Subacute Oral Toxicity Assesment of Ethanol Extract of Mariposa christia vespertilionis Leaves in Male Sprague Dawley Rats

  • The term Butterfly tea refers to decoction of Mariposa christia vespertilionis leaves which is widely consumed by cancer patients throughout Malaysia and has gained a huge popularity among Malaysians, not only cancer patients but also researchers to discover the real potential of this plant. Herein, the study is aimed at evaluating the possible toxicity in 28-day subacute oral toxicity of ethanolic extract M. christia vespertilionis in male Sprague Dawley rats. The 28-day subacute toxicity study was conducted to detect the no-observed adverse effect level (NOAEL). In this study, a total of 30 rats were divided into the control, 5% DMSO (vehicle), low dose (75 mg/kg), medium dose (125 mg/kg) and high dose (250 mg/kg) groups. The extract was administered daily from day 1 until day 28. At the end of the study, the animals were humanely sacrificed and assessed for the effect extract of Mariposa christia vespertilionis leaves on body weight and relative organ weights and haematological, biochemical and histopathological parameters. The haematological and serum biochemical parameters for the assessment of kidney and liver injuries were carried out. Results of haematological and serum biochemistry results showed no changes in the control and treated groups. In the histopathology, evaluation of kidney tissues in all treated groups showed no significant (p > 0.05) lesions. In contrast to kidney, liver tissues showed significant differences (p < 0.05) in lesions observed in low dose (430 mg), medium dose (700 mg) and high dose (1480 mg) groups with very mild, mild and mild to moderate lesion of hepatic necrosis, in the respective groups, and very mild hepatic degeneration and hepatitis were scored in all three groups.

  • Experimental infection method for Edwardsiellosis was studied to imitate histopathologically natural infections in the Tilapia Oreochrimis niloticus. Prior to the bacterial challenge, the intestine of tilapia was damaged by 0.2ml of 30% hydrogen peroxide introduced through a silicon tube which was inserted 4 to 5cm into the intestine from anus. E. tarda was mixed a 10% Gum arabia and administered into the stomach by a cannula 20 hours after the hydrogen peroxide treatment. Bacterial doses used were from $4.2{\times}10^6$ to $6.4{\times}10^\;CFU$/fish. Fish 72 hours after the challenged showed swollen and erosious lesion and focal necrosis with bacterial-eaden inflammatory cell in the Liver, Spleen, Kidney. Fish 96 and 120 hours after bacterial challenge were moribund showing swollen Liver, Spleen, Kidney and focal accumulation of macrophages and production of granulomas in the infected lesions.

'Experimental Study on the Effects of Alismatis Lhizoma on Hyperlipidemia' (택사(澤瀉)가 백서(白鼠)의 고지혈증(高脂血症)에 미치는 영향(影響)에 관(關)한 연구(硏究))

  • In order to investigate the effect of Alismatis Lhizoma on hyperlipidemia, experimental studies were performed on hyperlipidemia rats. Hyperlipidemia model (controll group) was induced by 1% cholesterol fed-diet for 8 weeks. Sample I group fed with 1% cholesterol and 4% Alismatis Lhizoma diet for 8 weeks. Sample II group fed with 1% cholesterol and 8% Alismatis Lhizoma diet for 8 weeks. The contents of serum total cholesterol, triglyceride, free fatty acid, phospholipid, HDL-cholesterol and LDL-cholesterol were measured, and fat accumulation in liver and the change of elastic and collagenous fiber in aortic wall were observed. The results were summurized as follows ; 1. The content of total cholesterol in the serum compared with control group tended to be decreased in sample group, and then sample I group showed a significant value. 2. The content of triglyceride in the serum compared with control group tended to be decreased in sample group, and then sample I group showed a significant value. 3. The content of free fat acid in the serum compared with control group tended to be decreased in sample group, but did not show a significance. 4. The content of phospholipid in the serum compared with control group tended to be decreased in sample group, and then sample I group showed a significant value. 5. The content of HDL-cholesterol in the serum compared with control group tended to be increased in sample group, and then sample II group showed a significant value. 6. The content of LDL-cholesterol in the serum compared with control group tended to be decreased in sample group, and then sample I group showed a significant value. 7. The lipophagy in liver compared with control group tended to be repressed in sample group. 8. The change of elastic and collagenous fiber lesion in tunica media of aortic wall, compared with control group tended to be repressed in sample group. According to the above results, it is assumed that Alismatis Lhizoma has a valid effect on hyperlipidemia. And yet, it needs to make further researches that sample I group showed more significant value than sample II group.

