Effects of Mastitis on Buffalo Milk Quality

  • The objectives of this study were to compare the effectiveness of different indicators of mammary inflammation in buffalo and to evaluate the association of the indicators with buffalo milk yield, composition, and rennet coagulation properties. This study was carried out at four buffalo farms in central Italy using a total of 50 lactating buffalo. Milk from each buffalo was tested at the beginning, middle, and end of lactation. To evaluate the relationship between mastitis markers and milk components, three classes were defined for each of the following markers: total somatic cell count (TSCC), differential somatic cell count (DSCC), and bacteriological results The regression coefficient for the reference method and the alternative method of determining TSCC was 0.81, indicating that the method routinely used to analyze buffalo milk consistently underestimated actual TSCC. The milk samples positive for udder-specific bacteria also had higher TSCC values than the samples that were negative for bacteria ($872{\times}10^3$/ml vs. $191{\times}10^3$/ml). In samples that were positive for udder-specific bacteria, polymorphonuclear leukocytes (PMN) made up greater than 50% of the cells. Moreover, only 1% of the samples in the lowest TSCC class were positive for bacteria. The correlation between TSCC and PMN was stronger (0.70), and PMN values in buffalo milk increased significantly when the TSCC class changed from low (38%) to medium and high (56% and 64%). Milk yield was negatively related to TSCC. Significant changes in lactose (4.87%, 4.80% and 4.64%) and chloride content (0.650 mg/ml, 0.862 mg/ml and 0.882 mg/ml) were also observed with increasing TSCC values. Higher TSCC was associated with impaired rennet coagulation properties: the clotting time increased, while the curd firming time ($p{\leq}0.05$) and firmness decreased. We concluded that in buffalo as in dairy cows, TSCC is a valid indicator of udder inflammation; we also confirmed that a value of $ 200{\times}10^3 cells/ml should be used as the threshold value for early identification of an animal affected by subclinical mastitis. In addition to its association with significantly decreased milk yield, a TSCC value above this threshold value was associated with changes in milk composition and coagulating properties.

Effect of Feeding Strategies on Milk Production of Holstein Dairy Cows Managed by Small-Farmers Alpine Grassland in Korea

  • The main objective of this experiment was to investigate effects of two different feeding systems on body weight, milk yield, milk composition, and mineral and fatty acids content of Holstein dairy cows' milk. Sixteen of 25 months-old Holstein dairy cows were assigned to two groups (n=8) to study effects of the feeding system for 150 days. Two feeding systems were compared for five months; Group 1 was housed indoors and mainly fed a concentrate diet, Group 2 was maintained outdoors for five-seven hours/day on various kinds grass in a pasture. The experiment was conducted June-October 2017. Results revealed the indoor-fed cows had higher body weight, that was significant compared with the outdoor-based feeding system of Holstein dairy cows (p<0.05). Indoor-raised milking cows had higher milk yield (32.45 kg) as compared with pasture-raised milk yield (26.44 kg). Cows fed indoors significantly increased milk yield, total protein content, lactose, citric acid level, and lowered level of total solid and free fatty acids relative to the pasture-fed milking cows (p>0.05). There were higher levels of mineral content and fatty acid content in the milk of indoor-fed dairy cows than the pasture-raised dairy cows (p>0.05). Our study results demonstrated the potential benefits of the indoor feeding system for increased body weight, milk yield, mineral and fatty acids content summer through autumn when low pasture growth rates and quality may otherwise limit production.

Postpartum Reproductive Management Based on the Routine Farm Records of a Dairy Herd: Relationship between the Metabolic Parameters and Postpartum Ovarian Activity

