Screening methods for drought and salinity tolerance with transgenic rice seedlings

  • Abiotic stress is one of the major serious limiting factors in rice (Oryza sativa) and caused rice production losses. It is important to precisely screen valuable genetic resources for improving stress tolerance and understanding tolerance mechanism to abiotic stresses. Because there are differences of experiment designs for screening of tolerant plant in several studies related to abiotic stress, this study has performed to provide the rapid and efficiency screening method for selection of tolerance rice to drought and salinity stresses. Two week-old rice seedlings that reached about three leaf stage were treated with drought and salinity stresses and examined tolerant levels with tolerant and susceptible control varieties, and transgenic plants. To determine the optimum concentration for the selection of drought and salinity condition, tolerant, susceptible and wild-type plants were grown under three soil moisture contents (5, 10 and 20% water contents) and three NaCl concentrations (100, 200 and 250 mM) for 10 days at seedling stage. 200 mM NaCl concentration and 5% moisture content soil were determined as the optimum conditions, respectively. The described methodologies in this study are simple and efficiency and might help the selection of drought and salinity tolerance plants at the 3,4-leaf-seedling stage.

  • This study was carried out to find the botanical characters for 27 color rice genetic resources collected from domestic 17 lines (including three varieties) and exotic 10 lines. We could gain the major informations from these materials for development of color rice variety. These lines showed a various variations in characters measured. Especially, Denorado introduced from Philippine was longer in culm length, panicle length and 1,000 grains weight than others. Whereas grain length and width of Sinucninul line introduced from Japan were similar to check, Heukjinjubyo. Heterosis degree for major characteristics of Suwon#432 ${\times}$ Heukmi#940018 combination among $F_1$ hybrids superior to others in tillers per plant, panicle length and 1,000 grains weight. Therefore, these lines and their combinations were expected as color rice developing line through progeny test in the future.

  • Food productivity in North Korea is about 50% lower than in South Korea. In order to increase the productivity of major crops, it is necessary to develop early maturing, disease resistance, and high-yielding varieties and apply them early. Since the late 1990s, North Korea has been actively developing potatoes, rice and com as major food crops, and soybeans are considered important as a protein-supplying crop. Domestic cultivated varieties, which are expected to be most adaptable eco-climatologically, are mainly selected from soil with high nutrient soil. It is necessary to test separately for adaptability in low organic soil. So it is very necessary to apply technology to improve soil improvement through rotational crop selection in the middle and long-term. Therefore, this study was conducted to test the adaptability to low organic soils of domestic cultivars and to select varieties. In 2021 there are twenty two (22) varieties of soybeans were grown in low organic soil at the field of Chungbuk National University. This year twenty two (22) varieties of soybeans were also grown in low organic soil at the field of Chungbuk National University. Sowing was done on June 10, the planting distance was 70cm × 15cm, after opening the cotyledons fully, the soybeans were thinned and leaving two plants per hole. In addition, various types of growth characteristics and quantitative components were investigated to evaluate the adaptability to low organic soil of domestic varieties. This study was conducted to investigate the growth characteristics and quantitative components of soybean varieties grown in low organic soil. The flowering period of 22 varieties of soybeans was about 14 days from July 22 to August 4. The flowers of the beans were white, purple, light purple and the pubescence color was gray and brown where most of them were gray. The highest plant height was up to 130.4 cm and lowest was 20.3 cm, highest stem length was up to 119.5 cm and lowest was 15.3 cm. Highest first pod height (FPH) was up to 34.0 cm and lowest was 3.0 cm. Highest stem diameter was up to 15.76mm and lowest was 1.76 mm. Number of main stem nodes was up to 19 and at least 1. Number of branch was up to 10 and at least 0. The number of pod per plant was up to 121. Bacterial pustule has been spread in soybean field.

Genetic interaction of Sub1A and Pup1 in rice

  • Rice is one of the major staple food in Asia, covering around half of the world population. More than 40% of rice cultivation area are subject to abiotic stresses such as drought, submergence and phosphate deficiency. Pyramiding useful genes into elite variety is a promising strategy to develop tolerance varieties to multiple abiotic stresses. However, some genes are not functionally compatible when they are introgressed into the same elite variety. Here, we tested the functional compatibility of Sub1 and Pup1, major QTLs for tolerance to submergence and phosphate (P)-deficiency conditions, respectively. Phenotypic analysis revealed that IR64-Sub1 Pup1(SP1) plants harboring both Sub1 and Pup1 QTLs showed significant tolerance to submerged conditions, similarly in IR64-Sub1 (Sub1) plant, while SP1 plants failed to tolerate to P-deficiency conditions; only IR64-Pup1 (Pup1) showed strong P-deficiency tolerance phenotype. In submerged conditions, the expression levels of Sub1A and PSTOL1, major genes for Sub1 and Pup1 QTLs, respectively, were not significantly different in between Pup1 and SP1 plants. On the other hand, the expression of both Sup1A and PSTOL1 was significantly downregulated in P-deficiency conditions, suggesting that Sub1 and Pup1 repressed gene expression each other in P-deficiency conditions. These results suggest Pup1 does not compromise the Sub1 function in submerged conditions while Sub1 suppresses the function of Pup1 in (P)-deficient condition, possibly by regulating transcript level of Pup1. In conclusion, Sub1 and Pup1 are functionally compatible in terms of submergence tolerance but not in P-deficiency conditions. Further analysis need to be performed to elucidate how Sup1 suppresses the function of Pup1 in P-deficiency conditions.