  • In order to investigate the effect of Lycii Radicis Cortex on hyperlipidemia, experimental studies were performed on hyperlipidemia rats. Cytologic Features of an Angiosarcoma in Pleural Fluid - A Case Report - (흉수의 혈관육종의 세포학적 소견 - 1예 보고 -)

  • Angiosarcomas are rare tumors, seen most commonly in the skin and soft tissue of head and neck legion. But it has been described in numerous body sites including thyroid, breast, liver, spleen, bone, etc.. Their biological behaviors depend on the microscopic grade, site of origin, and multifocality. We report the unique cytological features of an angiosarcoma in pleural fluid. A 61-year-old woman presented with a 6 month history of dyspnea on exertion and chest pain. Chest computerized tomography(CT) revealed multiple subpleural small nodules in the right lung and widespread all space consolidation and pleural effusion in the left lung. CT of liver revealed multiple small low attenuated lesion. The smears obtained from pleural fluid showed hypocellularity with a hemorrhagic background. The tumor cells were highly pleomorphic oval or spindle in shape and presented singly, in loose groups, in knitted syncytial aggregates, and in acinar pattern. Their nuclei had vesicular chromatin with delineated, thick nuclear membranes and occasionally a large eosinophilic, prominent nucleolus. The cytoplasm was plump, thin or protected in spindly fashion. Almost ail tumor cells showed variable sized intracytoplasmic vacuoles and their nuclei were sometimes crescentic by a huge vacuole. Occasional binucleated tumor cells and mitotic figures were present. Cellular debris and streaky materials were identified. Needle biopsy specimen from the pleura revealed anastomosing slit-like spaces lined by pleomorphic tumor cells. The tumor cells showed a strong reactivity for CD31 and vimentin and focal weak reactivity for factor VIII-related antigen.

Antioxidant effects of Geagibokrounghwan in experimental mouse fed cholesterol (고(高)Cholesterol 식이 생쥐에 미치는 계지복령환의 항산화효과(抗酸化效果))

  • This study was made to investigate the antioxidative effects of Geagibokrounghwan on the hepatic and renal lesion induced by cholesterol in mouse. The normal group was fed basal diet and water ; control groups were fed basal diet containing 0.5% of cholesterol ; test groups were fed the Geagibokrounghwan extract($10m{\ell}/kg$) after fed basal diet containing 0.5% of cholesterol for 6 weeks. In the liver and kidney of control group, lipid peroxidation(LPO) was significantly increased, however, the activities of superoxidation dismutase(SOD) and catalase and the amount of glutathion(GSH) were significantly decreased. In the liver and kidney of test group, lipid peroxidation(LPO) was decreased significantly as compared with control group. Contrary to this, the activities of superoxide dismutase(SOD), catalase and the amount of glutathion(GSH) were significantly increased. These results indicate that Geagibokrounghwan revealed the antioxidant effects, which may reduce the hepatic and renal damage induced by cholesterol in mouse.

Study on the Inhibitory Effects of Diallyl Sulfide and/or Diallyl Disulfide in the Rat Hepntocarcinogenesis (Diallyl Sulfide와 Diallyl Disulfide에 의한 랫드의 간암발생 억제효과에 관한 연구)

  • This study examined the anti-cancer effects of diallyl sulfide(DAS) and/or diallyl disulfide(DDS), major components of garlic oil, with the DEN-PH model in rats, by the numbers and areas per cm$^2$ of induced glutathion S-transferase placental form(GST-P) positive foci and silver-stained nucleolar organizer regions(Ag-NORs) counts per nuclei in liver as indicator. Sprague-Dawley(SD) rats were given the diethylnitrosamine(DEN, 200 mg/kg, i.p.) as initiator and 2 weeks later, in experiment 1, rats were treated with DAS(200 mg/kg, i.g.) and/or DDS(50 mg/kg, i.g.) for 6 weeks, respectively and concomitantly and also were given the same dose of DAS and/or DDS prior to DEN treatment for 2 weeks, and in experiment II, rats were treated with potential cancer promoter, 2-acetylaminofluorene (2-AAF, 20 mg/kg, i.g.). The DAS and/or DDS were treated prior to 2-AAF for 8 weeks, respectively and concomitantly. Then the anti-promoting effects of DAS and/or DDS were assessed. All rats were subjected to the two-thirds partial hepatectomy(PH) at week 3 and sacrificed at week 8. In experiment I, DAS and/or DDS treatment only prior to DEN showed inhibition of the development of GST-P positive foci. In experiment II, DAS and/or DDS treatment prior to 2-AAF promotion showed obvious inhibition of the development of GST-P positive foci in numbers and areas and AgNORs counts. In conclusion, We found DAS and/or DDS had the preventive effects on the hepatocarcinogenesis in rats and the concomitant treatment had some additive effects compared with the each treatment and AgNORs counts correlated well with the preneoplastic hepatic lesion.

