In Vitro and In Vivo Bone-Forming Effect of a Low-Molecular-Weight Collagen Peptide

  • This study reveals that low-molecular-weight collagen peptide (LMWCP) can stimulate the differentiation and the mineralization of MC3T3-E1 cells in vitro and attenuate the bone remodeling process in ovariectomized (OVX) Sprague-Dawley rats in vivo. Moreover, the assessed LMWCP increased the activity of alkaline phosphatase (ALP), synthesis of collagen, and mineralization in MC3T3-E1 cells. Additionally, mRNA levels of bone metabolism-related factors such as the collagen type I alpha 1 chain, osteocalcin (OCN), osterix, bone sialoprotein, and the Runt family-associated transcription factor 2 were increased in cells treated with 1,000 ㎍/ml of LMWCP. Furthermore, we demonstrated that critical bone morphometric parameters exhibited significant differences between the LMWCP (400 mg/kg)-receiving and vehicle-treated rat groups. Moreover, the expression of type I collagen and the activity of ALP were found to be higher in both the femur and lumbar vertebrae of OVX rats treated with LMWCP. Finally, the administration of LMWCP managed to alleviate osteogenic parameters such as the ALP activity and the levels of the bone alkaline phosphatase, the OCN, and the procollagen type 1 N-terminal propeptide in OVX rats. Thus, our findings suggest that LMWCP is a promising candidate for the development of food-based prevention strategies against osteoporosis.

Mesogen을 포함하지 않은 액정 polyurethane의 합성과 열적성질에 관한 연구 (Preparation and Properties of Liquid Crystalline Polyurethanes Containing No Mesogenic Unit)

  • 액정 polyurethane은 DMF 용매중 1,4-bis(6-hydroxyhexoxy)benzene (BHB6)과 2,5-tolylene diisocyanate (2,5-TDI)를 중부가 반응에 의해서 합성하였다. 고유점도는 0.26, $0.42d{\ell}/g$였다. 생성된 polyurethane의 열적성질 및 구조확인은 DSC, 편광현미경, X-ray회절 그리고 $^1H$-NMR 및 FT-IR에 의해 확인하였다 특히 분자량이 서로 다른 polyurethane 2,5-TDI/BHB6의 열적성질 및 물성에 대해서 비교 검토하였다. 고유점도가 [${\eta}$]=0.26인 polyurethane 2,5-TDI/BHB6는 monotropic 액정성을 나타내었다. 이러한 것은 구조적으로는 mesogen을 포함하고 있지 않다. 예를 들면 LCPU-L(저분자량)은 $T_{I-LC}$(등방성액체상전이점-액정) 및 $T_{LC-K}$(액정-결정)은 $122^{\circ}C$부터 $89^{\circ}C$까지 관찰되었다.

Glycidol을 개시제로 이용한 L-lactide 개환중합 (Ring-Opening Polymerization of L-lactide with Glycidol as Initiator)

  • $Al(O-i-Pr)_3$을 촉매로 이용하고 중합 개시제로 glycidol을 사용하여 L-lactide를 개환중합하여 glycidol-poly(lactide)(Gly-PLA)를 얻었다. $^1H$ NMR 분석 결과 glycidol의 말단에 존재하는 수산기가 Gly-PLA에는 존재하지 않았고 이를 통해 glycidol의 말단에 존재하는 OH기가 개시제의 역할을 하여 중합이 진행되었음을 확인하였다. Llactide용액중합과 벌크중합을 진행하였으며, L-lactide/glycidol 몰비, 중합 온도와 시간에 따라 생성된 Gly-PLA의 분자특성을 관찰하였다. L-lactide/glycidol 몰비가 증가할수록 수율과 분자량은 증가하였다. 또한 L-lactide/glycidol 몰비가 증가할수록 저분자량에 해당하는 낮은 녹는점 피크는 줄어들고, 고분자량에 해당하는 높은 녹는점 피크가 증가하였다.

  • Low molecular weight carbohydrates, phycobilin pigments and cell structure using light and transmission electron microscopy were used to describe a new genus of unicellular red algae, Erythrolobus coxiae (Porphyridiales, Porphyrideophyceae, Rhodophyta). The nucleus of Erythrolobus is located at the cell periphery and the pyrenoid, enclosed by a cytoplasmic starch sheath, is in the cell center. The pyrenoid matrix contains branched tubular thylakoids and four or more chloroplast lobes extend from the pyrenoid along the cell periphery. A peripheral encircling thylakoid is absent. The Golgi apparatus faces outward at the cell periphery and is always associated with a mitochondrion. Porphyridium and Flintiella, the other members of the Porphyrideophyceae, also lack a peripheral encircling thylakoid and have an ER-mitochondria-Golgi association. The low molecular weight carbohydrates digeneaside and floridoside are present, unlike both Porphyridium and Flintiella, which have only floridoside. The phycobilin pigments B-phycoerythrin, R-phycocyanin and allophycocyanin are present, similar to Porphyridium purpureum. The cells have a slow gliding motility without changing shape and do not require substrate contact. The ultrastructural features are unique to members of the Porphyrideophyceae and recent molecular analyses clearly establish the validity of this new red algal class and the genus Erythrolobus.

