기름종개와 왕종개의 잡종복합군의 2배체와 3배체에 관하여 (Diploid-triploid Hybrid Complex of the Spined Loach Cobitis sinesis and C.longicorpus(Pices, Cobitidae))

  • 기름종개 C. sinensis와 왕종개 C. longicorpus가 함께 서식하는 낙동강상류의 여러 수역에서 이들사이의 잡종이라고 추측되는 개체들이 높은 반도 (43%)로 출현하였는데 그들은 모두 암컷이었다. 이들 잡종 개체들은 체측에 특이한 구름무늬 반문을 가지면서 형태적으로는 기름종개와 왕종개의 중간형질을 보여주고 그들의 염색체수는 2배체형 인 49개이거나 3배형인 73개이었다. 이들은 기름종개와 왕종개 사이에 잡종에서 유래된 후 이 종과는 독립된 단성복합집단(unisexual complex) 이라고 생각한다.

In vitro and In vivo Protein Qualities of Boiled Fish Extracts with Spicy Vegetables

  • To evaluate the quality of fish extracts with spicy vegetables (garlic, onion and ginger) in suppressing fishy oder, fish extracts of crucian carp, loach, bastard halibut and jacopever were processed at 100 $^{\circ}C$ for 6 hours, and their in vitro and in vivo protein qualities were determined . Protein and total lipid contents were closely related to the degree of discarding floated lipid on fish extracts and the kinds of added apicy vegetables . Boiling (10$0^{\circ}C$) , appeared to improve in vitro protein qualities slightly more than hydrocooking (11$0^{\circ}C$), but those with mild processing tended to result in better protein qualities than high temperature cooking (136-14$0^{\circ}C$). Spicy vegetables did not have remarkable effects on improving in vitro protein quality parameters. Fish extracts with 10% ginger were generally higher in in vitro protein quality than with the other vegetables . In spite of higher in vivo protein digestibility of fish extracts containing spicy vegetables processed under mild conditions(10$0^{\circ}C$), PERs of those extracts were not higher htan those of extranct processed at high temperature.

A human case of Echinostoma hortense (Trematoda: Echinostomatidae) infection diagnosed by gastroduodenal endoscopy in Korea

  • A human Echinostoma hortense infection was diagnosed by gastroduodenoscopy. An 81-year-old Korean male, living in Yeongcheon-shi, Gyeongsangbuk-do and with epigastric discomfort of several days duration, was subjected to upper gastrointestinal endoscopy. He was in the habit of eating fresh water fish. Two live worms were found in the duodenal bulb area and were removed using an endoscopic forcep. Based on their morphological characteristics, the worms were identified as E. hortense. The patient was treated with praziquantel 10 mg/kg as a single dose. The source of the infection in this case remains unclear, but the fresh water fish consumed, including the loach, may have been the source. This is the second case of E. hortense infection diagnosed by endoscopy in Korea.

유전자 변환 동물 생산을 위한 Plasmid Vector(pSS4)의 개발 (Development of a Plasmid Vector(pSS4) for the Use in Animal Transgenesis)

  • Transgenic animals have become an important tool in the basic and applied sectors of genetic and biomedical sciences. In particular transgenes provide clear-cut markers in the spatial and temporal analysis of developing embryos for the understanding of developmental mechanisms. For the long-term use of plasmid vector in a particular purpose it would be necessary to develop one's own vector system which can be properly expressed in eukaryotic system. Plasmids were constructed from ori region of pUC19 and early region of SV40 through various steps. LacZ gene coding for $\beta$-galactosides was fused to early gene of SV40 in translational in-frame. Poly(A) tailing site of SV40 was inserted at the 3' lacZ so that initiation, elongation and terminatin be controlled by SV40 transcription (pSS4). Biological function of the constructed pSS4 was demonstrated via microinjection of the plasmid into fertilized loach eggs and subsequent detection of $\beta$-galactosidase in developing embryos. The result indicate that the newly constructed pSS4 is functional in a eukaryotic system in vivo. Thus pSS4 may be used as an efficient tool for the study of embryogenesis and a basic carrier for various genes for animal transgenesis.

