Leucaena leucocephala and Gliricidia sepium Supplementation in Sheep Fed With Ammonia Treated Rice Straw: Effects on Intake, Digestibility, Microbial Protein Yield and Live-Weight Changes

  • Two experiments were conducted to determine the effects of Leucaena leucocephala (leucaena) and Gliricidia sepium (gliricidia) supplementation on intake, digestion, outflow rates, microbial protein yield and live-weight changes in sheep fed with ammoniated rice straw (ARS). In experiment 1, three rumen cannulated Japanese Corriedale wether (mean body weight of 35.6 kg) in $3{\times}3$ Latin Square Design were used. Animals were fed ad libitum ARS alone, or supplemented with 200 g of either leucaena or gliricidia. In experiment 2, twenty-four growing native Philippine sheep with average body weight of $13.5{\pm}0.25kg$ were used in a completely randomized design (CRD) and offered similar diets to those of experiment 1. Supplementation increased total dry matter intake and nutrient digestibility except for fiber (p<0.05) without affecting ARS consumption. Nitrogen balance revealed that absorbed and retained N was significantly higher in leucaena and gliricidia. The significant improvement in N utilization and more digestible OM intake brought about by the inclusion of leucaena and gliricidia to ARS resulted in increased (p<0.05) microbial N yield. Efficiency of microbial N supply in supplemented group was not significantly different, but higher (p<0.05) than the 24.92 g N/kg DOMR for ARS group. Liquid outflow rate was 7.8 and 6.8 %/h, while the solid phase of rumen digesta was 4.4 and 3.8 %/h for the leucaena and gliricidia group respectively, which were significantly higher than 5.30 and 2.50 %/h in the control diet. The increase in total DMI resulted to higher (p<0.01) growth performance and efficient feed utilization. Average daily gain (ADG) was 19.3, 34.6 and 33.9 g/d for the ARS, leucaena and gliricidia respectively. It is therefore concluded that addition of leucaena and gliricidia to ARS in could increase nutrient intake and digestibility, subsequently improving N utilization and livestock performance.

  • 석탄화력발전소를 포함한 다양한 산업설비에서 유해 대기오염물질이 배출되고 있으며, 이러한 오염물질은 인체 건강과 자연 생태계에 영향을 준다. 특히, 질소산화물($NO_x$)와 이산화황($SO_2$)은 인체 건강에 악영향을 주는 미세먼지($PM_{2.5}$) 형성에 원인물질로 알려져 있다. 이러한 $NO_x$$SO_2$ 배출을 저감하기 위해서 선택적 촉매 환원(SCR)과 습식 탈황 공정(WFGD)으로 결합된 혼합 시스템이 사용되고 있으나, 높은 설치비용 및 운전비용을 필요로 하며, 유지보수의 문제점, 기술적인 한계점을 가지고 있다. 최근에 이러한 혼합 시스템을 대체하기 위한 $NO_x$, $SO_2$ 동시 저감 기술이 연구되고 있으며, 제안된 기술들은 흡수, 고도 산화(AOPs), 저온 플라즈마(NTP), 전자 빔(EB) 등이 있다. 이러한 기술들은 강한 수용성 산화제 및 산화력을 가진 화학활성종에 의한 $NO_x$, $SO_2$$HNO_3$, $H2SO_4$ 형태로의 산화 반응, 기-액 계면에서 $HNO_3$$H2SO_4$ 흡수 반응, 화학 첨가제에 의한 중화 반응을 기본으로 하고 있다. 본 논문에서는 각각의 동시 저감공정에 대한 기술적인 특징과 대용량 처리 공정 응용을 위한 향후 전망을 정리하였다.