우레탄 필름에 인쇄된 신축 가능한 전극 패턴의 특성 (Properties of Stretchable Electrode Pattern Printed on Urethane Film)

  • Currently, functional patterns are formed by screen printing on stretchable films, and they are applied to wearable and stretchable devices. In this study, three types of silver paste were prepared using three polyester binders with different Tg and molecular weights in order to impart elasticity to the conductive pattern itself. Rheological properties and DSC measurements were performed for each silver paste. Then, each silver paste was screen printing and cured by an IR dryer to evaluate adhesive strength, pencil hardness, resistance and surface shape change according to strain. As a result, it was found that the silver paste using a binder with a low Tg and a high molecular weight has the smallest resistance change depending on the strain. Namely, it was found that it is most preferable to use a binder with a low Tg and a high molecular weight as the stretchable electrode.

Chitosan을 이용한 감식초의 청징화 (Clarification of Persimmon Vinegar Using Chitosan)

  • The effects of chitosans with two different molecular weights (150 and 37 kDa) on the clarification of persimmon vinegar were investigated. The coagulated solids increased with increasing chitosan concentrations. Chitosan treatment had no effect on pH and acidity. Turbidity, browning, contents of tannin and soluble solids decreased with increasing chitosan concentrations. An increase in chitosan concentration resulted in an increase in color L* value and a decrease in a* and b* values. Reduction in turbidity and tannin contents somewhat differed depending on the molecular weights of chitosan. In sensory evaluation, chitosan treatment did not affect color and flavor over all concentrations tested. However, astringent taste was noticeably strong by treatment with 500mg/L chitosan, resulting in low overall acceptability. Therefore, the most effective clarification of persimmon vinegar was achieved by treatment with 400mg/L chitosan, irrespective of molecular weight.

Polymer Bland에서의 상평형에 관한 연구 (Study on the Phase Diagram for the Polymer Bland)

  • We study on the phase diagram of the polymer blend. For this purpose, one PS (polystyrene) and two PI(polyisoprene) were employed whose molecular weights were low enough to make the experimental determinations possible. The weight-average molecular weight(Mw) of PS was 2514, and Mws of two PIs were 2700. Interaction energy density (IED) of the Flory-Huggins lattice theory was defined as a function of temperature and composition, and the consequent equations for the binodal, and critical points were derived. By fitting the experimental binodal points to the derived binodal curve with a nonlinear regression method, the expression for the IED was determined. And the expression for the IED obtained from this study was compared with those reported in the literatures. Also were discussed the importance of accuracy in the expression the IED, and the IED's dependency on the temperature, composition and molecular weights.

  • Streptomyces tendae JC412 secreted two forms of protease(ST -1 and ST -2) when grown in OSY medium (oatmeal 1.5%. soybean meal 2%. dried yeast 1 %) supplemented with glucose(0.5%) and KH2PO4(0.05%). Initial pH of the culture medium was adjusted to 10.0 with NaOH and incubated at $27^{\circ}C$ on a rotary shaking incubator (180rpm). High- molecular-weight protease ST-1 was heat labile. whereas low molecular protease ST-2(22.000 Da) was reported to be heat stable. (omitted)

  • 이산화탄소/프로필렌 옥사이드 공중합을 통하여 고분자 사슬 내에 친수성기와 소수성기가 공존하는 저분자량의 블록 공중 합체를 합성하였다. 고활성의 촉매를 사용한 이산화탄소/프로필렌 옥사이드 공중합 반응에 단말기로 -OH기를 갖는 폴리(에틸렌 글리콜)(PEG)을 분자량 조절제로 투입하여 블록 공중합체를 합성하였다. 단말기 한쪽 끝에만 -OH기를 갖는 폴리(에틸렌 글리콜)을 투입하였을 때는 PEG-block-PPC (폴리(프로필렌 카보네이트)) 다이블록 공중합체가 얻어지고, 단말기 양쪽 끝 모두 -OH기를 갖는 폴리(에틸렌 글리콜)을 투입하였을 때는 PPC-block-PEG-block-PPC 트리블록 공중합체가 얻어진다. 제조된 블록 공중합체는 $^1H$-NMR 스펙트럼을 통하여 구조 분석을 하였고 GPC를 통하여 분자량을 측정하였다.

난소화성 획분이 높은 난소화성 덱스트린의 제조 방법 (Methods for Preparing Indigestible Dextrin with High Indigestible Fraction)

  • 높은 난소화성 획분을 갖는 난소화성 덱스트린은 열처리 덱스트린의 효소 가수분해액을 에탄올 또는 강산성 양이온 교환수지(UBK 530)로 처리함으로써 제조하였다. 난소화성 획분, 식이섬유 및 수출을 고려하여 ${\alpha}-amylase$와 amyloglucosidase를 반응시킨 가수분해액으로부터 난소화성 덱스트린을 제조하기 위한 에탄올 처리의 최적 조건은 에탄올을 30%(w/w) 효소 반응액에 고형분 대비 5배로 첨가하여 상온에서 3시간 동안 방치시키는 공정이었다. 포도당을 포함하는 저분자 당류와 고분자 당류를 강산성 양이온 교환수지에 의해 분리한 결과 내열성 ${\alpha}-amylase$와 amyloglucosidase에 의한 초기 효소 반응액은 43.6%의 DPI(glucose)과 51.5%의 DP4+(maltotetaos이상)를 나타낸 반면, 양이온 교환수지에 의해 초기 효소 반응액의 50%까지 분취한 난소화성 덱스트린은 7.1%의 DPI(glucose)과 91.2%의 DP4+(maltoteoaose이상)를 나타내었다. 따라서, 초기 효소 반응액의 난소화성 획분 44.5%는 저분자 당류의 분리 후 78.9%까지 증가하였다.
