적소두증유약침(赤小豆蒸溜藥鍼)이 비만(肥滿)쥐의 지질구성(脂質構成), 간장기능(肝臟機能), 항산화효과(抗酸化效果) 및 분자생물학적(分子生物學的) 특성(特性)에 미치는 영향(影響) (Effects of distilled Phaseoli Semen rubra Herbal-Acupuncture on lipid composition, liver function, antioxidant capacity and molecular biological aspects in obese rats induced high fat diet)

  • Effects of Phaseoli Semen rubra Herbal-acupuncture at zusanli(ST-36), Quchi(LI-11) and Sanyinjiao(Sp-6) on lipid composition, liver function, oxidative capacity and molecular biological aspects were investigate in high fat diet induced obese rats. Forty male Sprague-Dawley rats weighing about 400g were divided into 4 groups according to body weight and raised four weeks with control, zusanli(ST-36), Quchi(LI-11) and Sanyinjiao(Sp-6) Herbal-acupuncture groups. 1. Plasma total cholesterol and LDL-cholesterol showed a low values in the (ST-36) and (LI-11) Herbal-acupuncture groups and HDL-cholesterol showed a high values in the (ST-36) Herbal-acupuncture groups. 2. Plasma triglyceride and glucose showed a low values in the (ST-36) and (LI-11) Herbal-acupuncture groups. 3. The contents of plasma free fatty acids showed a tendence to decrease in the Herbal-acupuncture groups, however in the Herbal-acupuncture groups, the values showed no significantly different. 4. The activities of AST and ALT showed no significantly different in all treatment groups. 5. The contents of plasma ${\beta}-lipoprotein$ and free fatty acids showed a tendency to decrease in the Herbal-acupuncture groups compared to those of control group. In the Herbal-acupuncture groups, the values of (ST-36) and (LI-11) Herbal-acupuncture groups showed a low in the acupuncture groups. 6. Liver total cholesterol and triglyceride showed a low values in the (ST-36) and (LI-11) Herbal-acupuncture groups. 7. Contents of plasma TBARS showed a low values in the (ST-36) and (LI-11) Herbal-acupuncture groups, however contents of liver TBARS showed no significantly different among treatment groups. 8. Values of liver glutathione peroxidase and catalase activity showed a tendency to increase in the (ST-36) and (LI-11) Herbal­acupuncture groups. Values of liver super oxide dismutase activity showed a high in the (ST-36) Herbal-acupuncture groups compared to those of other groups. 9. Expression of apo-B mRNA in liver cell showed a low in (ST-36) Herbal-acupuncture groups, however expression of apo-E mRNA showed a high in Herbal-acupuncture groups and $TNF-{\alpha}$ mRNA in adipose cell showed no different among all the treatment groups. Expression of Leptin mRNA showed a tendance to decrease in (ST-36) Herbal-acupuncture group. 10. Histological character of liver, those of (ST-36) Herbal-acupuncture group showed a good, however other treatment groups showed slight vasodilation and slight fat accumulation. These results indicated that Phaseoli Semen rubra Herbal-acupuncture at (ST-36) and (LI-22) suppressed adipose tissue mass and lipid peroxidation and increased antioxidant system.

깻잎, 쑥, 참취의 건분 및 에탄올 추출물이 흰쥐의 지방대사와 항산화능에 미치는 영형 (Effect of Dried Leaf Powders and Ethanol Extracts of Perilla Frutescens, Artemisia Princeps Var. Orientalis and Aster Scaber on Lipid Metabolism and Antioxidative Capacity in Rats)