국가환경시료은행 시료 채취, 분쇄, 저장과 개선방안 고찰 (Sampling and Cryogenic Pulverization and Storage of Environmental Samples and Improvement of Operating Procedures in National Environmental Specimen Bank)

  • Environmental Specimen Banks (ESBs) are playing pivotal role in monitoring the effect of environmental pollution on the ecosystem based on the retrospective analysis of the representative samples collected regularly and stored in cryogenic condition. In Korea, National Environmental Specimen Bank (NESB) was established in 2009 and the standard operating procedures (SOPs) for sampling, and cryogenic milling and storage had been prepared during 2007-2010. Since then, the tentative SOPs for the seven kinds of specimens (shoots of Red Pine (Pinus densiflora) and Korean Pine (Pinus koraiensis), leaves of Mongolian Oak (Quercus mongolica) and Zelkova Tree (Zelkova serrata), eggs of Feral Pigeon (Columba livia var. domestica), muscles and organs of Common Carp (Cyprinus carpio), and Freshwater Bivalve (Unio (Nodularia) douglasiae)) have been put to test in the field and laboratory as well against the practicality and feasibility. The SOPs were improved by reflecting the findings from the research and the following discussion regarding the selection of specimen (Feral Pigeon suffering from a control management), sample size (a problem of decreasing number of sampling trees related to increasing sampling time) and period (a problem related to a bud growth), and sampling methods etc.. In addition, barcoding system for the management of the specimen information, and monitoring system of the cryogenic storage to regulate the optimum temperature and the liquid nitrogen level were also developed for the efficient and effective control of the samples. Lastly, the safety guide and emergency protocol were augmented to guarantee a safe work environment with the cryogenic facility. These improvements of the SOPs are expected to contribute to more stable operation of the NESB.

땅콩의 근류형성(根瘤形成)과 질소고정(窒素固定)에 대한 근류균(根瘤菌) 접종방법(接種方法)의 영향(影響) (Nodulation and N2 Fixation in Groundnut as Affected by Inoculation Method)

  • 항생제 내성 변이균주 JB7 $nal^rspe^r$와 CB756 $str^rrif^r$을 이용하여 살균하지 않은 밭토양에서 접종방법이 근류균의 증식과 근류형성 및 질소고정에 미치는 영향을 조사하였다. JB7 $nal^rspe^r$균주(菌株)를 종자당 $10^2$에서 $10^8$세포수준으로 접종(接種)했을때 접종농도(接種濃度)의 증가와 함께 근류형성(根瘤形成)이 증가하였다. 접종방법별(接種方法別)로는 YMB배양 액체접종(液體接種)이 Peat slurry나 Peat입자(粒子) 접종(接種)보다 더 많은 근류(根瘤)를 형성(形成)했고, Peat slurry와 입자(粒子) 접종간(接種間)에는 차이가 없었다. 토양(土壤) g당 $10^3$ 세포(細胞)의 근류균(根瘤菌)을 함유(含有)하는 토양(土壤)에 JB7 $nal^rspe^r$와 CB756 $str^rrif^r$을 혼합(混合)하여 접종(接種)했을때 모든 근류(根瘤)가 접종균(接種菌)에 의하여 형성되었으며, JB7 $nal^rspe^r$균주(菌株)에 의한 근류점유율이 월등히 높았다. 품종간에 근류점유율의 차이는 없었다. 접종균류(接種菌株)는 근권에서 점진적으로 증식되었으며, JB7 $nal^rspe^r$가 더 큰 폭으로 증가되었다. 땅콩의 수량은 무접종구보다 접종구가 높았으나 질소처리구(窒素處理區)와 접종구간에는 차이가 없었다.

Detection of Superior Markers for Polymerase Chain Reaction Diagnosis of Breast Cancer Micrometastasis in Sentinel Lymph Nodes

  • Breast cancer is the most prevalent type of cancer among women around the world, and mortality is primarily caused by micro-metastatic disease. The complex mechanisms of breast cancer invasion and metastasis are intrinsically related to the malignant cell type so that early detection of micro-metastases can help prolongation of survival for patient. The aim of the present research work was evaluation of the expression status of mammoglobin protein as a candidate molecular marker in the negative sentinel lymph node (SLN). Fifty tumor specimens, and 50 normal adjacent breast tissue samples from the same patients were selected on the basis of having more than 10% tumor content for RNA extraction from SLNs. Tumor samples and normal adjacent breast tissue were archived in the form of frozen fresh tissue in liquid nitrogen. Real-time PCR was performed on a Bioner life express gradient thermal cycler system. Mammoglobin gene overexpression in breast cancer metastasis was investigated. Single marker results were mammaglobin 66.7% and CK19 50.0%, with 58.3% for the two in combination. Due to improved outcome with at least 3 genes (83.3%), it seems, triple marker evaluation will be most likely useful for detecting micro-metastases instead of studying separate genes.

