Endothelial Cells Isolated from the Bovine Corpus Luteum Synthesize Prostaglandin $F_{2{\alpha}}$ Receptor

  • The corpus luteum is a transient endocrine gland essential for regulation of the ovarian cycle as well as for establishing and maintaining pregnancy. Prostaglandin $F_{2{\alpha}}$ (PGF) initiates functional and structural regression of the corpus luteum and therefore is an important regulator of the estrous cycle. It is a matter of debate whether the endothelial cells of the bovine corpus luteum express PGFR, the cognate receptor for PGF. Therefore, the aim of this study was to assess the expression of PGFR in bovine endothelial cells. Endothelial cells were isolated from the bovine corpus luteum of the mid-luteal stage using magnetic beads and cultured in vitro. We demonstrate that this isolation procedure generates a pure culture of endothelial cells as confirmed by synthesis of Factor VIII and lack of expression of $3{\beta}$-hydroxysteroid dehydrogenase. By RT-PCR, Western blot and immunofluorescence analyses, we further show that the cultured endothelial cells produced PGFR. This model system can be utilized to provide an experimental system to investigate the role of PGF on endothelial cells during the reproductive cycle.

  • 황체내 TNF$\alpha$는 큰황체세포를 포함한 황체내 세포들의 사망을 일으키고, 황체내 혈관의 퇴화와 프로게스테론 농도의 감소를 초래하는 등 황체의 구조적 및 기능적 퇴화를 유발하는 것으로 밝혀졌다. 반면 이와는 대조적으로 TNF$\alpha$는 황체발달의 초기에는 황체자극효과를 가지고 있다는 보고도 있다. 황체내에서 TNF$\alpha$의 중요성에도 불구하고, 아직 이 물질의 분비원이 대식세포라는 보고들과 대식세포는 주된 분비원이고 내피세포에서도 소량 분비된다는 보고 등 이견이 있다. 이를 알아보기 위해 저자들은 돼지에서 비임신기와 임신기의 성숙황체와 용해황체를 이용하여 대식세포와 TNF$\alpha$ 면역조직화학을 실시한 결과, 성숙황체내에서 TNF$\alpha$의 주된 분비원은 대식세포이고 내피세포도 일부 분비하는 것을 확인하였으며, 황체용해가 진행되면서 내피세포가 TNF$\alpha$의 분비와 반응에 다양성과 역동성을 보이고 있음을 추정할 수 있었다.

  • This paper was carried out to observe prostaglandin $F_{2{\alpha}}$-induced morphological changes in the placenta. White mice received intramuscular injections of $PGF_{2{\alpha}}$(containing dinoprost tromethamine 0.5mg/ml, Upjohn Co.) in once or twice with doses of 0.1ml on 10th to 18th day of their respective pregnancies, the histological changes of the placentae and ovaries were observed with light microscope. Abortion within 21 to 51 hours following $PGF_{2{\alpha}}$ administration occurred in the pregnant mice. Vacuolization of trophospongial cells, giant cells and myometrium under decidua basalis, spherical acidophilic inclusion bodies of various sixtes in trophospongial cells and giant cells. hydropic degeneration and necrosis of labyrinthine trophoblasts and yolk cells, and infltration of neutrophils in the placenta and the uterus were observed. In addition, there were decrease in glycogen of the placental labyrinth and the visceral yolk sac, but increase in glycogen deposit of mesometrial myometrium. Atrophy and increase in number of large lipid droplets of the luteal cells and proliferation of fibroblasts were also recognized in the corpora lutea.

생쥐 난소에서 Glucosamine-6-Phosphate Deaminase (GNPDA)의 발현 (Expression of Glucosamine-6-Phosphate Deaminase (GNPDA) in Mouse Ovary)

  • 생쥐 난소에서 glucosamine-6-phosphate deaminase (GNPDA)의 발현을 조사하였다. Western blot상에서 Mr. 31 kDa의 항원을 검출하였으며 출생 후2주에 급격한 발현의 증가가 확인되었다. 난소절편의 면역염색 결과 협막세포와 간충조직에서는 균질한 발현을 보인 반면 난포내 발현양상은 난포의 발달에 따라 다르게 나타났다. 일부 1차 난포의 난자에서 GNPDA의 발현이 관찰되었으나 강소형성 난포의 난자에서는 관찰되지 않았다. 황체화 과립세포에서의 발현은 황체발달에 따라 증가하였으며 황체퇴화 부위에서 강한 신호가 검출되었다. 난포발달에 따른 GNPDA발현의 차이는 GNPDA가 생쥐 난소 조직 재구성에 관여하고 있음을 암시한다.

