Effects of d-${\alpha}$-tocopherol supplements on lipid metabolism in a high-fat diet-fed animal model

  • High-fat diet up-regulates either insulin resistance or triglycerides, which is assumed to be related to the expression of peroxisome proliferator-activated receptor (PPAR)-${\alpha}$ and PPAR-${\gamma}$. The beneficial effects of vitamin E on insulin resistance are well known; however, it is not clear if vitamin E with a high-fat diet alters the expression of PPAR-${\alpha}$ and PPAR-${\gamma}$. We investigated the effects of d-${\alpha}$-tocopherol supplementation on insulin sensitivity, blood lipid profiles, lipid peroxidation, and the expression of PPAR-${\alpha}$ and PPAR-${\gamma}$ in a high-fat (HF) diet-fed male C57BL/6J model of insulin resistance. The animals were given a regular diet (CON; 10% fat), a HF diet containing 45% fat, or a HF diet plus d-${\alpha}$-tocopherol (HF-E) for a period of 20 weeks. The results showed that the HF diet induced insulin resistance and altered the lipid profile, specifically the triglyceride (TG) and total cholesterol (TC) levels (P < 0.05). In this animal model, supplementation with d-${\alpha}$-tocopherol improved insulin resistance as well as the serum levels of TG and very-low-density lipoprotein-cholesterol (VLDL-C) (P < 0.05). Moreover, the treatment decreased the levels of malondialdehyde (MDA) in the serum and liver while increasing hepatic PPAR-${\alpha}$ expression and decreasing PPAR-${\gamma}$ expression. In conclusion, the oral administration of d-${\alpha}$-tocopherol with a high-fat diet had positive effects on insulin resistance, lipid profiles, and oxidative stress through the expression of PPAR-${\alpha}$ and PPAR-${\gamma}$ in a high-fat diet-fed male mice.

  • In order to study the effect of source and level of the commonly used dietary fats on growth and metabolism of rats fed on low protein diet (rice diet) the weaning white rats were fed on various different experimental diets (see tables 1 and 2) during 11 weeks. The observations were made as follows : 1. Growth: (see table 3 and figures 1-9) In all dietary fats, among the 3 levels, 5% fat level is the best. Especially, the perilla oil group was remarkably good. 10% and 20% fat levels impaired the growth, consequently the growth rates of both 10% and 20% fat level groups were worse than those of Basal group (no fat added). However, 10% and 20% fat levels did not impaired the growth of VII group (10% soy flour added) In 5% fat level, the growth was good in sequence of perilla oil, tallow, sesame oil, soy oil and lard. 2. Feed consumption: (see table 3) In 20% fat level, the feed consumption was lowered. Generally, the feed consumption rate was proportional to the growth rate. In feed efficiency, 5% fat level was the best. 3. Liver weight: (see table 4) In liver weight per 100 G body weight, 20% fat level was the largest. This may be due to the poor body growth and liver fat accumulation. 4. Liver nitrogen: (see table 4) Generally, lower fat level groups showed liver nitrogen. Liver nitrogen is low in the groups of 20% fat level. 5. Liver fat: (see table 4) Generally, higher fat level groups showed higher liver fat. 6. Serum cholesterol: (see table 5) Generally, higher fat level groups showed higher serum cholesterol. Lard, sesame oil, and tallow groups showed higher level and soy oil and perilla oil groups showed lower level. Especially, perilla oil group showed remarkably lower level and VII group (10% soy flour added) showed lower level than VI group (same fat but no soy flour added).

The effects of Brassica juncea L. leaf extract on obesity and lipid profiles of rats fed a high-fat/high-cholesterol diet

  • BACKGROUND/OBJECTIVES: Obesity is a global health problem of significant importance which increases mortality. In place of anti-obesity drugs, natural products are being developed as alternative therapeutic materials. In this study, we investigated the effect of Brassica juncea L. leaf extract (BLE) on fat deposition and lipid profiles in high-fat, high-cholesterol diet (HFC)-induced obese rats. MATERIALS/METHODS: Male Sprague-Dawley rats were divided into four groups (n = 8 per group) according to diet: normal diet group (ND), high-fat/high-cholesterol diet group (HFC), HFC with 3% BLE diet group (HFC-A1), and HFC with 5% BLE diet group (HFC-A2). Each group was fed for 6 weeks. Rat body and adipose tissue weights, serum biochemical parameters, and tissue lipid contents were determined. The expression levels of mRNA and proteins involved in lipid and cholesterol metabolism were determined by reverse transcription polymerase chain reaction and western blot analysis, respectively. RESULTS: The HFC-A2 group showed significantly lower body weight gain and food efficiency ratio than the HFC group. BLE supplementation caused mesenteric, epididymal, and total adipose tissue weights to decrease. The serum levels of triglyceride, total cholesterol, and low-density lipoprotein cholesterol were significantly reduced, and high-density lipoprotein cholesterol was significantly increased in rats fed BLE. These results were related to lower glucose-6-phosphate dehydrogenase, acetyl-coA carboxylase, and fatty acid synthase mRNA expression, and to higher expression of the cholesterol $7{\alpha}$-hydroxylase and low density lipoprotein-receptor, as well as increased protein levels of peroxisome proliferator-activated receptor ${\alpha}$. Histological analysis of the liver revealed decreased lipid droplets in HFC rats treated with BLE. CONCLUSIONS: Supplementation of HFC with 3% or 5% BLE inhibited body fat accumulation, improved lipid profiles, and modulated lipogenesis- and cholesterol metabolism-related gene and protein expression.

