Growth signaling and longevity in mouse models

  • Reduction of insulin/insulin-like growth factor 1 (IGF1) signaling (IIS) extends the lifespan of various species. So far, several longevity mouse models have been developed containing mutations related to growth signaling deficiency by targeting growth hormone (GH), IGF1, IGF1 receptor, insulin receptor, and insulin receptor substrate. In addition, p70 ribosomal protein S6 kinase 1 (S6K1) knockout leads to lifespan extension. S6K1 encodes an important kinase in the regulation of cell growth. S6K1 is regulated by mechanistic target of rapamycin (mTOR) complex 1. The v-myc myelocytomatosis viral oncogene homolog (MYC)-deficient mice also exhibits a longevity phenotype. The gene expression profiles of these mice models have been measured to identify their longevity mechanisms. Here, we summarize our knowledge of long-lived mouse models related to growth and discuss phenotypic characteristics, including organ-specific gene expression patterns.

패션 제품의 수명 연장을 위한 디자인 가이드라인 - 여성 정장을 중심으로 - (Design guidelines for extending the longevity of fashion products - Focused on women's formal wear -)

  • The environment has increasingly attracted attention and fashion brands need to use new growth models by developing eco-friendly products, along with the drastic climate change. This study drew design guidelines from the factors of clothing disposal and reuse to propose ways to extend the longevity of clothing. It sets the design goals for the longevity extension of clothing as flexibility, originality, durability, and adjustability and drew a specific design guideline. The design methods used to achieve such goals are as follows. First, the design that is flexible in terms of physical changes needs to increase its activity and to be changeable, by applying pleats, rubber bands and elastic materials to the parts with many physical changes and movements. Second, it is necessary to reinforce the brand identity, create design that is flexible in terms of fashion and design very rare and attractive products, for the goal of original design beyond fashion. Third, it is necessary to increase the quality of clothing and improve the durability which can be decreased by washing and wearing. Fourth, it is necessary to create the design that can produce various styles, preserve the state of clothing and maintain its hygienic conditions by using removable detailed designs, shape-transformation designs and the designs which can be adjusted to climate changes and states, for the goal of adjustable design with better functionality. The findings provide ideas for fashion experts to pay more attention to the extending the longevity of clothing products and to develop eco-friendly designs and strategies.

코호트 사망률을 이용한 장수채권 가격산출 (Longevity Bond Pricing by a Cohort-based Stochastic Mortality)

  • 본 논문은 평균회귀 2요인 사망률 모형에 코호트 효과를 반영한 개선된 확률론적 사망률 모형을 제시한다. 한국 남자의 사망률 자료를 바탕으로 가중평균최소제곱법과 메트로폴리스 알고리듬을 이용하여 사망률 모형을 추정한 결과 코호트 효과를 반영하는 것이 모형 적합도를 향상시킴을 발견하였다. 국민연금공단과 같은 연금사업자가 자신의 장수위험을 금융시장에 순차적으로 전가하는 수단으로서 옵션방식 이자지급 장수채권의 활용을 제안하고 발행채권의 가격 산출방법을 제시하는 것이 본 논문이 기여하는 점이다. 특히 생존지수에 의해 이자지급 현금흐름이 결정되는 장수채권 가격산출을 위하여 코호트 효과가 매우 중요한 요소임을 강조하였다.

  • 할미꽃 신품종 '연홍'은 P. davurica와 P. koreana를 종간교잡하여 2003년에 전라남도농업기술원에서 육성되었고 유전적으로 자가불화합성의 특성을 가지고 있다. '연홍'의 화색은 진분홍이고 약은 진노랑, 주두는 진분홍, 꽃잎은 6매이며 엽은 녹색이다. 한편 개화특성은 년 2회 개화성이다. 주당 화경은 26.5개, 화경장과 절화장은 각각 58.6 cm와 46.8 cm, 포의 폭은 11.5 cm, 엽장은 34.4 cm이다. 꽃과 포의 절화수명은 각각 8.4일과 34일로 절화용으로 이용이 가능하다.

Autophagy and Longevity

  • Autophagy is an evolutionally conserved cytoplasmic degradation system in which varieties of materials are sequestered by a double membrane structure, autophagosome, and delivered to the lysosomes for the degradation. Due to the wide varieties of targets, autophagic activity is essential for cellular homeostasis. Recent genetic evidence indicates that autophagy has a crucial role in the regulation of animal lifespan. Basal level of autophagic activity is elevated in many longevity paradigms and the activity is required for lifespan extension. In most cases, genes involved in autophagy and lysosomal function are induced by several transcription factors including HLH-30/TFEB, PHA-4/FOXA and MML-1/Mondo in long-lived animals. Pharmacological treatments have been shown to extend lifespan through activation of autophagy, indicating autophagy could be a potential and promising target to modulate animal lifespan. Here we summarize recent progress regarding the role of autophagy in lifespan regulation.

