Quantitative analysis of cordycepin in Cordyceps militaris under different extraction methods

  • Cordyceps militaris (CM) is one of the most important medicinal mushrooms known to possess various biological activities. Cordycepin (CP) is a bioactive compound present in the fruiting bodies of CM and is known to have anti-tumor, anti-metastatic immunomodulatory and anti-inflammatory activities. In this study, we aim to analyze CP quantitatively under various CM extraction conditions. CP was measured using high-performance liquid chromatography, quantified using a reversed phase column using a gradient elution system of water and acetonitrile, and detected with a UV absorbance wavelength of 260 nm. The CP content of CM was the highest in 100% ethanol extract of the fruiting bodies and 60% ethanol extract of the mycelium. This study provides an efficient analysis method to determine the optimal extraction conditions for CP that can be used as a basis for developing functional foods and pharmaceutical products derived from CM.

  • Shotcrete (or Sprayed concrete) has been used as an important support material in New Austrian Tunnelling Method (NATM). Since the mid of 1990, permanent shotcrete tunnel linings such as Single-shell, NMT (Norwegian Method of Tunnelling) has been constructed in many countries for reducing the construction time and lowing construction costs instead of conventional in-situ concrete linings. Among essential technologies for successful application of permanent shotcrcte linings, high performance shotcrete providing high strength, high durability, better pumpability has to be developed in advance as an integral component. This paper presents the Ideas and first experimental attempts to increase early strength and bond strength of wet-mixed Steel Fiber Reinforced Shotcrete(SFRS) in sump water condition. In order to increase early strength, a new approach using high-early strength cement with liquid alkali-free accelerator has been investigated From the results, wet-mix SFRS with high-early strength cement and alkali-free accelerator exhibited excellent early strength improvement compared to the ordinary portland content and good bond strength even under sump water condition.

  • The hydration for the system of fluorogypsum and 20wt% blast furnace slag was investigated at liquid/soild ratio of 0.45 for 1, 3, and 7 days by using 3 kinds of accelerators such as K2SO4 Al2(SO4)3 $.$16-18H2O and AlK(SO4)2$.$12H2O After curing the hardened specimen was characterized by the compressive strength the content of combined water XRD, DTA and SEM It was found that the activating effect was increased in the order of K2SO4

  • Charicteristics of piggery waste and treatment processing sludges for reuse were investigated. If it was thoroughly regulated in disinfectants, antibiotic substances and heavy metals, raw piggery waste can be gratified in criteria for fermentative compost (liquid) for flowers cultivation. Also, Because it is satisfied with various criteria of heavy metals and fertilizer contents for reuse except water content, primary pre-treatment sludge is very good material for composting. If provated goods on heavy metals are used in coagulation & dewatering process, coagulation & dewatering sludges are suitable for criteria of special waste regulation and by-product compost. This study proves that, if they are accomplished with suitable composting and mature process, piggery waste and processing sludges are free from microbiological problems as well as criteria of composting.

Preparation of Instant Powdered Soup using Canned Oyster Processing Waste Water and Its Characteristics (굴통조림 부산액을 이용한 인스턴트 분말수프의 제조 및 특성)

  • To utilize canned oyster processing waste water effectively, this study was carried out to prepare instant powdered soup using the waste water (IPSW), Instant powdered souu from oyster hot-water extracts (IPSE) was prepared by mixing hot-water extracts powder (15 g) with table salt (5 g), cream powder (19 g), milk replacer (12 g), wheat flour (20 g), corn flour (15 g), starch (5 g), glucose (7.5 g) and onion powder (1.5 g). In preparing IPSW, mixed powder from wash water and boiling liquid waste, instead of powder from hot-water extracts and table salt, was added (powder from boiling liquid waste: powder from wash water= 12: 8) and other additives were added in proportion to those in the IPSE, The IPSW consists mainly of carbohydrates (about $72\%$). It was not different from the IPSE. The volatile basic nitrogen, viable cell counts, coliform group of the IPSW contains 33.4 mg/100g, $2.2\times10^4CFU/g$, <180 MPN/100g, respectively, and its water activity has 0.257. So it was a hygienically safe and conservable instant food. The main fatty acids of IPSW were 16: 0 and 18: 1n-9. Its chemical score of protein was $61.4\%$ and its main inorganic matter was iron. According to a sensual evaluation, in contrast to the IPSE, the IPSW had a bit lower aroma but better taste, It was concluded from the above chemical and sensory evaluation that even the boiling liquid waste which had been mostly abandoned because of its high table salt content can be used as a good material for instant powdered soup if it's powdered and mixed adequately with powder from wash water, and its table salt content is properly adjusted.

