Development of a Novel Long-Range 16S rRNA Universal Primer Set for Metagenomic Analysis of Gastrointestinal Microbiota in Newborn Infants

  • Metagenomic analysis of the human intestinal microbiota has extended our understanding of the role of these bacteria in improving human intestinal health; however, a number of reports have shown that current total fecal DNA extraction methods and 16S rRNA universal primer sets could affect the species coverage and resolution of these analyses. Here, we improved the extraction method for total DNA from human fecal samples by optimization of the lysis buffer, boiling time (10 min), and bead-beating time (0 min). In addition, we developed a new long-range 16S rRNA universal PCR primer set targeting the V6 to V9 regions with a 580 bp DNA product length. This new 16S rRNA primer set was evaluated by comparison with two previously developed 16S rRNA universal primer sets and showed high species coverage and resolution. The optimized total fecal DNA extraction method and newly designed long-range 16S rRNA universal primer set will be useful for the highly accurate metagenomic analysis of adult and infant intestinal microbiota with minimization of any bias.

  • We consider a Step Life Testing which is deviced for a two-component serial system with the considerably long life time. In the modelling stage we discuss the bivariate exponential distribution suggested by Block and Basu as the bivariate survival function for the two-component system, and develope the cumulative exposure model introduced by Nelson so that it can be used under the bivariate function. We consider inference on the component life time when the components are at work in the system by combining the information from system life test and that from the component tests carried out separately under the controlled environment. In data analysis, maximum likelihood estimators are discussed with the initial value obtained by an weighted least square method. Finally we discuss the optimal time for changing the stress in the simple step stress life testing.

Experimental Study on the Diagnosis and Failure Prediction for Long-term Performance of ESP to Optimize Operation in Oil and Gas Wells (유·가스정 최적 운영을 위한 ESP의 장기 성능 진단 및 고장 예측 실험 연구)

  • In general, electric submersible pumps (ESPs), which have an average life of 1.0 to 1.5 years, experience a decrease in performance and a reduction in life of the pump depending on oil and gas reservoir characteristics and operating conditions in wells. As the result, the failure of ESP causes high well workover costs due to retrieval and installation, and additional costs due to shut down. In this study, a flow loop system was designed and established to predict the life of ESP in long­term operation of oil and gas wells, and the life cycle data of ESP from the time of installation to the time of failure was acquired and analyzed. Among the data acquired from the system, flow rate, inlet and outlet temperature and pressure, and the data of the vibrator installed on the outside of ESP were analyzed, and then the performance status according to long-term operation was classified into five stages: normal, advice I, advice II, maintenance, and failed. Through the experiments, it was found that there was a difference in the data trend by stage during the long­term operation of the ESP, and then the condition of the ESP was diagnosed and the failure of the pump was predicted according to the operating time. The results derived from this study can be used to develop a failure prediction program and data analysis algorithm for monitoring the condition of ESPs operated in oil and gas wells.

  • High temperature materials in service are subjected to mechanical damage due to operating load and metallurgical damage due to operating temperature. Therefore, when designing or assessing life of high temperature components, both factors must be considered. In this paper, the effect of tensile hold time on high temperature fatigue crack growth and long term prior thermal aging heat treatment on creep rupture behavior were investigated using STS 316L and STS 316 austenitic stainless steels, which are widely used for high temperature components like in automotive exhaust and piping systems. In high temperature fatigue crack growth tests using STS 316L, as tensile hold time increased, crack growth rate decreased in relatively short tensile hold time region. In long term aged specimens, cavity type microcracks have been observed at the interface of grain boundary and coarsened carbide.

Study on the Development of High-efficiency, Long-life LED Fog Lamps for the Used Car Market

  • LED lighting,considered to be a new growth industry, has attracted a great deal of attention due to its higher illumination and longer life time than existing light sources. In this study, high-efficiency and long-life LED fog lamps for automobiles were developed, which can substitute the existing 27 W halogen fog lamps for a used car market. For this purpose, the number of LED modules, the body, heat sink, and the output of the fog lamp were first optimized through a numerical analysis. Then, a 10 W-class LED fog lamp was prototyped based on the optimized numerical model, and the performance of the fog lamp was successfully verified through the experiments.

  • Patients undergoing radiation therapy for head and neck cancer (HNC) experience significant early and long-term side effects. The likelihood and severity of complications depends on a number of factors, including the total dose of radiation delivered, over what time it was delivered and what parts of the head and neck received radiation. Late side effects include: permanent loss of saliva; osteoradionecrosis; radiation recall myositis, pharyngoesophageal stenosis; dental caries; oral cavity necrosis; fibrosis; impaired wound healing; skin changes and skin cancer; lymphedema; hypothyroidism, hyperparathyroidism, lightheadedness, dizziness and headaches; secondary cancer; and eye, ear, neurological and neck structures damage. Patients who undergo radiotherapy for nasopharyngeal carcinoma tend to suffer from chronic sinusitis. These side effects present difficult challenges to the patients and their caregivers and require life-long strategies to alleviate their deleterious effect on basic life functions and on the quality of life. This review presents these side effects and their management.

