Analysis of Related Compounds from Commercial Atenolol Raw Materials and Preparations by High-Performance Liquid Chromatography (HPLC를 이용한 시판 아테놀롤 원료 및 제품 중 유연물질의 분석)

  • Atenolol and related compounds found in raw materials and commercial products were analyzed by reversed-phase high-performance liquid chromatography. A mixed solution of phosphate buffer (3.4 g/l, pH 3.0), tetrahydrofurane and methanol (800:20:180, v/v/v) including sodium octanesulfonate (1 g/l) and tetrabutylammonium-hydrogensulfate (0.4 g/l) was used as mobile phase at the flow rate of 0.25 ml/min. Detection was carried out at UV 226 nm. Atenolol related compounds, such as bis ether, tertiary amine and blocker acid were identified by comparing the retention time of the standard. The within-day and between-day precisions of the separated compounds were less than 1.2% and 3.4%, respectively. The contents of related compounds of the tested samples were under the limit prescribed in the European Pharmacopoeia. The pattern of the related compounds showed that atenolol raw materials and products could be classified in three different groups, indicating that the materials originated from different source or treated in different way.

A Study on the Force Balance of a Main Oxidizer shutoff Valve (산화제 개폐밸브의 힘평형에 관한 연구)

  • A MOV(Main Oxidizer shutoff Valve) controls the flow rate of liquid oxygen into the rocket combustor by opening and shutting operations piloted by a pneumatic force. In order to improve the effective design for sealing parts of poppet and piston assemblies, the poppet assembly has been designed to be just contacted with the piston assembly. However, to avoid a gap at the poppet/piston contact surface and to evaluate the MOV operating performance, an analyze on the force balance during the closing motion have been performed. For the accuracy of the analysis, the friction forces and the hydraulic forces have been respectively obtained by experiments and CFD analysis. Through the analysis, some important design parameters such as the spring constant, poppet friction and orifice size in the force balance have been introduced and the required operation performance of the MOV has been proved feasible.

  • This research aims to develop polymer composites which can be used for PEMFC bipolar plate by injection molding process. Considering the moldability and stiffness, we used LCP(Liquid crystal polymer) as base resin. In order to improve electrical conductivity and mechanical properties, we chose carbon black, and both synthetic graphite and expanded graphite. The composites with different recipe are prepared for injection molding of PEMFC bipolar plate and CAE(Computer Aided Engineering) analysis was performed to predict melt flow and fiber orientation We did successfully fabricate the ASTM specimens by injection molding, and measure the electrical conductivity of the samples by using four point probe device. We measured mechanical properties such as flexural strength, flexural modulus and Izod impact strength. Conclusively, the electrical conductivity increased with increasing additive concentration, but both flexural strength and Izod impact strength decreased due to the brittle nature of carbon-based additives.

  • The In recent years, the need for more fuel-efficient and lower-emission vehicles has driven the technical development of alternative fuels such as LPG (Liquefied Petroleum Gas) which is able to meet the limits of better emission levels without many modifications to current engine design. LPG has a high vapor pressure and lower viscosity and surface tension than diesel and gasoline fuels. These different fuel characteristics make it difficult to directly apply the conventional gasoline or diesel fuel pump. In this study, experiments are performed to get initial performance and efficiency of the fuel pump under different condition of the temperature and composition of fuel. The characteristics of vane type fuel pump were investigated to access the applicability on LPLi engine.

Qualitative and Quantitative Analysis of Thirteen Marker Components in Traditional Korean Formula, Samryeongbaekchul-san using an Ultra-Performance Liquid Chromatography Equipped with Electrospray Ionization Tandem Mass Spectrometry

  • For efficient quality control of the Samryeongbaekchul-san decoction, a powerful and accurate an ultra-performance liquid chromatography (UPLC) coupled with electrospray ionization (ESI) tandem mass spectrometry (MS) method was developed for quantitative analysis of the thirteen constituents: allantoin (1), spinosin (2), liquiritin (3), ginsenoside Rg1 (4), liquiritigenin (5), platycodin D2 (6), platycodin D (7), ginsenoside Rb1 (8), glycyrrhizin (9), 6-gingerol (10), atractylenolide III (11), atractylenolide II (12), and atractylenolide I (13). Separation of the compounds 1 - 13 was performed on a UPLC BEH $C_{18}$ column ($2.1{\times}100mm$, $1.7{\mu}m$) at a column temperature of $40^{\circ}C$ with a gradient solvent system of 0.1% (v/v) formic acid aqueous-acetonitrile. The flow rate and injection volume were 0.3 mL/min and $2.0{\mu}L$. Calibration curves of all compounds were showed good linearity with values of the correlation coefficient ${\geq}0.9920$ within the test ranges. The values of limits of detection and quantification for all analytes were 0.04 - 4.53 ng/mL and 0.13 - 13.60 ng/mL. The result of an experiment, compounds 2, 6, 12, and 13 were not detected while compounds 1, 3 - 5, and 7 - 11 were detected with 1,570.42, 5,239.85, 299.35, 318.88, 562.27, 340.87, 12,253.69, 73.80, and $115.01{\mu}g/g$, respectively.

