Regulation of bone formation by high glucose in PDL cells

  • Insulin-dependent or Type 1 diabetes mellitus (IDDM) has been associated with an increased severity of periodontal disease. Since periodontal ligament (PDL) cells play a significant role in maintenance and regeneration of mineralized tissue, the success of procedures, such as guided tissue regeneration, is directly related to the ability of these cells to augment mineralized tissue. In this study, we investigated the time- and dose-dependent effect of high glucose on the proliferation and collagen synthesis of human periodontal ligament (PDL) cells. PDL cells were treated with high glucose (22mM, 33mM, 44mM) for 1 or 2 days. High glucose significantly inhibited proliferation of PDL cells as a time- and dose-dependent manner as evidenced by MTT assay. PDL cells were cultured in high glucose media (22mM, 33mM, 44mM) for 24 h. The ratio of collagen content to total protein was evaluated, and the gene expression of type I collagen was assessed by RT - PCR. The high concentration of glucose inhibited collagen synthesis, a marker of bone formation activity. This study indicated high glucose concentration could alter the metabolism of periodontal ligament cell, leading to alveolar bone destruction.

Appropriate Dimension of Freeway Lane Marking using Computer Based Laboratory Experiment (컴퓨터 기반 실내실험을 통한 고속도로 차선의 적정 규격)

  • PURPOSES : It is well known that experts determined the current standard dimensions of freeway lane markings. However, rigorous engineering rationale could be insufficient regarding whether or not the standard dimensions account for how visible the markings are to the driver. In this study, we seek to optimize the dimensions of freeway lane markings to improve their visibility to drivers. METHODS : The study was conducted as follows. First, alternative lane marking dimensions were selected which could be installed in a test construction site. Second, a video recording was made while driving on the test construction site. Third, subjects were shown the recorded video and then instructed to indicate their preference from among the various lane markings. Lastly, t-tests were applied to assess the statistical significance of differences in the preferences expressed. RESULTS : According to the t-test results, there was no significant difference in the preferences expressed regarding the lane marking widths. However, with regard to the dimensions of freeway lane marking, which represents line marking lengths, gap lengths, and widths of marking, the subjects expressed a preference for specific dimensions such as 6 m:12 m,13 cm, 8 m:12 m,10 cm and 6 m:12 m,10 cm. CONCLUSIONS : In considering the dimensions of freeway lane markings and their relation to visibility by the driver, it was found that dimensions such as 6 m:12 m,13 cm, 8 m:12 m,10 cm and 6 m:12 m,10 cm.

CHEMICAL ABUNDANCE ANALYSIS OF M31 AND M33 BASED ON THE SPECTRUM OF HII REGIONS (HII 영역 분광자료를 통한 M31과 M33의 화학원소 결정)

  • Chemical evolution of galaxies can be understood by studying the spatial distribution of heavy elements. We selected two nearby galaxies, M31 and M33 and investigated spectrum of their HII regions: a) the elec-tron densities have been derived from the [S II] 6717/6731 ratio along with the most recent atomic constants (Hyung & Aller 1996); b) the electron temperatures were determinated from the Pagel's empirical method. Nebula Model (Hyung 1994) has been employed to predict the spectral line intensities which gives the proper chemical abundances. The model would predict the line intensities correctly only when various input parameters such as the effective central star temperatures, gravity log g, model atmosphere as well as the geometry and the nebula physical condition are appropriate. Thus, the determination of chemical abundances of O, S, N of the two nearby galaxies M31 and M33 has been done, which shows a radial dependance of O/H and N/H: decrease with the distance, or increasing electron temperature due to the elemental deficiency. Abundances of M31 appear to be enhanced than those of M33.

Evaluation of Organ and Effective Dose using A PC-Based Monte Carlo Program in AEC Mode and Fix Mode for the whole spine antero-posterior radiography (전 척추 전.후 방향 검사 시 AEC Mode와 Fix Mode에서 PC-Based Monte Carlo Program을 이용한 장기선량 및 유효선량 평가)

