액상칼슘 첨가 김치의 숙성 중 품질에 미치는 영향 (Efface of Liquid Calcium Addition on the Quality of Kimchi during Fermentation)

  • 본 연구에서는 액상칼슘을 첨가하여 김치 숙성에 미치는 영향을 조사하였다. 그 결과 액상칼슘 첨가구들에서 숙성 중 pH감소가 지연되었고 산도는 더 낮게 나타났으며 액상칼슘 $3.5\%$ (v/w)첨가구보다 $7.0\%$ (v/w)첨가구에서 더 효과적이였다. 총균수 및 젖산균수를 조사한 결과 숙성 8일째까지 액상칼슘 첨가구들이 낮게 나타났으나 8일 이후에는 더 많은 수를 나타내었다. 모든 구간에서 L값과 b값은 조금씩 감소하였으며 a값은 증가하는 경향으로 나타났다. 대조구의 칼슘함량은 $50{\sim}60\;mg\%$이며 액상칼슘 A 농도 $3.5\%$ (v/w)와 $7.0\%$ (v/w) 첨가구에서 $103{\sim}110\;mg\%$$145{\sim}163\;mg\%$로 액상칼슘 B 농도 $3.5\%$ (v/w)와 $7.0\%$ (v/w) 첨가구에서는 $140{\sim}151\;mg\%$$210{\sim}220\;mg\%$으로 나타나 액상칼슘 B를 $7.0\%$ (v/w) 첨가하면 고칼슘 김치의 제조가 가능한 것으로 생각되었다.

Simultaneous Determination of Alkaline Earth Metal Ions by a Conventional High Performance Liquid Chromatographic System

  • A simultaneous determination method of alkaline earth metals was attempted with the conventional high performance liquid chromatographic system. Four cations, namely, magnesium, calcium, strontium and barium ion, were injected directly as aqueous solution into an eluent containing copper chloride solution and and were successfully separated and determined on a separating column (Zipax SCX, 4.6 mm i.d. ${\times}25$ cm length, Du Pont, USA) by using a variable wavelength UV detector. The linear calibration curves were obtianed in the range from $1.0{\times}10^{-4}M$ to $5.0{\times}10^{-4}M$ and the correlation coefficient of the calibration curve for each metal of magnesium and calcium in tap water. Alkaline earth metals were determined with the conventional high performance liquid chromatographic system.

  • The effect of calcium lignosulfonate (CLS) on the early hydration characteristics for clinker minerals was investigated. In the presence of CLS and unsufficient gypsum The hydration of $C_3$A lowered CLS adsorption to form ettrin-gite and the residual CLS in the liquid phase accelerated the solubility of C4AF hydration. As the result unreacted $Fe^{3+}$ in the liquid phase would be precipitated gelatinously on $C_3$ hydrates and the hydration of $C_3$ could be retarded. But by addition of optimum gypsum into the cement with CLS the presence of $Fe^{3+}$ in the liquid phase were lowered and $C_3$ hydration would be normallized.

면실박 단백질로부터 가수분해물 제조 및 철분, 칼슘 결합 펩타이드의 분리 (Isolation of Iron and Calcium-Binding Peptides from Cottonseed Meal Protein Hydrolysates)

  • 면실박으로부터 단백질을 추출한 후 단백질 가수분해효소인 Flavourzyme으로 가수분해를 실시하여 면실박 단백질 가수분해물을 얻었고, 가수분해 정도는 trinitrobenzenesulfonic acid 방법과 Sodium dodecyl sulfate-polyacrylamide gel electrophoresis를 통해 측정하였다. 면실박 단백질 가수분해물은 한외여과에 의하여 3 kDa 이하로 cut-off하였고, Q-Sepharos fast flow, Sephadex G-15, reversed-phase high performance liquid chromatography를 이용하여 Fe, Ca-binding 펩타이드를 분리하였다. 그 결과 철분과 칼슘 결합력이 가장 높은 분획 51을 얻을 수 있었고, 이렇게 얻어진 Fe, Ca-binding 펩타이드는 향후 기능성 식품 소재로써 활용될 수 있다고 판단된다.

