$N_2\;Fixation\;and\;H_2$ Production by a Phototrophic Bacterium, Chromatium sp. (광합성세균 Chromatium sp.의 질소고정과 수소생산)

  • Chromatium sp., densely populated on the bacterial plate of Lake Kaiike throughout the seasons, possibly fix N2 and concurrently produce H2 N2 Fixation and H2 production by Chromatium sp. were performed under photoautotrophic growth condition, and of which rates were much higher and showed expontial growth phase. Bacterial plater samples from Lake Kaiike collected on July 27, 1994 were used to know the effect of the light or H2S on N2 fixation and H2 productin by the bacteria. At low light intensity (250 lux), low rates of N2 fixation and H2 production were detected after 18 hours. However, high rates of the production were observed under the condition of high light intensity (1000 lux). On the other hand, a very low rate of N2 fixation was observed without an addition of H2S, while the bacterial rapilly increased N2 fixation and H2 production after adding H2S and the highest rate was observed in case of adding 20mg H2S-S/$m\ell$ to the bacterial plates.

Effect of Light Intensity on Growth and Flowering of Several Korean Native Wild Flower Plants (자생화훼식물의 광도차에 따른 생육반응)

  • Experiment was performed to investigate the effect of light intensity on growth and flowering status of Korean native Dianthus superbus var. longicalycinus, Sedum middendorifianum, Hemerocallis aurantiaca, Iris ensata var. spontanea and Aquilegia buergeriana var. oxysepola for effective landscaping. Number of floret and flower stalk per a plant were markedly increased under full sun(about 100,000lux), and decreased with decreasing light intensity in all tested plants. This result indicate that it will be cultivated in the open for promote landscaping values. However, leaf area and plant height were more increased under half sun(22,000lux) than open(100,000lux) or shade(7,700lux) in all plants except Sedum middendorifianum. Especially, Hemerocallis aurantiaca had same number of shoot and leaf size, even if planted under shade condition to show a wide adaptation to various light intensities.

Response od Striped Puffer to the White Light (백색광에 대한 까치복의 반응)

  • The purpose of this study is to find the light intensity which induced maximum gathering rate and to observe the variation of the gathering rate both in daytime and tat night by using Striped puffer, Fugu xanthopterus (Temminck et Schlegel). An experimental tank(360L$\times$50W$\times$55H cm) was set up in a dark room. An illumination system was attached to the end of one side of the tank to control horizontal light intensity. Eight artificial light sources were prepared by combination of three light bulbs (10W, 60W, 100W) and eight filters. During the experiment water depth was maintained 50 cm level in the tank. The tank was marked into six longitudinal sections each being 60cm long to observe the distribution of fish. The fish were acclimatized in dark condition for 50 minutes prior to the main experiment. Upon turning on the light, the number of fish in each section was counted 60 times every 30 seconds, and the gathering rate was obtained from the average number of fish in each section. The light intensity inducing maximum gathering rate was 298.56 lux (188.44-444.96 lux) at daytime and 298.56 lux (188.44-444.96 lux) at night. The variation of the gathering rate of fish in illumination time was increasing trend fluctuately and did not show any distinctive difference between day and night.

Combination of berberine and ciprofloxacin reduces multi-resistant Salmonella strain biofilm formation by depressing mRNA expressions of luxS, rpoE, and ompR

  • Bacterial biofilms have been demonstrated to be closely related to clinical infections and contribute to drug resistance. Berberine, which is the main component of Coptis chinensis, has been reported to have efficient antibacterial activity. This study aimed to investigate the potential effect of a combination of berberine with ciprofloxacin (CIP) to inhibit Salmonella biofilm formation and its effect on expressions of related genes (rpoE, luxS, and ompR). The fractional inhibitory concentration (FIC) index of the combination of berberine with CIP is 0.75 showing a synergistic antibacterial effect. The biofilm's adhesion rate and growth curve showed that the multi-resistant Salmonella strain had the potential to form a biofilm relative to that of strain CVCC528, and the antibiofilm effects were in a dose-dependent manner. Biofilm microstructures were rarely observed at $1/2{\times}MIC/FIC$ concentrations (MIC, minimal inhibition concentration), and the combination had a stronger antibiofilm effect than each of the antimicrobial agents used alone at $1/4{\times}FIC$ concentration. LuxS, rpoE, and ompR mRNA expressions were significantly repressed (p< 0.01) at $1/2{\times}MIC/FIC$ concentrations, and the berberine and CIP combination repressed mRNA expressions more strongly at the $1/4{\times}FIC$ concentration. The results indicate that the combination of berberine and CIP has a synergistic effect and is effective in inhibiting Salmonella biofilm formation via repression of luxS, rpoE, and ompR mRNA expressions.

