랫드의 간암 발생과정에서 분리한 자연살해세포의 활성측정 및 특성연구 (Study on natural killer cell activity and its characteristics during hepatocarcinogenesis in rats)

  • The purposes of this study were to set up the method of the natural killer(NK) cell activity assay using the flow cytometer and to examine the characteristics and distribution of the NK cell during rat hepatocarcinogenesis. Forty five male 6 week-old specific pathogen free(SPF) Sprague-Dawley rats were randomly divided into three groups. Group I was the non-treated control and given normal diet and water. Group II was treated with diethylnitrosamine(DEN, 200mg/kg, i.p.) and partial hepatectomy. Group III was treated with DEN, partial hepatectomy and 0.05% phenobarbital sodium in water from 3 to 16 weeks. All animals were examined the morphology of the large granular lymphocyte(LGL), the LGL percent of the total lymphocytes and the LGL conjugation rate with YAC-1 cell in peripheral blood, spleen and liver. Moreover, activity of the LGL isolated from peripheral blood lymphocytes was determined using the flow cytometer. As results, LGL were observed in the peripheral blood, spleen and liver. LGL were observed the relatively faintly staining basophilic cytoplasm with granules, and eccentric, often kidney-shaped nuclei in Giemsa stain. Its size was $11{\sim}13{\mu}m$. LGL percentage of the isolated lymphocytes in peripheral blood, spleen and liver were 1.8~2.3%, 1.3~1.4% and 0.87~0.99%, respectively. LGL conjugation rate with YAC-1 cell was shown to be peripheral blood(9.3~10.3 %) > spleen(7.7~8.7%) > liver(5.6~7.0%). The activity of the LGL isolated from peripheral blood lymphocytes in Group I, II and III was 33.7%, 30.5% and 35.4%, respectively. However, all values were not significantly between groups.

Endothelial Cell Proliferation and Vascular Endothelial Growth Factor Expression in Primary Colorectal Cancer and Corresponding Liver Metastases

  • Background: Colorectal carcinoma (CRC) is one of the major causes of cancer death worldwide. Data from the literature indicate differences between the proliferation rate of endothelial cells relative to the morphology growth type, possibly due to origin of specimens (autopsy material, surgery fragments) or quantification methods. Vascular endothelial growth factor (VEGF) is a factor that stimulates the proliferation of endothelial cells. It is expressed in more than 90% of cases of metastatic CRC. Aim: The aim of this study was to evaluate the endothelial cell proliferation and VEGF expression in primary tumors and corresponding liver metastases. Materials and Methods: Our study included 24 recent biopsies of primary tumors and corresponding liver metastases of CRC cases. CD34/Ki67 double immunostaining and RNA scope assay for VEGF were performed. Results: In the primary tumors analysis of VEGFmRNA expression indicated no significant correlation with differentiation grade, proliferative and non-proliferative vessels in the intratumoral and peritumoral areas. In contrast, in the corresponding liver metastases, VEGFmRNA expression significantly correlated with the total number of non-proliferative vessels and total number of vessels. CD34/Ki67 double immunostaining in the cases with poorly differentiated carcinoma indicated a high number of proliferating endothelial cells in the peritumoral area and a low number in the intratumoral area for the primary tumor. Moderately differentiated carcinomas of colon showed no proliferating endothelial cells in the intratumoral area in half of the cases included in the study, for both, primary tumor and liver metastasis. In well differentiated CRCs, in primary tumors, a high proliferation rate of endothelial cells in the intratumoral area and a lower proliferation rate in the peritumoral area were found. A low value was found in corresponding liver metastasis. Conclusions: The absence of proliferative endothelial cells in half of the cases for the primary tumors and liver metastases in moderately differentiated carcinoma suggest a vascular mimicry phenomenon. The mismatch between the total number of vessels and endothelial proliferation in primary tumors indicate that a functional vascular network is already formed or the existence of some mechanisms influenced by other angiogenic factors.

Culinary Cinnamon and Clove Powder Ameliorate Fatty Liver Formation Induced by Ethanol Supplementation in Zebrafish

  • Culinary herbs and spices have received much attention since they contain high concentrations of bioactive ingredients for antioxidant and anti-inflammatory activities. Protection effect of the herb and spice against acute alcohol consumption has been investigated using zebrafish as a vertebrate model. During 30 days bathed in water containing 1% Et-OH and the designated herb or spice, the survival rate of the Et-OH group was decreased sharply (up to 20% at 10 days). The cinnamon-fed group showed the highest and longer survival rate up to 80% up to for 30 days under the presence of Et-OH, while clove-fed group showed 40% survival rate for 25 days. Et-OH group serum exhibited the weakest antioxidant ability from ferric ion removal ability (FRA) assay; FRA ability was increased in the cinnamon-fed group up to 414%, while the clove and laurel group increased 256% and 309%, respectively. Histologic observation and Oil-red O staining showed hepatic tissue damage was severe in the Et-OH group. The cinnamon- or clove-fed group showed much ameliorated hepatic tissue morphology with minimized steatosis. The cinnamon- or clove-fed group showed lower serum GOT and GPT levels than the Et-OH group. Among hepatic tissue extract, the clove-fed group exhibited the lowest level of GOT and GPT. These results suggest that consumption of cinnamon and clove might be beneficial to attenuate progress of acute fatty liver change by alcohol consumption.