  • This study was performed to investigate the effects of dried leaf powders and ethanol extracts of Perilla frutescens(perilla leaf), Artemisia princeps var. orientalis(mugwort) and Aster scaber(chamchui) on lipid metabolism and antioxidative capacity in rats. Forty-nine male Sprague-Dawley rats weighing 105.9$\pm$1.7g were blocked into seven groups according to body weigth and raised for four weeks with diets containing 5% dried powders of perilla leaf, mugwort and chamchui, or ethanol extracts from the same amount of each dried leaf powder. Food intake was higher in the control and chamchui powder groups than the other remaining groups. Weight gain was not significantly different among all experimental groups. Food efficiency ratio was highest in the perilla leaf powder group. Plasma total lipid and cholesterol, liver total lipid and triglyceride levels were highest in the perilla leaf powder group. In contrast, dried powders of mugwort and chamchui showed hypolipidemic effects in plasma and liver. Plasma and liver TBARS levels in both dried powder and ethanol extract groups of the three plants were lower than control. There was also no significant difference between corresponding dried powder and ethanol extract groups. Catalas and GSH-Px activities in erythrocyte and liver were not different among all the experimental groups. However, SOD activities were significantly different among the esperimental groups. In erythrocyte. SOD activiteis of all dried powder and ethanol extract groups except the perilla leaf powder group were higher than control, and the chamchui power group showed the highest activity among them. In liver, all the plant groups showed higher SOD activities than control. There was no significant difference between corresponding dried powder and ethanol extract groups. In conclusion, the dried chamchui powder and liver. All the dried plant powder and ethanol extract were effective in decreasing the TBARS levels of liver and particularly plasma. Among the antiocidative enzymes, SOD activity was most responsive to the experimental diets. All the plant groups showed increases in liver SOD activities and there were more increments in dried powder groups. Since the effects of dried powder groups on lipid metabolism were better than those of extract groups, it is plausible that the high dietary fiber level in dried powders was effective. Antioxidative effects were not significantly different between corresponding dried power and extract groups, and it was thought that ethanol extracts from three plants also had similar antioxidative effects as dried powderers.

구인약침(蚯蚓藥鍼)이 비만쥐의 지질강하능, 항산화능, 전염증성 Cytokines 농도에 미치는 영향 (The Effects of Lumbricus Pharmacopuncture on the Lipid Lowering, Anti-oxidative Activity and Concentration of Proinflammatory Cytokines in Rat Fed High Fat Diet)

  • Objectives : This study was designed to investigate effects of Lumbricus pharmacopuncture (LP) on the lipid lowering, anti-oxidative activity and concentration of proinflammatory cytokines in rat fed high fat diet. Methods : Sprague-Dawley male rats were fed high fat diet for 8 weeks and experimental groups were divided into 4 groups as follows : Control, Lumbricus Jungwan ($CV_{12}$) pharmacopuncture (T I), Lumbricus Joksamni ($ST_{36}$) pharmacopuncture (T II), Lumbricus Jungwan ($CV_{12}$) and Joksamni ($ST_{36}$) pharmacopuncture (T III). Results : The levels of plasma FFA, TG, total cholesterol, LDL-C, TBARS, IL-$1{\beta}$, IL-6, TNF-$\alpha$ and liver total cholesterol, TG, TBARS, SOD, catalase, IL-6 in more than one LP group were significantly lower than those of Control groups. The level of IL-10 in one of LP groups was significantly higher than that of Control group. Conclusions : As the results were shown, LP treatment have considerable effects on lipid lowering, anti-oxidative activity and concentration of proinflammatory cytokines in rat fed high fat diet.

Antioxidant action of soy isoflavones on oxidative stress and antioxidant enzyme activities in exercised rats

  • BACKGROUND/OBJECTIVES: Isoflavones are widely believed to be beneficial to human health, in relation to their antioxidant potentials. Exercise can cause an imbalance between reactive oxygen species (ROS) and antioxidants. This study was conducted in order to investigate the ability of isoflavones in amelioration of oxidative stress induced by exercise. MATERIALS/METHODS: Male Sprague-Dawley rats were assigned to one of four groups: isoflavone-free with no exercise (CON-sd), isoflavone-free with exercise (CON-ex), isoflavone-supplemented with no exercise (ISF-sd), and isoflavone-supplemented with exercise (ISF-ex). Animals exercised on the treadmill for 30 minutes per day, five days per week. TBARS as a marker of oxidative stress and antioxidant enzyme activity, including SOD, GSH-px, and catalase were determined in liver tissue. Serum lipid profile was also examined. RESULTS: A significant effect of isoflavone alone was observed on abdominal fat pad mass. ISF-ex had significantly less abdominal fat pad than CON-ex. Both exercise and isoflavone treatment had significant effects on lowering plasma triglyceride (TG), thus, the ISF-ex group had a significantly lower TG level than the CON-sd group, by 30.9%. However, no differences were observed in plasma cholesterol, HDL-C, and cholesterol/HDL-C ratio. Exercise, isoflavone, and exercise-isoflavone interaction effects were significant on thiobarbituric acid reactive substances (TBARS) (P = 0.001, 0.002, and 0.005, respectively). The CON-ex group showed a higher TBARS level than the other three groups. By contrast, in the ISF-ex group, TBARS was restored to the level of the ISF-sd or CON-sd group. Isoflavone had a significant effect on superoxide dismutase (SOD) (P = 0.022) and catalase activities (P = 0.049). Significantly higher SOD and catalase activities were observed in ISF-ex than CON-ex. SOD and catalase activities showed an inverse pattern of TBARS. Taken together, isoflavones increased the activities of SOD and catalase with concomitant decreases in TBARS, indicative of decreased oxidative stress. CONCLUSIONS: Isoflavone supplementation enhances antioxidant action with attenuation of exercise-induced oxidative stress, as measured by decreases in TBARS, and inhibits body fat accumulation and plasma TG increase. Antioxidative effects ascribed to isoflavones may be partially exerted via enhancement of antioxidant enzyme activities.

