Gene expression of two glyceraldehyde-3-phosphate dehydrogenase (GAPDH) paralogs was examined during Edwardsiella tarda challenge and heavy metal exposures in mud loach (Misgurnus mizolepis; Cypriniformes) kidney and spleen. Transcription of the two mud loach GAPDH paralogs (mlGAPDH-1 and mlGAPDH-2) was significantly modulated by these stimulatory challenges in an isoform-dependent manner. Based on the real-time RT-PCR analysis, the mlGAPDH-2 transcripts were more preferentially induced by E. tarda challenge, whereas the mlGAPDH-1 transcripts were proven to show more inducibility in response to heavy metal exposure using Cd, Cu, Mn and Zn at $5{\mu}M$. Their isoform-specific response patterns were closely in accordance with the TF binding profiles in promoter and intron-1 of the two mlGAPDH isoforms, in which the mlGAPDH-2 has more binding sites for immune-related transcription factors than mlGAPDH-1 while the mlGAPDH-1 possesses exclusively metal responsive elements in its intron. Collectively, the mlGAPDHs are potentially involved in cellular pathways independent of glycolysis and the two GAPDH paralogs might undergo functional diversification or subfunctionalization at least at the transcription level.
This study was undertaken to determine the effects of HCG and trout pitutary on sexual maturation and ovulation in the Korean loach, Misgurnus anguillicaudatus. Sexual maturation was observed in the fish treated with 250+125 IU HCG within 2-4 days following injections. Total ovary weight and GSI were significantly greater in the hormone-treated fish than in the control fish. Furthermore, in the hormone-treated fish body weight increased by about 4-9 percent in 2 days. The water content of the ovulated eggs of the fish treated with 250+125 IU HCG was also greater than that in the control. Changes in both macroscopic & microscopic appearnce of Korean loach ovaries are describled, as well as changes in the gonadosomatic index. Oocytes pass through seven cytologically changes in the relative stages. By examining these stages, it was found that oogenesis occures in two broad phases. The previtellogenic phasebegins as a new oogonia continues to arise following spawning. These rapidly develop into early perinucleolus oocytes, which in turn develop into resting stage oocytes. Most oocytes remain in this stage when they develop into late perinucleolus oocytes. The vitellogenic phase begins as these late perinucleolus oocytes become transformed into early maturing oocytes through the accumulation of yolk. The cytoplasm completely fills with yolk as oocytes reach the late maturing stage. Shortly before spawning the final hyaline stage of developing is reached. Changes in the microscopic appearances of the ovaries were well correlated with changes in both gonadosomatic index and macroscopic appearance.
On September in 2010, myxosporidian parasites were found on the fins of Chinese muddy loach, Misgurnus mizolepis, Several cysts of approximately 2 mm diameter were observed on the fins. Numerous spores were found in the cyst. The total length of the spore was 13.0 to 15.8 ${\mu}m$, and the width of spore was 6.0 to 7.5 ${\mu}m$. As the results, the myxosporidians were finally identified as Thelohanellus misgurni (Kudo, 1919) on the basis of the presence of characteristic cysts and size of spores.
This study was carried out to investigate the histomorphological changes of ovaries obtained from 100 of 1-year-old female Korean loach(misgurnus anguillicaudatus). The light microscopic and ultrastructural changes ofooplasm and follicular membranes of oocytes, were observed by lightand transmission electron microscope during the reproductive cycle. All data were collected from November in 1991 to May in 1992. The results obtained in this study were as follows: The size of the nucleoli and number of the yolk granules increased as the oocytes grown. Yolk granules were loosely deposited in the oocytes as crystalline granules. Due to the presence of large early and late maturing oocytes, their ovaries were enlarged, transparent, granular and yellowish in color. The lattice was broken down at hydration, leaving the egg transparent. As the percentages of fish in LMO and RO stage increased from March to April, mean gonadosomatic index(GSI) values(18.49%) increased. Zona radiata change a squamous into cuboid shape in EMO stage. Processes from the granulosa cells and from the oocyte, microvilli grow and make contact with other in the pore canals of the zona radiata during vitellogenesis, but are withdrawn as the zona radiata becomes more compact and devoid of pore canals during oocyte maturation. Seasonal changes in the microscopic appearance of the ovaries were well correlated with those in both GSI and macroscopic appearance.
The authors have studied the distribution of alkaline phosphatase in the pharynx, esophagus, stomach (or intestinal bulb), anterior and posterior portions of intestine of three kinds of fishes. The results obtained are as follows: 1. Alkaline phosphatase activity of basal cells in the pharyngeal epithelium of loach and snakehead fish showed moderately positive reaction, and basal cells in the esophagel epithelium of loach and eel showed also moderately positive reaction. 2. Goblet cells of pharynx, esophagus, intestinal bulb and intestinal mucosa, and gastric glandular cells of the above fishes showed negative reaction for alkaline phosphatase. 3. Strongly positive reaction for alkaline phosphatase was observed at both intestinal bulb and the free border of intestinal epithelium, but weak positive reaction at the free border of posterior portion of loach intestine.
This stduy was carried out to obtain basic information available on ovulation, spawning, fertilization rate, hatching rate, and deformity rate after hCG injection in cyprinid loach, Misgurnus mizolepis. The results obtained in these experiments were as follows: 1. Ovulation and spawning occurred simultaneously and spawing was completed within 1 hour after ovulation. 2. More than 80% of fertilization rates appeared within 12 hours at 21$^{\circ}C$, 8 hours at $25^{\circ}C$, and 4 hours at 29$^{\circ}C$, respectively, following the onset of spawing. Afterwards, the fertilization rates of released eggs sharply decreased in three different water temperatures. 3. More than 70% of hatching rates appeared within 8 hours at 21$^{\circ}C$, 6 hours at $25^{\circ}C$, and 2 hours at 29$^{\circ}C$, respectively, following the onset of spawing. Afterwards, hatching rates of spawned eggs abruptly decreased in three different water temperatures. 4. The deformity rates of hatched larvae were high at $25^{\circ}C$, 8 hours following the onset of spawing. 5. Based on the developmental ability of oocytes, the optimum time of fertilization was 4 hours (stage 5) following the onset of spawing.

