The Effect of Light Intensity on the Growth and Chlorophyll Fluorescence Parameters of Three Ardisia Genus Native to Korea

  • This study investigated the growth and chlorophyll fluorescence reactions of three Ardisia genus grown under various indoor light intensity conditions with the aim of evaluating their suitability as indoor plants. Young seedlings of A. crispa (Thunb.) A.DC., A. pusilla DC., and A. japonica (Thunb.) Blume were used in the experiment. The plants were cultivated indoors for 10 weeks under different light intensities: 10, 50, 100, and 200 PPFD (μmol·m-2·s-1), and their growth was compared with that of plants cultivated in a greenhouse during the same period (mean value 236.8±20.4 PPFD at noon). Also, chlorophyll fluorescence analysis was investigated with a portable PAM fluorometer. The indoor plants were maintained at 12/12 h photoperiod, temperature at 25±1℃, and humidity at 55±3%. Irrigation frequency (once every three days) was the same for the indoors and the greenhouse. The results of growth in three Ardisia plants showed that almost all parameters except leaf number and chlorophyll content had similar levels regardless of light intensity. A. crispa and A. pusilla plants grown in 200 PPFD were investigated to have low chlorophyll contents. Meanwhile, chlorophyll fluorescence parameters differed based on light levels. In A. crispa, the Fv/Fm (0.77), DIo/RC (0.47) and Fm/Fo (4.77) parameters tended to be poor at 200 PPFD compared to those at other light intensities. Similarly, the DIo/RC, Fm/Fo, and Pi_Abs parameters of A. pusilla plant (200 PPFD) are 0.45, 4.48 and 2.42, respectively, which can be considered stress. The analysis of fluorescence in A. japonica showed that all parameters except ETo/RC had similar levels regardless of light intensity. The ETo/RC parameter was 0.49 and 0.72 in the control plants and plants 200 PPFD, respectively, which was lower than those in plants at other light intensities. Therefore, it seems that the relatively high light intensity acted as a stressor for Ardisia plants.

Cold Pressor Response to Seasonal Variation in Winter and Summer (국소한냉자극이 전신 및 국소혈액순환에 미치는 영향 -제 2 보 : 동계 및 하계의 계절변화에 따른 한냉반응-)

  • A possibility whether the appearance of adaptation to cold climate during winter could occur or not in Taegu area was evaluated by comparing the data obtained in winter with that obtained by the same method in summer. Circulatory response was induced by the immersion of one hand in the cold water. The systemic and local responses in the blood circulation from the immersed hand and the unimmersed opposite hand were observed simultaneously. In addition Galvanic skin resistance(GSR) that is influenced by the activity of autonomic nervous system and the vascular tonicity was recorded. The experiment was performed by examining sixty healthy college students in winter and fifty in summer, whose mean age was 21.0, mean weight $60.6{\pm}0.90\;kg(male)$ and $48.3{\pm}0.98\;kg(female)$. The cold stimulus was applied by immersing the left hand into the cold water of $5^{\circ}C$ for 3 minutes, and the response was observed on immersed left hand and unimmersed right hand simultaneously. The observation was made through determining mean blood pressure, heart rate, amplitude of photoelectric capillary pulse (APCP) and GSR. The results obtained are as follows: The mean blood pressure was elevated during the cold stimulation. The increase of blood pressure in summer was more remarkable than in winter. At the recovery period the blood pressure was decreased to the control level in winter but the decrease below the control level was observed in summer. The increase of heart rate in summer was more remarkable than in winter during the cold stimulation. At the recovery period heart rate in both winter and summer was decreased below the control level. During the cold stimulation the APCP was decreased on both hands in winter. However it was more prominent on left hand indicating additional direct cold effect on immersed hand. In summer, the decrease of APCP during immersion was less remarkable than that in winter, but the regain of APCP was faster than that in winter at the recovery period. And the prompt increase of APCP over the control level has been obtained at the 3 minutes of the recovery period. The GSR was remarkably increased on immersed hand but slightly decreased on unimmersed opposite hand during the cold stimulation. Thus the finding on immersed hand indicates that the local direct effect of cold water is more prominent than the systemic effect, where as the finding on unimmersed hand indicates that the circulatory response to painful stress elicited by the cold stimulation is more prominent than cold temperature itself. In summary, it seems that the systemic circulatory response to the local cold stimulation of the one hand is arised more from the secondary elicited pain sensation and less from the low water temperature. On the contrary to the report of Kim et $al^{39)}$, the adaptation phenomena in blood pressure to the relatively mild cold climate in winter was not observed in this study. The difference of circulatory response observed in this study between winter and summer may be due to the difference of the magnitude of subjective sensation of the cold water stimulation by the seasonal changes in air temperature.