  • The aim of this study was 1) to confirm the practical efficiency of a routine milk P4 monitoring system for postpartum reproductive management of a dairy herd, and 2) to evaluate the relationship between the blood metabolic profiles, milk quality and body weight of individual cows in the farm records, which may reflect the postpartum nutritional condition, and the time of postpartum resumption of ovarian activity of dairy cows. A total of 116 Holstein cows was used in the present study. First, during the period of Experiment 1, postpartum reproductive management based on weekly measured milk P4 concentration from individual cows was conducted. Compared with the reproductive records of the past two years without P4 monitoring, although the day from calving to first AI did not change, both the number of AI until pregnant (with P4; 1.9 times vs. without P4; 2.9 times) and the days open (with P4; 95.1 days vs. without P4; 135.8 days and 133.8 days) were significantly decreased. In Experiment 2, the measurement of blood constituents such as albumin, blood urea nitrogen, packed cell volume, ammonia, glucose, total cholesterol, non-esterified, AST and $\gamma$-GTP was performed on the blood samples taken once approximately 14 days postpartum, to monitor both health and nutritional conditions. The milk constituent parameters, such as milk protein (MP), milk fat (MF), SNF and lactose, collected from the monthly progeny test of individual cows, were used to monitor the postpartum nutritional status. Furthermore, the data obtained from the routine measurements of body weight were used to calculate the rate of peripartum body weight loss. The resumption day of the postpartum estrous cycle was assumed from the milk P4 profiles of individual cows. There was no clear relationship between each parameter from blood examination and those from resumption time. However, the cows had low values of MP, and SNF, which significantly affected the resumption of the postpartum estrous cycle. Similarly, a higher rate of body weight loss indicated a significant delay (more than 1 month) in the resumption of the postpartum estrous cycle, compared with the groups that had a medium or lower rate of body weight loss. The results of the present study demonstrated that the implementation of routine milk P4 monitoring-based postpartum reproductive management, together with milk quality parameters and routine BW data available in field conditions may be utilized as a practical approach for increasing the postpartum reproductive efficiency of a high yielding dairy herd.

  • 국산우유(國産牛乳)의 적정(適正) 살균조건(殺菌條件)을 구명(究明)하고자 $50^{\circ}C$로부터 $70^{\circ}C$까지 $5^{\circ}C$ 간격으로 가열처리(加熱處理)하고 처리유(處理乳)의 화학적조성(化學的組成) 및 미생물학적(薇生物學的) 성상(性狀) 변화(變化)를 확인함과 동시(同時)에 처리유(處理乳)의 보존성(保存性)을 시험(試驗)한 결과(結果) 다음과 같다. 1. $50^{\circ}C{\sim}70^{\circ}C$의 가열처리(加熱處理)에 의하여 우유(牛乳)의 pH 6.55로부터 6.42로 감소(減少)하였으며, 단백질(蛋白質), 지방(脂肪), 유당(乳糖) 및 회분(灰分)은 함량(含量)이 별로 변화(變化)하지 않았다. 2. 가열온도(加熱溫度)의 상승(上昇)에 따라 비(非) casein 태질소(態窒素) 및 여과성질소량(濾過性窒素量)은 감소(減少)하였으며 casein 태질소(態窒素)와 비단백태질소(非蛋白態窒素)는 증가(增加)하였다. 3. 가열온도(加熱溫度)의 상승(上昇)에 따라 우유중(牛乳中)의 Calcium 및 Vitamin C는 감소(減少)하였으며 가열처리유(加熱處理乳)의 인공소화율(人工逍化率)은 처리전(處理前)의 14.07%로부터 20.00%로 증가(增加)하였다. 4. $65^{\circ}C$의 가열살균(加熱殺菌)에 의하여 생균수(生菌數)는 $1.3{\times}10^4/m{\ell}$, 대장균수(大腸菌數)는 음성(陰性), 호냉성균수(好冷性菌數)는 $9.5{\times}10^2/m{\ell}$를 나타냈으며 99%의 살균율(殺菌率)을 표시(表示)하였다. 5. $65^{\circ}C$에서 가열살균(加熱殺菌)한 처리유(處理乳)는 $4^{\circ}C$의 냉장보존(冷藏保存)에서 화학적조성(化學的組成) 성상(性狀)의 변화(變化)가 적었다.

Isolation and Characterization of Cold-adapted Strains Producing ${\beta}-Galactosidase$

