This study was performed to investigate the effects of dried powder and juice of Aster scaber on lipid metabolism and antioxidative capacity in rats. Twenty one male Sprague-Dawley rats weighing 170.9$\pm$15.1g were blocked into 3 groups according to body weight and raised for 4 weeks with diets containing 5%(w/w) dried powder and juice from the same amount of Aster scaber. Food intake, body weight gain, food efficiency ratio and organ weights were not different among all the experimental groups. On lipid metabolism, both dried powder and juice supplementation decreased plasma total lipid, triglyceride(TG), total cholesterol and liver cholesterol levels by increasing fecal lipid excretions. Plasma thiobarbituric acid reactive substance(TBARS) concentrations were significantly decreased in dried powder and juice groups, whereas liver TBARS concentrations were not affected by experimental diets. Red blood cell(RBC) superoxide dismutase(SOD) activities were significantly increased by dried powder and juice supplementation, while activities of other antioxidative enzymes in RBC and liver were decreased or showed no changes. In conclusion, both Aster scaber dried powder and juice had the effects on lowering lipid levels by increasing fecal lipid excretions, and inhibiting of plasma lipid peroxidation in animals and the effects of juice were higher than that of dried powder. This study was performed to investigate the age-related changes of the lipid metabolism and thrombogenic capacity in Sprague-Dawley (SD) rats at the ages of 4, 8, 12, 16, 20 and 24 months old. Total lipid, triglyceride (TG) and total cholesterol in plasma and liver, HDL-cholesterol concentration, and eicosanoid contents in plasma were measured. Lipid peroxides were determined by the levels of thiobarbituric acid reactive substance (TBARS) in LDL fraction. Body weight was increased continuous until 16 months and decreased after 20 months. Epididymal fat pad (EFP) weight was increased continuously until 20 months and decreased at 24 months. Total lipid and TG concentrations in plasma were increased until 20 months and then rapidly decreased at 24 months but plasma cholesterol was increased continuously with aging. HDL-cholesterol level was increased continuously until 12 months, but decreased at 16 months and maintained there after. The TBARS levels in LDL fraction were the highest level at 24 months. Liver total lipid, TG, and total cholesterol concentrations were shown a tendency to increase with aging, and especially TG concentration was increased rapidly from 12 months to 16 months. Plasma thromboxane B$_2$ (TXB$_2$) and 6-keto-prostaglandin F$_1$ (6-keto-PGF$_1$) contents did not change with aging, but the ratio of TXB$_2$/6-keto-PGF$_1$ was increased with aging, especially from 8 to 12 months. These results showed that lipid levels in plasma and liver, TBARS levels in LDL fraction, and TXB$_2$/6-keto-PGF$_1$ ratio were increased with aging.
To investigate the effects of hot water soluble extract from Angelicae Gigantis Radix on lipid metabolism and antioxidant defense system in rats fed hypercholesterol diet, Sprague-Dawley male rats were fed either hypercholesterol diet with water or hot water soluble extract from Angelicae Gigantis Radix. Hypercholesterol diet contained 18 % beeftallow and 5 % cholesterol. After 4 weeks of experimental periods serum and liver obtained. The level of serum lipid and the activities of GOT and GPT was measured. The level of lipid and TBARS and the activity of GSH-Px was measured also. The results obtained were: 1) Food intake in the experimental group supplied hot water soluble extract from Angelicae Gigantis Radix was low significantly. 2) In the experimental group supplied hot water soluble extract from Angelicae Gigantis Radix of serum total lipid and total cholesterol was significantly low. 3) Serum GOT activity was significantly low in the experimental group supplied hotwater soluble extract from Angelicae Gigantis Radix. There was no difference of serum GPT activity between control and experimental group supplied hot water soluble extract from angelical Gigantis Radix. 4) There was no difference of lipid composition, TBARS level and GSH-px activity in liver between control and experimental group. The result of this study indicates that the hot water soluble extract from Angelicae Gigantis Radix was effective in decreasing the food intake of the experimental animals in the hypercholesterol diet and also effective in decreasing the levels of serum total lipid, total cholesterol and GOT activity. However it was not effective to change the level of lipid, TBARS and GSH-Px activity of liver.
