Quartz Crystal Microbalance 시스템을 이용한 저밀도 지질단백질측정용 면역센서의 개발 (Development of Quartz Crystal Microbalance-Based Immunosensor for the Determination of Low-Density Lipoprotein)

  • Immunosensor for the determination of LDL(Low-Density Lipoprotein), a good indicator for the diagnosis of atherosclerosis and hypercholesterolemia, was developed by using quartz crystal microbalance(QCM). The immunosensor consists of flow-through cell, oscillating circuit, oscilloscope, and frequency counter. FIA(Flow Injection Analysis) was applied to the QCM system for the measurement of LDL in liquid phase. Antibody showing binding affinity against LDL was immobilized on the gold electrode of a quartz crystal by covalent coupling via polyethylenimine / glutaredehyde. LDL was injected and bound to the antibody immobilized on the QCM immunosensor. The response of the immunosensor (F0 - F1) was found to be proportional to the LDL concentration from 200 $\mu\textrm{g}$/ml to 800 $\mu\textrm{g}$/ml. Operational conditions for the operation of immunosensor were also investigated in terms of sensitivity and non-specific binding.

갈근에서 분리한 Daidzin 및 Puerarin의 사람 Low Density Lipoprotein 대한 항산화 효과 (Antioxidant Activity of Daidzin and Puerarin toward Oxidation of Human Low Density Lipoprotein)

  • 갈근(Puerariae radix)을 MeOH 추출물에서 silica gel column chromatography를 이용하여 플라보노이드 화합물인 daidzin 및 puerarin를 분리하여 low density li-poprotein의 산화에 대하여 실험하였다. 이들 플라보노이드중 daidzin의 경우 100$\mu\textrm{g}$/$m\ell$에서 puerarin은 600$\mu\textrm{g}$/$m\ell$의 농도에서 5$\mu$M Cu$^{2+}$ 매개산화 LDL에 대하여 억제 효과가 좋았다. 이때 같은 농도의 diadzin과 puerarin를 첨가한 산화 LDL의 전기영동의 이동거리는 native LDL 보다는 약간 높았으나 oxidized LDL의 대조군 보다는 이동거리가 낮았다. 또 J774 및 macrophages 유도 산화 LDL에 있어서도 100$\mu\textrm{g}$ daidzin과 600$\mu\textrm{g}$/$m\ell$ puerarin을 첨가 하였을 때 억제효과 나타났다. LDL을 5$\mu$M Cu$^{2+}$존재하에서 산화시킬 때 동일 농도의 daidzin과 puerarin 을 첨가하면 conjugated dienes의 생성이 거의 억제되었다.

Inhibitory Effects of Garlic Oil on Human Low Density Lipoprotein Oxidation

  • 본 연구에서는 동맥경화의 원인으로 알려진 사람 low density lipoprotein (LDL)에 대한 garlic oil의 산화억제 효과에 대하여 실험하였다. Garlic oil을 HPLC로 분석한 결과 diallyl disulfide, diallyltrisulfide, diallyltetrasulfide and diallyl pentasulfide 등의 유황화합물이 많이 들어 있었다. 사람 LDL을 $Cu^{2+}$ 및 macrophages 유도 oxidized LDL로 산화시킬 때 garlic oil을 20~60${\mu}g$/mL 범위에서 첨가하며 실험하였을 때 용량 의존형으로 억제되었고, garlic oil을 60 ${\mu}g$/mL 첨가하였을 때 LDL의 산화가 거의 억제되었으며, 산화 LDL의 전기영동에 의한 이동거리도 60 ${\mu}g$/mL 농도에서 거의 억제되었다. 그리고 garlic oil은 동맥의 내피세포에서도 용량 의존형으로 억제효과를 나타내었다. 또한 garlic oil은 tocopherol, ascorbic acid 보다 약간 높은 억제효과를 나타내었다. 이상의 결과에 따르면 garlic oil은 macrophage 및 내피세포에서 LDL의 항산화 효과가 있어 LDL의 산화 억제효과가 있는 것으로 나타났다.