LNG 벙커링용 고효율 LNG 저장탱크 열해석 (Thermal analysis of LNG storage tank for LNG bunkering system)

  • IMO의 규제인 신조 선박에 대한 NOx 80% 감축의 2016년 발효를 앞두고, 청정에너지인 LNG연료 선박 및 벙커링 선박의 보급이 유럽 선진국들을 중심으로 추진되고 있다. LNG 저장탱크는 LNG 벙커링의 필수 설비로 현재의 액체질소 등을 저장하는 극저온 액체 저장탱크와 동일한 구조이며, IMO의 "C"형 가압탱크인 내외 용기로 구성된 2중 탱크에 진공펄라이트 단열재가 충전되는 형식이다. 그러나 이 단열방식은 진공작업이 어렵고 일 LNG 기화량이 2.0 % 내외가 되어 보다 고효율의 탱크가 요구되어 진다. 본 연구에서는 진공과 단열재를 분리하여 내외탱크에 고진공을 적용하고 외부 탱크에 우레탄폼을 가설시킨 탱크 단열 방식을 새로이 고안하여 열해석을 수행하였다. 해석결과 본 개발 탱크는 진공도가 $10^{-3}Torr$ 이하일 때 일 기화량이 0.03 % 이하로 매우 적게 유지될 수 있고, $10^{-4}Torr$ 이하가 되면 일 기화량이 0.11 %가 되었다. 진공이 파괴되는 경우에도 현재 진공펄라이트 단열은 일 4.9 %의 증발이 발생하나, 새 고안 탱크는 일 증발율이 4.12 %가 되는 매우 효율이 높고 안전한 LNG 탱크 단열방식이 되었다.

Comparison of Semen Characteristics, Frozen-Thawed Sperm Viability, Testosterone Concentration and Embryo Development between Yorkshire Boar A and B

  • This study was carried out to compare the semen characteristics, frozen-thawed sperm viability and testosterone concentration and in vitro fertilization (IVF) and development of in vitro matured pig oocytes between two Yorkshire boars. Semen and blood samples were collected once per week from October to November 2002 from two adult Yorkshire boars at 18 months of age with 170 kg body weight. Sperm were deep frozen in 5 ml maxi-straws with lactose-egg yolk and N-acetyl-D-glucosamine (LEN) diluent and stored in liquid nitrogen. Blood samples were obtained at 10 a.m. by inserting a 21 gauge, hypodermic needle attached to 10 ml syringe into surface veins in the ear. The concentration of testosterone was determined by Competitive Enzyme Immunoassay. Ovaries were collected from prepubertal gilts at a local slaughter house. Cumulus oocyte complexes were aspirated from antral follicles (3 to 6 mm in diameter). The medium used for oocyte maturation was modified TCM 199. After about 22 h of culture, oocytes were cultured without cysteamine and hormones for 22 h at $38.5^{\circ}C$, 5% $CO_2$ in air. For IVF, one frozen 5 ml straw was thawed at $52^{\circ}C$in 40 sec and was diluted with 20 ml Beltsville thawing solution at room temperature. Sperm were washed 2 times in mTLP-PVA and inseminated without preincubation after thawing. Oocytes were inseminated with $2{\times}10^7$/ml sperm concentration. Oocytes were coincubated for 6 h in 500 ${\mu}$l mTBM fertilization medium. At 6 h after IVF, oocytes were transferred into 500 ${\mu}$l NCSU-23 culture medium for further culture of 48 and 144 h. There were no significant differences in the semen volume, motility, normal acrosome morphology and sperm concentration of raw semen between A and B of Yorkshire boar. However, motility and normal acrosome of boar A were higher than those of boar B at 0.5, 2, 3, 4, 5 and 6 h incubations of frozen-thawed sperm. Testosterone concentration (3.75 ng/ml) of boar A was higher than that (2.34 ng/ml) of boar B. The rate of blastocyst formation (15.1%) of boar A was higher than that (10.4%) of boar B. In conclusion, serum testosterone concentration of boar showed very important role for the frozen-thawed sperm viability and the blastocyst formation of pig oocytes matured in vitro.

