유기용제 취급 근로자들의 Succinyl trialanine p-nitroanilide 가수분해 효소 활성에 관한 연구 (A study of Succinyl trialanine p-nitroanilide hydrolytic activity in workers exposed to organic solvents)

  • 유기용제 취급 근로자들의 간기능 장해의 새로운 지표로서 혈청 Succinyl trialanine p-nitroanilide (STN) 가수분해 효소 활성치에 관한 연구가 신발제조업 제화부서에 근무하는 종사자 114명을 대상으로 시행하여 다음과 같은 결과를 얻었다. 1. 전체 대상자의 간기능 검사치인 혈청 GOT, GPT, ${\gamma}GT$의 평균은 $22{\pm}12.32IU/l,\;20{\pm}9.65IU/l,\;28{\pm}21.35IU/l$였고, 혈청 STN 가수분해 효소 활성치는 $0.08{\pm}0.05$였다. 2. 혈청 STN 가수분해 효소 활성치는 남자에서 통계학적으로 유의하게 높았고(P<0.05), 연령군별로는 차이가 없었다. 3. 일일근무시간별 혈청 STN 가수분해 효소 활성치는 차이는 없었으나, 3년이상 근무한 근로자와 100ppm이상의 톨루엔에 노출된 근로자에서는 혈청 STN 가수분해 효소 활성치가 통계학적으로 유의하게 증가하였다(p<0.05). 4. 톨루엔 노출농도와 혈청 GOT, GPT, ${\gamma}GT$ 및 혈청 STN 가수분해 효소 활성치 사이에는 통계학적으로 유의한 상관관계를 나타냈다(p<0.01). 5. 혈청 STN 가수분해 효소 활성치에 영향을 미치는 요소중에서 톨루엔 노출농도가 가장 설명력이 높고, 통계학적으로 유의하였다(P<0.05).

만성골수성백혈병(慢性骨髓性白血病)에 있어서 $^{198}Au$교질(膠質)을 사용(使用)한 간비주사소견(肝脾走査所見) (Hepatosplenic Scanning with $^{198}Au$ Colloidal Gold in Chronic Myelogenous Leukemia)

  • 1975년(年) 1월(月)부터 1978년(年) 3월(月)까지 경북의대부속병원(慶北醫大附屬病院)에서 진단(診斷)된 만성골수성백혈병(漫性骨髓性白血病) 가운데 18례(例)에서 $^{198}Au$교질(膠質)을 사용(使用)하여 간비주사(肝脾走査)를 시행(施行)하여 다음과 같은 소견(所見)을 얻었다. 1) 만성골수성백혈병(漫性骨髓性白血病)의 남녀비(男女比)는 2:1이였고 연령적(年齡的)으로 30대(代)와 40대(代)가 많았다. 2) 간비주사(肝脾走査)에서 간(肝)의 공간점유병변(空間占有病變)을 의심(疑心)할 만한 소견(所見)은 없었다. 3) 비섭취(脾攝取)는 well visualization 군(群) 4례(例) poor visualization 군(群) 7례(例), nonvibualization 군(群) 7례(例)였고, visualization 군(群)에서 비내공간점유병변(脾內室間占有病變)을 의심(疑心)할 소견(所見)은 없었다. 4) 비(脾) well visualization 군(群)과 poor visualization군간(群間)에는 임상(臨床所見) 간기능검사소견(肝機能檢査所見) 및 혈액학적소견(血液學的所見)에서 큰 차이(差異)가 없었다. 5) 비(脾) nonvibualization 군(群)에서는 visualigation 군(群)에 비(比)해서 증상기간(症狀期間)이 비교적(比較的) 길었던 례(例)가 많았고, 혈소판감소증(血小板減少症) 및 골수(骨髓)의 거핵구감소증(巨核球減少症)의 례(例)가 많았으나 간기능검사소견(肝機能檢査所見)에서는 별차이(別差異)가 없었다. In vivo 시험에 의한 잉어체내 $^{14}C-endosulfan$의 대사 (In vivo Metabolism of Endosulfan in Carp (Cyprinus carpio))