Objectives : To investigate the anti-inflammatory effects of Prunella vulgaris pharmacopuncture in lipopolysaccharide (LPS)-induced inflammatory rat model. Methods : Sprague-Dawley rats were divided into 5 groups; normal control (n=8), LPS control (n=8), LPS+Prunella vulgaris pharmacopuncture at CV4 (CV4, n=8), LPS+Prunella vulgaris pharmacopuncture at ST36 (ST36, n=8), and LPS+Prunella vulgaris pharmacopuncture at CV12 (CV12, n=8). Pharmacopuncture was given every two days for 4 weeks followed by inflammation induction by peritoneal LPS injection (5mg/kg). Proinflammatory cytokines including interleukin-$1{\beta}$ (IL-$1{\beta}$), interleukin-6 (IL-6), tumor necrosis factor-$\alpha$ (TNF-$\alpha$), interleukin-10 (IL-10), thiobarbituric acid reactive substance (TBARS) from blood and liver tissue were compared before and 5 hrs after inflammation induction. Results : In CV4 and CV12 groups, plasma IL-$1{\beta}$, IL-6 and TNF-$\alpha$ levels increased by LPS injection, significantly decreased 5 hrs after injection (p<0.05). For CV12 group, plasma IL-10 concentration significantly increased (p<0.05). Liver IL-$1{\beta}$ and IL-6 levles significantly decreased in CV4 and CV12 groups (P<0.05), while normal and LPS control groups were not significantly different in TNF-$\alpha$ and IL-10 levels. Plasma TBARS concentration was significantly decreased in CV12 group, while there was no significant difference among LPS control and pharmacopuncture groups for liver TBARS concentration. Conclusions : Based on the present findings, Prunella vulgaris pharmacopuncture at CV12 may have a potentially preventive anti-inflammatory effect in an LPS-induced inflammatory rat model.
This study wa performed to investigate the effect of dired powder of chestnut and acorn on lipid metabolism, antioxidative capacity and antithrombotic effect in rats. Fifty-four male Sprague-Dawley rats weighing 199$\pm$17g were blocked into nine groups according to their body weight. Rats were raised with diets containing only flesh or flesh with inner skin of 5% and 10% dried nut powders for four weeks. Food intake, body weight gain, food efficiency ratio and organ weight were no different among the experimental groups. The plasma and liver lipid levels of all the nut diet groups were lover than those of the control group. The nut diets showed hypolipidenic effect in the plasma and liver. Plasma and liver thiobarbituric acid reactive substance (TBARS) concentrations were significantly decreased in all the nut diet groups. The plasma TBARS levels of the inner skin groups were significantly different from the control group dose-dependently. Superoxide dismutase(SOD) activity was significantly different among the experimental groups, and all the nut groups showed higher activity than the control group. There were significant differences in SOD activity between the chestnut and acorn groups and the chestnut groups showed higher erythrocyte SOD activity and the acorn groups showed higher liver SOD activity than the other groups. Whereas catalase and GSH-Px activities in the erythrocyte and liver of both nut groups showed a tendency to increase, they were not significantly different among the experimental groups. The bleeding time and whole blood clotting time tended to be extended by feeding both types of nut but they were not significantly different among the experimental groups. Production of TX $B_2$ and PG $F_{1{\alpha}}$ was no different among the experimental groups. These results suggest that chestnut and acorn diets have the effect of lowering plasma and liver lipid levels, inhibiting lipid peroxide formation and increasing antioxidative enzymes activity. Thus, it is plausible that chestnut and acorn could be recommended in the treatment and prevention of cardiovascular diseases.
Acetaminophen (N-acety1-p-aminophenol, paracetamol) is widely used as an over-the-counter analgesic and antipyretic drug. Intake of a over dose of acetaminophen may result in severe hepatic necrosis. In this study, we investigated the liver damage in mice using single dose (300 mg/kg) of acetaminophen and the possible protective effects of administration (50-200 mg/kg body weight) of SB-Ex on acetaminophen-induced liver damage in mice. The alanine aminotransferase (ALT), and aspartate aminotransferase (AST) activities were determined in the plasma of mice. The effect of SB-Ex on lipid peroxidation product thiobarbituric reacting substances (TBARS) and some antioxidant enzymes superoxide dismutase (SOD), catalase, d-aminolevulinate dehydratase (${\sigma}$-ALA-D) activities, and gluthathione peroxidase (GPx), were also evaluated in the mouse liver homogenate. Acetaminophen caused liver damage as evident by statistically significant increased in plasma activities of AST and ALT. There were general statistically significant losses in the activities of SOD, catalase, ${\sigma}$-ALA-D, and GPx and an increase in TBARS in the liver of acetaminophen-treated group compared with the control group. However, SB-Ex was able to counteract these effects. These results suggest that SB-Ex can act as hepatoprotectives against acetaminophen toxicity and is a good candidate for further evaluation as an effective chemotherapeutic agent.