포도씨유, 들깨유 및 옥수수유의 급여가 흰쥐의 체내 지질패턴 및 간조직의 지방산 조성에 미치는 영향 (The Effect of Grape Seed Oil, Perilla Oil, or Corn Oil-Containing Diet on Lipid Patterns in Rats and Fatty-Acid Composition in Their Liver Tissues)

  • 포도씨유의 생체 내 지질대사에 미치는 효과를 평가하고자 식이급원이 상이한 들깨유 및 옥수수유를 흰쥐에게 급여하여 체내 지질패턴 및 간조직의 지방산 조성을 분석한 결과는 다음과 같다. 일반식이군과 고지방 식이를 급여받은 쥐의 체중은 7주까지 꾸준히 증가하였다. 그러나 일반식이에 들깨유을 급여받은 쥐는 7주 후 체중이 감소하였고 다른 유지군을 급여받은 군들은 8주째부터 감소하였다. 식이섭취량 은 고지방 식이군 보다 일반식이군이 높았고 식이효율은 일반식이에 들깨유을 혼합하여 급여한 군에서 유의적으로 낮았다. 혈청 지질패턴 분석결과, 총지질 농도는 들깨유을 혼합한 일반식이군과 고지방 식이군에서 유의적으로 감소하였고 중성지방과 인지질 농도는 일반식이군에게 들깨유 급여시 유의적으로 낮았다. HDL-C은 일반식이군이 고지방 식이군보다 유의적으로 높게 나타났으나 일반식이군에 포도씨, 들깨유 및 옥수수유을 혼합하여 급여한 군간에 유의적인 차이는 없었던 반면 고지방 식이군와 들깨유을 급여한 군에서 HDL-C이 유의적으로 증가하였다. LDL-C은고지방식이군과 일반식이군간에 유의적인 차이가 나타났는데, 특히 고지방식이에 들깨유을 급여한 군에서 유의적으로 낮았고 포도씨유 급여군도 감소하는 경향이었다. HDL-ChDL-C 수치가 고지방식이 군에서 다른 식이지방에 비하여 PHF군에서 유의적으로 높았던 것은 혈중 지질패턴을유익하게 바꾸어준 결과이며, 이로 인하여 동맥경화인자인Al 지수가 CHF군에 비해 PHF군과 GSHF군에서 유의적으로 감소되었다. 간조직의 총지방, 총콜레스테를 및 중성지방 함량은 일반식이군은 들깨유군에서 가장 낮았고 그다음이 포도씨유군 및 옥수수유군 순으로 관찰되었다. 간조직의지방산 조성 분석결과 고지방식이군은 포화지방산은 감소하였고 단일불포화 지방산과 다가불포화 지방산은 일반식이군에 비해 증가하였다. 따라서 본 연구결과 유지 급원에 따라함유하는 지방산 조성의 비율도 달라져 식이 급원에 의해 생체내 지방산 조성에 큰 영향을 미치는 것으로 생각된다.

  • To observe the effect of the different level of PUFA and marginal tocopherol supplement on HDL-chol, tissue tocopherol content and fatty acid composition, the rats were supplied either safflower oil or conconut oil with or without tocopherol supplement to the experimental diet. Plasma tocopherol level was not greatly influenced by the different dietary fat and similar effect was observed in the liver but not in the adipose tissue. HDL-chol level was reduced in the high PUFA diet regardless of tocopherol content. No effect by tocopherol supplement was observed in the fatty acid composition of liver and adipose tissue lipid in both dietary PUFA levels . There was also no increase in the content of tissue polyenoid acid by tocopherol in the high PUFA diet . Fatty acid composition of tissue lipid was rather more influenced by dietary fat. Lauric and myristic acid contents were higher in the low PUFA diet and linoleic acd and total polynoic acid content were higher in the high PUFA diet. With tocopherol supplement tocopherol /PUFA ratio of tissue was increased but the ratio of high PUFA diet was significantly lower than that of low PUFA diet. Marginal tocopherol supplement could not reduce the peroxidizability index of high PUFA diet.

