This study was performed to investigate the effect of a raw diet (RD) on blood glucose and immune function in non-diabetic (normal) and streptozotocin (STZ)-induced diabetic rats. Male Sprague-Dawley rats were assigned to four groups (normal control, normal RD, diabetic control and diabetic RD). The control groups and the RD groups were fed an AIN-diet and RD for four weeks, respectively. Weight gain was statistically lower in the RD groups than in the controls. Fasting plasma glucose was significantly lower in the diabetic RD group than in the diabetic control group. The $CD4^+$ T-cell population was higher along with the $CD4^+/CD8^+$ ratio of the mesenteric lymph nodes in the normal RD group compared to the other groups. It can be concluded that RD may reduce the plasma fasting glucose concentration in diabetic rats and improve mesenteric lymph node immune function in normal rats.
An effect of carbon tetrachloride (CCl$_{4}$) was studied on the xanthine oxidase(XOD)activity of small intestine in male rats. Concomitantly a cause of increasing small intestine XOD was focused on an effect of actinomycin D and the kinetics of partial purified XOD frdm small intestine in CCl$_{4}$ intoxicated rats. An injection of CCl$_{4}$ to the rats showed an increase of small intestine XOD. In the pretreatment of actinomycin D before injection of CCl$_{4}$ to the rats, the XOD activities of small intestine were significantly decreased. In the partial purified enzyme preparation, the small intestine XOD in CCl$_{4}$ intoxicated rats showed the more increased Km and Vmax value with xanthine as substrate than that of control group.
Hypoglycemic efficacy of natural functional mixture(FM) and level of the diabete related hormones in streptozotocin(STZ)-induced diabetic rats were investigated. Male Sprague-Dawley rats were divided into three groups (normal, diabetic fed diets with/without FM). Supplement of FM did not affect the body weight and feed intake of STZ-induced diabetic rats. The increase in the weight of liver of STZ-induced diabetic rats was weakened by supplement of FM, whereas the weight of kidney and heart was not affected. Blood glucose level was slightly, and glucose tolerance of post-feeding was significantly improved by functional mixture. The mixture significantly reduced the elevated HbA1C level of diabetic rats by 15%, and it increased the level of insulin and C-peptide in blood and decreased glucagon level. Therefore, we conclude that FM in this study has a potency of prevention and treatment of diabetes mellitus.
This study was carried out to examine how dietary protein and calcium levels in rats fed fat-enriched diet affect the total lipid and cholesterol contents of blood and tissues. Male Sprauge-Dawley rats weighing approximately 200g were fed six purified diets which contained 18%(w/w) beef tallow, 1% (w/w) cholesterol, two source of protein, casein or isolated soy protein (ISP) and three levels of dietary calcium, 0.1%, 0.4% and 1.0%, first, for four weeks, and second, for eight weeks. The contents of the total lipid, cholesterol and triglyceride in blood, liver, heart and feces were determined. After four weeks feeding serum lipid and cholesterol concentrations significantly decreased in rats fed 1.0% (w/w) level calcium, regardless of dietrary protein sources. After eight weeks, these concentrations were significantly lower in the rats fed soy protein than in casein-fed rats. As dietary calcium level increased serum and tissue lipid and cholesterol contents were decreased and fecal lipid excretion increased. It is concluded that hypolipidemic and/or hypocholesterolemic effects of soy protein and calcium were partly due to decrease in lipid absorption.
Effects of fermented milk were tested in Sprague-Dawley male rats. In six treatment 150 rats were fed for 12 months: (1) normal control synthetic pellets+water, (2) cholesterol control synthetic pellets containing 1% of cholesterol and 500, 000 IU of vitamin D2/100 g (cholesterol pellets) +water, (3) cholesterol pellets+25% fermented milk, (4) cholesterol pellets+50% fermented milk, (5) cholesterol pellets+75% fermented milk, (6) cholesterol pellets+100% fermented milk. In 3, 6, 9, 12 months, rats were sacrificed for analysis of samples. Rats received the fermented milk had significantly lower (p<0.05) serum cholesterol levels and higher ratios of HDL-cholesterol to total cholesterol than did the water fed rats. Stomach, heart, abdominal aorta and kidney of rat from fermented milk group showed slighter calcification or necrosis than did those of rat from cholesterol control group. Weight gain, diet intake, and drink consumption were significantly different among groups. Results indicate that fermented milk had a helpful effect of experimental hypercholesteremia and atherosclerosis.
This study was performed to evaluate an single dose oral toxicity of a new hepatotherapeutic agent GODEX (HEPADIF-S) in Sprague-Dawley rats. Male and female rats were administered dosages of 5, 2.5, 1.25 ,0.625, 0.3125, and 0 g/kg B.W. of GODEX, respectively. After single oral administration of GODEX to rats, we observed them daily for 2 weeks. GODEX slid not induce any toxic signs in the mortalities, clinical signs, body weight changes, and gross necropsy findings of rats. Based on these results, it is concluded that GODEX may have no side effect and its LD$_{50}$ value may be over 5 g/kg B.W, in rats.s.