  • In this study, the distributions of internal climates such as air temperature, humidity, dust, ammonia gas, and air velocity were systematically measured at a mechanically ventilated broiler houses during summer season, with local weather data. The analysis was focused on the suitability, stability, and uniformity of internal climate, resulting in serious stress on chickens and decrease of productivity In the mechanically ventilated broiler house, the difference between measured and recommended air temperatures(suitability) was 10.4C in maximum during the summer time. The difference of air temperature in the house between day and night was $8.7^{\circ}C$ in maximum. And maximal hourly range of internal air temperature at 0.4m height from the floor was $3.7^{\circ}C$ suggesting it maintained thermal uniformity in the broiler house. The $NH_3$ and dust concentrations were pretty low because ventilation was fully performed. The air speed at chicken location was measured 2.2m/s and 1.7m/s, respectively without and with chicken existence.

Determining Spatial and Temporal Variations of Surface Particulate Organic Carbon (POC) using in situ Measurements and Remote Sensing Data in the Northeastern Gulf of Mexico during El $Ni\tilde{n}o$ and La $Ni\tilde{n}a$ (현장관측 및 원격탐사 자료를 이용한 북동 멕시코 만에서 El $Ni\tilde{n}o$와 La $Ni\tilde{n}a$ 기간 동안 표층 입자성 유기탄소의 시/공간적 변화 연구)

  • Surface particulate organic carbon (POC) concentration was measured in the Northeastern Gulf of Mexico on 9 cruises from November 1997 to August 2000 to investigate the seasonal and spatial variability related to synchronous remote sensing data (Sea-viewing Wide Field-of-view Sensor (SeaWiFS), sea surface temperature (SST), sea surface height anomaly (SSHA), and sea surface wind (SSW)) and recorded river discharge data. Surface POC concentrations have higher values (>100 $mg/m^3$) on the inner shelf and near the Mississippi Delta, and decrease across the shelf and slope. The inter-annual variations of surface POC concentrations are relatively higher during 1997 and 1998 (El Nino) than during 1999 and 2000 (La Nina) in the study area. This phenomenon is directly related to the output of Mississippi River and other major rivers, which associated with global climate change such as ENSO events. Although highest river runoff into the northern Gulf of Mexico Coast occurs in early spring and lowest flow in late summer and fall, wide-range POC plumes are observed during the summer cruises and lower concentrations and narrow dispersion of POC during the spring and fall cruises. During the summer seasons, the river discharge remarkably decreases compared to the spring, but increasing temperature causes strong stratification of the water column and increasing buoyancy in near-surface waters. Low-density plumes containing higher POC concentrations extend out over the shelf and slope with spatial patterns and controlled by the Loop Current and eddies, which dominate offshore circulation. Although river discharge is normal or abnormal during the spring and fall seasons, increasing wind stress and decreasing temperature cause vertical mixing, with higher surface POC concentrations confined to the inner shelf.

Physiological Responses to Drought Stress of Seven Evergreen Hardwood Species (상록활엽수 7수종의 건조스트레스에 대한 생리적 반응)