  • In summary, progesterone is probably the only naturallly occurring progestational agent of any significance. Small amounts may be synthesized by the cells of the follicle in the provulatory swelling phase; however, the major production is by the corpus luteum cells of the ovary during the luteal phase of the cycle. It is constantly produced in small amounts by the adrenal gland and by the testis in the male. In the adrenal and the testis, it probably serves as the precursor for corticoids and androgens. It is transported in the blood by a specific binding protein and metabolized and conjugated in the liver into sodium pregnanediol glucuronide which also circulates in the blood. Approximately 20% is excreted in the urine as sodium pregnanediol glucronide; pregnanolone represents a minor metabolic product. The pregnanediol which is excreted in the bile is enzymically hydrolyzed by the gut so that the pregnanediol recovered in the feces is in the free form.

Involvement of Ski Protein Expression in Luteinization in Rat Granulosa Cells

  • Ski protein is implicated in proliferation/differentiation in a variety of cells. We had previously reported that Ski protein is present in granulosa cells of atretic follicles, but not in preovulatory follicles, suggesting that Ski has a role in apoptosis of granulosa cells. The alternative fate of granulosa cells other than apoptosis is to differentiate to luteal cells, however, it is unknown whether Ski is expressed and has a role in granulosa cells undergoing luteinization. Thus, the aim of the present study was to locate Ski protein in the rat ovary during luteinization to predict the possible role of Ski. In order to examine the expression pattern of Ski protein along with the progress of luteinization, follicular growth was induced by administration of equine chorionic gonadotropin to immature female rat, and luteinization was induced by human chorionic gonadotropin treatment to mimic luteinizing hormone (LH) surge. While no Ski-positive granulosa cells were present in preovulatory follicle, Ski protein expression was induced in response to LH surge, and was maintained after the formation of corpus luteum (CL). Though Ski protein is absent in granulosa cells of preovulatory follicle, its mRNA (c-ski) was expressed and the level was unchanged even after LH surge. Taken together, these results demonstrated that Ski protein expression is induced in granulosa cells upon luteinization, and suggested that its expression is regulated post-transcriptionally.

Downregulation of the Expression of Steroidogenic Acute Regulatory Protein and Aromatase in Steroidogenic KGN Human Granulosa Cells after Exposure to Bisphenol A

  • Although increasing evidence of cause-and-effect relationship between BPA exposure and female reproductive disorders have been suggested through many studies, the precise biochemical and molecular mechanism(s) by which BPA interferes with steroidogenesis in the ovarian cells still remain unclear. Therefore, the purpose of this study was to discover the steroidogenic biomarker(s) associated with BPA treatment in human granulosa cell line, KGN. In this study, our results obtained via the analysis of steroidogenesis-related protein expression in KGN cells using quantitative polymerase chain reaction (qPCR) and western blot analyses revealed that the expression levels of steroidogenic acute regulatory (StAR) and aromatase decreased considerably and gradually after BPA treatment in a dose-dependent manner under BPA treatment. Further, remarkable decreases in their expression levels at the cellular levels were also confirmed via immunocytochemistry, and subsequent StAR and aromatase mRNA expression levels showed profiles similar to those observed for their proteins, i.e., both StAR and aromatase mRNA expression levels were significantly decreased under BPA treatment at concentrations ≥0.1 μM. We observed that follicle stimulating hormone upregulated StAR and aromatase protein expression levels; however, this effect was suppressed in the presence of BPA. Regarding the steroidogenic effects of BPA on KGN cells, controversies remain regarding the ultimate outcomes. Nevertheless, we believe that the results here presented imply that KGN cells have a good cellular and steroidogenic machinery for evaluating endocrine disruption. Therefore, StAR and aromatase could be stable and sensitive biomarkers in KGN cells for the cellular screening of the potential risk posed by exogenous and environmental chemicals to female reproductive (endocrine) function.