  • There are AEC mode and fix mode to exposure when the whole spine antero-posterior radiography is done by using DR equipment. This study compared the utility of fix mode to AEC mode, by evaluating organ dose and effective dose and by examining the quality of radiographic image. GE DEFINIUM 8000 and ART-200X Rando Phantom manufactured by Flukebiometical were used for this study. The Rando phantom was set in front of wall detector of X-rays equipment. AEC mode was set at 80kVp and Fix mode was set at 80kVp, 25mAs, 32mAs, 40mAs, and 50mAs. Whole spine AP image were aquired by combining C, T-L and L-S spine images obtained through 3 exposures. When obtaining C, T-L and L-S spine images, were checked for Air kerma (mGy) value calculated by UNFORS Xi meter attached at the phantom surface of center of radiation field. The effective and organ doses were compared by PCXMC program (PC-Based Monte Carlo Program). The quality of obtained radiographic image was evaluated visually by 3 radiologists using resolution chart. When the effective doses was calculated based on tissue weighting factor of ICRP-103, 1.278mSv was measured by AEC mode, and Fix mode measured 0.405mSv at 25mAs, 0.518mSv at 32mAs, 0.649mSv at 40mAs, and 0.810mSv at 50mAS. In addition, the organ dose measured with esposure at 25mAs by Fix mode was almost equivalent to the organ dose by AEC mode, at the esophagus, thyroid, oral mucosa, salivaly glands located at the cervical spine part, while the organ dose by Fix mode was in general lower than the organ dose by AEC mode at the other organs. When Fix mode at 32mAs, 40mAs, and 50mAs was compared to AEC mode for organ dose in 26 organs, AEC mode had higher measurement in 21 organs but not for than brain, trachea, thyroid, oral mucosa, and salivaly glands which are located at the cervical spine part. The image quality evaluated by resolution test chart was much higher with AEC mode than the quality with Fix mode at all exposure conditions. However, while the image quality of cervical spine exposured at 50mAs by Fix mode was lower than the quality of AEC mode, thoraco-lumbar spine and lumbo-sacral spine were calculated and the quality was similar to AEC mode. Scoliosis occurs mainly at thoraco-lumbar and lumbo-sacral spine, not at cervical spine. Compared to AEC mode, Using the appropriate protocol (80kVp, 50mAs) of fix mode for whole spine AP radiography was thought to be useful because the image quality of the thoraco-lumar and lumbo-sacral spine was similar on AEC mode, Also organ and effective doses can be decreased with Fix mode. Therefore, It is considered that fix mode can be used properly with AEC mode for whole spine AP radiography when considering patient's body posture.

  • It is known that the ranks of the semigroups $\mathcal{SOP}_n$, $\mathcal{SPOP}_n$ and $\mathcal{SSPOP}_n$ (the semigroups of orientation preserving singular self-maps, partial and strictly partial transformations on $X_n={1,2,{\ldots},n}$, respectively) are n, 2n and n + 1, respectively. The idempotent rank, defined as the smallest number of idempotent generating set, of $\mathcal{SOP}_n$ and $\mathcal{SSPOP}_n$ are the same value as the rank, respectively. Idempotent can be seen as a special case (with m = 1) of m-potent. In this paper, we investigate the m-potent ranks, defined as the smallest number of m-potent generating set, of the semigroups $\mathcal{SOP}_n$, $\mathcal{SPOP}_n$ and $\mathcal{SSPOP}_n$. Firstly, we characterize the structure of the minimal generating sets of $\mathcal{SOP}_n$. As applications, we obtain that the number of distinct minimal generating sets is $(n-1)^nn!$. Secondly, we show that, for $1{\leq}m{\leq}n-1$, the m-potent ranks of the semigroups $\mathcal{SOP}_n$ and $\mathcal{SPOP}_n$ are also n and 2n, respectively. Finally, we find that the 2-potent rank of $\mathcal{SSPOP}_n$ is n + 1.

  • The extraction curve of metal ions with Amberite LAl-chloroform has been found to be more steeper than with Amberite LAl-xylene or hexane, and the extraction ratio of Zn (II) in 2M HCl solution is 98%. The extraction ratio of As (III) in 9~11M HCl soln., Sb (III) in 2~4M HCl soln., and Fe (III) in 6~10M HCl soln. are 100%. The separated elements from Bi metal were determined by colorimetry, Zn (III) with dithizone, As (III) with Gutzeit method, Sb (III) with brilliant green and Fe (III) with thiocyanate.

Effect of pH, Electrolytes, and Molecular Weights of Sodium Alginate (Prepared from Sacchrina japonicas) on Gluten Surface Hydrophobicity (글루텐의 표면소수성에 미치는 전해질, pH 및 다시마(Sacchrina japonicas) 알긴산나트륨의 분자량의 영향)

  • Changes in gluten surface hydrophobicity, which play an important role in the functional characteristics of protein, were measured according to various protein concentrations, pH levels, electrolytes concentrations, and alginate molecular weights using 8-anilino-1-naphthalene sulfonic acid (ANS) as a fluorescent probe. Gluten surface hydrophobicity decreased as gluten concentration increased, reaching a maximum pH of 7.0. The effects of alginate molecular weights and alginate concentration on the surface hydrophobicity, emulsifying activity index (EAI), and emulsion stability index (ESI) of gluten-sodium alginate dispersion (GAD) were measured. Gluten surface hydrophobicity rapidly increased the asl NaCl concentration of gluten solution up to 300 mM and showed no significant increase above 300 mM. However, gluten surface hydrophobicity notably decreased until the concentration of CaCl2 and MgCl2 reached 30 mM, indicating no significant variations above 30 mM. GAD surface hydrophobicity increased as the concentration and molecular weight of sodium alginate increased, however, gluten concentration increased as the GAD surface hydrophobicity decreased. The EAI and ESI of GAD increased as both molecular weight and concentration of sodium alginate increased.

