살균성, Phenylthionocarbamate 유도체들의 정량적인 구조와 독성과의 관계 (Quantitative Structure-Toxicity Relationships (QSTRs) of Fungicidal Phenylthionocarbamate Derivatives)

  • 아직까지 시도된 바 없는 살균성 phenylthionocarbamate 유도체들의 phenyl-치환기가 변화함에 따라 TOPKAT 계산으로 예측된 다양한 급성 및 만성 독성값에 미치는 정량적인 분자구조와 독성과의 관계 (QSTRs)를 검토한 결과는 다음과 같다. (1) 기질분자의 구조변화에 따른 독성치와 그의 판별점수 (D.S.) 에 기초하여 분자 중 특정부분 (fragment)이 양 (+)의 값으로 독성에 기여하는 대체적인 순서는 Aro. C=C, -O-, -NH- 및 할로겐 (X) 원자의 순이었다. (2) 대부분의 화합물들은 매우 높은 돌연변이와 발암성이 예측되었으며 특히, 치환기의 위치에 관계없이 fluoro-치환체는 $B_2$상수에 따른 입체효과 ($(B_2)_{opt.}=1.54{\AA}$)에 의하여 모두 100% 돌연변이를 발현한 반면에 trifluorornethyl-치환체는 돌연변이 발현 가능성이 전혀 없었다. (3) 가장 높은 독성 발현조건은 phenyl-치환기에 대한 소정의 적정값으로 돌연변이성에는 $(B_2)_{opt.}=1.54{\AA}$, 발암성에서 수컷 rat와 mouse는$(R)_{opt.}=0.16$$(\pi)_{opt.}=0.16$ 그리고 rat의 경구독성은 $LD_{50};({\varepsilon}LOMO)_{opt}=-0.52e.v.$, chronic LOAEL; $(B_3){opt.}=1.54{\AA}$, 어독성은 ${LC_50}$; $(logP)_{opt.}=4.25$ 및 물벼룩에 대한 독성은 $EC_{50};({\sigma})_{opt}=-0.68$를 나타내는 경우 이었다. (4) 돌연변이성에는 할로겐 중, fluoro-기를 위시하여 nitro 및 methyl-기 등의 순서로 기여하였고 phenyl 고리 상, 치환기의 위치와 전자 수수관계에는 대체로 무관한 경향을 나타내었다.

  • This study was carried out to investigate the effect of red ginseng extract on development of pepsinogen 1 decrased pyloric glands in experimental stomach carcinogenesis in Sprague-Dawley male rats. Sequential quantitative analyses (by ABC immunohistochemical staining) were made of pepsinogen 1 decreased pyloric glands (PDPG) after treating rats first with a single dose (160 mg/kg) N-methyl-N'-nitrosoguanidine (MNNG) and then with N-ethyl-N'-nitro-N-nitrosoguanidine (ENNG) (100ug/ml of drinking water) as a second gastric carcinogen (or promoter).

  • SCi-B-Vac, the 3rd hepatitis B vaccine , was selected for clinical evaluation on the basis of toxicologic profiles in preclinical studies. These studies were performed to obtain information on its toxic signs, organs which are mainly affected, and to estimate its lethality in mice and rats given Sci-B-Vac through two routes of administratin. In male and female rats given a single intragastrical dose of Sci-B-Vac, we estimated that $LD_{50}$ values were over 2.00 ml/100g B.W. (10ng/ml), respectively.

  • The recombinant glycoprotein, LBD-005 (Lucky R & D Center, Biotechnology)stimulates the growth of stem cells and activates the development of the hematopoietic cell in a similar manner to the naturally occurring GM-CSF. This test was conducted to investigate if LBD-005 had any effect on fertility in male and female rats. Sprague-Dawley rats(88 of each sex) bred at KRICT, were dosed subcutaneously, at a volume of 2ml/kg body weight with LBD-005 at 0, 250, 500 or 1, 000mg/kg body weight. The male rats were dosed, from 6 weeks of asge, for 60 days prior to mating.