We investigated if Chlorella vulgaris (CV) has protective effects on cadmium (Cd) induced liver damage in male Sprague-Dawley (SD) rats. Forty rats, aged 5 weeks old and weighed 90-110g, were divided into a control (with Cd free water), 50 ppm of $CdCl_2$ in drinking water treated groups (Chlorella 0% diet group (Cd/CV0%), Chlorella 5% diet group (Cd/CV5%) or Chlorella 10% diet group (Cd/CV10%). All the rats had freely access to water and diet for 8 weeks. The results show that body weight gain and relative liver weight had significantly lower in Cd/CV0%-treated group than in Cd/CV-treated groups. Hepatic Cd contents showed significantly less by feeding CV (P<0.05). Cd/CV0%-treated rats had significantly (P<0.05) higher hepatic T-MTs, and Cd-MTs concentrations, compared to Cd/CV5% or Cd/CV10% treated rats. The MT I/II mRNA was expressed in the liver of all experimental rats. Its expression was more increased in Cd/CV5%- or Cd/CV10%-treated rats, compared to control and Cd-treated rats. Thus, this study suggested that CV would have a protective effect on Cd-treated liver injury by the reduction of Cd concentrations and stimulation of Cd-MT binds in the liver. However, more studies are needed to identify the proper mechanism of CV and liver toxicity.
This study investigated the in vivo insulin function of Polygonatum odoratum in normal and diabetic male Sprague-Dawley rats. Diabetes mellitus was induced by an i.p. injection of streptozotocin. Normal and diabetic rats were assigned to the diet groups of the control basal diet and Polygonatum odoratum diet. The animals were fed the diet and water ad libitum for 15 days. Initial and final body weights, total food intake and serum glucose and insulin levels were measured. An insulin suppression test was performed to elucidate the insulin function in the peripheral tissues. The results showed that the final serum glucose levels significantly decreased in the diabetic rats on the Polygonatum odoratum diet compared with the diabetic rats on the control diet. The final serum insulin levels were increased in the diabetic rats on the Polygonatum odoratum diet compared with the diabetic rats on the control diet. The in vivo function of the insulin increased in the diabetic rats on the Polygonatum odoratum compared with the diabetic rats on the control diet. These data indicate that Polygonatum odoratum may be beneficial in improving the in vivo insulin function in streptozotocin-induced diabetic rats.
This study was conducted to investigate the growth rate and hematological changes of the rats when supplemented with the powdered tubers of chufa(Cyperus esculentus) for four weeks. Nutritional composition of the powdered tuber of chufa is crude lipid(22.3%), crude protein(7.0%), crude fiber(8.8%), ash(5.1%), nitrogen free extract(48.9%) and water(7.9%), respectively. Sixteen Sprague-Dawley male $rats(173.1\pm3.8$ g) were randomly divided into two groups, control group and chufa group. Rats in control group were fed with American Institute of Nutrition(AIN)-76 diet and rats in chufa group were fed with modified AIN-76 diet contained the powdered tubers of chufa at the proportion of 26% of total diet in order to substitute the oil content of AIN-76 diet. Final body weight, gain of body weight and feed intake of rats in chufa group were larger than those of rats in control group significantly. But feeding efficiency of rats in chufa group was less than that of rats in control group significantly. All hematological values such as hemoglobin concentration and hematocrit and serological values such as glucose, total protein, albumin, AST, ALT, cholesterol, high density lipoprotein-cholesterol and triglyceride were not significantly different between control and chufa group, respectively. Thus powdered tuber of chufa showed good tastefulness and did not provoke any adverse effect to the health indicators of rats. Therefore tuber of chufa would be used as new adequate feed resource for animals and possibly as suitable food resource for humans.
The hypoglycemic effects of four edible plants (Angelicae tenuissimae (A. ten.), Pleurospermum kamtschaticum (P. kam.), Adenophora remotiflora (A. rem.) and Zanthoxylum schinifolium (Z. sch.)) in streptozotocin (STZ) -induced diabetic rats were investigated. Sprague-Dawley male rats weighing 190-230 g were induced diabetes mellitus by the STZ injection (45 mg/kg) into the tail vein and were divided into six groups ; normal, STZ-control and four edible plant groups (A. ten., P kam., A. rem. and Z. sch. groups). Normal and STZ-control groups were fed a AIN-93 diet and four groups of STZ-induced diabetic rats were fed one of each experimental diets containing 10% of the edible plant powder for 4 weeks. Diabetic rats showed the lower weight gain compared to the normal rats. In experimental groups except P. kam., AST activities were close to normal. A. ten. group were lowered ALT activities slightly. The plasma glucose levels of the diabetic experimental groups were significantly decreased at 4th week. The plasma insulin levels in diabetic experimental groups were not significantly different compared to the STZ-control group. The liver glycogen levels in STZ injected rats were significantly lower in compared to the normal rats. However no significant differences were found in response experimental plants intake in diabetic rats. The muscle glycogen were not significantly different among all the groups.