Responses of Pseudomonas sp. DJ-12 to Pollutant Stresses of Benzoate and 4-Chlorobenzoate

  • Aromatic hydrocarbons can be utilized as carbon and energy sources by some microorganisms at lower concentrations. However, they can also act as stresses to these organisms at higher concentrations. Pseudomonas sp. DJ-12 is capable of degrading 0.5 mM concentration of benzoate and 4-chlorobenzoate (4CBA). In this study, the exposure of Pseudomonas sp. DJ-12 to the pollutant stresses of benzoate and 4CBA at various concentrations was comparatively studied for its cellular responses, including survival tolerance, degradability of the aromatics, and morphological changes. Pseudomonas sp. DJ-12 utilized 0.5 to 1.0mM benzoate and 4CBA as carbon and energy sources for growth. However, the organism could not degrade the aromatics at concentrations of 3 mM or higher, resulting in reduced cell viability due to the destruction of the cell envelopes. Pseudomonas sp. DJ-12 cells produced stress-shock proteins such as DnaK and GroEL when treated with benzoate and 4CBA at concentrations of 0.5mM, or higher as sublethal dosage; Yet, there were differing responses between the cells treated with either benzoate or 4CBA. 4CBA affected the degradability of the cells more critically than benzoate. The DnaK and GroEL stress-shock proteins were produced either by 1mM benzoate with 5 min treatment or by higher concentrations after 10min. The proteins were also induced by 0.5mM 4CBA, however, it needed at least 20min treatment or longer. These results indicate that the chlorination of benzoate increased the recalcitrance of the pollutant aromatics and changed the conditions to lower concentrations and longer treatment times for the production of stress-shock proteins. of stress-shock proteins produced by the aromatics at sublethal concentrations functioned interactively between the aromatics for survival tolerance to lethal concentrations.

Enhanced macrophage uptake of radiolabeled liposome triggered by ginseng extracts

  • During tumor progression various immunosuppressive cells are recruited to a tumor microenvironment (TME). Tumor-associated macrophages (TAMs) are particularly abundant in TME. Based on their function, macrophages are categorized into two phenotypes: tumoricidal M1 and tumor-supportive M2. Generally, TAMs closely resemble M2-macrophages and lead to tumor growth. However, their phenotype can be changed by immune activator from M2 to M1 and thus promote tumor immunotherapy. Ginseng extracts are well known for its anti-tumor and anti-inflammatory effects from numerous reported studies. However, the mechanism of their effects is still not clear. Recently, some studies suggested that ginseng extracts induced immune activation as well as anti-tumor activities by a repolarization of activated macrophage from M2 phenotype to M1 phenotype. But, further verification about the mechanism as to how ginseng extracts can stimulate the immune response is still needed. In this study, we investigated whether ginseng extracts can alter the phenotype from M2 macrophages to M1 macrophages in mice by using a radiolabeled liposome. And we also evaluated the potential of radiolabeled liposome as a nuclear imaging agent to monitor the transition of phenotype of TAMs. In conclusion, the ginseng extracts seem to change the phenotype of macrophages from M2 to M1 like as lipopolysaccharide (LPS) in mice.

Disease Dispersal Gradients of Rice Blast from a Point Source (점접종원으로부터 벼 도열병 확산의 경사)

  • Rates of lesion development over time and disease gradients over distance for blast disease on the two rice varieties, Brazos and M-20 1 were significantly affected by two different cultural conditions, upland and flooded conditions. Flooding rice field plots lowered the rates of lesion increase and flattened the disease gradients for both varieties. Despite absence of statistically significant differences in the rate of lesion increase between four sampled distances from infection focus, rate of lesion development tended to be slightly greater as distance from the infection focus increases. Rate of lesion increase was greater with more susceptible variety M-201 than with Brazos. Disease gradient was steeper for M-201 than for Brazos. As blast disease progressed, disease gradients became flattened regardless of variety due to the infections originated from secondary foci. Between two empirical disease gradient models examined, Kiyosawa & Shiyomi model was fitted better over Gregory model. Rates of blast isopath movement under upland conditions were calculated as approximately 0.2m/day and 0.4 m/day for Brazos and M-201, respectively. The results in this study suggest that differences in varietal resistance to blast could be detected by measuring disease gradient as efficiently as by measuring infection rate.