  • Salinity stress is a major limiting factor in cereal productivity. Many studies report improvements in salt tolerance using model plants, such as Arabidopsis thaliana or standard varieties of rice, e.g., the japonica rice cultivar Nipponbare. However, there are few reports on the enhancement of salt tolerance in local rice cultivars. In this work, we used the indica rice (Oryza sativa) cultivar BR5, which is a local cultivar in Bangladesh. To improve salt tolerance in BR5, we introduced the Escherichia coli catalase gene, katE. We integrated the katE gene into BR5 plants using an Agrobacterium tumefaciens-mediated method. The introduced katE gene was actively expressed in the transgenic BR5 rice plants, and catalase activity in $T_1$ and $T_2$ transgenic rice was approximately 150% higher than in nontransgenic plants. Under NaCl stress conditions, the transgenic rice plants exhibited high tolerance compared with nontransgenic rice plants. $T_2$ transgenic plants survived in a 200 mM NaCl solution for 2 weeks, whereas nontransgenic plants were scorched after 4 days soaking in the same NaCl solution. Our results indicate that the katE gene can confer salt tolerance to BR5 rice plants. Enhancement of salt tolerance in a local rice cultivar, such as BR5, will provide a powerful and useful tool for overcoming food shortage problems.

Phenotypic and genotypic screening of rice accessions for salt tolerance

  • Rice (Oryza sativa L.) is one of the major crops that is seriously impacted by global soil salinization. Rice is among those crops where most of the high-yielding cultivars are highly sensitive to salinity. The key to a plant survival under NaCl salt stress is by maintaining a high $K^+/Na^+$ ratio in its cells. Selection for salinity tolerance genotypes of rice based on phenotypic performance alone is less reliable and will delay in progress in breeding. Recent advent of molecular markers, microsatellites or simple sequence repeats (SSRs) were used to find out salt tolerant rice genotypes. In the current experiment phenotyping and genotyping studies were correlated to differentiate different rice accessions for salinity tolerance. Eight rice accessions along with check plant Dongjin were screened by physiological studies using Yoshida solution with 50mM NaCl stress condition. The physiology studies identified four tolerant and four susceptible accessions based on their potassium concentration, sodium concentration, $K^+/Na^+$ ratio and biomass. 17 SSR markers were used to evaluate these rice accessions for salt tolerance out of which five molecular markers were able to discriminate tolerant accessions from the susceptible accessions. Banding pattern of the accessions was scored comparing to the banding pattern of Dongjin. The study identifies accessions based on their association of $K^+/Na^+$ ratio with molecular markers which is very reliable. These markers identified can play a significant role in screening large set of rice accessions for salt tolerance; these markers can be utilized to improve salt tolerance of commercial rice varieties with marker-assisted selection (MAS) approach.

Effects of Main Shaft Velocity on Turbidity and Quality of White Rice in a Rice Processing System

  • Purpose: The purpose of this study is to analyze turbidity and quality characteristics of white rice as a function of main shaft blast velocity and to verify the optimum processing conditions in the cutting type white rice processing system (CTWRPS). Methods: Sindongjin, one of the rice varieties, which used to be produced in Gimje-si, Jeollabuk-do, in 2015, was used as the experimental material. Turbidity and quality characteristics of white rice were measured at three different main shaft blast velocities: 25, 30, and 35 m/s. The amount of test material used for a single experiment was 20 kg, and after processing, whiteness was found to be $42.5{\pm}0.5$, following which, turbidity and quality characteristics were measured. Results: Turbidity decreased with increase in the shaft blast velocity, and as a result, was lowest at 35 m/s of shaft blast velocity among all the other experiment velocities. The trend of cracked rice ratios was similar to the turbidity. Broken rice ratio turned out to be less than 2.0% in all the test conditions. In the first stage of processing, the processing pressure decreased as the main shaft blast velocity increased. Additionally, in the second stage of processing, the processing pressure was at its lowest value at the main shaft blast velocity of 35 m/s. Energy consumption, too, decreased as the main shaft blast velocity was increased. Conclusions: From the above results, it is concluded that the main shaft blast velocity of 35 m/s is best for reducing turbidity and producing high quality rice in a CTWRPS.