산마늘추출물이 과산화지질급여 비만쥐의 지질강하, 항산화효과 및 염증매개물질의 생산에 미치는 영향 (Effects of Allium victorials Extract on Lowing Lipid, Anti-oxidation and Concentration of Inflammatory Mediators in Rats Fed High Oxidized Fat)

  • 본 연구는 산마늘추출물이 과산화지질을 급여한 비만쥐의 지질강하, 항산화효과 및 염증매개물질의 생산에 미치는 영향을 검토했다. 그 결과 혈장 FFA, TG, total cholesterol 및 LDL-cholesterol 농도는 산마늘추출물 처리군 들에서 감소했으며, 혈장HDL-cholesterol 농도는 산마늘추출물 처리군 들에서 증가했다. 간장 내 total cholesterol 농도 및 TG 농도는 산마늘추출물 처리군 들에서 감소하는 경향을 나타내었다. 혈장 및 간장의 TBARS 농도는 산마늘추출물 처리군 모두가 대조군보다 낮은 경향을 보였다. 간장 GSH-Px, SOD 및 CAT활성치모두가 산마늘추출물 처리군 들에서 증가하는 경향을 보였다. 혈장 NO, Ceruloplasmin 및 ${\alpha}1$-acid glycoprotein 농도는 산마늘추출물 투여군 들이 대조군보다 낮은 경향을 나타내었다. 이와 같은 결과는 산마늘추출물에 지질강하, 항산화 및 항염증작용에 효과를 나타내는 기능성물질이 내재하고 있음을 시사한다.

Effect of Colored Potato Flakes Against Acetaminophen-induced Liver Damage in Rats

  • We examined the hepatoprotective effects of colored potato flakes on acetaminophen (AAP)-induced liver damage in rats. F344/DuCrj (8 week-old) rats were fed a cholesterol-free diet with 54.9486 g of ${\alpha}$-corn starch/100g diet and were orally treated with 25% colored flakes of Kitamurasaki (KM: light purple), Northern Ruby (NR: red), and Shadow Queen (SQ: medium purple) potatoes co-administered with AAP (0.5 g/100 g diet) for 4 weeks. The hepatic thiobarbituric acid-reactive substances (TBARS) values in the KM, NR, and SQ groups were significantly lower (p<0.05) than those in the control groups with and without AAP. Furthermore, the hepatic catalase, Mn-superoxide dismutase (SOD), and Cu/Zn-SOD mRNA levels in the KM, NR, and SQ groups were higher than those in the control groups with and without AAP. The present findings suggest that colored potato flakes are useful as a prophylactic agent against oxidative liver damage.

추출 조건을 달리한 감잎과 녹차의 물 및 에탄올 추출물이 노령쥐의 지방대사와 항산화능에 미치는 영향 (Effect of Water and Ethanol Extracts of Persimmon Leaf and Green Tea Different Conditions on Lipid Metabolism and Antioxidative Capacity in 12-month-old Rats)

