKF117 was developed from the cross of NC82 with N. africana and KF0007-7 was developed from the cross of KF117 with NC82. The agronomic characteristics and disease resistance of KF120 was evaluated during 2006-2007 field performance test. It showed better growth characteristics and yield performance than standard cultivar KF109. It had 2 more leaves per plant, flowered 2 days later than KF109. The yield of cured leaf of KF120 was increased by about 5% compared to KF109. The chemical composition and physical properties of the cured leaf of KF120 were as much acceptable as those of KF109. KF120 showed high resistance to PVY compared to KF109. It showed a similar mode of resistance to bacterial wilt and black shank as was found in KF109.

Development of Hybrid Rice Facilitated by Cytoplasmic Genetic Male Sterility II. Responses of Korean Cultivars to the Chinese Cytoplasmic Genetic Male Sterile lines (웅성불임성을 이용한 수도잡종품종개발 II. 중공의 세포형질 유전적 웅성불임계통에 대한 몇가지 한국품종의 반응)

  • Chinese CGMS (Cytoplasmic-Genetic Male Sterility) is being transfered to Korean cultivars which will be used as CGMS maintainers. These maintainer lines along with the Chinese CGMS lines and their maintainer lines were crossed with another group of Korean cultivars which will be used as the fertility restorers, and their yield heterosis were examined. The yield heterosis was calculated as high as 113.0-221.1 % in terms of heterosis (F$_1$/Midparent) with the grand mean of 150.5%, and, 86.1-179.8% in terms of heterobeltiosis (F$_1$/better parent) with the grand mean of 125.3%. Among the female parent groups, Chinese mainteiners were the highest following by Chinese MS lines and the Korean maintainers were the lowest. Heterobeltiosis of yield components were 92.7%, 120.0%, 103.0% and 112.0% for number of panicles/unit area, number of grains/panicle, fertility per cent and 1000 grain weight, respectively. Path-coefficients of yield components to the yield were calculated. The highest one was number of grains per panicle ranging 0.8073-0.8649 followed by the 1000 grain weight ranging 0.2000-0.5032. The grain yield ratios to the grand mean were calculated in terms of combining ability. It was highest with the Chinese maintainers followed by Chinese MS lines and The Korean maintainers were lowest. This facts bring cautions to the breeding efforts to transfer the Chinese CGMS in to Korean maintainers.

A New F1 Hybrid Variety of Rapeseed (Brassica napus L.), 'Jomang' with Early Maturation, High Yield and High Oleic Acid (조숙, 다수성, 올레인산 고함유 유채 신품종 '조망')

  • 'Jomang' is a new $F_1$ hybrid variety of rapeseed (Brassica napus L.) with early maturation, high yield and high oleic acid. This hybrid variety was developed from the cross between Mokpo-CGMS (male sterile line) and 8516-B-5-6-5-3 (restore line) for the production of edible oil and biodiesel in 2006. Yield trials were conducted from 2007 to 2008, and regional adaptation trials were examined at four locations in 2008. 'Jomang' has green and parted leaf, yellowish flower and black seed coat. The ripening date of 'Jomang' is 1~4 days earlier than 'Sunmang' and 'Tammiyuchae'. The average seed yield of the regional adaptation trials was 406 kg/10a, which was 7% and 42% higher than those of 'Sunmang' and 'Tammiyuchae,' respectively. Total oil content of 'Jomang' was 44.5%. 'Jomang' has no erucic acid in fatty acid composition. However, oleic acid content is 68.2%, which is 3.4% and 5.1% higher than 'Tammiyuchae' and 'Sunmang,' respectively. Total glucosinolate content was 2.31 mg/g. Therefore, this variety is recommended as a leading variety at southwestern area including Jeonnam, Jeonbuk and Kyongnam provinces and Cheju island of Korea.

A New F1 Hybrid Variety of Rapeseed 'Suan' with Early Maturing and High Oleic Acid (조숙 올레인산 고함유 1대잡종 유채 신품종 '수안')

  • 'Suan' is a new $F_1$ hybrid variety of rapeseed (Brassica napus L.) with early maturing and high oleic acid. This hybrid variety was bred by the cross between Mokpo-CGMS (male sterile line) and 8516-B-5-6-5-3 (restore line) for the production of bio-diesel and edible oil in 2006. 'Suan' has green and parted leaf, yellowish flower, and black seed coat. 'Suan' is more tolerant to lodging and stem rot compared to 'Sunmang', check variety. The ripening date of 'Suan' is June 2nd which is 6 days earlier than 'Sunmang'. Yield trials were conducted from 2006 to 2007 and regional adaptation trials were examined at five locations each in 2008 and 2009. The average seed yield of regional adaptation trials was 381 kg/10a that was 4% higher than that of 'Sunmang'. Total oil content of 'Suan' was 44.3%. Oleic acid content was 68.3%, which is 5.2% higher than 'Sunmang' but, erucic acid was not detected. Total glucosinolate content was 2.31 mg/g. Therefore, this variety is recommended as a leading variety at southwestern area including Jeonnam, Jeonbuk and Kyongnam provinces of South Korea.

Characteristics Related to Photosynthesis and Grain Yield of $F_1$ Hybrid Rice (벼 1대잡종의 광합성 및 수량 관련 특성)

  • This study was carried out to investigate heterosis in photosynthetic characteristics and yield of F$_1$ hybrid rice using genetic cytoplasmic male sterile lines. Two F$_1$ hybrid rices, HR7028A/Yongmoonbyeo and IR54756A/Yongmoonbyeo, showed heterobeltiosis in dry weight and leaf area at the heading date and at the 20 day after heading. HR7028A/Yongmoonbyeo showed heterobeltiosis and IR54756A/Yongmoonbyeo showed heterosis in chlorophyll content and RuDPCase activity in flag leaf, T-sugar and starch content in stem, and $CO_2$ fixation at the 20 day after heading as well as at the heading date. HR7028A/Yongmoonbyeo showed heterobeltiosis in number of panicles and grains per panicle, and IR54756A/Yongmoonbyeo had heterobeltiosis in number of panicles per plant. As results, heterobeltiosis of HR7028A/Yongmoonbyeo in yield was 11% and that of IR54756A/Yongmoonbyeo was 4%.
