Phase II Clinical Study on the GEMOX Regimen as Second-line Therapy for Advanced Ovarian Cancer

  • Aim: To investigate the effectiveness and adverse effects of gemcitabine by fixed-dose rate infusion plus oxaliplatin (GEMOX regimen) as second-line therapy for advanced ovarian cancer. Methods: 64 patients with advanced ovarian cancer were divided into an experimental group (44 cases) and a control group (20 cases). The experimental group was treated with continuous intravenous infusion of gemcitabine at 1000 $mg/m^2$ with a fixed-dose rate of 10 $mg/m^2/min$, on days 1 and 8 and oxaliplatin at 100 $mg/m^2$ on day 1, IVGTT, repeated every 3 weeks. The control group was treated with intravenous infusion of gemcitabine at 1000 $mg/m^2$ within 30 min on days 1 and and oxaliplatin at 100 $mg/m^2$ on day 1, IVGTT, again repeated every 3 weeks. CT scans or MRI were used for review every 1-2 cycles. Results: The effective rate in the experimental group was significantly high than control group (43.2% vs 35.0%; P < 0.05), with no obvious difference of hematologic or non-hematologic toxicity between the two groups (P > 0.05). Conclusion: GEMOX regimen is very effective to treat advanced ovarian cancer, with low toxicity, good tolerance and improved life quality in patients.

Development of Algorithms for Correcting and Mapping High-Resolution Side Scan Sonar Imagery (고해상도 사이드 스캔 소나 영상의 보정 및 매핑 알고리즘의 개발)

  • To acquire seabed information, the mosaic images of the seabed were generated using Side Scan Sonar. Short time energy function which is needed for slant range correction is proposed to get the height of Tow-Fish to the reflected acoustic amplitudes of each ping, and that leads to a mosaic image without water column. While generating mosaic image, maximum value, last value and average value are used for the measure of a pixel in the mosaic image and 3-D information was kept by using acoustic amplitudes which were heading for specific direction. As a generating method of mosaic image, low resolution mosaic image which is over 1m/pixel resolution was generated for whole survey area first, and then high resolution mosaic image which is generated under 0.1m/pixel resolution was generated for the selected area. Rocks, ripple mark, sand wave, tidal flat and artificial fish reef are found in the mosaic image.

Kinetics of Malonyl-CoA Synthetase from Rhizobium trifolii and Evidences for Malonyl-AMP Formation as a Reaction Intermediate

  • The catalytic mechanism of malonyl-CoA synthetase from Rhizobium trifolii was investigated by the steady state kinetics and intermediate identification. Initial velocity studies and the product inhibition studies with AMP and PPi strongly suggested ordered Bi Uni Uni Bi Ping-Pong Ter Ter system as the most probable steady state kinetic mechanism of malonyl-CoA synthetase. Michaelis constants were $0.17{\pm}0.04 {\mu}M,\;0.24{\pm}0.18 {\mu}M\;and\;0.045{\pm}0.26 {\mu}$M for ATP, malonate and CoA, respectively. The TLC analysis of the $^{32}P-labelled$ products in reaction mixture containing $[{\gamma}-^{32}P]$ ATP in the absence of CoA showed that PPi was produced after the sequential addition of ATP and malonate. Formation of malonyl-AMP, suggested as an intermediate in the kinetically deduced mechanism, was confirmed by the analysis of $^{31}P-NMR$ spectra of AMP product isolated from the $^{18}O$ transfer experiment using $[^{18}O]$malonate. Two resonances were observed, corresponding to AMP labelled with zero and one atom of $^{18}O$, indicating that one atom of $^{18}O$ transferred from $[^{18}O]$malonate to AMP through the formation of malonyl-AMP. Formation of malonyl-AMP was also confirmed through the TLC analysis of reaction mixture containing $[{\alpha}-^{32}P]$ATP. These results strongly support the ordered Bi Uni Uni Bi Ping-Pong Ter Ter mechanism deduced from the initial velocity and product inhibition studies.

  • In this communication, we introduce an Ishikawa type iterative algorithm for finding the approximate solutions of a class of nonlinear set valued variational inclusion problems. We also establish a characterization of strong convergence of this iterative techniques.

  • In the future, a machine-tool will be more improved in the form of a knowledge evolution based device. In order to develop the knowledge evolution based machine-tool, this paper proposes the structure of standard M2M(Machine To Machine) and the scheme of agent communication in environment. The communication agent such as dialogue agent has a role of interfacing with another machine for cooperation. To design of the communication agent module in M2M environment, FIPA(Foundation of Intelligent Physical Agent) and ping agent based on JADE(Java Agent Development Framework) or FIPA-OS(Open Source) are analyzed in this study. Through this, it is expected that the agent communication can be more efficiently designed and the knowledge evolution based machine-tool can be hereafter more easily implemented.