Extracts of Codonopsis lanceolata Decrease $CCl_4$-Induced Hepatotoxicity on High Fat Diet in Rats

  • This study was designed to investigate the effect of Codonopsis lanceolata extracts on carbon tetrachloride (CCl₄)­induced hepatotoxicity in rats fed high, medium or low fat diet. The liver weight was increased not by the level of fat in diet but by administration of CCl₄. Activities of serum aspartate aminotransferase (AST) and alkaline phosphatase (ALP) were significantly increased by CCl₄, the activities of AST and alanine aminotransferase (ALT) were significantly decreased in case of CCl₄ treatment with C. lanceolata extracts. In high fat diet with CCl₄ administration, the content of total lipid in liver was significantly increased, which was significantly decreased by the administration of C. lanceolata extracts. Administration of C. lanceolata water extracts remarkably prevented the tissue injury such as lipid degeneration, infiltration of inflanunatory cells and necrosis caused by CCl₄, but those methanol extracts did not affect histopathological changes of the liver as water extract. The data suggest that intake of C. lanceolata extracts would prevent lipid accumulation and hepatotoxicity induced by high fat diet and CCl₄ abministratim, and the water extract of C. lanceolata is supposed to be more effective than those methanol extract.

폐경 후 비만 여성의 식이 및 운동 교육이 체지방향 및 혈중지질농도에 미치는 효과 (Effects of Nutrition and Exercise Education on Fat Mass and Blood Lipid Profile in Postmenopausal Obese Women)

  • Obesity is an independent risk factor fur coronary artery disease in the postmenopausal women, which may be mediated by alteration of blood lipid metabolism. We are aimed to evaluate the effects of low energy diet, restriction of high fat foods and exercise education on weight, fat mass and blood lipid profile. Fifteen postmenopausal obese women were studied. Subjects received detailed advice about how to achieve a reduction of weight by a low-energy diet (1,200kcal), restriction of high fat food and increased exercise in every 4 wks during 8 wk. To evaluate the effectiveness for education programs, dietary fat habit and daily nutrient intakes, exercise were tested before and after intervention. Anthropometry, computerized tomography, and blood lipid profile were assessed before and after intervention. According to the nutrition education, energy intake (from 1776.1 ${\pm}$ 28.2 kcal to 1268.7 ${\pm}$ 115.2 kcal, p < 0.001) and percent of energy from fat (21.9%, p < 0.01) were significantly decreased. However, the index of nutritional quality was over 1.0. On the contrary, exercise (from 341.3 ${\pm}$ 222.1 kcal to 569.4 ${\pm}$ 309.8 kcal, p < 0.05) was increased and dietary fat habit scores (from 30.8 ${\pm}$ 5.2 to 36.1 ${\pm}$ 3.0, p < 0.01) were improved. Also, body weight (4.2%, p < 0.001), BMI (4.5%, p < 0.001), body fat mass (3.4%, p < 0.05) and waist to hip ratio (1.1%, p < 0.05) were decreased. Among abdominal fat, visceral fat (26.1%, p < 0.05) and subcutaneous fat (14.8%, p < 0.01) were decreased. But there was no difference in visceral fat to subcutaneous fat ratio. We observed HDL-cholesterol increase (11.7%, p < 0.05), triglyceride reduction(14.8%, p < 0.05) and atherogenic index improvement (from 2.7 ${\pm}$ 0.7 to 2.3 ${\pm}$ 0.7, p < 0.05) However, there was no change in LDL-cholesterol and total cholesterol. Theses results showed that low energy diet, restriction of high fat foods and exercise education could result in reduction of body weight, fat mass, visceral and subcutaneous fat, and in improvement of blood lipid profile in the postmenopausal obese women.

고지방식이 비만모델에서 방기황기탕(防己黃芪湯) 합(合) 영계출감탕(苓桂朮甘湯)의 항비만 효과 (Anti-obesity Effects of Banggihwnggi-tang-hap-yeonggyechulgam-tang in High Fat Diet Induced Obese Mice Model)