  • [ ${\beta}-Galactosidase$ ] is extensively employed in the manufacture of dairy products, including lactose-reduced milk. Here, we have isolated two gram-negative and rod-shaped coldadapted bacteria, BS 1 and HS 39. These strains were able to break down lactose at low temperatures. Although two isolates were found to grow well at $10^{\circ}C$, the BS 1 strain was unable to grow at $37^{\circ}C$. Another strain, HS-39, evidenced retarded growth at $37^{\circ}C$. The biochemical characteristics and the results of 16S rDNA sequencing identified the BS 1 isolate as Rahnella aquatilis, and showed that the HS 39 strain belonged to genus Buttiauxella. Whereas the R. aquatilis BS 1 strain generated maximal quantities of ${\beta}-galactosidase$ when incubated for 60h at $10^{\circ}C$, Buttiauxella sp. HS-39 generated ${\beta}-galactosidase$ earlier, and at slightly lower levels, than R. aquatilis BS 1. The optimum temperature for ${\beta}-galactosidase$ was $30^{\circ}C$ for R. aquatilis BS-1, and was $45^{\circ}C$ for Buttiauxella sp. HS-39, thereby indicating that R. aquatilis BS-1 was able to generate a cold-adaptive enzyme. These two cold-adapted strains, and most notably the ${\beta}-galactosidase$ from each isolate, might prove useful in some biotechnological applications.

국내 유가공산업의 R&D활성화를 위한 연구 동향과 방향 (Research Trends and Future Directions for R&D Vitalization of Domestic Dairy Industry)

  • 최근 한반도를 둘러싼 주변 환경이 크게 변화하는 가운데 특히 중국의 유제품 소비가 급증하고 있기 때문에 국내유가공산업은 향후 50년을 설계해야 하는 중요한 기로에 서 있다. 소비 측면에서 유제품의 시장 동향을 살펴보면 신선우유 소비는 감소하고, 발효유 등 유가공품 소비는 증가하고 있다. 원유사용량은 2010년 1,939천 톤을 예상되며, 반면 우유 공급량은 예년도 원유수요량으로 판단할 때 35천 톤 가량 공급 부족이 예상된다. 현재 다자간 협상 중인 미국 및 EU와 FTA 협정이 발효될 경우, 낙농산업분야의 상당한 피해가 예상되고, 수입 개방에 따라 국산 원유의 생산 감축으로 이어질 것이다. 현재 유가공업체가 농가에서 수취하는 원유는 미생물 함량과 체세포수가 매우 낮은 양질의 우유이기 때문에 초고온살균법 대신 유럽처럼 우유 고유 풍미 및 영양성분을 유지한 저온살균우유 생산을 유도하는 정책적 연구가 필요하다. 국내 치즈 시장은 65,423톤 규모로 시장 규모는 매년 확대되고 있으나 대부분을 수입에 의존하고 있는 실정이다. 자연치즈 산업 활성화와 더불어 반드시 해결할 문제로서 부산물(유청) 처리와 "목장형유가공(On-Farm Processing)"의 활성화이다. 이를 위해 규제를 완화하고 몇 가지 당면 문제를 해결한다면 대형 유가공업체와 동반성장이 가능하다고 생각된다. 국내 발효유 시장은 꾸준히 성장을 기록한 유일한 품목으로서 위, 간, 장 건강을 위한 기능성 발효유 제품의 매출이 크게 성장하였다. 인체에 유용한 프로바이오틱 유산균을 이용한 발효유 제품의 연구 개발은 중요한 연구 테마이다. 초유는 위장관 감염증의 치료나 예방에 사용될 수 있는 면역우유 제조에 사용될 수 있으며, 포유동물의 초유를 이용한 다양한 우유 제품의 연구 개발도 가능하다. 우유를 나노분자가공방법과 기능성나노식품소재의 첨가로 제품의 영양과 기호성을 향상시킬 수 있는 우유 제품을 개발할 수 있을 것이다. 국내 기능성 유제품의 효능 및 효과에 대한 표시기준 개정이 유가공업계가 안고 있는 최대의 당면 문제이다. 원유 수급 불균형은 구조적인 문제로서 우유의 적극적인 용도 관리가 선진국형 관리 체계로 바뀌어야 한다. 즉 내수용, 수출용, 수급 조절용으로 세분하는 것이다. 그리고 행정의 전문화를 위해 낙농연구 개발 지휘본부(control tower)의 설립을 건의하는 바이다. 외국의 "낙농위원회(Dairy Board)"처럼 수출용 유가공제품의 연구 개발을 전체적으로 총괄 지휘할 수 있는 기관이 필요하다. 마지막으로 유가공분야 인재육성 정책을 적극 검토해야 한다. 유가공 전공자 및 기술자에 대한 유학 또는 연수 기회를 확대함으로써 학생들의 의욕과 사기를 진작시켜야 한다.