Storing Digital Information in Long-Read DNA

  • There is urgent need for effective and cost-efficient data storage, as the worldwide requirement for data storage is rapidly growing. DNA has introduced a new tool for storing digital information. Recent studies have successfully stored digital information, such as text and gif animation. Previous studies tackled technical hurdles due to errors from DNA synthesis and sequencing. Studies also have focused on a strategy that makes use of 100-150-bp read sizes in both synthesis and sequencing. In this paper, we a suggest novel data encoding/decoding scheme that makes use of long-read DNA (~1,000 bp). This enables accurate recovery of stored digital information with a smaller number of reads than the previous approach. Also, this approach reduces sequencing time.

A Study on Patterns on Korean Medical Containers (한의약기(韓醫藥器)문양 연구)

  • Patterns are symbolic expressions of life style or sentiment of people as a product of culture. Furthermore, it is a type of language that the shape or implication does not change much over time. Patterns shown on the Korean medical containers are related to human life and health that they are not very diverse and splendid but closed used in real life. The medical containers include the alcohol container or tea-ware in broad sense, but under this study, it would be handled only for the Korean medical containers directly used to protect human health and treatment of diseases. They would be the Medicine Mortar and Pestle that is used to grind the herbal medical ingredients, pots that hold the liquid decoction, containers to keep the medical ingredients in general conceptual use for medical purposes. The patterns inscribed are classified in animal pattern, plant pattern or alphabet pattern. Turtle, crane, ginseng, and life patterns have the contemplation to pray for long life without disease or good fortune, while plum pattern, dragon pattern and the like are used in the medical containers used mainly in royal families with the social status and dignity shown. As such, patterns have not only ornament elements but also the symbolic implication to represent long life without illness for human. It provides the basic data of medical wisdom contained in the pattern and willingness of human to protect life from illness. On the basis of such research, it would require ensuing studies to make comparison and analysis with the medical containers with the patterns in the adjacent countries in the days to come.

  • Organic light emitting diode(OLED) has been developed fast from 1963 when electric light emitting phenomenon was discovered. PMOLED(passive matrix OLED) is producted earlier than AMOLED(active matrix OLED). PMOLED is mainly mounted at sub display, but AMOLED is mounted at main display. Nowadays AMOLED is expanded to PMP(portable multimedia players), navigation and TV market. Even thought OLED's market is opening to many applications, OLED's life is worried until now. If we know about OLED's real life, we need time to test so much time over 20,000hrs. Realistically, there is difficult to test such as long time with products from the information-technology sector having a short life cycle. In this paper, we study about OLED's accelerated test to reduce life test by current. We can design OLED's accelerated life model by the result of test. The model consists of design variables like ratio of light emitting, organic material structure, condition of aging, etc. In conclusion, this model can be applied to study about organic material, machine and manufacturing process etc, and also it's possible to develop a method of manufacturing process & materials, so we need to study on the subject of this paper continuously.

  • The study objected to the 120 persons out of 360 persons who have registered under the association of the dental technicians in Pusan to grasp contents about improvement of life long education of the present day of November, 1987, and its results, which we have acquired by this study were as follows : 1. A sense of satisfaction toward the system of life long education appeared as unsatisfaction and its unsatisfaction decreased from the rate of member's attendance or exam. marks, the contents of education, and place of education. 2. The problem of the exam. mark which derived by the presence by the substitute appeared which 46.7 percent(56 persons), lack of opportunity for the recompletion at branch association appeared which 19.2%(23 persons), given 4. exam. marks at the over all scientific contest was appeared 14.2 percent(17 persons), relation which legal treatment when ones didn't completed exam. mark was appeared 10.8 percent(13 persons) and lastly, appeared 8.3 percent (10 persons) with time of delivery of the exam mark card. 3. The frequency of the opening for the life long education which related with the rate of members attendance was fixed as nowadays two times per annually, but they perferred the opening per academic blanch once out of the firest half year and the latter half of the year and, they liked the openings at the first week or second week and preferred the holding on saturdays and sundays. 4. The subject of the life long education have appeared the opinion that the thesis was lop-sided toward the allocation of time schedule of lecture appeared the opinion of which they perferred the rate of an hour of cultural subject and two hours of major studies, and invitation for an instructor they preferred one dentist, one member, one dean for the cultural subject, and there are lots of opinions which they wished display of materials. 5. A sence of satisfaction about the place they perferred was decreased such as ; an auditorium concerned of the colleges was 46.7 Percent(56 Persons), hotels 26.7 Percent(32 Persons), an auditorium of the general hospital 20 percent(24 persons) 6. And lastly, there are lots of opinions who have required the necessities to constitute for the academic branch.