A novel Tc1-like transposable element has been identified as a new DNA transposon in the mud loach, Misgurnus mizolepis. The M. mizolepis Tc1-like transposon (MMTS) is comprised of inverted terminal repeats and a single gene that codes Tc1-like transposase. The deduced amino acid sequence of the transposase-encoding region of MMTS transposon contains motifs including DDE motif, which was previously recognized in other Tc1-like transposons. However, putative MMTS transposase has only 34-37% identity with well-known Tc1, PPTN, and S elements at the amino acid level. In dot-hybridization analysis used to measure the copy numbers of the MMTS transposon in genomes of the mud loach, it was shown that the MMTS transposon is present at about $3.36{\times}10^4$ copies per $2{\times}10^9$ bp, and accounts for approximately 0.027% of the mud loach genome. Here, we also describe novel MMTS-like transposons from the genomes of carp-like fishes, flatfish species, and cichlid fishes, which bear conserved inverted repeats flanking an apparently intact transposase gene. Additionally, BLAST searches and phylogenetic analysis indicated that MMTS-like transposons evolved uniquely in fishes, and comprise a new subfamily of Tc1-like transposons, with only modest similarity to Drosophila melanogaster (foldback element FB4, HB2, HB1), Xenopus laevis, Xenopus tropicalis, and Anopheles gambiae (Frisky).
Monogeneans(Phylum Platyhelminthes) have been known as common parasites onto the skin and gills of cultured freshwater fishes. Infestation with these parasites result in a great loss in aquaculture industry. Some classification studies on these parasites have mostly been conducted in Japan but rarely in Korea. For the purpose of classifying monogenean parasites infesting two Korean freshwater fishes, carp(Cyprinus carpio) and loach(Misgurnus anguillicaudatus), samples captured from March 1998 to April 2000 were examined. Here we report that three different species of Monogeneans, Diplozoon nipponicum, Dactylogyrus extensus from carp and Gyrodactylus paralatus from loach, respectively. Of these the latter two parasites have never been identified in Korea.
In this study, we measured the morphometric and histological changes in the cyprinid loach, Misgurnus anguillicaudatus, during the early period of growth. Eyes, yolk length, yolk height, and yolk volume of the larva decreased for 16 days post hatching (DPH) (P<0.05). During 60 DPH (P>0.05), the most anterior extension of the head ${\times}$ the posterior end of the supraoccipital, the most anterior extension of the head ${\times}$ the origin of the dorsal fin, the most anterior extension of the head ${\times}$ the origin of the pectoral fin, the posterior end of the supraoccipital ${\times}$ the origin of the pelvic fin, and the origin of the dorsal fin ${\times}$ the ventral origin of the caudal fin gradually decreased, whereas the most anterior extension of the head ${\times}$ the dorsal origin of the caudal fin, the origin of the dorsal fin ${\times}$ the origin of the anal fin, the origin of the dorsal fin ${\times}$ the origin of the pectoral fin, and the insertion of the dorsal fin ${\times}$ the origin of the pelvic fin gradually increased (P<0.05). In the cyprinid loach, the retina is composed of six layers: the epithelial layer, ganglion cell layer, inner nuclear layer, inner plexiform layer, outer limiting membrane, and rod and cone layer (RCL). After hatching, part of the RCL gradually increased in density. The kidney and midgut epithelium were already formed in the cyprinid loach just after hatching and grew gradually in subsequent days.
We designed a 16-week feeding experiment to test the effects of dietary supplementation with distillers' dried grain (DDG) on the growth, feed utilization, and body composition of juvenile muddy loach Misgurnus anguillicaudatus. Seven isonitrogenous and isocaloric diets were formulated to contain 0% DDG (Control); 10%, 20%, and 30 % DDG from rice (diets RM10, RM20 and RM30); and 10%, 20%, and 30% DDG from rice and wheat flour (diets RWM10, RWM20 and RWM30), respectively. Three replicate groups of juvenile muddy loach averaging $1.5{\pm}0.10$ g were fed one of the diets to satiation twice daily. Survival of juvenile muddy loach fed the RWM20 and RWM30 diets was higher than that of the control group (P<0.05), while weight gain tended to decrease when fish were fed diets containing DDG. Weight gain of fish fed the RWM10 diet was greater than that of fish fed other diets, but the RM30 diet resulted in less weight gain than did the control (P<0.05). The feed and protein efficiency ratios of fish fed the RM10, RM20, RM30, RWM10, RWM20 and RWM30 diets were all similar to the control (P>0.05). Neither daily feed intake nor proximate composition of the whole body was affected by dietary DDG (P>0.05). The compositions of isoleucine, leucine, lysine, threonine, and valine were all elevated in fish fed the RM30, RWM10, RWM20 and RWM30 diets relative to those of other groups (P<0.05). In contrast, methionine + cystine and phenylalanine + tyrosine were all lower in fish fed the RM30, RWM10, RWM20 and RWM30 diets, as compared to those of other groups (P<0.05). Our results suggest that DDG is suitable as a partial replacement for soybean meal and wheat flour, and could be used at a rate of up to 20% for rice, or 30% for rice and wheat flour, for optimum growth performance of juvenile muddy loach.
