This study was carried out to establish improved techniques on organogenesis from callus culture of Gladiolus. Organogenesis from the callus was effective in the half strength of MS solid medium without 2,4-D at 15 $^{\circ}C$ under 24 hours of daylength. Formation of adventitious root was most effective in the liquid shaking culture, and adventitious shoot induction was effective in the liquid stationary culture. From these results, we could find optimal culture conditions for redifferentiation from callus, in addition, liquid shaking culture revealed as more useful when compared with that of solid culture method for the redifferentiation of callus in Gladiolus `Topaz'.
The removing copper from CCA, CCFZ treated wood were tested with Tyromyces palustris. The shacking culture, solid culture, and stationary culture methods were tested to removal of copper in CCA, CCFZ treated wood. The steam pressure method and oxalic acid method were tested as pretreatment. To investigate of copper removal mechanism, the oxalic acid that produced by T. palustris and combination with copper was examined on stationary culture that contained CCA and CCFZ treated wood chips. Oxalic acid increased copper removing rate of T. palustris as pretreatment. Stationary culture had most removal effectiveness among the culturing methods. The removal rate of copper on bioreactor was 61% on 7 days. Copper-oxalate was detected on liquid culture, which show the oxalate from liquid of culture combined with copper from treated wood. It would be related to the removal of copper from treated wood by T. palustris.
Various parameters such as solvent selection, concentration, solid/liquid ratio, soaking time, temperature, stationary, shaking conditions, and repeated extractions were investigated in order to determine the optimum extraction conditions of ${\beta}$-glucosidase from bagasse fermented by mixed culture of Aspergillus niger NRC 7A and Aspergillus oryzae NRRL 447. Among various solvents tested, non ionic detergents gave the best results than the inorganic or organic salt solutions and distilled water. The optimum conditions for extraction of ${\beta}$-glucosidase were 30 min soaking time at $40^{\circ}C$ under shaking condition at 150 rpm, with solid/liquid ratio 1:15 (w/v), which yielded $2882.74{\pm}95.52U/g$ fermented culture (g fc) of enzyme activity. With repeated washes under the above optimum conditions, the results showed that enzyme extracted in the $1^{st}$ and $2^{nd}$ washes represents about 90% of the total activity.
In order to clarify the best cultural conditions of Rhizopus niveus the effects of aeration, pH and various nutrients, such as different carbon and nitrogen sources, vitamins, and growth substances, on the mycelial growth were studied through liquid culture, and amylase activities of the fungus at different cultural periods were measured. Soluble starch, xylose and galactose are excellent sources of carbon for growth of the fungus. Sorbose and lactose are utilized slightly for growth. peptone, ammonium sulfate and alanine are excellent nitrogen sources for growth, tyrptophane nad potassium nitrate are utilized slightly for growth and sodium nitrite is not utilized. Thiamine nad gibberellin are excellent growth substances for the fungal growth, and biotin, nicotinamide and indole acetic acid (IAA) are also effective. Rhizopus niveus grows better at rotatory culture than at stationary culture and earlier growth of the fungus increases remarkably at rotatory culture. Optimum pH than at pH3. Growth increases linerly with an increase of soluble starch content up to 100g per liter medium, but 5 grams of ammonium sulfate per liter is the optimum nitrogen concentration for growth, if Pfeffer's medium is employed. Amylase activities of Rhizopus at different cultural periods showed that the maximum amylase production takes place after the cell population has reached its peak in the culture. Dextrinogenic amylase production has reached maximum at stationary phase, and maximum saccharogenic maylase production takes place in the pahse of negative gorwth acceleration.
The effect of nutrient nitrogen on the degradation of pentachlorophenol (PCP) by Phanerochaete chrysosporium in a liquid culture was investigated. PCP disappeared at almost the same rate in both nutrient nitrogen-sufficient (NS) and -limited (NL) sttionary cultures. However, more pentachloroanisole (PCA) was accumulated in the NS culture than in the NL culture. The effect of nitrogen on the degradation of PCA was also tested in both cultures. PCA disappeared faster in the NL culture than in the NS culture, indicating that the lower accumulation of PCA during the degradation of PCP in the NL culture was due to the faster degradation of PCA in the NL culture than in the NS culture. In another experiment, PCA was added to shaking cultures rather than stationary cultures to search for any other metabolite(s). While no other metabolite but PCA was found in the NS stationary culture, 2,4,5,6-tetrachloro-2,5-cyclohexadiene-1,4-dione(TCHD) was found as the only indentifiable product in the NL shaking culture. Thus, PCP would appear to be metabolized to TCHD via PCa or directly oxidized to TCHD by lignin peroxidase. Since all the above results indicate that no innocuous metabolite was formed during the degradation of PCP by the fungus, it is quite feasible to use the fungus in the biotreatment of PCP.