  • This study was performed to investigate effects of dried leaf powders, water, 75% and 95% ethanol extracts of persimmon leaf and green tea on lipid metabolism, lipid peroxidation and antioxidative enzyme activity in 12-month-old rats. Fifty-four male Sprague-Dawley rats weighing 542$\pm$4.5g were blocked into groups according to their body weight and were raised for four weeks with the diets containing 5%(w/w) dried leaf powders of persimmon(Diospyros kaki Thunb) and green tea(Camellia Sinensis O. Ktze), water or 75% and 95% ethanol extracts from same amount of each dried tea powder. Food intake was not significantly different among all groups, but weight gain of green tea powder group was significantly lower than that of control group. Plasma and liver lipid levels of all the tea diet groups were lower than those of control group. Especially, 75% ethanol extract of persimmon leaf decreased total lipid and triglyceride concentrations in plasma and 95% ethanol extract of persimmon leaf decreased liver total lipid level. However, there was no difference between 75% ethanol extracts groups and 95% ethanol extracts groups in lipid metabolism. Superoxide dismutase(SOD) and catalase activities in erythrocyte were remarkably increased by all the green tea diets. SOD, catalase and glutathione peroxidase activities in liver were increased by the feeding of ethanol extracts from green tea and persimmon leaf powder. Liver xanthine oxidase activity was not different among all groups. Plasma Thiobarbirutic acid reactive substance(TBARS) concentrations of all the green tea diet groups were significantly low. It was thought that high flavonoids in green tea inhibited plasma lipid peroxidation by promoting SOD, catalase activities in erythrocyte. 95% ethanol extract of persimmon leaf also inhibited plasma lipid peroxidation by high vitamin E and beta-carotene. Persimmon leaf powder decreased liver TBARS concentration by vitamin E, betacarotene and vitamin C and by increasing activities of antioxidative enzymes with flavonoids. In conclusion, dried leaf powders, water, 75% and 95% ethanol extracts of persimmon leaf and green tea were effective in lowering lipid levels and inhibiting lipid peroxidation in 12-month-old rats. Above all, ethanol extracts of persimmon leaf decreased plasma and liver lipid levels and persimmon leaf powder effectively inhibited liver lipid peroxidation. Extracts of green tea leaf inhibited plasma lipid peroxidation. In lowering lipid levels and inhibiting lipid peroxidation, ethanol extracts were more effective than water extracts, but there was no difference between 75% ethanol extracts and 95% ethanol extracts in lipid metabolism. 둥굴레 추출물 및 분획의 과산화지질 생성 저해효과 (Anti-lipid Peroxidative Effect of Extracts and its Fractions Polygonatum odoratum)

  • The rhizoma of Polygonatum odoratum (Liliaceae) promotes the production of body fluid and relives dryness symptoms and has hypoglycemic effect. In order to evaluate the prevention of lipid peroxidative efficacy of P. odoratum, its extracts (Et$_2$O and MeOH ex.) and its fractions ($H_2O$, 20% MeOH, 40% MeOH, 60% MeOH and 100% MeOH fr.) were measured by TBARS assay on rat liver S9 and human erythrocyte ghost membrane. The order of anti-lipid peroxidative effect was as follows: Et$_2$O ex. >40% MeOH fr. >60% MeOH fr.>100% MeOH fr. Based on these results, we conclude that membrane lipid peroxidation is inhibited in vitro by addition of P. odoratum ether extract and its gradient MeOH fractions excepts $H_2O$ fr., and which have significant hepatoprotective activity in $CCl_4$-treated rats.

뽕나무와 꾸지뽕나무의 수피 수용성 추출물이 콜레스테롤 함유식이 투여 흰쥐의 지질농도 및 과산화지질 농도에 미치는 영향 (Effect of Stem Bark Extract from Morus alba and Cudrania tricuspidata on the Concentrations of Lipid and Tissue Lipid Peroxidation in the Cholesterol-Fed Rats)