  • The purpose of this research is that infant artificial feeding products is used in clonic with the study on characteristic, ingredients and indication of geneal and special modified milks. The result is as follows. 1. The main ingredients of four company products-Maeil , Namyang, Pasteur, Aebout is similar but the functional is different 2. General infant formula is divided into 100days, 5-6months, 12months, 24months and 36months out of consideration for growth and development of infant. 3. The special infant formula published in nation is as follows. Maeil soy A, Maeil MF1, Namyang hope doctor and Maeil HA is available for diarrhea. Maeil HA, Maeil HA-21 and Namyang hope allergy is available for hypoallergy. Maeil soy A is available for diarrhea of milk allergy. Maeil MF1 or Namyang hope doctor is available for acute bacterial or viral temporal diarrhea. Maeil HA is available for allergic chronic diarrhea. Maeil HA and Namyang hope allergy as eHP-formula is available for chronic diarrhea for lactose intolerance and milk allergy. Maeil-21 as pHP-formula for neonates with allergy family, allergic symptoms such as atopic dermatitis, asthma except digestive system.

개방식우사내 Fan과 Sprinkler설치가 착유우의 생리적 변화 및 유생산성에 미치는 영향 (Effects of Sprinkler and Fan Cooling System in Loose Barn on the Physiological Parameter and Milk Productivity in Holstein Cows During Summer)

  • 고온다습한 여름철에 우사내 fan + sprinkler에 의한 방서대책이 Holstein착유우의 생리적 반응, 유생산성 및 유조성분 등에 미치는 영향을 조사한 결과는 다음과 같다. 1 외기 온도가 가장 높은 15:00시에 측정한 대조구와 처리구의 건구 온도, 상대 습도, 이슬점 온도 및 수증기압 등은 각각 30.15 : 30.4$0^{\circ}C$, 71.44 : 70.82%, 24.32 : 24.26$^{\circ}C$, 30.51 : 30.50mbar로 fan + sprinkler설치가 기상 조건에 영향을 미치지는 않았다. 2. 피부온도는 대조구(34.02$^{\circ}C$)에 비하여 fan + spinkler 처리구(32.96$^{\circ}C$)가 유의하게 저하되었다(P<0.05). 또한 직장온도도 대조구가 41.21$^{\circ}C$, 처리구가 39.53$^{\circ}C$로 대조구에 비해 처리구에서 유의하게 낮았다(P<0.05). 3. 혈청 Cortisol 농도는 대조구가 1.44$\mu\textrm{g}$/dL로 처리구의 0.90$\mu\textrm{g}$/dL 보다 유의하게 높았다(P<0.05). 4. 평균 산유량은 대조구가 23.66kg, 처리구가 25.87kg으로 대조구에 비해 처리구가 유의하게 증가하였으며(P<0.05), 시험축을 유기별로 나누어 유량을 조사한 바 대조구 보다 처리구가 비유초기, 중기 및 후기에서 각각 8.16%, 2.83% 및 12.14%의 증가를 보였으나 유의차는 없었다. 5. 우유중 유당, 유단백 및 무지고형분 등의 함량 및 생산량은 대조구와 처리구간에 차이가 없었다. 그러나 유지방 함량의 경우 대조구가 3.22%, 처리구가 3.37%로 처리구가 0.15% point 증가했으나 유의차는 없었다. Fan + sprinkler가 설치된 시설에서 사육된 젖소에서 생산된 우유내 체세포수는 대조구에 비해 26.63%가 감소되었으나 유의차는 발견할 수 없었다. 이상의 시험 결과에서 볼 때 fan + sprinkler를 이용한 방서 방법은 고온 stress를 완화시켜 유생산성의 증가에 기여할 수 있는 하나의 방법이라고 본다.

A New Synthetic Medium for Lactic Lactococci: Application to Marine lactic Acid Bacteria