Correlation of Bevacizumab-induced Proteinuria with Therapeutic Effects in Patients with Colorectal Cancer (직결장암 환자에서 Bevacizumab에 의한 단백뇨 발현과 치료효과와의 상관관계 분석)

  • Background: Bevacizumab-induced proteinuria is known to occur when vascular endothelial cell receptors are blocked, which leads to decreased protein filtration. Although several studies have analyzed the correlation between therapeutic effect of bevacizumab and proteinuria, no conclusion has been established. Methods: In this retrospective study, colorectal cancer patients who received bevacizumab and urinary protein check from January 2015 to December 2016, were included. The incidence of proteinuria and the grade according to Common Terminology Criteria for Adverse Events (CTCAE) 4.0 were evaluated after bevacizumab administration. The primary objective was to correlate proteinuria with overall response rate (ORR) and time to progression (TTP). Primary lesion, metastasized organs, surgery or radiation therapy, chemotherapy were investigated for analysis of risk factors for proteinuria development. Results: A total of 149 patients included in the analysis. Proteinuria occurred 19.5% (n=29) in the study patients; 20 in grade 1, 7 in grade 2, and 2 in grade 3. ORR was 55.2% in the proteinuria group and 51.7% in the non-proteinuria group. There was no difference between two groups (p=0.89). The TTP through the survival curve was similar in both groups (10 months, p=0.97). The risk of proteinuria was high in patients who had liver metastasis (p=0.02) and no surgery (p=0.01). Conclusions: These result indicates that bevacizumab-induced proteinuria expression was not correlated with the therapeutic effect on patients with colorectal cancer. Further analysis is required to find out the correlation between proteinuria and therapeutic effects. The risk of proteinuria was increased from patients who had liver metastasis, and no surgery.

Increase of Tc-99m RBC SPECT Sensitivity for Small Liver Hemangioma using Ordered Subset Expectation Maximization Technique (Tc-99m RBC SPECT에서 Ordered Subset Expectation Maximization 기법을 이용한 작은 간 혈관종 진단 예민도의 향상)

  • Purpose: RBC blood pool SPECT has been used to diagnose focal liver lesion such as hemangioma owing to its high specificity. However, low spatial resolution is a major limitation of this modality. Recently, ordered subset expectation maximization (OSEM) has been introduced to obtain tomographic images for clinical application. We compared this new modified iterative reconstruction method, OSEM with conventional filtered back projection (FBP) in imaging of liver hemangioma. Materials and Methods: Sixty four projection data were acquired using dual head gamma camera in 28 lesions of 24 patients with cavernous hemangioma of liver and these raw data were transferred to LINUX based personal computer. After the replacement of header file as interfile, OSEM was performed under various conditions of subsets (1,2,4,8,16, and 32) and iteration numbers (1,2,4,8, and 16) to obtain the best setting for liver imaging. The best condition for imaging in our investigation was considered to be 4 iterations and 16 subsets. After then, all the images were processed by both FBP and OSEM. Three experts reviewed these images without any information. Results: According to blind review of 28 lesions, OSEM images revealed at least same or better image quality than those of FBP in nearly all cases. Although there showed no significant difference in detection of large lesions more than 3 cm, 5 lesions with 1.5 to 3 cm in diameter were detected by OSEM only. However, both techniques failed to depict 4 cases of small lesions less than 1.5 cm. Conclusion: OSEM revealed better contrast and define in depiction of liver hemangioma as well as higher sensitivity in detection of small lesions. Furthermore this reconstruction method dose not require high performance computer system or long reconstruction time, therefore OSEM is supposed to be good method that can be applied to RBC blood pool SPECT for the diagnosis of liver hemangioma.

A Study on the Safety of Korean Domestic Fermented Meju (한국산 재래식 발효메주의 안전성에 관한 연구)

  • For this experiment frist the fermented Soybean Meju was prepared by domestic ordinary method of Korean and concentrated Meju suspensoidal extract was made for toxicological test sample on mice and rat. Even the oral dose of 10g extract per kg of mice had not any acute toxicity. So the median lethal dose(LOD50) is lilely to be much greatr or not to be. In comparison of the average weight gain rate for 4 weeks the control and test groups of 0.308g Ex/rat kg and 9.24g Ex/rat kg daily were the much the same as about 19% but 3.08g Ex/rat kg group waas exceptionally 2.7% higher than control For this period all test groups did not show any abnormality in appearance and anatomic findings. In serum GPT GOT alkaline phosphatase and blood urea nitrogen value there were no difference significantly between control and the test groups of 0.308g/rat kg and 3.08g/rat kg but 9.24g/rat kg group was some what higher than control. Especially in cholesterol alkaline phosphatase and blood urea nitrogen value 9.24g/rat kg group or 28days had 100mg/이 37.5u 32mg/이 respectively which were increased significantly. The pathological findings of all test groups especially 9.24g/kg group did not showed any characterized sign of acute or subacute liver damage or intestine lesion. So it is supposed that there is not any tocixants originated from some molds in fermented Korean domestic Meju which is prepared traditionally in winter.