Analysis of Fish Expression Vectors for Construction of Two MARs Expression Vector System in Fish Cell Line

  • In previously study we isolated several fish matrix attachment regions (MARs) capable of replicating the plasmid by itself. In this study we construct a fish expression vector pBaEGFP(+) containing mud loach ${\beta}$-actin promoter EGFP as reporter gene and SV40 signal. To analyze the effects of the fish expression vector respectively. The fish ARS containing constructs pBaEGFP(+)-ARSs were transfected cells with pBaEGFP(+)-ARS101 and pBaEGFP(+)-ARS223 reduced 10 days to 25 days and then was constant to 30 days after transfection while that of the control vector without ARS element was basal level. The intensity of both constructs showed about 30fold of the intensity compared with the control vector on 30days after transfection individually .E. coli back-transformation analysis shows that pBaEGFP(+)-ARS223 and pBaEGFP(+)-ARS905 maintain in episomal state at least 30 days after transfection. The result indicates that both may be able to replicate the vector in BF-2 cell. Therefore the matrix-attached ARSs enhancing expression of the reporter gene might be useful as a component o the expression vector for transgenic studies.

미꾸라지(Misgumus mizolepis G.)의 심장에 존재하는 Immunoreactive Atrial Natriuretic Peptide의 특성에 관하여

  • 포유류의 심방에 주로 존재하여 체액 및 전해질 대사의 조절과 혈압 조절에 관여하는 atrial natriuretic peptide(ANP)가 담수어류인 미꾸라지(Misgumus mizolepis G.)의 심장 및 기타조직에도 존재하는지 알아보고 이들의 분자적 특성 및 온도차에 의한 함량변동을 조사하였다. 미꾸라지의 심장과 동맥구의 추출액은 모두 희석농도에 따라 표준곡선에 평행하는 ANP immunoreactivity를 나타냈다. 미꾸라지의 심장과 동맥구 모두에서 저분자량 및 고분자량의 immunoreactive ANP(irANP)가 관찰되었다. 한편 미꾸라지의 심장내 irANP함량은 61.56 $\pm$ 5.08 pg/mg이었으며, 심장외 조직인 동맥구는 60.59 $\pm$ 5.04 pg/mg으로 거의 동량의 irANP를 함유하였다. 미꾸라지를 저온상태에 적응시킨 경우에는 심장보다도 동맥구에서 더 유의한 irANP의 함량변동을 보였다(p < 0.01), 미꾸라지의 심장 및 동맥구에 존재하는 irANP는 주로 고분자량의 Pro-ANP가 관찰되므로써 미꾸라지의 심장과 동맥구 모두가 irANP를 조직내에서 합성할 것임을 시사하였다. 아울러 온도는 미꾸라지의 경우에서 irANP의 함량변동에 영향을 주는 요인임을 알 수 있었다.

Effects of Herbicide on the Environmental Ecosystem in Subtropics

  • Herbicides play a very important role in modern agriculture. However, the herbicide applied to the agricultural field may accumulate in the field, converting the advantages to environment pollution. Many small animals in the ecosystem such as alderfly, earthworm, butterfly, loach, frog, firefly, some birds and aquatic organisms have been known to disappear gradually. In addition, several behavior of herbicides including adsorption by soil, movement by water, photodecomposition, volatilization to air, absorption by plant, metabolism by soil microorganisms and so on, are proceeded while the herbicide remained in the environment. In this review, fate and behavior of herbicides in the environment and their effect on ecosystem after their application are focused on four aspects : the first is the absorption and metabolism of herbicides by plant; the second is the residues of herbicides in soil and water environments: the third is the accumulation and release of herbicides in aquatic organisms and the fourth is the translocation of herbicides in model agricultural ecosystem. Many factors may affect the behavior and fate of herbicides after their application, climatic conditions and soil properties seem to be the most important. Therefore, the fate and behavior of herbicide in Taiwan, located on subtropical region, may differ from those in Korea.

동면 어류의 지방질과 중성 및 인지질 조성의 시기적 변화(I) - 미꾸라지 근육 지질 조성의 변화- (Seasonal Variation in Lipids and Fatty Acid Composition of Neutral and Phospholipids of Hibernant Fishes(I) - Variation of Muscle Lipid Compositions of Loach (Misgurnus mizolepis) -)

