A plasmid vector containing two reporter genes, mer-lux and lac-GFP, was transformed to both Escherichia coli and Pseudomonas putida. Their cellular activities and biofilm characteristics were investigated in flow-cell units by measuring bioluminescent lights and fluorescent levels of GFP. Bioluminescence was effective to monitor temporal cell activities, whereas fluorescent level of GFP was useful to indicate the overall cell activities during biofilm development. The light production rates of E. coli and P. putida cultures were dependent upon concentrations of HgCl2. Mercury molecules entrapped in P. putida biofilms were hardly washed out in comparison with those in E. coli biofilms, indicating that P. putida biofilms may have higher affinity to mercury molecules than E. coli biofilms. It was observed that P. putida expressed GFP cDNA in biofilms but not in liquid cultures. This may indicate that the genetic mechanisms of P. putida were favorably altered in biofilm conditions to make a foreign gene expression possible.
To establish an efficient and reliable microspore culture system for pepper (Capsicum annuum L.), the effect of light intensity used for donor plant's growth, microspore isolation methods, the composition of culture medium, and culture period in light on the production of embryos were investigated. The viability of microspores taken from the plants grown under the light intensity of 10,000 lux was almost same as that from the lower (5,500 lux) light intensity, and the embryo induction and development were a bit higher when donor plants were grown under the lower light intensity. This result implies that lower light intensity does not interfere with the embryo induction and development. However, it was very difficult to prepare microspores for culture since only a small number of flower buds could be harvested from plants grown under the light intensity of 5,500 lux. Microspore isolation methods greatly affected microspores viability; that is, when microspores were isolated by blending rather than maceration, the greater number of viable microspores were easily generated (about 13 times). Among media used for microspores culture in this study, MN medium was most efficient for embryo induction and development. Total number of embryos and the number of cotyledonary embryos were highest when microspores were cultured in dark for 4 weeks, and then in light for one week. These results will be provide valuable information to set up efficient microspore culture system of hot pepper with a high frequency of embryo production, which are applicable to gene transformation and mutagenesis.
Optima for temperature, salinity and light intensity for Nitzschia closterium, Chlorella salina, I내초교냔 galbana and Tetraselmis subcordiformis, which are widely used in bivalve hatcheries of Shandong Province in China, were estimated. The temperature optimum was 23 $^{\circ}C$ for N. closterium and I. galbana and 25 and 27$^{\circ}C$ for C. salina and T. subcordiformis, respectively. That for salinity was 23${\textperthousand}$ for N. closterium and T, subcordiformis, but was 33${\textperthousand}$ for C. salina and I. galbana. In general, all the four microalgae grew faster under 6,000 lux than under 4,000 lux. Growth of N. closterium was faster at $25^{\circ}C$ and dropped abruptly >$25^{\circ}C$, and that of C. salina and T. subcordiformis progressively increased upto $25^{\circ}C$ but dropped beyond 27$^{\circ}C$. T. subcordiformis was the most eurythermal among the 4 species. For mass culture of microalgae in Korea, N. closterium and C. salina are suitable during spring and autumn but C. salina and I. galbana during summer. T. subcordiformis is suitable for culture throughout the year.
Currently, plastic bags are being used for sawdust cultivation of shiitake mushroom. However, due to serious environmental problems caused by the use of plastic bags, we studied the efficacy of bottle cultivation method to replace the sawdust bag method. Small detachable plastic bottles (400 g capacity) filled with Quercus spp. sawdust and wheat bran (4:1 w/w) media were incubated for 80 and 120 days. The weight loss (%) of the media was higher for the NIFoS 2464 strain at an approximate light intensity of 300 Lux than light intensity of 500 Lux; the light intensity was associated with the loss of sawdust medium-weight during the cultivation period. The highest yield was observed when the strain was cultivated for 80 days under dark conditions, 40 days under 500 Lux light, and air circulation fan speed of 30 rpm. When incubated for 120 days, mushroom yield in the bottle media was higher at 40 days of light exposure than 20 days of light exposure. In the bottle media incubated for 80 days under dark conditions, the mushrooms fruited due to repetitive water spraying on the top of the media and light stimulation during the fruiting period. The media could be separated from the bottles because the media shrank after the first harvest. These separated plastic bottles could be re-used for mushroom cultivation, thereby reducing the amount of plastic waste.

This study carried out an experiment to identify subject's work effectiveness and energy saving effect using LED light. Towards this end, this study configured nine various lighting environments in order to control PWM (Pulse Width Modulation) and illuminance (lux), which are the characteristics of LED light. The PWM ratio of LED light was set as R:G:B=1:1:1, R:G:B=4:1:5, and R:G:B=8:7:7, respectively, and illuminance (lux) was set as 400 lx, 700 lx, and 1000 lx, respectively. In addition, the indoor environment was set temperature $20-24^{\circ}C$, humidity 50%-60%, and the amount of clothing 1. This study analyzed work effectiveness and energy consumption in nine lighting environments, each. Error correction was performed for work effectiveness analysis, and cumulative power consumption was measured in each lighting environment for energy consumption analysis. According to experiment results through the lighting environments suggested in this study, accuracy and spent time effectiveness were good in 700lux and higher than 400lux. For spent time, the best effectiveness was revealed in the suggested PWM ratio, R:G:B=8:7:7. The lowest power consumption on each illuminance (lux) was revealed in the order of R:G:B=8:7:7, RGB=1:1:1, and R:G:B=4:1:5. Therefore, pulse-width modulation effect is proposed in this paper was found to affect the efficiency and energy saving.