  • This is a case report of invasive aspergillosis of the right maxillary antrum and the left lung in a 43 year-old male patient with diabetes mellitus and liver cirrhosis. The results were as follows. 1. Invasive aspergilliosis should be considered in compromised patients who have nasal symptomatology, infraorbital swelling and pain. 2. Invasive aspergillosis is a rapidly progressive mycotic infection of the sinus which may extend to the orbit, nasal cavity and cheek. 3. Surgical intervention is particularly urgent in orbital aspergillosis in patient in whom the eyesight is still preserved. 4. Diagnosis depends upon pathological demonstration of tissue invasion by organism with the typical morphology of aspergillus species. 5. Long-term antifungal drugs should be administrated postoperatively in pathint with invasive aspergillosis.

  • In attempt to develop a drug delivery system using serum albumin microspheres, bovine serum albumin microspheres containing antitumar agent. Cytarabine, were prepared. The shape, surface characteristics, size distribution, behavior of in vivo distribution, drug release behavior, and degradation of albumin microsphers in animal liver issue homogenate and proteolytic enzyme were investigated. The shape of albumin microspheres was spherical and the surface was smooth and compact. The size distribution of the albumin microspheres was effected by dispertion forces during emulsification and albumin concentration. Distribution of albumin microspheres after imtravenous administration in rabbit was achieved immediately. In vitro, albumin microsphere matrix was so hard that it retained most of cytarabine except initial burst during the first 10 minutes, and the level of drug release during the initial burst was affected by heating temperature, drug/albumin microsphere matrix was so hard that it retained most of cytarabine except initial burst during the first 10 minutes, and the level of drug release during the initial burst was affected by heating temperature, drug/albumin concentration ratio and size distribution. After drug release test, the morphology of albumin microspheres was not changed. Albumin microsphere matrix was degraded by the animal liver issue homogenate and proteolytic enzyme. Morphology, Morphometry, Growth Performance and Carcass Characteristics of Pekin, Nageswari and Their F1 Crossbred Ducks under Intensive Management

  • This study investigated the morphological features, growth, and meat yield performance of Pekin (P), Nageswari (N), and their reciprocal F1 crossbreds (P♂×N♀ and N♂×P♀). A total of 301-day-old ducklings were reared in four different pens up to 20 weeks of age under intensive management conditions. Feeding and management practices were similar for all individuals throughout the experimental period. The morphology and plumage pattern of F1 crossbreds were similar to those of indigenous Nageswari ducks because of the dominant inheritance of the extended Black allele (E locus). Genotype had significant differences (P<0.05) among the four genotypes in morphometric measurements, except wing and shank length. Growth performance was highly significant among the four genotypes (P<0.001) from one-day to 12 weeks of age. The average live weights of P, N, P♂×N♀ and N♂×P♀ crossbred genotypes at 12 weeks of age were 2038.35±29.74, 1542.44±33.61, 1851.85±28.59 and 1691.08±27.80 g, respectively. Meat yield parameters varied significantly (P<0.05) among the different genotypes for all studied traits, except for liver and gizzard weight. Moreover, no significant differences (P>0.05) were observed between P and P♂×N♀ crossbred for important meat yield traits such as hot carcass weight, dressing%, back half weight, drumstick with thigh weight and breast meat weight. Remarkably, the P♂×N♀ crossbreed possesses 50% native inheritance, which contributes to better adaptation in a hot-humid environment. Our results revealed that the P♂×N♀ genotype could be suitable for higher meat production with better adaptability in the agro-climatic conditions of Bangladesh.

Silymarin, a flavonoid antioxidant, protects streptozotocin-induced lipid peroxidation and β-Cell damage in rat pancreas

