  • ${\beta}-Galactosidase$ activity of Aspergillus phoenicis was studied using ONPG and lactose as substrate. It increased monotonically during the exponential growth phase and dropped rapidly at the beginning of the stationary one. It exhibited high tolerable temperature and acidic optimal pH which provides certain advantages from the industrial view point. Enzyme of ${\beta}-galactosidase$ had more subsrate affinity for ONPG than for lactose and its apparent maximum activity was also higher with the former as substrate. Activity of this enzyme depended upon the conditions of immobilization. Optimum crosslinking reaction was occurred at pH 7.2 and 0. 35 vol. % of glutaraldehyde concentration.

The Influence of Self-directed Learning & Critical Thinking Disposition on Clinical Competence in Nursing Students (간호학생의 자기주도적 학습, 비판적 사고성향이 임상수행능력에 미치는 영향)

  • Purpose: The purpose of this study was to investigate self-directed learning and critical thinking disposition which influence clinical competence in nursing students. Method: As a descriptive study, a total of 271 third year students were recruited from a nursing college in Daegu. A structured questionnaire was self-administered from June 7, 2011 to June 10, 2011. The data were analyzed using SPSS/WIN version 12. Results: In self-directed learning, there were significant differences in gender (t=2.26, p=.025), satisfaction of major (F=7.77, p=.001), and satisfaction of clinical experience (F=10.39, p<.001). Critical thinking disposition differed in gender (t=2.82, p=.005). Moreover gender (t=4.00, p<.001), satisfaction of achievement (F=6.50, p=.002), satisfaction of major (F=4.24, p=.015), and satisfaction of clinical experience (F=9.54, p<.001) differed with clinical competence. Clinical competence was positively related to self-directed learning (r=.45, p<.001) and critical thinking disposition (r=.51, p<.001). According to the result of multiple regression, critical thinking disposition (t=5.80, p<.001), satisfaction of achievement (t=3.33, p=.001), gender (t=2.93, p=.004) and self-directed learning (t=2.35, p=.019) were significant factors of clinical competence explaining 33.0% of the variances. Conclusion: Critical thinking disposition, satisfaction of clinical experience, gender and self-directed learning had a positive effect on clinical competence in nursing students. To enhance clinical competence for nursing students, it is necessary to develop self-learning teaching strategy and curriculum.

  • The protease produced by a Bacillus pumilus CN8 strain was purified by DEAE-Cellulose-52 ion exchange. It has a molecular weight of approximately 96,920 Dalton. In the present study, this protease showed strong activity over a broad range of pH (6.5-9.5) and temperature from $40^{\circ}C$ to $60^{\circ}C$, and the protease performed the maximal activity at pH 7.3 at $42^{\circ}C$. The effect of metal ions on protease activity showed that $K^+$ could slightly increase the protease activity, and other ions such as $Zn^{2+}$, $Fe^{2+}$, $Na^+$, $Ca^{2+}$, $Mg^{2+}$ had no significant activation or inhibition to the protease (P> 0.05), and the more important is that $Cu^{2+}$, $Mn^{2+}$, $Sn^{2+}$, $Cd^{2+}$ had a strong inhibitory effect on the protease activity.

Isolation of Protoplasts from Rhizopus nigricans (Rhizopus nigricans로부터 원형질체의 분리)

  • Conditions for isolation of protoplasts from spores and mycelia of Rhizopus nigricans were studied. Larger amount of protoplasts was obtained from swollen spores in liquid medium contained with 5% of 2-deoxy-D-glucose for 4 hours than from mycelia. Enzyme mixture of Novozym 234(2%) and ${\beta}-glucuronidase(5000\;unit/ml)$ was most effective for the isolation of protoplasts from swollen spores and from mycelia. The solution of 0.6 M $MgSO_4$ or mannitol and pH 6.0 showed good results as the osmotic stabilizer and the optimal condition of pH of the enzyme solution for the isolation of protoplast from the swollen spores, respectively. At this condition, $8.0{\times}10^6\;cells/ml$ of protoplasts was obtained from swollen spores by digestion with lytic enzyme mixture for 2 hours.

  • Chinese shroud through literature are as follows. 1. Taetae, SimeI, P'oo, Hansam, Ko, Mal, Nukpaek , Kwatu, Ch'ungi, Pokkn, Myokmok, Ri, Aksu, Mo and m were used the most in China. 2. The cloths of Chinese shroud were p'o, Paek , Kyon and Kum. The colors of the Chinese shroud were Hyon, Hun and white. 3. The size of the Chinese shroud is as follows . The size of the Ch'ungi was similar to the size of jujube kernel, the length of Myokmok was one Chk two Chn or one Chk two Chn or one Chk five Chn, the length of Aksu was one Chjk two Chn and it's width was five Chn. The chil of Mo reached the hands and the length of Swae was three Chk and the length of m was five Chn. 4. In Chinese shroud, , cotton was put in P'oo. Aksu was tide by the strings at two corners. Myokmok was tied by the strings of four corners. The tip of the m was divided and Mo warpped the whole body. 5. The clothes of Soryom was nineteen Ch'ing. The clothes of Taeryom in Kum were one hundred Ch'ing in the Chinese. The impliment of Soryon were Kum, kyo, SangeI, SaneI, Ch'im , Yok and Kyon in the Chinese shroud. In the case of the implement of TAeryom, the chinese shroud had Kum , Kyo, SangeI, Sane, Ch'im and Yok.

