Listeria monocytogenes is a gram-positive, facultative anaerobe food pathogen responsible for the listeriosis that mostly occurs during the low-temperature storage of a cold cut or dairy products. To understand the systemic response to a wide range of growth temperatures, L. monocytogenes were cultivated at a different temperature from 10℃ to 42℃, then whole cell proteomic analysis has been performed both exponential and stationary cells. The specific growth rate increased proportionally with the increase in growth temperature. The maximum growth rate was observed at 37℃ and was maintained at 42℃. Global protein expression profiles mainly depended on the growth temperatures showing similar clusters between exponential and stationary phases. Expressed proteins were categorized by their belonging metabolic systems and then, evaluated the change of expression level in regard to the growth temperature and stages. DnaK, GroEL, GroES, GrpE, and CspB, which were the heat&cold shock response proteins, increased their expression with increasing the growth temperatures. In particular, GroES and CspB were expressed more than 100-fold than at low temperatures during the exponential phase. Meanwhile, CspL, another cold shock protein, overexpressed at a low temperature then exponentially decreased its expression to 65-folds. Chemotaxis protein CheV and flagella proteins were highly expressed at low temperatures and stationary phases. Housekeeping proteins maintained their expression levels constant regardless of growth temperature or growth phases. Most of the growth related proteins, which include central carbon catabolic enzymes, were highly expressed at 30℃ then decreased sharply at high growth temperatures.

Listeria monocytogenes is a well-known high-risk foodborne pathogen that grows at refrigeration temperature and is responsible for outbreaks of listeriosis. We report here the incidence of L. monocytogenes in fresh chicken carcasses and present genetic diversity of L. monocytogenes isolates. In this study, 25 g of chicken carcasses from markets in Korea were examined according to the FDA method, and presumptive isolates were confirmed by multiplex PCR assay. L. monocytogenes isolates were analyzed by Pulsed-Field Gel Electrophoresis using restriction enzymes, ApaI and AscI, to obtain strain-specific DNA fragments profiles. Antimicrobial resistance of L. monocytogenes strains against generally used antibiotics (Penicillin G, Kanamycin, Tetracycline, Vancomycin, Cephalothin, Rifampicin, Erythromycin, Ampicillin, Gentamicin, Streptomycin, and Chloramphenicol) were analyzed by NCCLS protocols to examine the presence of antimicrobial resistance in natural L. monocytogenes. Of a total 274 chicken samples, 81 samples (29.6%) were positive for L. monocytogenes. Listeria innocua (50.1%), Listeria welshimeri (6.9%), and Listeria grayi (11.3%) were also detected. PFGE analysis, using restriction enzymes ApaI and AscI, showed 27 pulsotypes of L. monocytogenes. Antimicrobial resistance analysis confirmed the existence of antimicrobial resistance for penicillin G and tetracycline in isolated L. monocytogenes strains.
The incidence of zoonoses in Korea has recently increased. But the study for high risk group such as pig farmers to zoonoses has not been conducted in Korea. Thus we reviewed the articles in order to obtain basic data for zoonoses among pig farmers, especially in rural communities. Pigs are one of the most important domestic livestock in Korea not only from economic standpoint but also from standpoint of food. Pigs also represent a potential reservoir for many novel pathogens, therefore may transmit these to humans via direct contact, vectors such as mosquitos, or contaminated meat. The zoonoses associated with pigs can be classified into bacterial pathogen, viruses and so on. Bacterial zoonoses include brucellosis, leptospirosis, listeriosis, enterohemorrhagic Escherichia coli infection, pasteurellosis, salmonellosis, yersiniosis, tuberculosis, anthrax, necrobacillosis, swine erysipelas, erysipeloid, melioidosis, Streptococcus suis infection, Clostrium difficile infection, and campylobactor infection. Viral zoonoses consist of Japanese encephalitis, swine influenza, Nipah virus, Reston ebolavirus, and hepatitis E virus infection. Other type of zoonoses include actinomycosis, toxoplasmosis and Taenia solium infection. These zoonoses were important in Korean health policy but lately they have been overlooked. For effective health policy, we need to study zoonoses associated with pigs, and clinicians and veterinarians must care deeply about these zoonoses.
본 연구에서는 위해 수준이 상대적으로 클 것으로 추정되는 가열하지 않고 그대로 섭취하는 신선 채소류를 대상으로 이들 식품군에서 많은 발생하는 Listeria monocytogenes에 대한 국내 오염수준 모니터링 자료와 신선 채소류의 섭취량, L. monocytogenes의 양-반응 모델(dose-response models)을 바탕으로 위해 수준을 추정할 수 있는 모델을 개발하였고, 이를 바탕으로 위해 수준을 평가하였다. 개발된 모델을 @RISK를 이용하여 시뮬레이션 한 결과 신선 채소류의 섭취를 통한 L. monocytogenes에 의한 1인1일당 리스테리아증 발생가능성은 평균 $8.76{\times}10^{-16}$로, 연간 천만명당 발생환자수는 $3.23{\times}10^{-6}$로 추종되었다. 또한 본 연구에서 개발된 모델을 바탕으로 추후 더욱 발전된 미생물위해평가(MRA) 연구와 결과를 도출하기 위한 제언을 추가하였다.