  • $^{14}C-{\alpha}-endosulfan$(100 nM, $41\;{\mu}g$)을 공시어의 각 조직(간, 신장, 소화관) 추출액에 처리하여 대사물질의 생성과 이에 미치는 cofactor의 영향을 시험관내 반응으로 비교한 결과, 105,000 g soluble fraction에서 phase I system에 해당되는 cofactor의 첨가시 공시어의 간 조직에서 가장 높은 대사물질 생성율을 보였고 phase II system에서는 NADPH를 cofactor로 첨가하였을 때 간과 신장조직에서 높은 대사물질 생성율을 보였다. 하지만 소화관 조직에서는 GSH 단독처리나 GSH+NADPH의 혼합처리시 높은 대사물질 생성율을 보였다. 또한 microsomal fraction에서는 phase I system에 해당되는 cofactor의 첨가시 공시어의 소화관 조직에서, phase II system의 경우 NADPH 단독처리나 NADPH+GSH 혼합처리시 대사물질의 생성이 가장 많았다. 이러한 결과로 보아 공시어의 간과 신장에서는 MFO에 의한 대사작용이, 소화관에서는 GST에 의한 대사작용이 활발하다고 할 수 있었다. 공시어의 각 조직에서 높은 생성율을 보인 대사물질은 EA(endosulfan alcohol), EE(endosulfan ether), EHE(endosulfan hydroxyether)이었으며 in vivo 시험에서와 마찬가지로 endosulfan sulfate는 검출되지 않았다. 이 실험에서 ${\alpha}-endosulfan$은 잉어체내에서 ${\beta}-endosulfan$으로 이성화되었다가 EA로 가수분해되거나 직접 EA로 가수분해되어 다시 EE나 EHE로 산화되며 EHE는 EL(endosulfan lactone)로 산화되는 것으로 제안할 수 있었다.

GM-CSF가 방사선 치료시 발생한 호중구감소증에 미치는 영향 (Effect of Granulocyte-Macrophage Colony-Stimulating Factor (GM-CSF) on Neutropenia Occuring during Radiotherapy)

  • Purpose : To assess the efficacy of recombinant human granulocyte-macrophage colony-stimulating factor(GM-CSF) in the neutropenia by radiotherapy. Materials and Methods : Eleven patients with various solid tumor were treated with a daily subcutaneous dose of GM-CSF(3-7microgram/kg) for 5days during the radiotherapy. Before and during the course of the study all the patients were monitored by the recording of physical examination, the complete blood count with differential and reticulocyte count and liver function test. Eight patients received prior or concurrent chemotherapy. Results : In 10 patients, the neutrophilic nadir was significantly elevated and the lenght of time that Patients had a neutrophil count below $10^3/mm^3$ a threshold known to be critical to acquiring infective complications was shortened following GM-CSF injection. A significant rise (two fold or greater) of neutrophil count was seen in 10 of 11 patients. In most patients, discontinuation of GM-CSF resulted in a prompt return of granulocyte counts toward baseline. However the neutrophil count remained elevated over $10^3/mm^3$ during radiation therapy, and radiotherapy delays were avoided. Other peripheral blood components including monocytes and platelets also increased after GM-CSF treatment. No significant toxicity was encountered with subcutaneous GM-CSF treatment. Conclusion : GM-CSF was well tolerated by subcutaneous route and induced improvement in the neutropenia caused by radiotherapy.

경부 결핵성 임파선염 (Tuberculous Cervical Lymphadenitis)