  • Lactococcal cells are nutritionally fastidious and thus, generally cultured either in milk or M17 medium (Terzaghi and Sandine, 1975). In this study, Lactococcus cremoris wild-type (KH) and its less­proteolytic mutant (KHA1) cells were grown on the M17 medium or with modified M17 medium by replicated parallel experiments. The modified M17 medium had the same composition as M17 medium, except that lactose was replaced by glucose. Analyses of culture-broth samples, in which the M17 and the modified M17 media were used, were conducted by high-performance liquid chromatography (HPLC). But, working with these media created noisy problems in analyses of samples. Therefore, a new semi-synthetic medium was developed on the basis of nutritional requirements (Morishita et al., 1981). The composition of the semi-synthetic medium determined on the basis of the nutritional requirements and the composition of milk, is presented in Table 1. The composition of M17 medium is also presented and compared in the table. L. cremoris KH and KHA1 cells were grown again on the new synthetic medium containing glucose or lactose. The broth samples were then drawn and analyzed by HPLC. Clearer separations of fermented products were achieved from the new medium than those with the M17 and the modified M17 media. In comparison with the M17 or the modified M17 media, growth on the new medium was good (Kim et al, 1993). Additional fermentations were also carried out at a controlled pH of 7.0, where enhanced growth of lactococcal cells was obtained. In the fermentations, samples were also analyzed for the concentrations of sugar and lactic acid. The results showed that the new synthetic medium was as good as or better than the M 17 and the modified M 17 media. This is because casein hydrolysate in the synthetic medium provided a ready supply of amino acids and peptides for L. cremoris KH and KHA1 cells. Lactic acid bacteria (LAB) including Lactococcal cells have been known to be an effective means of preserving foods, at the same time as giving particular tastes in fields of dairy products. LAB also have always occupied an important place in the technology of sea products, and marine LAB have known to be present in traditional fermented products (Ohhira et al, 1988). To apply the new synthetic medium to marine LAB, two different LAB were isolated from pickled anchovy and pollacks caviar and were grown on the new media in which various concentrations of NaCl $(3, 5, 7 and 10\%)$ added. They were also grown on the medium solution in natural seawater $(35\%o\;salinity)$ and on the solution of natural seawater itself, too. As seen in Fig. 1, Marine LAB were grown best on the synthetic medium solution in natural seawater and the higher concentrations of NaCl were added to the medium, the longer lag-phase of growth profile appeared. Marine LAB in natural seawater were not grown well. From these results, the synthetic medium seems good to cultivate cells which are essential to get salted fish aged. In this study, it showed that the new synthetic medium provided adequate nutrition for L. cremoris KH and KHA1 cells, which have been used as cheese starters (Stadhouders et al, 1988). Using this new medium, the acid production capability of starter cultures could be also measured quantitatively. Thus, this new medium was inferior to the M17 or the modified M17 medium in culturing the cheese starters and in measuring fermentation characteristics of the starter cells. Moreover, this new medium found to be good for selected and well-identified marine LAB which are used in rapid fermentations of low-salted fish.

Mammary alveolar cell as in vitro evaluation system for casein gene expression involved in glucose level

  • Objective: Glucose is an essential fuel in the energy metabolism and synthesis pathways of all mammalian cells. In lactating animals, glucose is the major precursor for lactose and is a substrate for the synthesis of milk proteins and fat in mammary secretory (alveolar) epithelial cells. However, clear utilization of glucose in mammary cells during lactogenesis is still unknown, due to the lack of in vitro analyzing models. Therefore, the objective of this study was to test the reliability of the mammary alveolar (MAC-T) cell as an in vitro study model for glucose metabolism and lactating system. Methods: Undifferentiated MAC-T cells were cultured in three types of Dulbecco's modified Eagle's medium with varying levels of glucose (no-glucose: 0 g/L, low-glucose: 1 g/L, and high-glucose: 4.5 g/L) for 8 d, after which differentiation to casein secretion was induced. Cell proliferation and expression levels of apoptotic genes, Insulin like growth factor-1 (IGF1) receptor, oxytocin receptor, ${\alpha}S1$, ${\alpha}S2$, and ${\beta}$ casein genes were analyzed at 1, 2, 4, and 8 d after differentiation. Results: The proliferation of MAC-T cells with high-glucose treatment was seen to be significantly higher. Expression of apoptotic genes was not affected in any group. However, expression levels of the mammary development related gene (IGF1 receptor) and lactation related gene (oxytocin receptor) were significantly higher in the low-glucose group. Expressions of ${\alpha}S1-casein$, ${\alpha}S2-casein$, and ${\beta}-casein$ were also higher in the low-glucose treated group as compared to that in the no-glucose and high-glucose groups. Conclusion: The results demonstrated that although a high-glucose environment increases cell proliferation in MAC-T cells, a low-glucose treatment to MAC-T cells induces higher expression of casein genes. Our results suggest that the MAC-T cells may be used as an in vitro model to analyze mammary cell development and lactation connected with precise biological effects.