PMSG를 투여한 미성숙랫드에 발생한 자궁축농증 발생례 (Pyometra in the PMSG-treated Immature Female Rats)

  • Nine immature 30-day-old female rats were injected sc at 0800 hr with pregnant mare serum gonadotrophin(PMSG) to induce ovulation and mating. Fifty-six hours later the animals were placed with mature male rats overnight (one female and one male). Five of 9 immature female rats treated with PMSG were pregnant and allowed to maintain the pregnancy to term. Three of 5 pregnant rats were failed to maintain pregnancy to term. Two of 5 pregnant rats seemed to be developed normally and increased abdominal enlargement as pregnancy progresses, but did not occurred parturition on day of 43 or 48 of pregnancy, respectively. On day 44 or 49, pregnant rats were killed and examined uterus and ovaries. There was no fetus but approximately 50∼60ml. of mucopurulent fluids were accumulated in the uterine cavity and 40 or 42 corpora lutea persisted in the ovaries. Pyometra was developed after coitus in PMSG-treated immature female rat.

흰쥐의 성별, 체중 및 실험사료의 첨가 방법이 대사 에너지 이용성에 미치는 영향 (The Effects of Feed Supplementation Method, Sex and Weight of Experimental Animal on Metabolizable Energy Value)

  • This study was to investigate the validity of energy measurement system by using different body size and sex of rats and feed supplementation methods. There appeared no statistical differences in energy(AME, AMEn) utilization between male and female rats with average 200g body weight, and between male rat of 200g and 300g body weight. Glucose replacing corn in the basal diet or replacing diet itself at a level of 30% by weight appeared to have the same metabolizable energy value, suggesting that either methods to supplement test ingredient brought about the same results. Consequently, following suggestions could be made from results of present study : first, Sprague-Dawley rate of either sex weighing 200-300g may be used as an animal model to obtain in vivo metabolizable energy from dietary carbo-hydrates or sea foods. Second, the testing ingredients may be added at the expence of corn in the basal diet at 30% level.

  • The antioxidant effects of aqueous green tea extracts with $\alpha$-tocopherol and lecithin dissolved in lard were investigated on the lipid metabolism in serum and liver of Sprague-Dawley male rats. Elderly male rats were fed for 8 weeks by different experimental diets containing 10% lard(w/w), 0.5% aqueous green tea extracts, 0.05% $\alpha$-tocopherol and 1.0% lecithin. By the addition of the mixture of the antioxidants, the level of total cholesterol. HDL-cholesterol and triglyceride in serum and liver were not changed. The activities of glutathione peroxidase were significantly increased in the case of addition of the antioxdant mixtures but the activities of superoxide dismutase and glutathione S-transferase were not affected.

흰쥐에서 JG-381의 만회경구독성시험 (Single Oral Toxicity of JG-381 in Rats)

  • The single oral toxicity of JG-381 was studied in Sprague-Dawley rats of both sexes. In this study, rats were administrated orally with dosages of 267, 400, 600, 900 and 1350 mg/kg of JG-381. We daily examined number of deaths, clinical signs, body weights and gross findings for 14 days after JG-381 administration. When we administered different doses of 267, 400, 600, 900 and 1350 mg/kg, we found 1, 4, 4, 5 and 5 male rats died and 3, 5, 4, 5 and 5 female rats died within 1 day after administration, respectively. Some clinical signs (decrease locomotor activity, salivation, soft stool, prone position, lacrimation, crouching position, convulsion, ataxic gait, incontinence of urine) were also observed during the experimental period. Our findings suggest that oral L $D_{50s}$ (95% confidence limit) for male and female rats are 327 mg/kg (270~396 mg/kg) and 250 mg/kg (256~264 mg/kg), respectively.y.