  • 동면 어류인 미꾸라지를 일정시기(산란기전:5월, 산란기후:8월, 동면직전:11월, 동면직후:3월) 별로 채취한 다음 자.웅별 전육질부를 시료로 취하여 지방질과 중성 및 인지질 조성을 분석, 검토하였다. 중성지질이 구성지질의 대부분을 이루었고 다음은 인지질, 당지질순이었으며 그 조성과 변화 pattern이 자.웅이 거의 비슷한 경향을 나타내었다. 3월 이후 중성지질의 조성비가 점차 많아져 8월에는 90% 가까이 증가하였고 이후 점차 감소하여 이듬해 3월이 가장 많았다. 중성지질과 인지질 중에는 각각 triglyceride와 phosphatidyl choline 조성이 가장 많았으며 그 함량은 8월에 최고치를, 3월에 최소치를 나타내었다. 중성지질의 주요 지방산은 16:0, 16:1, 18:1, 18:2, 18:3($\omega$3) 등으로 이중 16:1 조성은 인지질보다 월등히 많았고 20:4($\omega$6), 20:3($\omega$3), 22:6($\omega$3) 등 고도 polyene산은 훨씬 적게 분포되었다. 중성지질의 지방산중 16:0 조성은 연중 거의 변화가 없었으나 16:1, 18:1, 18:2, 18:3($\omega$3)은 3월보다 5월이 2~4% 정도 높았고 고도 polyene산 조성은 그만큼 적었다. 이중 16:1 조성은 5월 이후 계속 줄어든 반면, 18:1, 18:2 w성은 11월까지 거의 비슷하였다. 또, 8월에는 18:0, 14:1, 17:1 조성의 증갈 포화 및 monoene산 조성이 다소 높아지고 고도 polyene산 조성은 더욱 줄어들어 불포화도가 최소치를 나타내었다. 11월부터는 그 반댈 포화 및 monoene산 조성이 점차 줄어들어 동면 직후인 3월에는 18:0, 16:1, 18:1 등 대부분의 포화산과 monoene산 및 18:2 조성이 더 큰 폭으로 떨어졌고 20:1, 20:4($\omega$6), 20:5($\omega$3), 22:6($\omega$3) 조성비는 그 만큼 더 높아지는 경향이었다. 인지질은 16:0, 16:1, 18:1, 18:2, 20:4($\omega$6), 20:5($\omega$3)이 주요 지방산이었고 3월과 5월 사이에 포화산과 monoene산 조성은 거의 차이가 없었으나 polyene산 조성은 중성지질과 비슷하게 변화였다. 8월에는 16:0, 18:0 등 주로 포화산 조성이 높아지고 고도 polyene산 조성은 그만큼 줄었으나 11월부터는 16:0, 18:0과 18:1, 18:2, 18:3($\omega$3) 조성이 다시 줄고 대부분의 고도 polyene산 조성은 점차 많아져 이듬해 3월에는 중성지질과 마찬가지로 불포화도가 더 큰 폭으로 커지고 특히 18:2, 18:3($\omega$3)과 20:4($\omega$6)와 22:6($\omega$3)의 변화가 두드러졌다.

당뇨병 환자의 대체요법 경험실태에 대한 조사 연구 (A Survey of Utilization of Complementary Alternative Medicine in Diabetes Mellitus)

  • In our country, patients with diabetes mellitus are searching for complementary treatments to recover from illness, while they received treatments from the doctor. However, have been evaluated or investigated systematically. This study was carried out to explore application of complementary treatments for patients with DM. For this survey, questionnaires were developed by researchers and the data was collected from July to October of 1999. Among the 223 subjects, there is one general hospital, one oriental hospital, 15 area C.H.P, ahd 2 area health centers. The results were as follows: 1. The total number of cases was 223 and the average age was 62.85 years old and average duration of DM was 8.1 years. The number of patients who had experience with alternative therapies was 145 (65%). The number of those who have not undergone treatments was 78 (35%). 2. The 43.5% of experienced CAM were advised family and relatives. Only 30.3% approved the effect of CAM and 52.5% said that If another a new CAM is introduced, they will try it. 4. Ninety three kinds of complementary treatments were used. Among the used items, 63.7% was various types of plants, 21.6% was animal material and 14.7% was the mixed group. As a single item, Bombyx Mori (Silkworm) was the most frequently used (10.5%) followed by the bean, mushroom, Morus bombycis (mulberry), Ginseng, Commelina Communis (Dalgaebi), Chinese medicine, root of Rosa rugosa (Haedangwha). 5. Among the used items, Trichosanthes kirilowii Max. Eucommia ulmoides Oliver, Commelina Communis, Aralia elata, pine needle, mulberry fruit, root of Rosa rugosa. Ginseng, Lycii Fructus, Dioscorea radix, Polygonatum odoratum, Cassia tora L, Bombyx Mori, loach, Crucian carp were based upon the pharmacological function of effect for control of diabetes mellitus symptom. 6. In the analysis of the relationships between the general characters of the patients with new complementary alternative medicine try and hospital treatment; 1) The shorter group suffered from DM (p=.038), poor Self-MBG (p=.037) and wanted to try new complementary alternative medicine. 2) The group of DM education experience were carried out hospital treatment well (p=.045). In conclusion, further study will be required for the patients experience using alternative therapies as the D-M in terms of holistic view of patients.