  • For the effective operation of manufacturing system, FMS(Flexible Manufacturing System) and CIM(Computer Integrated Manufacturing) system are developed. In these systems, a machine tool is the target of integration in last 3 decades. In nowadays, the conventional concept of machine tools is changing to the autonomous manufacturing device based on knowledge-evolution through applying advanced information technology in which open architecture controller, high speed network and internet technology are contained. In this environment, a machine tool is not the target of integration but the subject of cooperation. In the future, a machine tool will be more improved in the form of a knowledge-evolution based device. In order to develop the knowledge-evolution based machine tools, this paper proposes the structure of knowledge evolution in M2M(Machine To Machine) and the scheme of a dialogue agent among agent-based modules such as a sensory module, a dialogue module, and an expert system. The dialogue agent has a role of interfacing with another machine for cooperation. To design the dialogue agent module in M2M environment, FIPA-OS and ping agent based on FIPA-OS are analyzed in this study. Through this, it is expected that the dialogue agent module can be more efficiently designed and the knowledge-evolution based machine tools can be hereafter more easily implemented.

  • It is known that the ranks of the semigroups $\mathcal{SOP}_n$, $\mathcal{SPOP}_n$ and $\mathcal{SSPOP}_n$ (the semigroups of orientation preserving singular self-maps, partial and strictly partial transformations on $X_n={1,2,{\ldots},n}$, respectively) are n, 2n and n + 1, respectively. The idempotent rank, defined as the smallest number of idempotent generating set, of $\mathcal{SOP}_n$ and $\mathcal{SSPOP}_n$ are the same value as the rank, respectively. Idempotent can be seen as a special case (with m = 1) of m-potent. In this paper, we investigate the m-potent ranks, defined as the smallest number of m-potent generating set, of the semigroups $\mathcal{SOP}_n$, $\mathcal{SPOP}_n$ and $\mathcal{SSPOP}_n$. Firstly, we characterize the structure of the minimal generating sets of $\mathcal{SOP}_n$. As applications, we obtain that the number of distinct minimal generating sets is $(n-1)^nn!$. Secondly, we show that, for $1{\leq}m{\leq}n-1$, the m-potent ranks of the semigroups $\mathcal{SOP}_n$ and $\mathcal{SPOP}_n$ are also n and 2n, respectively. Finally, we find that the 2-potent rank of $\mathcal{SSPOP}_n$ is n + 1.

Utilization of biofilter/photo-catalytic reactor system for the simultaneous treatment of hydrogen sulfide and toluene from waste-air

  • Transient behavior of biofilter/photo-catalytic reactor system was observed to eliminate both hydrogen sulfide and toluene from waste air at its four sampling ports. The biofilter was packed with a equivolume mixture of granular activated carbon(GAC) and compost as packing media on which Thiobacillus sp. IW and Burkholderia cepacia G4 were inoculated and were fixed. The biofilter/photo-catalytic reactor system was run for eight stages of operation under various operating conditions. As a result the removal efficiencies of hydrogen sulfide and toluene began to decrease from 100% after the inlet loads of hydrogen sulfide and toluene surpassed ca. 100 $S-g/m^{3}/h$ and $161g/m^{3}/h$, respectively, and were rapidly decreased to 60% after the inlet loads of hydrogen sulfide and toluene were increased to 200 $S-g/m^{3}/h$ and $644g/m^{3}/h$, respectively.

MicroRNA-100 Resensitizes Resistant Chondrosarcoma Cells to Cisplatin through Direct Targeting of mTOR

  • Chondrosarcomas are malignant cartilage-forming tumors of bone which exhibit resistance to both chemotherapy and radiation treatment. miRNAs have been well demonstrated to regulate gene expression and play essential roles in a variety of biological processes, including proliferation, differentiation, migration, cell cycling and apoptosis. In this study, we obtained evidence that miR-100 acts as a tumor suppressor in human chondrosarcomas. Interestingly, cisplatin resistant chondrosarcoma cells exhibit decreased expression of miR-100 compared with parental cells. In addition, we identified mTOR as a direct target of miR-100. Overexpression of miR-100 complementary pairs to the 3' untranslated region (UTR) of mTOR, resulted in sensitization of cisplatin resistant cells to cisplatin. Moreover, recovery of the mTOR pathway by overexpression of S6K desensitized the chondrosarcoma cells to cisplatin, suggesting the miR-100-mediated sensitization to cisplatin dependent on inhibition of mTOR. In summary, the present studies highlight miR-100 as a tumor suppressor in chondrosarcoma contributing to anti-chemoresistance. Overexpression of miR-100 might be exploited as a therapeutic strategy along with cisplatin-based combined chemotherapy for the treatment of clinical chondrosarcoma patients.