The influences of dietary supplement of citron tea on the hepatocellular chemical carcinogenesis have been studied by examining placental glutathione S-transferase(GST-P) positive foci area in a liver tissue, contents of total cytochrome P450, thiobarbituric acid reactive substances(TBARS) and glucose 6-phosphatase(G6Pase) in hepatic microsome and glutathione S-transferase(GST) activity. Weaning Sprague-Dawley male rats were fed AIN76 diet with or without citron tea supplement. Rats of CTR and CTR+ groups were fed diet without citron tea supplement while CDI and CDI+ groups were fed diet with citron tea supplement for the entire experimental period(13 weeks). Rats of CDP and CDP+ groups were fed diet without citron tea supplement for the first 7 weeks and swiched to citron tea containing diet for the last 6 weeks of experimental period. CTR+, CDI+ and CDP+ groups were carcinogen treated group. Diethylnitrosamine(DEN) was used as a carcinogen initiator and injected to the rats of carcinogen treated groups as a single dose of 200 mg/kg body weight intraperitoneally after 4 weeks of feeding. 2-Acethylaminofiuorene(AAF) was used as a carcinogen promoter and supplied in the diets of carcinogen treated rats as 0.02% content for the last 6 weeks starting from 2 weeks after DEN injection. Rats were sacrificed after 13 weeks of feeding. Liver/body weight ratio and GST activities were increased by carcinogen treatment. However, they were not changed by citron tea supplement. Total cytochrome P450 contents were not changed by carcinogen treatment or citron tea supplement. TBARS contents of carcinogen treated rats showed tendency to decrease by citron tea supplement. G6Pase activity decreased by carcinogen treatment and citron tea supplement. The area of GST-P positive foci detected in carcinogen treated rats were decreased by citron tea supplement and not affected by the timing and the duration of citron tea supplement. These results suggest that citron tea has suppressive effects on hepatocellular chemical carcinogenesis probably through antioxidant compounds by decreasing TBARS contents.
Objevtives & Methods : Effects of cultivated wild ginseng pharmacopuncture at BL18 and LI11 on lipid composition, cytokine level, liver function, anti-oxidative capacity and histological characters were investigated in diet-induced obese rats. Forty male Sprague-Dawley rats weighing about 400g were divided into 4 groups of control, BL18, LI11 and BL18 plus LI11 pharmacopuncture groups and raised for 4 weeks. Results : 1. Plasma ${\beta}$-lipoprotein, free fatty acids level and TNF-${\alpha}$ levels significantly decreased in the pharmacopuncture groups compared to those of no treatment group. Plasma and liver total cholesterol, triglyceride, glucose and thiobarbituric acid reactive substance (TBARS) levels were also significantly lower than those of no treatment group. There was, however, no difference in TBARS level among pharmacopuncture groups. Liver total cholesterol level of BL18 pharmacopuncture group was lower than those of the other two pharmacopuncture groups. In LDL-cholesterol level, BL18 pharmacopuncture and BL18 plus LI11 pharmacopuncture groups only had significantly lower levels than that of no treatment group. 2. There was no significant difference between cultivated wild ginseng pharmacopuncture groups and no treatment group in IL-6, alanine transaminase (ALT), aspartic acid transaminase (AST) levels. 3. Compared with \ those of no treatment group, pharmacopuncture groups had significantly higher levels of HDL-cholesterol, superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and catalase activities. There was, however, no significant difference among pharmacopuncture groups. 4. Histological characters of heart, kidney and liver of BL18 pharmacopuncture group were similar to those of normal rats. Conclusions : These results indicate that cultivated wild ginseng pharmacopuncture at BL18 and LI11 may suppress adipose tissue mass and lipid peroxidation and activate antioxidant system.

Effect of Korean red ginseng (KRG) on the level of serum and liver lipids and lipid peroxidation was investigated in the rats fed high fat diet. Content of serum total cholesterol was significantly decreased (P<0.05) in KRG I group and KRG II group. Content of HDL-cholesterol was significantly increased by 69.75% and 39.15% in KRG I and KRG II group compared to control group, respectively. Atherogenic index (hi) was also significantly decreased by 74.76% and 37.38% in KRG I and KRG II groups compared to control group, respectively. Serum triglyceride content was significantly decreased (p<0.05) in only KRG II group. Antioxidative activity of KRG on the lipid peroxidation of serum and tissues in rats was also studied in vivo by measuring the formation of thiobarbituric acid reactive substances (TBARS). Contents of TBARS in the serum of both KRG groups were significantly decreased (p<0.05) and that of nonheme iron in serum was significantly increased (p<0.05) in a dose-dependent manner, which suggested that lipid peroxidation contents are inversely correlated with serum nonheme iron content. Content of TBARS in liver was significantly decreased (p<0.05) in KRG I and KRG II groups, without any influence in other tissues. Content of TBIARS in liver microsomal fractions stimulated by Fe$^{2+}$/ascorbate was significantly decreased (p<0.05) in KRG I and KRG II groups, whereas this observation did not occur in liver mitochondrial fractions. When the effect of KRG on TBARS content in the liver fractions of homogenates, microsomes, and mitochodria stimulated by Fe$^{2+}$/ascorbate was tested in vitro experimental model, TBARS of liver three fractions was significantly decreased at 6 mg/mL KRG compared with those of control. These results suggested that KRG powder have hypocholesterolemic effect as well as antioxidative effect in the serum and liver of the rats fed high fat diet.