  • Objectives This study is to investigate anti-obesity effects of Banggihwanggi-tang-hap-yeonggyechulgam-tang (BY), an herbal formula, in high fat diet induced obese mice model. Methods Fourty five male C57Bl/6J mice were randomly assigned to normal group fed with normal research diet (Nor, n=9), high fat diet control group treated with water (Veh, n=9), high fat diet group treated with orlistat (Oris; n=9, Orlistat 40 mg/kg), high fat diet group treated with low concentraion BY (BYL; n=6, BY 0.87 g/kg) and high fat diet group treated with high concentration BY (BYH; n=6, BY 1.74 g/kg). Results Seven weeks later, antioxidative capacity, body weight, epididymal fat pad and liver weight, reactive oxygen species (ROS), peroxynitrite ($ONOO^-$), alanine aminotransferase (ALT), aspartate aminotransferase (AST), total cholesterol, triglyceride, high density lipoprotein (HDL), low density lipoprotein (LDL), superoxide dismutase (SOD), catalase, glutathione peroxidase (Gpx), heme oxygenase (HO)-1 and histology of liver were evaluated. In the BYH group, 1,1-diphenyl-2-picrylhydrazyl and 2,2'-azinobis (3 ethybenzothiazoline-6-sulfonic acid) radical scavenging activity were more than L-ascorbic acid. Body weight gain were significantly less than Veh group. Epididymal fat pad and liver weight gain were significantly less than Veh group. ROS and $ONOO^-$ were significantly less than with Veh group. ALT and AST were significantly less than with Veh group. Total cholesterol, triglyceride and LDL were significantly less, HDL were significantly more than Veh group. SOD, catalase, Gpx, HO-1 significantly increased compared with Veh group. Injury on liver was lesser than Veh group. Conclusions It can be suggested that BY has anti-obesity effects in high fat diet induced obese mice model.

고지방식이에서 일반식이로의 전환이 유방암의 성장 및 전이에 미치는 영향 (Effect of dietary changes from high-fat diet to normal diet on breast cancer growth and metastasis)

  • 본 연구에서는 일반식이로의 식이 제한이 고지방식이로 촉진된 유방암의 성장과 전이에 미치는 영향과 그 기전을 알아보았다. 8주간 고지방식이를 섭취시킨 후 유방암을 유방 조직에 이식하거나 꼬리 정맥으로 통해 전이시켰으며, 암 생성 및 전이 후 HFC군은 고지방식이를, DR군은 일반식이로 전환시켜 사육하였다. 본 연구 결과, 원발성 유방암의 경우, HFC군과 DR군의 체중에는 차이가 없었으나 간과 비장, 신장 주변의 지방세포의 무게에서 유의한 차이를 보였다. DR군이 HFC군에 비해 유방암의 개시를 지연시켰으며, 유방암의 성장과 무게를 유의하게 감소시켰다. 일반식이로의 전환에 의한 유방암 성장 억제는 세포 분열 억제와 세포사멸을 조절하는 인자의 발현 감소에 의한 caspase-3의 활성 촉진에 의한 것으로 나타났다. 또한, 식이 제한은 폐로 전이된 유방암의 수를 유의미하게 감소시켰다. 본 실험 결과, 일반식이로의 제한은 고지방식이로 촉진된 유방암의 성장과 전이를 억제하는 효과를 나타내며, 이는 유방암 세포의 성장 억제와 사멸 유도에 의한 것으로 나타났다. 따라서, 유방암의 성장이나전이를 억제하기 위해서는 총 섭취 열량을 조절하고, 지방의 섭취량을 줄이는 균형 잡힌 식이를 섭취하도록 해야 할 것으로 사료된다.

Effects of caloric restriction with varying energy density and aerobic exercise on weight change and satiety in young female adults

  • This study examines the combined effects of caloric restriction on body composition, blood lipid, and satiety in slightly overweight women by varying food density and aerobic exercise. Twenty-three women were randomly assigned to one of two groups for a four-week weight management program: the high-energy density diet plus exercise (HDE: n = 12, $22{\pm}2$ yrs, $65{\pm}7$ kg, $164{\pm}5$ cm, $35{\pm}4%$ fat) and low-energy density diet plus exercise (LDE: n = 11, $22{\pm}1$ yrs, $67{\pm}7$ kg, $161{\pm}2$ cm, $35{\pm}4%$ fat) groups. Subjects maintained a low-calorie diet (1,500 kcal/day) during the program. Isocaloric ($483{\pm}26$ for HDE, $487{\pm}27$ kcal for LDE) but different weight ($365{\pm}68$ for HDE, $814{\pm}202$ g for LDE) of lunch was provided. After lunch, they biked at 60% of maximum capacity for 40 minutes, five times per week. The hunger level was scaled (1: extremely hungry; 9: extremely full) at 17:30 each day. Before and after the program, the subjects' physical characteristics were measured, and fasting blood samples were drawn. The daily energy intake was $1,551{\pm}259$ for HDE and $1,404{\pm}150$ kcal for LDE (P > 0.05). After four weeks, the subjects' weights and % fat decreased for both LDE (-1.9 kg and -1.5%, P < 0.05) and HDE (-1.6 kg and -1.4%, respectively, P < 0.05). The hunger level was significantly higher for HDE ($2.46{\pm}0.28$) than for LDE ($3.10{\pm}0.26$) (P < 0.05). The results suggest that a low-energy density diet is more likely to be tolerated than a high-energy density diet for a weight management program combining a low-calorie diet and exercise, mainly because of a reduced hunger sensation.