  • Tuberculous cervical lymphadenitis is one of common cause of neck mass in young adult in Korea. Tuberculous cervical lymphadenitis known as scrofula was being treated by the 'Royal Touch' in the 5th century and by surgery in the 17th century, yet the principle of the treatment is still controversal. We report the clinical evaluation and therapeutic result about 121 tuberculous cervical lymphadenitis. The result were as follows: 1) The annual incidence(1985-1994) was 30.5 % (37 cases: 1985-1986), 17.7 % (21 cases: 1990-1992). 19.8%(24 cases: 1994). 2) The age of highest incidence was 20-29 year old age group in 41.3% (50 cases) and female predominated over male by 1.8 : 1. 3) The duration of disease was less than 6 months, in 85.9% (104 cases). 4) The most commonly involved LN group was superficial cervical group in 69.4% (84 cases), and difference between Rt & Lt, was not significant. 5) The most common symptom and local finding(P/E) were, painless swelling of LN in 74.3 % (90 cases) and single mass in 59.5 % (72 cases). 6) In seasonal variation, 85.9% (104 cases) was spring and summer. 7) Procedures except biopsy for evaluation were chest PA, AFB smear & culture(sputum), Mantoux test, USG, CT and, Associated extracervical tuberculous lesions were lung, axilla, breast. 8) In operation method (involving biopsy), Excision was 68.5 % (83 cases), neck dissection was 6.6% (8 cases). 9) The Modality, Duration and side effect of antituberculous medication: INH-Rifampin­Ethambutol was 66.1 % (88 cases), duration was 1 year in 84.3% (102 cases), side effects were severe GI trouble (24.8%), liver function damage (3.3%). 10) 3 cases recurred on the same site after 2 yrs(2 cases) and 4 months(1 case) and its treatment was curretage or I & D, with antituberculous medication.

소청룡탕합옥병풍산(小靑龍湯合玉屛風散)이 백서(白鼠)의 알레르기 비염(鼻炎)에 미치는 효과 (The Effects of Sochungyong-tang plus Okbyeong-san Extract on Allergic Rhinitis of Rats)

  • Background and Objectives : Although the incidence of allergic rhinitis has increased specific therapeutics with a certain medicine is not well developed yet. In therapeutics of allergic rhinitis, limited medicines just have been used in the clinin. Sochungyong-tang has been a widespread medicine for allergic rhinitis. However, okbyeongpung-san has been taken for raising immunity. In medical treatment of allergic rhinitis, raising immunity is a necessary part of the process. In this reason, this study aimed to find out therapeutic effects of combination of sochungyoung-tang and okbyeongpung-san extract on allergic rhinintis of nasal tissue in rats. Materials and Methods : The Sprague-Dawley male rats, about 200g of body weight, were divided into three groups: the normal group, the control groups, and the sample groups. With the Levine and Vaz method, the allergic reaction was induced with ovalbumin as antigen in the intraperitoneal route. Rats of the sample groups were treated with 2cc of sochungyoung-tang plus okbyeongpung-san extract once a day for 28days. I observed historical changes in nasal tissue. I also found changes in the segment of lympocyte and neutrophil of Leukocyte and erythrocyte. I used the statistical methods of post Hoc test(p<0.05) Results : The increase of the goblet cell and mucous secretion in the sample groups was rare when compared to the control groups. Effects of sochungyoung-tang plus okbyeongpung-san extract on the liver function were also studied in rats. Treatment of sochungyoung-tang plus okbyeongpung-san extract did not affect GOT and GPT. The number of lymphocyte was decreased, while the number of erythrocyte was increased in sample groups. Conclusion : This study shows that sochungyoung-tang plus okbyeongpung-san plus okbyeongpung-san extract has considerable effects on allergic rhinitis.

The Relationship between Urinary 8-hydroxydeoxyguanosine and Polycyclic Aromatic Hydrocarbons Exposure in Working Environment

  • To investigate the exposure effect of polycyclic aromatic hydrocarbons (PAHs), we analyzed the relationship between urinary 8-hydroxydeoxyguanosine (8-OHdG) concentration and PAHs exposure. The study population contained 44 workers in steel-pipe coating and paint manufacture industries. We measured airborne total PAHs as an external dose, urinary 1-hydroxypyrene (1-OHP) as an internal dose of PAHs exposure, and urinary 8-OHdG as an effective dose of oxidative DNA damage. There was significant correlation between the urinary concentration of l-OHP and the environmental concentration of PAHs, pyrene, urinary cotinine, AST, and GGT. The mean of urinary 8-OHdG was $17.07\pm1.706{\mu}g/g$ creatinine in workers exposed to airborne PAHs. There was significant correlation between the urinary concentration of 8-0HdG and the airborne concentration of PAHs. From the results of stepwise multiple regression analysis about 8-OHdG, significant independents was total PAHs. In this study, there were significant correlation between the urinary concentration of 8-OHdG and the airborne concentration of PAHs. The urinary 1-OHP was effective index as a biomarker of airborne PAHs in workplace. But it was influenced by non-occupational PAHs source, smoking and biomarkers of liver function test.