  • 콜레스테롤 함유식이(대조군)에 뽕나무(M군) 및 꾸지뽕나무(C군) 줄기껍질 수용성 추출물을 첨가하여 흰쥐에 급여시킨 후 혈청과 간장 지질 농도 및 각 조직중의 지질과산화물 함량에 미치는 영향을 검토하였다. 혈청 중성지질 농도는 대조군에 비교해서 M군에서는 유의적으로 감소하였고, C군에서는 감소경향을 보였다. C군에서 혈청 총 콜레스테롤 및 인지질 농도 및 M군에서 혈청 중성지질 농도는 대조군에 비해서 각각 저하하였다. 간장 콜레스테롤 농도는 대조군에 비해 C군 및 M군에서 저하경향을 나타내었으나, 중성지질 및 인지질 농도는 각군간에 유의적인 차이는 없었다. 흰쥐 각 조직의 생체막 지질과산화물 생성 정도의 지표인 TBARS의 상대적 농도는 뇌, 신장, 심장, 간장의 순으로 나타났다. 신장의 지질 과산화물 함량은 대조군에 비교해서 C군 및 M군에서는 유의적으로 감소하였으나, 뇌 및 간장의 지질 과산화물 농도는 오히려 유의적으로 증가하였다. 간장의 비헴철 함량은 대조군에 비해서 C군 및 M군에서 유의적으로 증가하였으며, 이러한 증가는 간장 지질 과산화물 함량 증가와 깊이 관련되는 것으로 시사되었다. 흰쥐 조직으로부터 분획한 microsome에 $Fe^{2+}$/ascorbate를 첨가하여 $37^{\circ}C$에서 반응을 시키지 않은 것(0 hr)으로부터 지질 과산화물을 측정한 결과에서는 각 군간에 유의적인 차이는 없었으나, $Fe^{2+}$/ascorbate를 첨가하여 1시간 반응시킨 간장 및 뇌 조직 microsome의 지질 과산화물은 대조구에서 현저히 증가하였다. 한편 간장 microsome에 $Fe^{2+}$/ascorbate 첨가 1시간 반응에서는 대조군에 비해 C군 및 M군에서 유의적으로 감소하였으나, 뇌 조직에서는 이러한 효과가 없었다. 이상의 결과에서 뽕나무 및 꾸지뽕나무 줄기껍질 수용성 추출물에는 혈청 중성지질 저하 작용 뿐만아니라 신장 및 간장 microsome의 지질과산화물 농도 저하작용이 인정되었다.

녹차 열수추출물이 동$\cdot$식물성 단백질과 고콜레스테롤을 급여한 흰쥐의 지질대사와 항산화효과에 미치는 영향 (Effects of Hot Water Soluble Extract from Green Tea on the Lipid Metabolism and Antioxidant Effect in Rats fed Animal or Vegetable Protein and a Hypercholesterol Diet)

  • 본 연구는 동, 식물성 단백질 식이에 고콜레스테롤을 공급한 식이와 열수 추출한 녹차급여가 지질대사와 항산화계에 미치는 효과를 알아보고자 한국 사람들이 보통 전통차로 음용하는 조건인 $0.5\%$ 녹차열수 추출물을 동물성 단백질인 카제인,식품성 단백질인 대두 단백질과 고콜레스테롤을 체중이 약 180g이 되는 Sprague-Dawley종 수 컷 흰쥐에 공급하여 지방대사와 항산화계에 미치는 영향을 보고자하였다. 4주간 실험식이로 사육한 후 혈청지질성분과, 간의 지질성분, 간의 항산화 효소 활성과 과산화물인 TBARS의 양을 측정하였다. 그 결과는 다음과 같다. 1. 체중증가량과 FER은 전반적으로 대두단백질 군보다 카제인 군이 높았고 카제인 군에서는 녹차열수추출물을 공급하였을 때 체중증가량과 FER이 유의적으로 감소하였다(P < 0.05). 식이섭취량은 유의적인 차이를 보이지 않았다. 2. 혈청 중의 총콜레스테롤, LDL-콜레스테롤 농도는 전반적으로 대두단백질 군보다 카제인 군이 높았으나 카제인군의 LDL-콜레스테롤 농도만이 녹차열수추출물을 공급하엿을 때 유의하게 낮게 나타났다(P < 0.05). 3. 간의 중성지방 함량은 전반적으로 대두단백질 군보다 카제인 군이 높았으나 녹차열수추출물을 공급했을 때 대두단백질 군에서만 중성지방 함량이 유의하게 낮게 나타났다(P < 0.05). 4. 간의 항산화효소의 활성은 모든 군에서 유의한 차이가 없었으나 TBARS 함량은 녹차열수추출물을 공급했을 때 카제인 군에서만 유의하게 낮게 나타났다(P < 0.05). 이결과로 보면 식물성 단백질군 보다는 동물성 단백질군에 녹차 열수 추출물을 공급하였을 때 사료효율, 체중감소, 혈청 LDL-콜레스테롤과 간의 TBARS(thiovarbituric acid reactivesubstance) 생성을 낮추는 효과를 보여주었다. 간의 중성지방의 경우는 대두단백질 군에서만 녹차열수추물 공급에 따라 낮아지는 효과가 있었다(P < 0.05). 그러므로 녹차 열수 추출물이 지방대사와 항산화계에 미치는 영향은 식물성단백질 보다는 동물성 단백질이 공급된 식이에 건강에 바람직한 영향을 미치는 것으로 나타났다.