  • This research aims to analyze and compare the drought resistance of 7 species of landscape trees commonly grown in Korea. The 7 species are: Camellia japonica, Rhaphiolepis indica, Quercus glauca, Machilus thunbergii, Daphniphyllum macropodum, Dendropanax morbifera and Cinnamomum camphora. In order to analyze their drought resistance, the samples were left without irrigation for 30 days (05/09/2016 ~ 05/10/2016), during which period their respective drought resistor, relative water content, electrolyte elution figures and proline content were measured. As the non-irrigation proceeded, C. camphora was the first to wither, followed by D. morbifera, then D. macropodum, then M. thunbergii, then Q. glauca, then R. indica then finally C. japonica. Of the 7 species, Q. glauca, C. japonica and R. indica can be considered highly drought resistant, since they survived for longer than 3 weeks without irrigation. Relative water content (RWC) plummeted dramatically after the first 15 days of non-irrigation. Whereas RWC readings of C. camphora, D. morbifera, D. macropodum and M. tunbergii dropped by 40% or more, the other 4 species reported a relatively low rate of decrease at 20% or lower. The Camellia japonica, the R. indica and Q. glauca, which were the species with relatively high drought resistance, showed low proline content and electrolyte elution figures, whereas those of C. camphora, D. macropodum, D. morbifera and M. tunbergii were higher. Analysis through the nonlinear regression analysis logistic model showed that non-irrigation proved fatal for the 7 sample species in a range of 22.7 to 37.6 days. The C. japonica, R. indica, Q. glauca and M. tunbergii demonstrated a high drought resistance of 30 days or longer, whereas C. camphora, D. morbifera and D. macropodum had a low resistance of 25 days or less to drought from lack of water. In conclusion, out of the 7 species of broad-leaved evergreen trees tested, C. japonica, R. indica and Q. glauca seem to be suitable for use as landscape trees, owing to their high drought resistance.

In Vitro Anticancer and Antioxidant Effect of Solvent Extracts from Tuna Dried at Low Temperature Vacuum. (저온진공건조 참치추출물의 in vitro 항암 및 항산화 효과)

  • We investigated the inhibitory effects of solvent extracts from dried tuna on the growth of cancer cell lines (HT1080 human fibrosarcoma and HT-29 human colon cancer cells) and $H_2O_2$-induced oxidative stress. Inhibitory effects of acetone with methylene chloride (A+M) and methanol (MeOH) extracts on the growth of HT1080 and HT-29 cancer cells increased in a dose dependent manner (p<0.05). The inhibitory effect was more significant on the growth of HT1080 cells, and A+M extracts had a higher inhibitory effect compared to MeOH extracts. The treatments of hexane, 85% aq. methanol, butanol and water fractions significantly inhibited the growth of both cancer cells (p<0.05). Among the fractions, hexane and 85% aq. methanol fractions showed higher inhibitory effects. In order to determine the protective effect on $H_2O_2$-induced oxidative stress, a DCHF-DA (dichlorodihydrofluorescin diacetate) assay was conducted. All fractions, including crude extracts of dried tuna, appeared to significantly reduce the levels of intracellular reactive oxygen species (ROS) with dose responses (p<0.05). Among the fractions, BuOH and 85% methanol fractions showed a higher protective effect on the production of lipid peroxides. These results indicate that the consumption of tuna may be recommended as a potent functional food for preventing cellular oxidation and cancer.

Studies on the Germination Characters of Korean Ginseng (Panax ginseng C.A. Meyer) Seed (고려인삼종자(高麗人蔘種子)의 발아특성(發芽特性)에 관(關)한 연구(硏究))

  • This study was conducted to define the optimal conditions for embryo growth during seed stratification and for breaking dormancy as well as seed germination of stratified ginseng seeds. The experiments were also carried out to detect some materials which were expected to induce seed dormancy in the ginseng seeds. The results summarized as follows; 1. The growth of embryo during seed stratification was significantly inhibited by the existence of endocarp. The fastest embryo growth was resulted at $15^{\circ}C$ and an estimated optimal temperature for embryo growth was about $18^{\circ}C$. 2. There was no significant difference between the embryo growth and germination ratio of ginseng seeds which were sown in seed bed at Aug-5 without seed stratification and that of artificial seed stratification. 3. Embryo growth and germination ratio was significantly inhibited by high temperature treatment at $30^{\circ}C$ for 24 hours or respiration stress by immersing seeds in water for 10 days or more. 4. When the seed stratification was started at $10^{\circ}C$, growth of embryo in the ginseng seeds were almost stopped. But, when the seeds were stratified first at $20^{\circ}C$ for 50 days and next at $10^{\circ}C$ for 50 days, the embryo growth was significantly promoted compared with the embryo growth in the seeds which were stratified at $20^{\circ}C$ for 100 days. 5. The successive embryo growth after seed stratification was significantly accelerated at $10^{\circ}C$ but the seeds chilled at $5^{\circ}C$ for 100 days were resulted in the highest germination ratio as well as the shortest days for germination. 6. The successive embryo growth during chilling treatment and seed germination were significantly inhibited by immersing seeds in water just before chilling treatment or during chilling treatment and by interruption of chilling treatment with raising temperature to $20^{\circ}C$ for 20 days during chilling treatment. 7. The germination ratio of ginseng seeds which finished chilling treatment was highest at $10^{\circ}C$ and 62.5% was the estimated soil moisture for the best germination of ginseng seeds. The ginseng seeds were found to require high amount of oxygen for germination. 8. Only water soluble material in homogenized ginseng seeds showed a significant inhibiting effect on the seed germination of sesame, millet and soybean. Water soluble material dissolved from undehisced ginseng seeds showed stronger inhibiting effect on the seedling growth of sesame than material from dehisced ginseng seeds. Extraction temperature did not influence the inhibiting effect of the material dissolved from ginseng seeds on the seedling growth of sesame. 9. Water soluble materials dissolved from the berry pulps, leaves, fresh roots and dried roots also showed a significant inhibiting effect on the seedling growth of sesame. 10. Water soluble materials dissolved from the ginseng seeds, leaves and fresh roots showed a significant inhibiting effect on the germination of true fungi and the growth of spawn but the growth of phytopathogenic bacteria was not. 11. Among the water soluble materials dissolved from ginseng seeds, the materials of low molecular weight less than 3,000 were resulted a significant inhibiting effect on the seedling growth of sesame and the materials of high molecular weight also showed an inhibiting effect.