  • The measurement of icing conditions needs to be carried out accurately by the ice detector system of an aircraft. Ice detector systems should be installed in locations not affected by backwash, rotor downwash or moving doors or other equipment. Various analyses were carried out in order to find the proper locations sufficiently far from these interfering effects. In this study, the optimum position of the ice detector was assessed using computer simulation, with respect to different flight modes, flow velocities and the amount and distribution of liquid water around the sensor.

  • The most efficient method for reducing the organic content of dilute liquid waste is by aerobic-biological treatment. Basically, the organisms responsible for treatment possess the ability to decompose complex organic compounds and to use the energy so liberated for their bodily functions: reproduction, growth, locomotion and so on. That part of organic matter used to produce energy is converted to the essentially stable end products of carbon dioxide, water and ammonia, while the remainder is converted to new cells which can be settled and thus removed from the liquid before the waste is discharged to the receiving body water. Oxygen must be supplied continuously during the aerobic process. In the field of sewage treatment the Warburg respirometer is used mainly for the measurement of the oxygen uptake of samples. In this experiment the Warburg constant volume respirometer was used to determine the oxygen uptake by bacteria in the presence of various glucose concentrations. The rate of oxygen uptake by the bacteria was expressed as the respiratory quotient. The result indicated that the oxygen uptake was proportional to the glucose concentration. Oxalic Acid from Lentinula edodes Culture Filtrate: Antimicrobial Activity on Phytopathogenic Bacteria and Qualitative and Quantitative Analyses

  • The culture filtrate of Lentinula edodes shows potent antimicrobial activity against the plant pathogenic bacteria Ralstonia solanacearum. Bioassay-guided fractionation was conducted using Diaion HP-20 column chromatography, and the insoluble active compound was not adsorbed on the resin. Further fractionation by high-performance liquid chromatography (HPLC) suggested that the active compounds were organic acids. Nine organic acids were detected in the culture filtrate of L. edodes; oxalic acid was the major component and exhibited antibacterial activity against nine different phytopathogenic bacteria. Quantitative analysis by HPLC revealed that the content of oxalic acid was higher in the water extract from spent mushroom substrate than in liquid culture. This suggests that the water extract of spent L. edodes substrate is an eco-friendly control agent for plant diseases.

토양내 총 NAPL과 공기접촉 NAPL의 측정을 위한 분별 NAPL 분배 추적자 기술의 개발

  • Gaseous partitioning tracer test has been used for determining the volume and spatial distribution of residual non-aqueous phase liquid (NAPL) in the unsaturated soils. In this study, an experimental method for measuring the content of gas-exposed NAPL as well as that of total NAPL in a sand during air sparging was developed. Two different gaseous phase NAPL-partitioning tracers were used; n-pentane, with very low water solubility, was used as the tracer that partitions into NAPL that is only in contact with the mobile gas, and chloroform, with fairly good water solubility, was selected for detecting total NAPL content in the sand. Helium and difluromethanewere used as the non- reactive tracer and water-partitioning tracers, respectively. Using n-decane as a model NAPL (NAPL saturation of 0.018), 25.6% of total NAPL was detected by n-pentane at the water saturation of 0.68. Oniy 9.1% of total NAPL was detected by n-pentane at the water saturation of 0.84. This result implies that the quantity of gas-exposed NAPL increased about three times when the water saturation decreased from 0.84 to 0.68. At the water saturation of 0.68, more than 90% of total NAPL was detected by chloroform while 65.8% of total NAPL was detected by chloroform at the water saturation of 0.84. Considering that the removal rate of NAPL during air sparging for NAPL-contaminated aquifer is expected to be greatly dependent upon the spatial arrangement of NAPL phase with respect to the mobile gas, this new approach may provide useful information for investigating the mass transfer process during air-driven remedial processes fer NAPL-contaminated subsurface environment.

Component Characteristics of Canned Oyster Processing Waste Water as a Food Resource (식품소재로서 굴통조림 가공부산액의 성분 특성)

  • As a part of basic investigation for utilization of canned food processing by-products, a food components of the canned oyster processing waste water such as boiled and released water(BRW), wash water(WW) were investigated and compared with hot-water extracts from oyster. From the results of measuring heavy metal conte수, viable cells and coliform group, the canned oyster processing waste waters might not invoke health risk in using food resource. The contents of taste compounds (free amino acids, ATP related compounds, TMA (O) and total creatinine) of BRW and WW accounted for about 254% and 95%, respectively, in comparison with those of control (hot-water extract from oyster). The BRW showed a very high content of salt in comparing to the WW and control. In descending order, the values of whiteness index was WW, control and BRW. Sensory scores for color, oyster flavor intensity and saline taste were not significantly different between WW and control. But, BRW had the highest score in oyster flavor intensity, while had the lowest score in color and saline taste. But, the color and saline taste of BRW might be able to control by some pretreatment (concentration and drying in mild condition, desalination and recipe control etc). These results indicated that BRW and WW generated from various step during canned oyster processing could be a potential food resource by controlling of saline taste and color intensity.
