Cryopreseryation and Germination of Native Aquilegia Species Seeds by Predehydration Treatment (건조 전처리에 의한 자생 Aquilegia속 식물 종자의 초저온 저장과 발아)

  • Predehydration effects for cryopreservation in -196$^{\circ}C$ liquid nitrogen were studied in Korean native Aquilegia buergeriana var. oxysepala Kitamura and A. flabellata var. pumila Kudo seeds. Aquilegia species seeds were adjusted to moisture contents between 3.2 and 9.7% by air dry treatments. Seeds were placed in paper envelopes after submerged in liquid nitrogen and rewarming in 38 $^{\circ}C$ water. Seeds moisture contents by duration of drying were identified as controlling factors in the survival of Aquilegia species seeds for cryopreservation. Aquilegia species seeds having approximate 5% moisture content were able to withstand cooling to -l96$^{\circ}C$. Undehydrated seeds of Aquilegia buergeriana var. oxysepala Kitamura after being cryopreservated in liquid nitrogen have a 10.9% of moisture content and show 52.5% in germination. But, Aquilegia buergeriana var. oxysepala Kitamura seeds dehydrated by drying for 60 min. to have 6.0% of moisture content before cryopreservation show 84.7% in germination test. Properly dehydrated seeds after being stored in liquid nitrogen showed over 60% in germination rate and also shows an uniform sprouting time,11~13 days in average. Any morphologically deformity in germinating beds has not been observed. Results from this study suggest that Aquilegia species seeds can cryopreservation in liquid nitrogen if the seed moisture content is controled by a proper amount of dehydration.

Fluoride Removal Using Ready-Mixed Concrete Sludge (레미콘 슬러지를 이용한 불소제거)

  • The purpose of this study was performed to investigate the optimum conditions of pH, concrete sludge, seed dosage, mixing intensity, operation time in treating fluoride-containing wastewater as $CaF_2$ using the ready-mixed concrete sludge. Considering fluoride removal, water content, that pH 6, concrete sludge dosage of 10 g/L, Seed dosage ($CaF_2$) of 2 g/L, mixing intensity of 100 rpm and operation time of 60 min were found to be optimum. Correspondingly, removal of fluoride and water content was about 85% and 64%, respectively. Increase in amount of seed dosage did not affect fluoride removal efficiency. but the result that the water content is decreased was shown up in occuring the solid-liquid separation well.

Analytical Determination of Vitamin B12 Content in Infant and Toddler Milk Formulas by Liquid Chromatography Tandem Mass Spectrometry (LC-MS/MS)

  • The development of a sample preparation method and optimization of the analytical instrumentation conditions were performed for the determination of the vitamin B12 content in emulsified baby foods sold on the Korea market. After removal of the milk protein and fats by chloroform extraction and centrifugation, the vitamin B12 was water extracted from the sample. Following filtration of the solution through a nylon filter, the water-soluble extract was purified by solid-phase extraction using a Liquid Chromatography Tandem Mass Spectrometry (LC-MS/MS). The solution eluted from the cartridge was dried under a stream of nitrogen gas and reconstituted with 1 mL of water. The sample solution was injected into an LC-MS/MS system after optimizing the mobile phase for vitamin B12 detection. The calibration curve showed good linearity with the coefficient of correlation (r2) value of 0.9999. The limit of detection was 0.03 µg/L and the limit of quantitation was 0.1 µg/L. The method of detection limit was 0.02 µg/kg. The vitamin B12 recovery from a spiking test was 99.62% for infant formula and 99.46% for cereal-based baby food. The sample preparation method developed in this study would be appropriate for the rapid determination of the vitamin B12 content in infant formula and baby foods with emulsified milk characteristics. The ability to obtain stable results more quickly and efficiently would also allow governments to exercise a more extensive quality control inspection and monitoring of products expected to contain vitamin B12. This method could be implemented in laboratories that require time and labor saving.

Marker compounds contents of Salvia miltiorrhiza Radix depending on the cultivation regions

  • Salvia miltiorrhiza Radix is cultivated in Korea and China and is traditionally used to treat cardiovascular diseases. In this study, we developed and validated a quantitative analysis method for S. miltiorrhiza Radix using high-performance liquid chromatography (HPLC). Identification was performed using ultra performance liquid chromatography-tandem mass spectrometry. For quantitative analysis, we used seven marker compounds. Separation conditions for HPLC were optimized using an ODS column with gradient conditions of 1% formic acid in distilled water and 1% formic acid in acetonitrile, with a flow rate of 0.8 mL/min and a detection wavelength of 280 nm. This method showed good linearity ($R^2=0.9998$), precision (relative standard deviation ${\leq}3.3%$), accuracy (recovery of 94.16-102.89%), limit of detection ($7.53{\mu}g/mL$), and limit of quantification ($23.71{\mu}g/mL$). This approach successfully quantified marker compounds in S. miltiorrhiza Radix. The individual marker compounds were identified by comparing the molecular masses and retention times with does standard compounds. Marker compound contents of S. miltiorrhiza Radix were investigated with different cultivation regions. Seven marker compounds were detected and quantified in all samples. Among them, salvianolic acid B showed the highest contents and it ranged from 4.13 to 7.15%. The salvianolic acid B content (7.15%) of marker compound was the highest in Bonghwa, and the tanshinone IIA content (1.90%) was the highest in Pohang. The results of marker compounds and developed method were intended to provide a favorable reference for the study of S. miltiorrhiza Radix from different regions of Korea.