  • An experimental study was performed for the combustion-field visualization of the burner which burns the liquid hydrocarbon fuel atomized by an ultrasonic oscillator. Configurations of the flame and combustion-field were caught by both high-speed camera and thermo-graphic camera, and those images were analyzed in detail through a post-processing. As a result, the combustion-field grew and reaction-temperature rose due to the strengthening of combustion reaction with the increasing flow-rate of carrier-gas. In addition, a phenomenon of flame flickering was discussed through the comparative analysis of the variational behaviors between the visible flame and IR (Infrared) flame-field.

  • Design parameters required for Methane/oxygen bipropellant small-rocket-engine were derived through a theoretical performance analysis. The theoretical performance of the rocket engine was analyzed by using CEA and optimal propellant mixture ratio, characteristic length, and optimal expansion ratio were calculated by assuming chemical equilibrium. A coaxial-type swirl injector was chosen because of its outstanding atomization performance and high combustion efficiency compared to other types of injector and also a bell nozzle with 80% of its full length was designed. The rocket engine configuration with 1.72 MPa of chamber pressure, 0.18 kg/s in total propellant mass flow, and O/F ratio of 2.7 was proposed as a ground-firing test model.

Pressure Control of Staged Combustion Liquid Rocket Engine (다단연소사이클 액체 로켓엔진의 압력제어에 대한 연구)

  • For the control of pre-burner combustion pressure, the open angle of the TTR (Throttle for Thrust Regulation) valve was varied from $143^{\circ}$ to $185^{\circ}$ while testing cold flow, ignition, and combustion. The major performance variables of rocket engines and hydraulic performance of the TTR valve regarding the open angle were verified. However, the controllability of pre-burner combustion pressure was not verified due to the limitations of the test. Comprehensive research will be done after addressing these problems.

Milk Fat Substitution by Microparticulated Protein in Reduced-fat Cheese Emulsion: The Effects on Stability, Microstructure, Rheological and Sensory Properties

  • Fat reduction in the formulation of cheese emulsion causes problems in its flowability and functional characteristics during spray-dried cheese powder production. In order to eliminate these problems, the potential of using microparticulated whey protein (MWP) in cheese emulsions was examined in this study. Reduced-fat white-brined cheese emulsions (RF) with different dry-matters (DM) (15%, 20%, and 25% excluding emulsifying salt) were produced using various MWP concentrations (0%-20% based on cheese DM of emulsion). Their key characteristics were compared to full-fat cheese emulsion (FF). MWP addition had no influence on prevention of the phase separation observed in the instable group (RF 15). The most notable effect of using MWP was a reduction in apparent viscosity of RF which significantly increased by fat reduction. Moreover, increasing the amount of MWP led to a decrease in the values of consistency index and an increase in the values of flow behavior index. On the other hand, using high amounts of MWP made the emulsion more liquid-like compared to full-fat counterpart. MWP utilization also resulted in similar lightness and yellowness parameters in RF as their full-fat counterparts. MWP in RF increased glossiness and flowability scores, while decreased mouth coating scores in sensory analyses. Fat reduction caused a more compact network, while a porous structure similar to FF was observed with MWP addition to RF. In conclusion, MWP showed a good potential for formulation of reduced-fat cheese emulsions with rheological and sensorial characteristics suitable to be used as the feeding liquid in the spray drying process.

Development and validation of LC-MS/MS for bioanalysis of hydroxychloroquine in human whole blood

  • This study aimed to analyze a high-performance liquid chromatography (HPLC) separation using a pentafluorophenyl column of parent drug hydroxychloroquine (HCQ) and its active metabolite, desethylhydroxchloroquine (DHCQ) applying to determine bioequivalence of two different formulations administered to patients. A rapid, simple, sensitive and specific liquid chromatography-tandem mass spectrometry (LC-MS/MS) method has been developed and validated for bioanalysis of HCQ and its metabolite DHCQ in human whole blood using deuterium derivative $hydroxychloroquine-D_4$ as an internal standard (IS). A triple-quadrupole mass spectrometer was operated using electrospray ionization in multiple reaction monitoring (MRM) mode. Sample preparation involves a two-step precipitation of protein techniques. The removed protein blood samples were chromatographed on a pentafluorophenyl (PFP) column ($50mm{\times}4.6mm$, $2.6{\mu}m$) with a mobile phase (ammonium formate solution containing dilute formic acid) in an isocratic mode at a flow rate of 0.45 mL/min. The standard curves were found to be linear in the range of 2 - 500 ng/mL for HCQ; 2 - 2,000 ng/mL for DHCQ in spite of lacking a highly sensitive MS spectrometry system. Results of intra- and inter-day precision and accuracy were within acceptable limits. A run time of 2.2 min for HCQ and 2.03 min for DHCQ in blood sample facilitated the analysis of more than 300 human whole blood samples per day. Taken together, we concluded that the assay developed herein represents a highly qualified technology for the quantification of HCQ in human whole blood for a parallel design bioequivalence study in a healthy male.