본 연구는 13과 37종 양치식물의 엽상체(지상부)와 근경의 메탄올 추출물을 대상으로 식품의 오염 및 변질의 원인이 되는 Bacillus subtils, 식중독의 원인이 되는 Escherichia coli, 식중독 및 선회병(listeriosis)의 원인이 되는 Listeria monocytogenes, 피부 여드름의 원인이 되는 Propionibacterium acnes에 대한 항균활성을 탐색하고 항균력이 우수한 천연소재를 선발하기 위하여 시행하였다. $5mg{\cdot}mL^{-1}$의 농도의 추출물을 대상으로 각 미생물에 대하여 50% 이상의 항균활성을 나타낸 양치식물 추출물을 선발하였으며, 선발된 추출물을 2mg/disc의 농도로 조절하여 agar diffusion법으로 항균력을 측정하여 15mm 이상의 억제환을 나타내는 추출물을 선발하였다. 상기의 방법으로 선발된 추출물을 $0.125{\sim}2.000mg{\cdot}mL^{-1}$의 농도로 조절하여 72시간 동안 각 미생육의 생육에 미치는 영향을 분석하여 항균활성이 우수한 양치식물의 추출물을 선발하였다. 연구의 결과, B. subtils의 생육억제에는 야산고비의 성엽과 쇠고비의 근경 추출물, E. coli의 생육 억제에는 설설고사리의 성엽과 나도히초미 및 더부살이고사리의 근경 추출물, L. monocytogenes의 생육억제에는 관중의 성엽과 공작고사리, 꿩고비 및 쇠고비의 추출물이 효과적이었다. 한편 대부분의 양치식물 추출물은 P. acnes의 생육억제 효과가 미비하였으나, 관중과 참지네고사리는 성엽과 근경 모두에서 P. acnes의 생육억제 효과가 매우 우수하였으며, 별도의 제균처리를 하지 않은 관중과 참지네고사리의 메탄올 조추출물의 $MIC_{50}$은 성엽의 추출물에서 66.8과 $80.4{\mu}g{\cdot}mL^{-1}$, 근경의 추출물에서는 8.8과 $54.2{\mu}g{\cdot}mL^{-1}$로 나타났다.
Listeria monocytogenes is an important foodborne pathogen that comprises four genetic lineages: I, II, III, and IV. Of these, lineage II is frequently recovered from foods and environments and responsible for the increasing incidence of human listeriosis. In this study, the phylogenetic structure of lineage II was determined through sequencing analysis of the ascB-dapE internalin cluster. Fifteen sequence types proposed by multilocus sequence typing based on nine housekeeping genes were grouped into three distinct sublineages, IIA, IIB, and IIC. Organization of the ascB-dapE internalin cluster could serve as a molecular marker for these sublineages, with inlGHE, inlGC2DE, and inlC2DE for IIA, IIB, and IIC, respectively. These sublineages displayed specific genetic and phenotypic characteristics. IIA and IIC showed a higher frequency of recombination (${\rho}/{\theta}$). However, recombination events had greater effect (r/m) on IIB, leading to its high nucleotide diversity. Moreover, IIA and IIB harbored a wider range of internalin and stress-response genes, and possessed higher nisin tolerance, whereas IIC contained the largest portion of low-virulent strains owing to premature stop codons in inlA. The results of this study indicate that IIA, IIB, and IIC might occupy different ecological niches, and IIB might have a better adaptation to a broad range of environmental niches.
Background: Listeria monocytogenes is a gram-positive bacterium that causes listeriosis mainly in immunocompromised hosts. It can also cause foodborne outbreaks and has the ability to adapt to various environments. Peptide uptake in gram-positive bacteria is enabled by oligopeptide permeases (Opp) in a process that depends on ATP hydrolysis by OppD and F. Previously a putative protein Lmo2193 was predicted to be OppD, but little is known about the role of OppD in major processes of L. monocytogenes, such as growth, virulence, and biofilm formation. Objectives: To determine whether the virulence traits of L. monocytogenes are related to OppD. Methods: In this study, Lmo2193 gene deletion and complementation strains of L. monocytogenes were generated and compared with a wild-type strain for the following: adhesiveness, invasion ability, intracellular survival, proliferation, 50% lethal dose (LD50) to mice, and the amount bacteria in the mouse liver, spleen, and brain. Results: The results showed that virulence of the deletion strain was 1.34 and 0.5 orders of magnitude higher than that of the wild-type and complementation strains, respectively. The function of Lmo2193 was predicted and verified as OppD from the ATPase superfamily. Deletion of lmo2193 affected the normal growth of L. monocytogenes, reduced its virulence in cells and mice, and affected its ability to form biofilms. Conclusions: Deletion of the oligopeptide transporter Lmo2193 decreases the virulence of L. monocytogenes. These effects may be related to OppD's function, which provides a new perspective on the regulation of oligopeptide transporters in L. monocytogenes.