  • In this study, using the governing equation for thermohydrodyamic lubrication involving the homogeneous mixture of incompressible fluid derived by based on the principle of continuum mechanics, it is discussed the effects of water within engine oil on the performance of high speed journal bearing of a turbocharger. The governing equations are the general equations being able to be applied on the mixture of Newtonian fluid and non- Newtonian fluid. Here, the fluid viscosity index, n of power-law non-Newtonian fluid is supposed to be 1 for the application of the journal bearing in a turbocharger lubricated with the mixture of two Newtonian fluid, for example, water within engine oil. The results related with the bearing performance are shown that the bearing friction is to decrease and the side leakage and bearing load increase as increasing the water content in an engine oil.

  • It is well-known fact that dimensional stability of wood is greatly enhanced by acetylation of wood. This dimensional stability results from bulking of the reacted acetate within the cell wall, which reduces further swelling when the modified woods corne into contact with water or water vapor. The purpose of this research was to determine the water absorption and dimensional stability of the acetylated solid wood in liquid water and in humidity tests. Radiata pine, Siberian larch, and Japanese larch were acetylated. Antiswelling efficiency(ASE) and equilibrium moisture content(EMC) of acetylated wood, at 95% RH and $20^{\circ}C$ after 4weeks, was quite dependent on WPG, but was not dependent on species.

Analysis of γ-Aminobutyric Acid Content in Fermented Plant Products by HPLC/UV

  • ${\gamma}$-Aminobutyric acid (GABA) content in fermented plant products and their main plant materials (aerial part of Acanthopanax sessiliflorus, fruit of Crataegus pinnatifida, and whole plant of Morus alba) was determined by high-performance liquid chromatography. GABA was quantified using a reverse-phase column with a gradient elution program (water:acetonitrile =90:10 to 0:100 for 40 min). UV detection was conducted at 280 nm. GABA content was measured in fermented plant products (15.07 mg/g), aerial part of A. sessiliflorus (4.49 mg/g), fruit of C. pinnatifida (10.59 mg/g), and whole plant of M. alba (2.31 mg/g). The presence of GABA in fermented plant products, including A. sessiliflorus, C. pinnatifida, and M. alba is important in industrial application for health supplements.

Effect of Addition of Egg Yolk Lecithin on the Lipid Oxidation of a Water/canola Oil Emulsion (달걀 노른자위 레시틴의 첨가가 물/카놀라 기름 에멀션의 지방질 산화에 미치는 영향)

  • Effect of the addition of egg yolk lecithin at a concentration of 350 mg/kg on iron-catalyzed autoxidation and chlorophyll-photosensitized oxidation of a water/canola oil emulsion (W/O) during storage at $25^{\circ}C$ was studied based on headspace oxygen consumption and hydroperoxide production. Changes in the phospholipid (PL) composition of the emulsion were determined by high performance liquid chromatography. Headspace oxygen consumption and hydroperoxide content of the emulsion increased with storage time, and addition of egg yolk lecithin did not have any significant effect on these parameters during iron-catalyzed autoxidation and chlorophyll-photosensitized oxidation of the emulsion. PL content of the emulsion decreased during both oxidations, and the degradation rate was higher during autoxidation than during photosensitized oxidation. Phosphatidylcholine content ratio tended to increase during autoxidation. The results suggest that egg yolk lecithin in canola oil emulsion behaves differently during iron-catalyzed autoxidation and chlorophyll-photosensitized oxidation.

Application Level of Anaerobic Digestion Waste Water from Methane Fermentation of Pig Manure on Rice (벼에 대한 돈분뇨 혐기성 소화액비의 시용기준 연구)

  • This study was conducted to evaluate the effect of the proper application level of anaerobic digestion waste water on rice. The waste water was from methane fermentation of pig manure to use as a liquid manure. The mixture treatment of 70% liquid manure and 30% chemical fertilizer (LM 70%+CF 30%) and 100% liquid manure (LM 100%) treatment were higher number of tiller than other treatments at the both tillering and heading stages of rice. The yields of LM 70%+CF 30% and LM 100% treatments were a little higher than that of NPK treatment, but the mixture treatment of 50% liquid manure and 50% chemical fertilizer (LM 50%+CF 50%) was a little lower yield than NPK treatment. The periodic changes of the $NH_4-N$ and $NO_3-N$ contents of the NPK and the LM 50%+CF 50% treatments in paddy soil were a little higher than those of other treatments at the early stage of rice. The $NH_4-N$ contents of NPK and the LM 50%+CF 50% treatments in irrigation water quality were higher than those of other treatments, however there was no difference in $NO_3-N$ content among the treatments. The $NH_4-N$ and $NO_3-N$ contents of non fertilizer treatment in infiltration water quality were leached a little higher than those of other treatments. It may be due to poor growth of rice following to reduce the nutrient uptake by rice and to increase relatively the nutrient leaching to the ground water. The proper application level of anaerobic digestion waste water as a liquid manure could be suggested to apply LM 70%+CF 30%. All treatments were the same amount of nitrogen content for the standard application amount on rice.

  • Lyocell yarns were treated with NaOH, liquid ammonia, high pressure steam and polycarboxylic acids, and dyed with five reactive dyes. The water content of the samples was also measured by the centrifugal and chromatographic techniques. The total pore volume w as substantially increased by NaOH treatment, while decreased by high pressure steaming. The pore size distributions for the dyed samples were affected by the dyes used.