Recombination and Expression of VP1 Gene of Infectious Pancreatic Necrosis Virus DRT Strain in a Baculovirus, Hyphantria cunea Nuclear Polyhedrosis Virus (전염성 췌장괴저바이러스 DRT Strain VP1유전자의 Baculovirus Hyphantria cunea Nuclear Polyhedrosis Virus에 재조합과 발현)

  • Expression of the cDNA of the VP1 gene on the genome RNA B segment of infectious pancreatic necrosis virus (IPNV) DRT strain in E. coli and a recombinant baculovirus were carried out. The VP1 gene in the pMal-pol clone (Lee et al. 1995) was cleaved with XbaI and transferred into baculovirus transfer vector, pBacPAK9 and it was named pBacVP1 clone. The VP1 gene in the pBacVP1 clone was double-digested with SacI and PstI and then inserted just behind T5 phage promoter and the $6{\times}His$ region of the pQE-3D expression vector, and it was called pQEVPl. Again, the $6{\times}$His-tagged VP1 DNA fragment in the pQEVP1 was cleaved with EcoRI and transferred into the VP1 site of the pBacVP1, resulting pBacHis-VP1 recombinant. The pBacHis-VP1 DNA was cotransfected with LacZ-Hyphantria cunea nuclear polyhedrosis virus (LacZ-HcNPV) DNA digested with Bsu361 onto S. frugiperda cells to make a recombinant virus. One VP1-gene inserted recombinant virus was selected by plaque assay. The recombinant virus was named VP1-HcNPV-1. The $6{\times}$His-tagged VP1 protein produced by the pQEVP1 was purified with Ni-NTA resin chromatography and analyzed by SDS-PAGE and Western blot analysis. The molecular weight of the VP1 protein was 94 kDa. The recombinant virus, VP1-HcNPV-1 did not form polyhedral inclusion bodies and expressed VP1 protein with 95 kDa in the infected S. frugiperda cells, which was detected by Western blot. The titer of the VP1-HcNPV-1 in the first infected cells was $2.0{\times}10^5\;pfu/ml$ at 7 days postinfection.

Effect of Stroke Prevention Program for The Elderly in The Rural Area (농촌지역 노인을 위한 중풍예방 프로그램의 효과)

  • The goal of this study was to examine how effective stroke prevention program is for body mass index, blood pressure, blood glucose, and serum lipids of the elderly in the rural areas. The stroke prevention program included health care education, nutrition guidance and exercise. The program was provided for 12 weeks(36 sessions) from May to August, 2014. Participants were 55 elderly people in the rural areas; 28 people in the experimental group and 27 in the control group. The results of before and after the experiment showed that body mass index(p<.001), TC(p<.001), TG(p=.047), and LDL(p=.010) were significantly better in the experimental group compared to the control group. Also, after the program, the figures of body mass index, fasting blood glucose, TC, TG, and HDL in the control group were decreased, while those of LDL were increased. Study findings indicate that the stroke prevention program is effective and can be recommended as a precaution against the stroke of the elderly in the rural area by preventing risk factors beforehand.

Modified Differential Protection for Transformers in Wind Farms

  • The liberalization of electricity market and environmental concerns are the major driving forces for the development of Distributed Generation (DG). The mode of grid-connected wind power generation is becoming popular and has matured as a reliable DG technology. The voltage generated by the wind generator is stepped up to the higher voltage by the transformers before connecting to the grid. Operating algorithm of the differential relays for transformer protection used in the wind farms need to be modified to take care of the dynamic nature of fault current caused by the intermittent nature of the wind power. An algorithm for the differential relay is proposed in which dual slope characteristics are adjusted with varying fault level situation according to the wind generator in service as well as with the wind speed. A case study conducted for a typical wind farm shows that the proposed method avoids mal-operation of the differential relay in varying wind power conditions.

Studies on the Blood Chemical Values in Thoroughbred Foals (Thoroughbred종 망아지의 혈액화학식에 관한 연구)

  • The considerable researches have been done on the change,1 associated with age in the blood picture of domestic animals, but a survey has not been of the blood picture in Thoroughbred fears in Korea. Therefore, the purpose of this study is to outline the range of values in clinically not\\\\`- mal Thoroughbred foals. Some blood chemical parameters from 3 days to 8 weeks of age were exam-ined to obtain normal values in Thoroughbred foal7. Blood samples were obtained from the jugular vein in 11 Thoroughbred foals and collected at 3 days, and at 1,2,4 and 8 weeks of age, respectively. The results obtained ill this study were summarized as follows: The packed cell volume (PCV) at 4 weeks of age was significantly lower than that at 3 days of age (P<0.01) and that al 2 and 8 weeks of age were straightly lower than that at 3 days of age (P<0.05). The total protein (TP) at 8 weeks of age was higher than that at 3 days, 1 and 2 weeks of age (P<0.05). The activity of aspartate ami-notransferase (AST) at 2,4 and 8 weeks of age were significantly higher than that at 3 days of age (P<0.01). The total bilirnbin showed a tendency to decrease gradually according to the weeks and that at 2,4 and 8 weeks of age were significantly lower than that at 3 days of age (P<0.01). The BUN at 1 and 8 weeks of ago were lower than that at 3 days of age (P<0.05). The concentration of Ca at 1 week of alee was higher than that at 3 days of age (P<0.05). The vitamin A and vitamin E showed a tendency to decrease gradually from 3 days to 4 weeks of age and increased slightly at 8 weeks of age.