Changes in Circulatory and Respiratory Activities Observed on Men in an Engine Room of a Navy Ship (함정 기관실내 활동의 순환 및 호흡 기능에 대한 영향)

  • Circulatory and respiratory activities were observed in men exposed to the environment of engine room of a cruising Republic of Korea Navy ship and compared to the control values obtained in an ordinary laboratory room on land. The environment of an engine room of cruising navy ship was presumed to be a multiple stress acting on men. The environment of the engine room included high temperature $(35-42^{\circ}C)$, low relative humidity (20-38% saturation), vibration (about 7 cycles per second), rolling and pitching of ship and noises. Sixteen men were divided into two groups consisted of each 8 subjects. Subjects of sea duty group had experience of continuous on board duty averaging 3.5 years. Men of land duty group had no experience of on board activity. On land observations were made on one day prior to the boarding and leaving the port and four days after landing. In between observations in the engine room were made on the first, 5 th, 9 th, 12 th, and 14 th day of on board activity. The whole experimental period lasted for 20 days. Measurements on circulatory and respiratory parameters were at standing resting state (after 30 minutes standing in the case of on land study and 15 minutes in engine room study) and within one minute after cessation of on the spot running of which rhythm was 30/min. and lasted for 5 minutes. Oxygen consumption and pulmonary function test were done in the period of two minutes from the 3rd to 5th minutes of running. The following results were obtained. 1. Body temperature showed no change regardless of group difference or on land or on board measurements. 2. Pulse rate increased markedly after boarding the ship id both groups. Pulse rate increased from the first day on board at rest and after exercise as compared to the on land control value. This increase in pulse rate was more marked after exercise. Sea duty group showed less increase in pulse rate at rest than the land duty group. Standing and resting pulse rate of sea duty group on lam was 81 and increased to 87 at the 5th day on board and remained smaller than the land duty group throughout the period on board. Control standing and resting pulse rate of land duty group on land was 76 and reached 89 at the 9th day on board and thereafter decreased a little. Pulse rate of land duty group at rest on board remained greater than that of sea duty group throughout the period on board. 3. Systolic blood pressure of sea duty group increased after boarding the ship and remained higher than the control value on land. In the land duty group, however, systolic blood pressure decreased during the period on board the ship. Diastolic blood pressure decreased in both groups. 4. Resting breathing rate of land duty group increased and remained higher than the control value on land. In sea duty group, however, resting breathing rate showed a transient increase on the 1st day on board and decreased thereafter to the control value on land and kept the same level throughout the period of cruise. Absolute value of breathing rate in the sea duty group was greater than the land duty group both at rest and after exercise. 5. There was a lowering of breathing efficiency in both groups. Thus, increases in tidal volume and minute ventilation volume and decreases in maximum breathing capacity, vital capacity, capacity ratio and air velocity Index were observed after boarding the ship. An increase in ventilation equivalent was also observed in both groups. The lowering of breathing efficiency was more marked in the land duty group than the sea duty group. 6. Energy expediture increased in both groups during their stay on the ship and was more marked in the sea duty group. 7, Lactate concentration in venous blood at rest and after exercise increased after boarding the ship and no group difference was observed.