Studies on the Temperature Response and Critical Day-length Affecting the Heading Date of Major Cultivating Rice Varieties in Recent Korean Paddy Field (농가재배 주요 벼 품종들의 출수에 영향을 미치는 온도 반응과 한계일장 구명 연구)

  • Rice is sensitive to day-length and short-day plants. It has a characteristic that the photosensitivity response required for flower bud differentiation decreases under long-day conditions. To identify critical photoperiod required for flower bud differentiation of major cultivation rice varieties, the average temperature was fixed at 28 ℃, and the day length was set at 12 hours and 10 minutes intervals from 13 hours to 14 hours 30 minutes. The critical photoperiod for each cultivar was set to day-length, where the daily cumulative response [(X(Critical Photoperiod)-Y(Set day-length))/(X(Critical Photoperiod)-12:00(Optimal Day-length)) × (28.0(Set Temperature)-10(Minimum Temperature))/(29.2(Maximum Temperature)-10(Minimum Temperature))] was the same for each day-length conditions. The flower bud differentiation time of all varieties was 32 days before heading at the average temperature of 28 ℃ conditions. The critical photoperiod of the early maturing type, such as Woonkwang, Odae, Koshihikari, Jopyeong, were 19:20, 18:14, 18:58, 17:30, respectively. Medium maturing type, such as Daebo, Haiami, Samdeok, were 16:08, 16:15, 16:55, respectively. Mid-late maturing type, such as Saenuri, Sindongjin, Chucheong, Samkwang, Ilpum, Saeilmi, Hwangkeumnuri, Dongjinchal, Ilmi, Hopum, Yeonghojinmi, were 15:58, 15:56, 16:36, 16:44, 15:35, 16:26, 15:33, 16:20, 16:29, 16:13, 15:41.

Arsenic Concentrations of Groundwater and Rice Grains in Bangladesh and Phytoremediation (방글라데시의 지하수와 쌀의 비소오염 및 식물정화법)

  • While groundwater is the major source for drinking and irrigation purposes, arsenic (As) contamination of groundwater is a serious issue in Bangladesh. With a view to reduce As contamination in drinking water the guideline value recommended for Bangladesh is 0.05 mg/L. We assessed groundwater As in an As-affected Sadar Upazilla (small administrative unit) in the District (administrative unit) of Chapai Nabwabganj during 2006, where 50% hand tube well water were above the recommended limit (0.05 mg/L) during dry season. Almost 20% tube well waters were above the recommended limit during rainy season, perhaps due to the dilution of water table. The groundwater in Bangladesh contaminates surface soils and plants thereby As entering the food chain. In 2005, we examined the As levels in different rice varieties grown in different Districts of Bangladesh and the As concentrations in rice grain ranged from 0.07~1.12 mg/kg while the concentrations in 3 rice varieties were above the recommended limit (1 mg/kg rice grain) and the maximum concentration was 1.12 mg/kg rice grain in the rice variety BR 11. With few exceptions, the As content of rice grain in Bangladesh is not considered to be concentration of greater health concern as yet. We also observed enhanced root uptake, efficient root-to shoot translocation, and a much elevated tolerance through internal detoxification all contribute to As hyperaccumulation in a plant, ladder brake fern (Pteris vittata L.). But the phytoremediation technique might not be an appropriate tool to reduce the As calamity in the vast areas of Bangladesh. To mitigate the As problem of Bangladesh, better coordination among governmental agencies and many other organizations will be required to combat the disaster.

Variation of Properties of Lipid Components in Rice Endosperm Affected on Palatability (식미에 영향을 미치는 쌀 배유조성 지질성분특성 변이)

  • In order to understand the difference in rice eating quality, two rice varieties having different eating qualities were examined from the correlation between palatability score (Toyo value) and components of lipid in rice endosperm. We also analyzed the relationship between lipid properties and physicochemical characteristics of milled rice. Major fatty acids of milled rice starch-lipids, known to exist as inclusion complexes with amylose in starch granules, were palmitic (38.25% in Gopum and 39.75% in Palgong) and linoleic aicds (33.13% in Gopum and 31.93% in Palgong). In addition, the unsaturated fatty acid contents of rice endosperm showed a significant relationship with palatability. Plant sterols in two rice samples were identified, Gopumbyeo of good eating quality had high contents of squlaene and cycloartenol and similar contents of campesterol, stigmasterol and sitosterol. There were significant differences in pasting properties of rice flours and starches between the two cultivars. These results suggested that rice lipids impact on pasting properties after cooking, subsequently relating to palatability.