A Study on White LED Lighting of Interior Space for the Readability by Age (연령대별 가독(加讀)기능을 위한 실내공간의 백색 LED조명환경 연구)

  • The experimental study of the white LED lighting focused on the readability in interior space according to age group is conducted realizing importance of the lighting environment and necessity of lighting study depending on the user situation. The results of study represent that a teenager does not show sensitive reaction to change of the color temperature and illuminance. The people in 20s and 30s prefer neutral white LED lighting and are very sensitive to change of the light environment. The people in 50s and 60s prefer high illumination and cool white LED lighting series. People of all ages highly appraise 6000K of 700 lux and 4000K in the illuminance range of 700 lux to 1000 lux for readability. The white LED lighting of color temperature 4000K is estimated as a suitable environment for readability in a wide range of illumination.

Effect of the light qualities on the growth characteristics and yield in the cultivation of Lyophyllum ulmarium (만가닥버섯 재배시 광 종류에 따른 생육 특성 및 수량)

  • This experiment was carried out to clarify the effect of light qualities on the growth characteristics and yield of fruiting body in the cultivation of Lyophyllum ulmarium. The intensity of illumination by light qualities was in the order of white light(2,270Lux), yellow light(1,750Lux), blue light(460Lux) and red light(400Lux). An investigation of fruiting body showed these results that the pileus size and stipe diameter of fruiting body on CBM(Chungbuk mushroom)-1757 were much larger than Hypsizigus marmoreus, and an effect of yellow light seemed to be better than those of another light. In comparison with Hypsizigus marmoreus, the growth duration of CBM-1757 was shortened by 8 days which included 2 days for mycelial culture, 1 day for first pinning requirement and 1 day for growth. The growth duration in yellow light illumination was about 70 days showing the tendency of 2~4 days reduction. There were no differences in results such as number of effective stem and fresh weight. The yield of fruiting body per bottle in CBM-1757(95.6g) was little higher than Hypsizigus marmoreus(94.8g). By a white light's standard, the yields of blue and red light illumination were decreased by 2~9%, but that of yellow light illumination was increased by 8%. The chromaticity results showed that brightness, red and yellow coloration of CBM-1757 were higher than those of Hypsizigus marmoreus, and yellow light treatment was more effective than another light.

Varietal Difference in Heading Date of Rice by Night Illumination (야간 조도에 따른 벼 품종별 출수 반응)

  • This experiment was conducted to study the varietal and ecological difference of rice in heading date by night illumination. The degree of delay in heading date and days from heading initiation to full heading (DHIF) of rice were in the order of mid-late maturing cultivars (MLC)> medium maturing cultivars (MEC) >early maturing cultivars (EAC) >very early maturing cultivars (VEC). However, the MEC showed significant variation in days to heading; Kwanganbyeo delayed significantly, but Ansanbyeo did not. The comparative sensitivity of different rice ecotypes per lux in delayed heading was 0.09-0.10, 0.14-0.26, 0.16-0.58, and 0.35-0.54 day in VEC, EAC, MEC, and MLC, respectively; the shortest ion Odaebyeo (0.09 day), moderate in Daeanbyeo (0.35 day), and the longest in Kwanganbyeo (0.58 day). Difference of DHFI was 0.-1 day in VEC, -l-2 days in EAC, 0-l6 days in MEC and 6-18 days in MLC under 50-70 lux compared to 2 lux; Daejinbyeo (-1 day), Hwaseongbyeo (6 days), Chucheongbyeo (9 days), Kwanganbyeo (16 days), and Ilpumbyeo (18 days).

Interaction Between the Quorum Sensing and Stringent Response Regulation Systems in the Enterohemorrhagic Escherichia coli O157:H7 EDL933 Strain

  • Quorum sensing and the stringent response are well-known regulation systems for the expression of virulence genes in enterohemorrhagic Escherichia coli (EHEC). However, how these two systems interact is not well known. E. coli strains with mutations in two regulation systems, ${\Delta}luxS$ (ECM101) and ${\Delta}luxS{\Delta}relA{\Delta}spoT$ (ECM201), and the ${\Delta}luxS$ complement strain to ECM201 (ECM202) were created from EHEC O157:H7 EDL933 to investigate how the regulatory systems interact. The phenotypic changes of the mutant strains were characterized and compared with the wild type. The mutant strains exhibited no obvious growth defects, although acid resistance and cellular cytotoxicity were decreased significantly in all the mutant strains. Phenotypic characterization revealed that mutations in the stringent response system (ECM201 and ECM202) influenced the metabolic (defective utilization of arabinose and L-sorbose) and enzymatic activities (decreased trypsin activity, and increased ${\alpha}$-glucosidase activity). In contrast, the quorum sensing system mutant (ECM101) did not display these phenotypes. The motility of the quorum sensing system mutant (ECM101) was unchanged, but mutation in the stringent response system influenced the motility. Our results suggest that quorum sensing interacts with the stringent response regulation system.