Effects of the Various Dietary Additives on Growth and Tolerance of Abalone Haliotis discus hannai against Stresses (다양한 사료첨가제 공급에 따른 전복의 성장과 스트레스에 대한 내성 효과)

  • Effects of the various dietary additives on growth and tolerance of abalone Haliotis discus hannai to the stresses were determined in the 16-week feeding trial. Seventy juvenile (an initial body weight of 4.2 g) abalone per container were randomly distributed into 21, 50 L plastic rectangular containers each. The six kinds of experimental diets were prepared: control (CON) with no additive, by-product of green tea (BPG), extract of figs (EF), extract of green tea (EG), commercially available product of Hearok (PH), and Haematococcus (HC). In addition, dry sea tangle (ST) was prepared to compare the efficiency of the experimental diets. Fishmeal, soybean meal and shrimp head meal were used as the protein source, and dextrin, sea tangle powder and wheat flour, and soybean oil and fish oil were used as the carbohydrate and lipid sources, respectively in the experimental diets. The experimental diets were fed to abalone once a day at a satiation level with a little leftover. The feeding trial lasted for 16 weeks. At the end of the 16-week feeding trial, abalone was exposed to the different types of stresses (air exposure, and sudden changes of rearing temperature and salinity). Survival of abalone fed the sea tangle was highest. However, weight gain of abalone fed the EF, EG and PH diets was significantly (P<0.05) higher than that of abalone fed the BPG diet or dry sea tangle. Shell length of abalone fed the all experimental diets was significantly (P<0.05) higher than that of abalone fed the dry sea tangle. Accumulated mortality of abalone fed the sea tangle was low when exposed to the different types of stresses. Also, relatively low mortality was achieved in abalone fed the HC and EF diets. In considering these results, it can be concluded that the various sources of additives is effective to improve production of abalone, and Haematococcus and extract of figs can be considered as dietary additives to improve resistance of abalone against the different types of stresses.

Low temperature plasma deposition of microcrystalline silicon thin films for active matrix displays: opportunities and challenges

  • The spectacular development of AMLCDs, been made possible by a-Si:H technology, still faces two major drawbacks due to the intrinsic structure of a-Si:H, namely a low mobility and most important a shift of the transfer characteristics of the TFTs when submitted to bias stress. This has lead to strong research in the crystallization of a-Si:H films by laser and furnace annealing to produce polycrystalline silicon TFTs. While these devices show improved mobility and stability, they suffer from uniformity over large areas and increased cost. In the last decade we have focused on microcrystalline silicon (${\mu}c$-Si:H) for bottom gate TFTs, which can hopefully meet all the requirements for mass production of large area AMOLED displays [1,2]. In this presentation we will focus on the transfer of a deposition process based on the use of $SiF_4$-Ar-$H_2$ mixtures from a small area research laboratory reactor into an industrial gen 1 AKT reactor. We will first discuss on the optimization of the process conditions leading to fully crystallized films without any amorphous incubation layer, suitable for bottom gate TFTS, as well as on the use of plasma diagnostics to increase the deposition rate up to 0.5 nm/s [3]. The use of silicon nanocrystals appears as an elegant way to circumvent the opposite requirements of a high deposition rate and a fully crystallized interface [4]. The optimized process conditions are transferred to large area substrates in an industrial environment, on which some process adjustment was required to reproduce the material properties achieved in the laboratory scale reactor. For optimized process conditions, the homogeneity of the optical and electronic properties of the ${\mu}c$-Si:H films deposited on $300{\times}400\;mm$ substrates was checked by a set of complementary techniques. Spectroscopic ellipsometry, Raman spectroscopy, dark conductivity, time resolved microwave conductivity and hydrogen evolution measurements allowed demonstrating an excellent homogeneity in the structure and transport properties of the films. On the basis of these results, optimized process conditions were applied to TFTs, for which both bottom gate and top gate structures were studied aiming to achieve characteristics suitable for driving AMOLED displays. Results on the homogeneity of the TFT characteristics over the large area substrates and stability will be presented, as well as their application as a backplane for an AMOLED display.