Lipoprotein Lipase-Mediated Uptake of Glycated LDL

  • The glycation process plays an important role in accelerated atherosclerosis in diabetes, and the uptake of atherogenic lipoproteins by macrophage in the intima of the vessel wall leads to foam cell formation, an early sign of atherosclerosis. Besides the lipolytic action on the plasma triglyceride component, lipoprotein lipase (LPL) has been reported to enhance the cholesterol uptake by arterial wall cells. In this study, some properties of LPL-mediated low-density lipoprotein (LDL) uptake and the effect of LDL glycation were investigated in RAW 264.7 cell, a murine macrophage cell line. In the presence of LPL, $^{125}I$-LDL binding to RAW 264.7 cells was increased in a dose-dependent manner. At concentrations greater than $20\;{\mu}g/ml$ of LPL, LPL-mediated LDL binding was increased about 17-fold, achieving saturation. Without LPL, both very low-density lipoprotein (VLDL) and high-density lipoprotein (HDL) were ineffective in blocking the binding of $^{125}I$-LDL to Cells. However, LPL-enhanced LDL binding was inhibited about 50% by the presence of VLDL, while no significant effect was observed with HDL. Heat inactivation of LPL caused a 30% decrease of LDL binding. In the presence of LPL, the cells took up 40% of cell-bound native LDL. No significant difference was observed in cell binding between native and glycated LDL. However, the uptake of glycated LDL was significantly greater than that of native LDL, reaching to 70% of the total cell bound glycated LDL. These results indicate that LPL can cause the significant enhancement of LDL uptake by RAW 264.7 cells and the enhanced uptake of glycated LDL in the presence of LPL might play an important role in the accelerated atherogenesis in diabetic patients.

인삼 사포닌 분획이 혈청 지단백질 분포에 미치는 영향 (Effect of Saponin Fraction of Panax ginseng C.A. Meyer on Blood Serum Lipoprotein Distribution of Cholesterol Fed Rabbits)

  • 고려 인삼 뿌리에서 얻은 사포닌 분획이 고cholesterol 식이로 2주내지 4주간 사육한 토끼의 혈청 지단백질 분포에 미치는 영향을 조사하였다. 고cholesterol 식이를 인삼과 함께(시험군) 또는 단독(대조군)으로 2~4주간 투여한 토끼의 혈청 극저밀도 지단백질(VLDL)과 저밀도 지단백질(LDL)이 다같이 크게 증가하는 반면 고밀도 지단백질(HDL)의 농도는 저하 하였다. 그러나 VLDL과 LDL 농도의 증가율이나 HDL 농도의 저하율은 인삼을 투여한 토끼군에서 현저히 적었으며 인삼사포닌의 고cholesterol 혈증 개선작용이 VLDL의 LDL 농도를 낮추고 HDL의 농도를 높이기 때문인 것으로 생각된다.

  • 최근 지방 섭추의 증가에 따른 혈관계 질병이 증가 추세에 있다. 이러한 동맥경화 및 고지질의 질병은 지질 단백질(lipoprotein)과 관련하여, LDL 및 산화 LDL의 특성을 중심으로 고찰하였다. 인체의 혈장에 함유된 LDL 함량의 증가는, 동맥경화와 직결되는 것을 의미하며, 이러한 LDL은 매우 hydrophobic한 특성을 가진 550Kd의 단일 polypeptide인 Apo B-100라는 단백질이, 지질성분인 triglyceide, phospholipid 및 cholesterol와 결합되어 있다. 최근 이러한 LDL은 산화(oxidation)되는 경우, 정상적인 LDL-receptor pathway를 따르지 않고, macrophang와 결합하므로서, foarn cell을 형성하여 동백경화가 촉진되는 것으로 알려지고 있다.