Present studies are carried out to find media components and culture methods for in vitro propagation of Athyrium niponicum and to establish the optimal economic masspropagation systems. Among pinnae, petiole and rhizome segments only rhizome segments produced young plants. Rhizome segments showed vigorous plant regeneration on 1/2MS medium and supplement to 1% sucrose and 50 $mg{\cdot}L^{-1}$$NaH_2PO_4$ were promoted the plant regeneration from rhizome segments. Kinetin was better than BA for plant regeneration and combination with 2 ${\mu}M$ kinetin and 5 ${\mu}M$ IBA was most efficient for plant regeneration. Solid or liquid medium with or without 0.1% qactivated charcoal in modified 1/2MS medium (1% sucrose, 50 $mg{\cdot}L^{-1}$$NaH_2PO_4$, 2 ${\mu}M$ kinetin, 5 ${\mu}M$ IBA, pH 5.8) were used to find the optimal culture methods. The plant regeneration from rhizome segments were most vigorous on solid medium without activated charcoal. The addition of activated charcoal were inhibited the plant regeneration from rhizome segments not only on solid medium but also liquid stationary or suspension culture.
The effect of cadmium ions on ligninolytic and decolourizing activities in cultures of two white-rot fungi, Cerrena unicolor and Trametes versicolor, were examined. Cadmium was added to the shallow stationary cultures growing on a liquid mineral medium. Both examined strains sorbed Cd ions in the first 24 hr of incubation. An appreciable stimulation of the activity of extracellular laccase (LAC) and inhibition of the extracellular manganese-dependent peroxidase (MnP) were simultaneously observed when 25 mgL-1 and 50 mgL-1 of cadmium ions were added to the cultures. On the other hand, the addition of cadmium ions also resulted in stimulating the decolorization activity of C. unicolor to decolorize Remazol Brilliant Blue R (RBBR) in the cultures, but decreasing it in the culture of T. versicolor, which is compared to the inhibition of MnP activity in this fungus. Our data indicate that the presence of Cd(II) ions can affect the ligninolytic activity of white-rot fungi. It was found that C. unicolor is a strain resistant to the presence of Cd ions in the liquid culture media, and has a potential to use this strain for bioremediation of sites contaminated with both heavy metals and aromatic pollutants.

The main objectives of this research were to study the cultural and nutritional characteristics of Tricholoma matsutake and to establish its liquid culture system. The optimum growth of T. matsutake was observed in HA and TMM agar media. Similarly highest growth was observed in PDB and TMM liquid media. The optimal temperature for the mycelial growth was $25^{\circ}C$. The most suitable carbon source was dextrin among 12 different carbon sources tested. Yeast extract and peptone were best nitrogen sources among 17 different sources tested. The optimum mineral salts were $Fe_{2}(SO_{4})_{3}{\cdot}H_{2}O$ and KCl among 9 different sources tested. Shaking culture gave higher mycelial growth compared to stationary culture. Similarly, optimum medium amount for shaking culture was 100 ml per 250 ml flask. The highest mycelial growth was obtained when $5{\sim}7$ mycelial discs were inoculated in 100 ml of medium and incubated for $8{\sim}9$ weeks, respectively. The highest proportion of mycelial growth was observed at 40 : 1 ratio of medium to inoculum volume in 8 l air-lift fermenter.
Lactic acid bacteria were isolated from silage, which produce high level of hydrogen peroxide in cell culture supernatant. The 16S rDNA sequences of the isolate matched perfectly with that of Lactobacillus fermentum (99.9%), examined by a 16S rDNA gene sequence analysis and similarity search using the GenBank database, thus named L. fermentum CS12-1. L. fermentum CS12-1 showed resistance to low pH and bile acid. The production of hydrogen peroxide by L. fermentum CS12-1 was confirmed by catalase treatment and high-performance liquid chromatography. L. fermentum CS12-1 accumulated hydrogen peroxide in culture broth as cells grew, and the highest concentration of hydrogen peroxide reached 3.5 mM at the late stationary growth phase. The cell-free supernatant of L. fermentum CS12-1 both before and after neutralization inhibited the growth of enterotoxigenic Escherichia coli K88 that causes diarrhea in piglets.
This study was carried out to investigate the efficient in vitro mass propagation methods for juvenile sporophytes of Matteuccia struthiopteris. Chopped segments of pinnae, petiole and rhizome were cultured on 1/2MS with 0.1% activated charcoal. Among these explant sources only rhizome segments produced young sporophytes, regenerating vigorously on 1/2 MS medium. Adjusting sucrose concentration to 2% and supplement to $50mgL^{-1}$$NaH_2PO_4$ in 1/2MS medium proved to be more efficient for plant regeneration. Various combinations of growth regulators such as kinetin, BA, NAA, and IBA were added to the growing media, and the best sporophyte regeneration was obtained by $1{\mu}M$ kinetin. The BA addition resulted in vigorous proliferation of meristematic tissues, but without differentiation to sporophytes. Three types of culture methods, solid using agar, liquid stationary, and liquid shaking culture, were employed with or without activated charcoal. The addition of 0.1% activated charcoal to modified 1/2MS media (2% sucrose, $50mgL^{-1}$$NaH_2PO_4$, $1{\mu}M$ kinetin, pH 5.8 and 0.8% agar) yielded highest sporophyte regeneration in liquid shaking culture.