Seminal Attributes and Semen Cryo-banking of Nepalese Indigenous Achhami (Bos indicus) Bull under Ex-situ Conservation

  • The study was conducted to evaluate the seminal attributes and cryobanking of Achhami (Bos indicus) bull semen. Of two Achhami bulls, 8 ejaculates from each bull were evaluated for seminal attributes. For semen freezing and cryo-banking, 4 ejaculates (having ≥2 mL semen volume, ≥75% of sperm motility and ≥1,000 × 106 cells/mL of sperm concentration) from each bull were used. Semen samples were diluted in egg-yolk-tris-citrate extender using a two-step dilution protocol, and were frozen in liquid nitrogen (LN2) vapour in a styrofoam box. The mean semen volume, colour, sperm mass activity, motility, viability, concentration, abnormal acrosome, midpiece and tail and, abnormal head of two Achhami bulls were 4.4 ± 0.5 mL vs. 2.5 ± 0.2 mL, 2.5 ± 0.1 vs. 2.4 ± 0.1, 3.5 ± 0.1 vs. 3.5 ± 0.1, 77.0 ± 1.1% vs. 78.3 ± 1.3%, 94.4 ± 0.5% vs. 91.0 ± 0.6%, 1137.7 ± 73.7 × 106 cells/mL vs. 1060.0 ± 44.3 × 106 cells/mL, 10.2 ± 0.5% vs. 10.3 ± 0.5% and 6.7 ± 0.5% vs. 8.2 ± 0.3%, respectively. The post-thawed sperm motility and viability were 53.0 ± 2.0% vs. 50.0 ± 0.0% and 80.2 ± 0.4% vs. 73.2 ± 0.7%, while evaluating by computer-assisted sperm analysis (CASA) system, the percentage of the progressive motility, fast motility, slow motility, local motility and immotile sperm were 75%, 68%, 7.4%, 16.6% and 8.6%, respectively. A total number of 620 doses semen straw were cryo-banked. Due to the acceptable post-thawed sperm motility and viability recorded, cryopreservation of Achhami semen is hereby recommended so as to preserve the Achhami breed. For further validation, the fertility will be observed from the produced frozen semen.

관상어용 기포발생기를 개조한 시료채취 장치와 기존의 시료채취기와의 성능 비교 (Comparative Study on the Efficiency Test Using Remodeled Bubble Generating Pump for Aquarium Fish and Established Air Sampling Pump)