사람 LDL의 지질과산화에 의한 geraniin의 항산화 효과 (Antioxidative Role of Geraniin in Lipid Peroxidation of Human LDL)

  • 본 연구는 동맥 경화의 원인으로 알려진 사람 oxidized low density lipoprotein (LDL)에 대한 geraniin의 산화 억제 효과에 대하여 실험하였다. 사람 LDL을 C $u^{2+}$유도 LDL로 산화 시킬 때 50와 100 $\mu\textrm{g}$/ml 농도의 geraniin를 첨가하여 TBARS을 측정한 결과 LDL에 대한 항산화가 높았으며 용량 의존형으로 나타났다. Geraniin를 20-100 $\mu\textrm{g}$/ml의 농도를 조절하여 전기 영동에 의한 이동상을 조사한 결과 100 $\mu\textrm{g}$/ml geraniin의 농도에서 거의 완전한 억제 효과를 보였다. 사람 LDL에 C $u^{2+}$로 유도하여 LDL를 산화시킬때 conjugated diene를 보면 geraniin를 100 $\mu\textrm{g}$/ml 첨가하였을 때 억제 효과가 높았다. 또한 geraniin은 동맥의 내피세포에 서도 그 농도에 따라 억제효과를 나타내었다. 그리고 phorbol myristate acetate를 처리한 macrophage 유도활성 산소의 소거 효과는 geraniin의 농도가 100 $\mu\textrm{g}$/ml일때 거의 소거하였다. 이상의 결과로 보아 geraniin는 $\alpha$-tocopherol, ascorbir acid 및 합성 항산화제인 probucol과 거의 비슷한 항산화 활성이 있어 동맥 경화의 예방에 효과적이라는 결론을 얻었다.

Effects of Wet Cupping (Al-Hijamah) on Cholesterol in a Sudanese Population

  • Background: Wet cupping (WC) is an efficient and cost-effective technique for removing metabolic waste from the bloodstream via the skin. The study aimed to examine the effect of WC on cholesterol levels including total cholesterol (TC), high-density lipoprotein cholesterol (HDL-C), and low-density lipoprotein cholesterol (LDL-C) in a Sudanese population. Methods: In this prospective cross-sectional study, 30 participants undergoing regular WC therapy were enrolled. Blood samples collected twice: pre-WC therapy (case group) and 10-14 days afterward (controls). Results: Of the participants, 56.67% were male and 43.33% were female, aged 24-69. Pre-WC TC and LDL-C levels were significantly higher than the post-WC control group (p = 0.001). Conversely, HDL-C levels decreased significantly in the pre-WC cases compared to controls (p = 0.001). No significant sex-based difference in mean cholesterol levels (p > 0.05). Conclusions: After WC, males and females experienced significant reductions in TC and LDL-C, and significant increase in HDL-C.

Functional Properties of Modified Low Density Lipoprotein and Degradation of Modified LDL by Human Monocyte-Macrophages

  • Human plasma low density lipoprotein(LDL) is the main carrier for cholesterol, and recent studies suggest the normal LDL can be readily oxidized by free radical and not interact with LDL receptor. Lipoprotein pariticles are consisted of lipid andprotein, and fatty acids of lipoproteins are prone to oxidation. LDL particles readily undergo oxidative modification by copper. From the results, oxidized LDL altered its biological properties. A marked increase in the electrophoretic mobility of LDl on agarose gel indicated that negative surface charge of the LDL particles was increased. Also, the results from the HPLC showed that oxidized LDL was degraded into several polypeptides nonenzymatically. Degradation tests which measured the amount of 5-IAF labelled oxidized LDL were carried out by monocyte and hepatocyte cell culture. Hepatocyte cell culture of modified LDL did not show consistent pattern. However, binding rate of modified LDL with HMDM(human monocyte derived macrophage) was enhanced with oxidation, but was retarded by addition of antioxidants(hyaluronic acid, vitamin A, vitamin E). Also comparisons of oxidized-LDL, acetyl-LDL and MDA-LDL showed significant differences in the chemical properteis and binding affinity to HMDM. Thus, modificaition of normal LDL altered its biological properties.