2022년 1월부터 11월까지 부산지역에서 유통 중인 신선 농산물 210건을 대상으로 리스테리아균 분포 현황 및 병원성 여부를 조사하였으며, 분리된 균주를 대상으로 혈청형 및 유전자 지문분석을 통해 역학적 연관성을 확인하였다. 조사 대상 신선농산물에서 총 40건의 리스테리아균이 검출되었으며, Listeria monocytogenes 등 4종의 리스테리아균이 검출되었다. 이 중 L. innocua가 22건, L. monocytogenes 10건, L. grayi 6건, L. rocourtiae 2건으로 나타났으며, 이 중 식중독을 유발하는 L. monocytogenes균은 팽이버섯에서만 검출되었다. L. monocytogenes의 병원성을 유발하는 유전자인 iap, prfA, inlA, inlC, inlJ 및 hly 6종에 대한 분석 결과, 총 10개 균주 중 6개 균주에서 iap 등 6종의 병원성 유발 유전자가 검출되었으며, 4개 균주에서 hly를 제외한 5종의 유전자가 검출되어 식중독 발생 잠재위험이 있음이 확인되었다. 지역 유통 식재료에 분포하는 L. monocytogenes의 유전학적 유사도 및 오염원 추이를 확인하기 위해 신선농산물에서 분리한 L. monocytogenes 10균주 및 2022년 부산지역 유통 가금류에서 분리한 L. monocytogenes 2균주를 대상으로 혈청형 분석 및 PFGE를 실시한 결과, 신선농산물 분리균주의 혈청형은 1/2a 및 1/2b 두 가지 serotype으로 확인되었으며, 가금류 분리균주는 모두 1/2a형으로 나타났다. 유전자지문 분석결과, 전체 균주의 유사도는 100-45.7%로 나타났고, 이 중 100% 상동성을 보인 균주들은 동일 생산농장 또는 동일지역 유래 팽이버섯에서 분리되어 오염원의 출처가 같음을 추측할 수 있었다. 신선농산물 분리균주와 유통 가금육 분리균주와의 유사도 확인 결과, 일부 팽이버섯 분리균주와 가금육 분리균주의 유사도가 90.9-84.6%로 나타났다. 농산물 및 축산물 생산시설간 오염원 이동 및 교차 가능성을 유추할 수 있었다.
L. monocytogenes는 Listeriosis를 일으키는 중요한 식중독 균으로 현재 국내 식품공전에서는 증균배양을 기초로 검출하며, 규격은 불검출로 관리하고 있다. 그러나 Listeria종 간의 혼합오염시 증균 과정에서 경쟁생육이 존재하여 L. monocytogenes 위음성의 가능성이 있다고 보고되고 있다. 국내 식품공전은 L. monocytogenes 증균을 위한 1차 배지로 규정되어 있으나 LEB 배지에서의 Listeria 종 간의 생육 연구는 보고된 바 없다. 본 연구는 식품에서 주로 검출되는 Listeria 속 4종(L. monocytogenes, L. innocua, L. ivanovii, L. seeligeri)을 LEB배지에 혼합배양하며 증균과정에서 생육의 차이가 존재하는 것을 확인하였다. 특히, L. innocua에 의해 L. monocytogenes의 생육이 저해되며, L monocytogenes가 L. innocua보다 초기균수가 2.0 log CFU/mL 이상 오염이 되어있어야지만 L. innocua보다 생육이 잘 되는 것을 확인하였다. Listeria 종 간의 혼합오염이 있을 경우 현재 검출법으로는 L. monocytogenes의 검출이 어려울 수 있다고 판단된다. 따라서 L. monocytogenes 검출율을 높이는 새로운 증균배지 개발의 필요성을 확인하였다. 향후 본 연구는 L. monocytogenes 검출률을 높여 국내 식품의 식품 안전에 기여 할 수 있으며 국내 식품 관리 규격 개정 시 기초가 되는 참고 자료로 활용 할 수 있을 것으로 생각된다.
Listeria sp. is one of the pathogenic bacteria causes the infection listeriosis, through mainly raw food such as fishery food, dairy food and vegetables. Listeria sp. is a Gram-positive, non-spore-forming, motile, and facultative anaerobic bacterium. Because of the tolerance of Listeria sp. to low temperature and high salt concentration, it is very difficult to prevent them contaminated in the food, which do not require heating, especially, such as raw fishery products. So prevention and removal of bacterial contamination at the food manufacturing stage is the best method. In this study, therefore, several natural products including Psidium guajava and Geranium thunbergii were screened to investigate the antibacterial activity against Listeria sp., with expectation of fewer side effects and fewer resistance problems. Significant effects of two extracts were confirmed by well diffusion assay, MIC assay, and growth inhibition assay. P. guajava and G. thunbergii showed MIC values at 64-256 ㎍/mL meaning strong antibacterial activities against 6 kind of Listeria sp. tested. And the growth of Listeria sp. in the liquid media was actually inhibited by the addition of these two extracts.