Caloric restriction and its mimetics

  • Caloric restriction is the most reliable intervention to prevent age-related disorders and extend lifespan. The reduction of calories by 10-30% compared to an ad libitum diet is known to extend the longevity of various species from yeast to rodents. The underlying mechanisms by which the benefits of caloric restriction occur have not yet been clearly defined. However, many studies are being conducted in an attempt to elucidate these mechanisms, and there are indications that the benefits of caloric restriction are related to alteration of the metabolic rate and the accumulation of reactive oxygen species. During molecular signaling, insulin/insulin-like growth factor signaling, target of rapamycin pathway, adenosine monophosphate activated protein kinase signaling, and Sirtuin are focused as underlying pathways that mediate the benefits of caloric restriction. Here, we will review the current status of caloric restriction.

Changes of physicochemical properties of seed longevity from a cross between japonica and weed rice

  • In previously study, we evaluated seed germination for longevity derived from a cross between 'Ilmi' and 'Dharial', a weed rice collected in Bangladesh. The strong germination trait originated from 'Dharial' was incorporated into 'Ilmi', through backcross method. The germination ratio was evaluated after two years of room temperature storage conditions. A high germination ratio of 80.5% in donor plant of 'Dharial' and 77.3% in an introgression lines was observed based on the two years of storage while the recurrent japonica cultivars, 'Ilmi' was failed in germination. In this study, we investigate changes in physicochemical properties of 'Ilmi' and introgression lines (ILs) stored at room temperature. We analyzed germination rate, texture of cooked rice, toyo glossiness value, pasting properties, amylose content, protein content and ${\alpha}-amylase$ content of 'Ilmi' and 5 introgressions every 4 months on the room temperature condition. Seed germinations were decreased by storage periods. Three ILs germination rate was slowly decreased more than 'Ilmi' and 2 another ILs after 4 months. Toyo glossiness value of 'Ilmi' and three ILs were no difference, but, 2 ILs gradually decreased every 4 months at storage periods. Pasting properties were affected by storage temperatures and periods of 'Ilmi' and ILs. The increase at breakdown was observed but setback was decreased by storage periods. Amylose content and protein content were no significant difference at storage periods, respectively. The ${\alpha}-amylase$ content was gradually increased during the storage periods. The introgression line could be useful to increase longevity and maintain quality during storage of japonica rice seed.

  • Learning from the failure of mechanical systems is a necessity, given that it is the understanding of how and why things fail that generates effective redesign. This subsequently enables the technology that surrounds us to become more reliable, safer, and more economical by extending component life and minimizing the wasteful decisions made to replace systems that am either sound for continued operation could be easily repaired. Considerations for cost-effective decision making, so as to promote healthy machinery, equipment, and structures, are discussed in terms of learning from failure analysis, improving via reliability engineering, and achieving longevity through integrated diagnostics.

  • Due to the fact that there are low birth rate and longer life longevity, schools have been shutting down and converted into welfare facilities. However, most of researches about utilizing closed down school are focused on case researches or architectural planning and design. Moreover, majority of existing welfare centers for the aged are not designed with the flow of human traffic in mind, which means there is no consideration of physical or psychological characteristics of elderly people. That fact leads to increasing number of shutting down welfare facilities. Therefore, this thesis will present the necessity of effective space allocation, extension and reconstruction for residents in aged care by analyzing space using index of space syntax with depth-map software.

Lindera obtusiloba Extends Lifespan of Caenorhabditis elegans

  • Lindera obtusiloba has been widely used as a traditional medicine for the treatment of lots of diseases, including abdominal pain, bruise, and hepatocirrhosis. Here in this study, we elucidated the lifespan-extending effect of methanolic extract of Lindera obtusiloba (MLO) using Caenorhabditis elegans model system. We found that MLO has potent lifespan extension activities under normal culture condition. Then, we determined the protective effects of MLO on the stress conditions such as osmotic, thermal and oxidative stress. To reveal possible mechanism of MLO-mediated lifespan, we further investigated the effect of MLO on the antioxidant enzyme activities and intracellular ROS levels. Our results demonstrated that superoxide dismutase and catalase activities were significantly up-regulated by MLO treatment, resulted in reduced intracellular ROS levels. In this work, we also tested whether MLO-mediated longevity activity was associated with aging-related factors such as food intake and growth. Our data revealed that both of pharyngeal pumping rate and body length were significantly shifted by MLO treatment, indicating these factors were involved in MLO's lifespan-extension effects. Although MLO induces reduction in food intake, the body movement of MLO-fed aged worms was not decreased, compared to untreated control worms, indicating MLO might extend lifespan without affecting healthspan.