소 미성숙난자의 체외성숙과 배발생에 황체의 형태가 미치는 영향 (Effect of Luteal Morphology of Donors on the Maturation and Subsequent Development in Vitro of Bovine Immature Oocytes)

  • 본 연구는 공여난소의 황체 형태가 소 미성숙난자의 체외 성숙과 발생에 미치는 영향을 감수분열과 수정란 생산을 비교하여 조사하였다. 공여난소를 황체의 형태에 따라 다음과 같이 4 group 으로 분류하였다. Group 1 ; 최근에 배란한 흔적으로 보이는 혈포가 있는 난소. Group 2 ; 황체는 적색 또는 갈색이며, 혈관이 황체의 가장자리에 국한되어 분포하고 있는 난소. Group 3 ; 황체는 오렌지 또는 황갈색이며, 혈관이 황체의 정상부 까지 분포한 난소. Group 4 ; 황체는 잃은 황색 또는 백색이며, 황체 정상부의 혈관분포가 사라진 난소. 미성숙난자의 체외성숙과 배발생을 위하여 각각 TCM 199과 TLP-PVA을 기본배지로 사용하였다. 각 group에서 채취한 미성숙 난자를 4, 14, 24시간 체외성숙시켰을 때 metaphase I과 metaphase II에 도달한 난자의 비율은 각 group 사이에 차이가 없었으며, 각 group 에서 metaphaseII난자는 체외성숙 후 14시간에 나타나기 시작하여 24시간에 성숙이 종료하였다. 체외 배발생 후 2세포기와 8세포기까지의 발생에는 공여난소 황체의 형태가 영향을 미치지 않았으나, 상실배와 배반포로 발생한 수정란의 비율은 group 1과 3에서 채취한 난자에서 group 2와 4에서 채란한 난자에 비하여 유의적으로 증가하였다 (p<0.05). 이러한 결과는 공여난소의 번식상태가 체외 수정란생산에 유의적인 영향을 미치는 것으로 암시하고 있다.

  • In order to study the follicular atretic mechanism in mammalian ovary, the relationship between pyknotic index (PI) of granulosa cells and the steroid concentrations in the follicular fluid of atretic follicle was investigated. Follicles were isolated from porcine ovary according to their sizes and the reproductive phases. Steroid concentrations were quantified by high-performance liquid chromatography. As PI increased, the concentration of progesterone was significantly increased(p<0.05), whereas testosterone and estradiol showed no significant changes in their concentration. As follicular size was increased, PI of follicular GC in the luteal phase was increased significantly(p<0.05) and the molar ratio of progesterone to testosterone was increased in the follicles of follicular phase. It can be concluded that progesterone accumulated in the follicular fluid as atresia of the follicles was progressed, and that PI of granulosa cells could be used as one of convenient and pratical criteria for the identification of follicular atresia.

Relationship between Plasminogen Activity and Plasminogen Inhibitor during the Culture of Porcine Oviduct Epithelial Cells

  • The present study was performed to identify changes of plasminogen activator (PA) and plasminogen activator inhibitor (PAI) in porcine oviduct epithelial cells (POECs) during the estrous cycle. POECs obtained from ovary in pre-ovulatory (Pre-Ov), early to mid-luteal stage (Early-mid L) and post-ovulatory stage (Post-Ov). For the examine of PA activity, $1{\times}10^5$ fresh cells of POECs were cultured in DMEM/Ham F-12 containing 10% FBS and 0.2% amphotericin under humidified atmosphere of 5% $CO_2$ in air and $38^{\circ}C$. The urokinase-type PA (uPA) was observed at 7 days of POECs culture. PA activity was measured with culture prolonged of 0, 3, 6, 12 and 24 h after culture of 7 days. The PA activity were high significantly (p<0.05) at 12 h of culture, but PA activity were decreased with culture periods increased. The PA activity in POECs of Post-Ov stage were higher significantly (p<0.05) than that of Early-mid L and Pre-Ov stage. When PAI-1 and PAI-2 were added during the POECs culture, the PA were observed significant low activity (p<0.05). The PA activity and protein expression were decreased by PA inhibitor. This results suggest that PAI-1 and PAI-2 have a suppressive action on change of PA activity during the estrous cycle of pigs. Specifically, this study using PA inhibitor was effect the PA activity and PAI expression in oviduct epithelial cells in pigs.