  • The present study is aimed at finding the influence of silymarin (a flavonoid) (25 mg/kg & 50 mg/kg) in streptozotocin (STZ)-induced diabetic rats. Type 2 diabetes was induced by single intraperitoneal injection of STZ (100 mg/kg) to 3 days old rat pups. Silymarin was administered for 15 days after the animals were confirmed diabetic (75 days after STZ injection). Blood glucose, glycosylated hemoglobin ($HbA_{1c}$), lipid peroxides (LPO) levels and reduced glutathione (GSH) contents in pancreas and liver were estimated following the established procedures. Biochemical observations were further substantiated with histological examination of pancreas. Blood glucose and $HbA_{1c}$ levels, which were elevated by STZ, were lowered to physiological levels by the administration of silymarin. The levels of LPO were significantly increased in STZ-induced diabetic rats. Silymarin reduced the LPO levels in both pancreas and liver. GSH contents which were reduced significantly in pancreas and liver of STZ-induced diabetic rats were brought back to near normal levels by silymarin treatment. Multifocal necrotic and degenerative changes of pancreas in STZ-diabetic rats were minimized to near normal morphology by administration of silymarin as evident by histopathological examination. Silymarin showed a dose dependent protective effect on STZ-induced $\beta$-cell damage. It could be attributed to the antioxidative and free radicals scavenging properties of the flavonoid. Thus, it may be considered as a natural antioxidant with potential therapeutic application in the treatment of type 2 diabetes.

Polyunsaturated Fat 및 Se 식이가 흰쥐의 혈청 비타민 E 및 Creatine Phosphokinase 와 간세포의 형태에 미치는 영향 (Effects of Dietary Polyunsaturated Fat and Se on Levels of Serum Vitamin E and Creatine Phosphokinase and Morphology of Rat Hepatocytes)

  • Vitamin E status affected by dietary high PUFA and Se was examined by biochemical and morphological means. Rats were fed four different diets(I : 15% p/s=1 control diet, II : 15% perilla oil diet, III : 15% perilla oil, vitamin mix -vitamin E, IV : 15% perilla oil, vitamin mix-vitamin E and salt mix -Se ) for $4\frac{1}{2}$ weeks. Various dietary treatments had no significant effects on body weight gains of rats. Activities of serum creatine phosphokinase known as an indicator of vitamin E deficiency were significantly higher( P < 0.001) in groups fed diets high in PUFA, regardless of the addition or omission of vitamin E from the vitamin mixture than those in control group. Vitamin E concentrations of serum and liver were affected by experimental diets and serum levels were more affected than those in liver. Electron microscopic observations of the liver revealed 1) the presence of swollen and degenerated mitochondria and lysosome-like body(II), and 2) markedly swollen and degenerated mitochondria, numerous lysosomes and decreased in size and number of microvilli along the bile canaliculus ( III, and 3) a remarkable accumulation of lipid droplets, nuclear pyknosis, degenerated mitochondria and increased number of lysosomes scattered along the cell junction in the hepatocytes (IV).

고지방식이 섭식 횐쥐의 간조직에 미치는 한방초콩환의 영향 (Effect of Herbal Chokong Pill on the Microstructure of Liver Cell in Rats Fed High Fat Diet)

  • 본 연구는 한방초콩환의 고지방식이를 섭식한 흰쥐의 간조직 지방제거 효과를 관찰하기 위하여 서목태(Rhynchosia nulubilis)를 식초에 15일간 절인 후, 5종류의 한약재(감초, 산조인, 백출, 황기, 산수유)를 첨가하여 한방초콩환을 제조하고, 고지방식이로 흰쥐의 비만을 유도하면서 고지방식이에 한방초콩환을 1%와 5% 첨가하여 9주 동안 투여한 후 간장의 무게, 혈청 효소활성도 및 간세포의 미세구조를 투과형전자현미경(TEM)으로 관찰하였다. HF군은 정상군의 간 무게에 비하여 약 35% 증가된 반면에 T5군의 경우는 HF군에 비하여 약 13% 감소하였다. 혈청 중 GOT 활성도는 정상군과 HF군이 비슷한 수준이었으나 T1군과 T5군에서는 HF군에 비하여 각각, 6.1%, 17.8% 감소하였으며 혈청 중 GPT 활성도 역시 정상군과 HF군이 거의 비슷한 수준이었으나 T1군과 T5군에서는 HF군에 비하여 각각 25.4, 32.4% 감소하였다. 간세포조직의 지방축적 정도 및 형태적 특징을 전자현미경으로 관찰한 결과, 정상군의 모양은 원형 내지 다각형을 나타내고 핵은 크고 둥글며 세포의 중앙에 위치해 핵질이 고르게 발달해 있을 뿐만 아니라 세포질에는 미토콘드리아를 비롯한 각종 세포 소기관이 고루 분포되어 있었으며 세포 전반에 걸쳐 지방구는 보이지 않았다. HF군은 세포전반에 걸쳐 지방구에 뒤덮여 세포의 형태 관찰이 불가능할 정도였으며 지방의 과잉 축적으로 인한 전자밀도가 낮아 백색을 나타내어 세포소기관의 막과 형태가 불분명한 지방간의 심각한 양상을 볼 수 있었다. T1군은 HF군에 비하여 지방구가 많이 소멸되었으며, T5군은 T1군에 비하여 지방세포의 크기와 수가 현저히 줄어든 것이 관찰 되었고, 더욱더 선명한 핵과 핵막 및 세포질의 전자밀도가 높아 지방질의 감소를 볼 수 있었다.