This study was conducted to investigate visual threshold of nigro (Cichlasoma nigrofasciatum) on white LED light. The visual threshold was obtained by analyzing electrocardiogram (ECG) of the nigro. 5 individuals (body weight: 15.62~45.49 g; TL: 8.9~12.4 cm) were trained for lights by an electric stimulus. And then the heart rate (beats/10s) before and after switching on the light were compared. Light intensity range was from 0.00 to 226.4 lux. Average heart rate was 10.36 beats/10s in the normal condition. When the fish perceived the light, the heart rate was decreased. Visual threshold of the fish was 2.59 lux.
Natural habitats for Calanthe species were generally established in forests of deciduous broad-leaved trees. The habitat areas had lower summer temperatures, more precipitation, and harrow humidity ranging 75~90% throughout the year than adjacent non-habitat areas. The most influential factor on the habitat establishment was light intensity. The light levels in the habitats are cut down by tree canopies, which also reduce temperature before the defoliation in autumn. The most frequent distribution of the habitat was in the areas with light intensity of 400~1,500 lux under tree canopies and of 30,000~50,000 lux in winter after defoliation. Calanthe was characteristically distributed in the broad-leaved forests where high levels of light are reduced in summer, and cold temperatures in winter are insulated by mulched layers of fallen leaves.
Purpose: This study was designed to investigate the present conditions of illumination techniques in dental clinics in order to contribute improvement of operating environment. This study also aimed exploring qualitative and quantitative luminous intensity required for color temperature in dental clinic illumination, which was a critical part of esthetic dentistry. Materials and methods: A total of forty-eight local dental clinics were selected for sampling a) luminous intensity, and b) color temperature. The author measured the luminous intensity and the color temperature with lux meter and color meter respectively between 12pm and 2pm. The dental unit chair placed in the general operation positions were kept the distance 60 cm then all dental units were measured three times and averaged. The author measured the luminous intensity and color temperature with both common dental operating light and then without operating light in different office environments. The study was conducted under three conditions: 1 artificial illumination in clear day light 2. artificial illumination in cloudy day light, and 3. artificial illumination alone. Results : The results obtained were as follows. 1. The average luminous intensity in dental clinic lighting was 425 lux which was not sufficient to produce the optimal shade of the patient's teeth. Furthermore, the average luminous intensity even in full operating lighting was 9532 lux which fell short of the required level of 10,000 lux. 2. The average color temperature of all dental clinics surveyed was 5169 K which met the optimal range. However, only 33.3% fell in the correct region between 5,000-5,500 K as 25% were over 5,500 K and 41.6% were below 5,000 K. As a result, 66.7% were under insufficient color temperature conditions. 3. The dental unit chair placed next to a window, hence exposure to natural lighting, had significantly higher luminous intensity and color temperature compared to the dental unit chair which didn't have a window or natural lighting. 4. The data analysis revealed that only 6.3% of the dental clinic were met the standard of the average luminous intensity and color temperature.
This research was done to identify the mass propagation method in winter daphne (Daphne odora Thunb) using its softwood cuttings and to investigate its plant characters established at different light and soil conditions as an indoor plant. Cuttings from winter daphne were taken and grown in different treatment consisted of rooting media (perlite, vermiculite, perlite + vermiculite (1 : 1) and commercial horticulture media soil), indole butyric acid (IBA) hormone concentrations (0, 100, 500, 1000 ppm and Rootone) and date of cutting. Transplants were grown at different light intensities (100, 1000, 2500 lux and control) and growing media. Results showed that cuttings grown in perlite + vermiculite (1 : 1) gave higher percentage (100%) rooting. Cuttings treated with Rootone and IBA 100 ppm showed good rooting growth and cutting taken in June, 25 gave the highest rooting (96.7%). The best plant growth obtained at 1000 lux (56~60 ${\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$) followed by 2500 lux (125~130 ${\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$) and scoria mixed with commercial horticulture media soil showed better growth of transplants.
The effects of light and storage conditions on the germination of Oenothera lamarckiana were investigated. Germination was induced by red light, but inhibited by green, blue or infrared. No germination occurred in the dark. Radicle was more photosensitive than other parts of the seed. The rate of germination was proportional to increase in Light intensity ranging from 10 to 3000 Lux. Photosensitivity occurred 6 hrs after water imbibition of air-dried seeds. Maximum photosensitivity was detected 1-2 days after seeding under the light condition at 300 Lux. Germination was almost completed 3 days after seeding. When seeds stored in the chilling and wetting condition were kept at constant temperature in the dark, they germinated well. Air-dried seeds, however, didn't germinate. Whereas, the seeds which kept at alternating temperature had the tendency that seeds stored at room temperature (25$^{\circ}C$) germinated better than those stored at low temperature (4$^{\circ}C$).