  • This study is designed to compare the performance of established samplers (personal air sampler and MiniVOL portable air sampler) commonly used in the air environment or work environment with that of the sampler made by remodeling the air bubble generator for aquarium fishes. Sampling method used in this study is the filter collection method for PM10 and total suspended particles (TSP), the liquid collection method for sulfur dioxide ($SO_2$) and nitrogen dioxide ($NO_2$), and the solid collection method for toluene, respectively. There is not a significant difference in the average concentration of TSP between the Gilian personal air sampler (1st, $0.316{\pm}0.095$; 2nd $0.191{\pm}0.090$; 3rd, $0.185{\pm}0.073mg/m^3$) and the remodeled sampler (1st, $0.317{\pm}0.106$, 2nd $0.201{\pm}0.050$; 3rd $0.189{\pm}0.081mg/m^3$). There are also not significant differences in the average concentration of PM10 among the Gilian personal air sampler ($0.058{\pm}0.006mg/m^3$), the remodeled sampler ($0.052{\pm}0.008mg/m^3$) and the MiniVOL portable air sampler ($0.054{\pm}0.007mg/m^3$). The average concentration of the SO2 by the established sampler and the remodeled one is $3.79{\pm}0.21ppb$ and $3.45{\pm}0.15ppb$, respectively. In addition, there are not sigmficant differences in the average concentration of the NO2 between the Gilian personal air sampler (1st, $0.325{\pm}0.068$; 2nd $0.341{\pm}0.206$; 3rd, $2.971{\pm}0.078{\mu}g/m^3$) and the remodeled sampler (1st, $0.300{\pm}0.062$; 2nd $0.332{\pm}0.144$, 3rd, $2.968{\pm}0.085{\mu}g/m^3$). There are not significant differences in the average concentration of toluene between the Gilian personal air sampler (1st, $0.499{\pm}0.072$; 2nd $0.598{\pm}0.112$; 3rd $2.284{\pm}0.077{\mu}g/m^3$) and the remodeled sampler (1st, $0.463{\pm}0.133$; 2nd $0.603{\pm}0.082$; 3rd $2.353{\pm}0.115{\mu}g/m^3$). From these results, we can conclude that the performance of the remodeled sampler is not different from that of established samplers. There is possibility that the remodeled sampler can be used as a alternative device for Gilian personal air sampler in area and personal air sampling.

퇴비단 여과액비와 농축액비를 이용한 양액재배가 토마토(Lycopersicum esculentum Mill.)의 생육 및 수량에 미치는 영향 (Effects of Compost Leachate and Concentrated Slurry on the Growth and Yield of Tomato(Lycopersicum esculentum Mill.) in Hydroponic Culture)

  • 본 연구는 돈분뇨 여과 및 농축액비를 이용한 양액재배의 가능성을 검토하기 위하여 여과액비와 농축액비와 부산물, 양액 혼합처리가 토마토 생육에 미치는 영향을 검토하였다. 퇴비화 과정 중 과수분 상태에서 배출되는 퇴비단 여과액비와 막분리 처리과정에서 한외 여과막을 통과하고 역삼투막 처리에서 역류되어 나오는 돈슬러리 농축액비를 공시재료로 하였다. 본 연구는 질소함량을 기준으로 액비와 부산물, 양액의 혼합하는 처리구를 두어 전기전도도와 pH를 조정하여 토마토의 양액재배를 실시한 결과를 요약하면 다음과 같다. 1. 여과액비와 농축액비는 다량 및 미량원소를 함유하고 부유물질(SS)이 낮아 양액재배시 관배수의 막힘문제 없이 활용이 가능하였다. 여과, 농축액비는 인산, 칼슘, 마그네슘 함량이 낮고 칼륨이 높은 양분불균형를 나타내었다. 2. 여과액비는 원예연 표준양액 대비 총수량 91%, 상품수량 70%를 나타내어 여과액비 단독급액으로 토마토의 유기 수경재배 생산이 가능하였다. 3. 막처리 농축액비 100% 처리구는 토마토의 지상부의 생육이 지연되고 과중이 감소되어 화학양액 대비 40%의 수량을 나타내었다. 4. 퇴비단 여과액비+부산물, 농축액비+부산물 혼합처리구의 토마토 수량은 대조구 대비 각각 87, 76%를 나타내어 여과액비 단독시용구의 수량에 미치지 못하였다. 5. 질소기준으로 여과액비와 농축액비에 양액을 50% : 50% 비율로 혼합하여 균형처방 처리한 경우 표준양액 처리와 대등한 수량을 나타내었다. 결론적으로 토마토 양액재배에서 생육과 수량을 고려할 때 액비와 양액의 50 : 50 혼합 재배시 수량이 유지되어 가축분뇨 유래 유기액비에 의하여 화학양액를 50% 대체할 수 있을 것으로 판단된다.