단치소요산가감방(丹梔逍遙散加減方)이 streptozotocin으로 유발된 고혈당 생쥐에 미치는 영향 (Effect of Danchisoyosangagambang Extract on the Hyperglycemic Mice Induced with Streptozotocin)

  • This study has been carried out to understand the effect of Danchisoyosangagambang (DC) on the hyperglycemic mice induced with streptozotocin(STZ). Experimental groups were made diabetic mice by intraperitoneal injection of STZ(60 mg/kg of body weight) tw ice by 24 h interval and then 120 mg/kg STZ was injected again 3 days after the earlier treatment. Control group was administered mice with 0.9 % saline(2 mL/kg), and experim ental groups were administered DC extract(DCA group, 10 mg/kg/day; DCB group, 30 mg/kg/day) after hyperglycemic induction for 6 weeks. The body weight of experimental groups was lower than control. The blood glucose concentration increased continuously, rea ching to 298.9 mg/dL after 6 weeks, however, experimental groups of the DCA and DCB groups significantly(p<0.0l) decreased in the 4, 5, and 6 weeks groups. Blood glucose tolerance test was not significant between control and experimental groups. We examined the blood transaminase activities to know the effect of herbal medicine on liver function. The GOT activities were lower in group DCB than in control. The GPT activities were lower in group DCA and DCB than in control. The content of triglyceride was significantly increased in group DCA compared to control. The SOD and catalase activities were higher in the group DCA compared to control. The results of immunohistochemical study, a few of insulin positive cells observed in the control and experimental group. These results suggest that administration of DC extract to the hyperglycemic mice decreased the blood glucose level.

수종의 한약재가 Streptozotocin으로 유발된 고혈당 생쥐에 미치는 영향 (The Effects of Several Herbal Medicine Concentrated Solution on the Hyperglycemic Mice Induced with Streptozotocin)

  • Objective : This study has been carried out to understand the effect of several herbal medicine concentrated solution on the hyperglycemic mice Induced with streptozotocin(STZ). Methods : The 60 mg/kg of STZ injection into mice twice by 24 h interval and then 120 mg/kg of STZ injection again 3 days after the earlier injection. Control group was subjected to natural recovery, however, treated groups were fed 0.2 ml of several herbal medicine-concentrated solution (PA (x2, several herbal medicine-concentrated solution 1 group); PB (x4 several herbal medicine-concentrated solution 2 group) daily for 6 weeks. Result : The weight of PA was higher than that of control, but weight of PB was lower than control. The blood level of control increased continuously, reaching to 350mg/dL after 6 weeks, however, PA and PB showed a fast reduction of blood glucose. In blood glucose tolerance test, PA and PB showed better resistance than control. The GOT level in significantly(p<0.05) decreased in PA and PB compared with control group. The BUN and creatinine levels are significantly(p<0.01) decreased in PA compared with control group. Feeding of several herbal medicine-concentrated solution in a concentration of PA had an efficient effect on regeneration or recovery of Langerhans islet and ${\beta}-cell$ damaged by STZ. More Langerhans islet and high insulin-immunohistochemical resistance were observed in PA compared with control, but they were higher in PB than in PA. The number of Langerhans islet ${\beta}-cell$ and Langerhans islet. Conclusions : The result from the six weeks of observation demonstrates that the several herbal medicine concentrated solution have a positive effect of lowering the level of blood sugar and they increased insulin concentration. They have an effect for recovery of pancreas tissue and recovery of kidney, liver function from a diabetes mellitus.