식초와 다슬기회분을 이용하여 제조한 초산칼슘의 품질 특성 (Quality Characteristics of Calcium Acetate Prepared with Vinegars and Ash of Black Snail)

  • 천연의 수용성 칼슘자원을 얻기 위하여 현미식초(Br),감식초(Pe) 및 사과식초(Ap)와 다슬기 회분으로 제조한 액상 및 고상의 초산칼슘(LCA및 SCA) 즉, 액상으로 Br-LCA, Pe-LCA, Ap-LCA와 고상으로 Br-SCA, Pe-SCA, Ap-SCA의 품질특성을 조사하였다. 초산칼슘 제조용 식초의 pH는 2.34∼3.06 범위였으며, 식초 100 mL과 반응하는 다슬기 회분의 양은 20.43∼23.50g, 고상 초산칼슘의 수율은 11.02∼13.01%(w/v)이었다. Ap-LCA 및 Ap-SCA의 색상은 연한 황색을 띠었으나 Br-LCA 및 Br-SCA는 갈색, Ap-LCA 및 Ap-SCA는 진한 갈색을 띠었다. Br-LCA, Pe-LCA 및 Ap-LCA의 칼슘함량은 각각 3.02, 2.06 및 2.30%(w/v)이었으며, Br-SCA, Pe-SCA 및 Ap-SCA의 칼슘함량은 각각 27.15, 16.31 및 19.48%(w/w)이었다. 고상초산칼슘의 용해도는 증류수에서 36.82∼39.92%(w/v), 소주에서 32.05∼39.04%(w/v), 진간장에서 13.12∼18.65%(w/v), 이온음료에서 38.35∼38.90%(w/v), 요구르트에서 33.47∼35.58%(w/v)이었으며 두유와 우유에서는 curd를 생성하였다. 신맛과 쓴맛은 표준품에 비하여 낮았으나 떫은 맛, 비린 맛 및 맛난 맛은 높았다.

습식분쇄하여 분무건조한 초미세 분말 칼슘의 품질특성 (Quality Characteristics of Spray Drying Microparticulated Calcium after Wet-grinding)

  • 해조칼슘을 습식분쇄한 초미세액상칼슘을 분무건조하여 부형제 종류에 따른 분말칼슘의 품질특성을 알아보고자 gum arabic, cyclodextrin, Na-caseinate를 첨가하여 품질특성을 비교하였다. 분무건조한 초미세분말칼슘의 수분함량은 2% 내외의 안정한 분말을 얻을 수 있었고, 색도는 원료인 해조칼슘보다 L값은 높았으며, b값은 감소하는 것으로 나타났다. 입자크기는 당류의 부형제 첨가로 초미세 크기로 분쇄되는 것을 확인할 수 있었으며, gum arabic을 첨가하여 제조한 초미세액상칼슘을 분무건조한 분말이 식초와 pH에 대한 용해도에서 가장 우수한 것으로 나타났다. 칼슘함량은 해조칼슘분말이 28%의 칼슘함량을 나타내었고, 부형제를 달리하여 제조한 초미세액상칼슘을 분무건조한 분말은 27% 내외로 비슷한 칼슘함량을 나타내어, 습식분쇄나 분무건조 공정에서 칼슘의 손실은 없는 것으로 나타났다. 흡습에 대한 안정성실험에서는 해조칼슘분말이 흡습에 안정하였으며, gum arabic을 첨가하여 분무건조한 분말이 흡습이 많이 발생하였다. 전자주사 현미경 관찰을 통하여 습식분쇄로 입자크기가 작아지는 것을 확인할 수 있었고, gum arabic을 사용하여 얻은 초미세칼슘분말이 단백질계나 사용하지 않았을 때보다 더 균일한 입자분포를 보였다. 따라서 습식분쇄한 초미세액상칼슘을 분말화하여 용해성등 품질특성이 개선됨을 확인하였으며 습식분쇄기술과 분무건조법을 식품가공기술로 활용할 수 있는 가능성을 확보하였다.

펄프 섬유의 세포벽 미세공극 충전 (Cell Wall Micropore Loading of Pulp Fibers)

  • The unique cell wall micropores of pulp fiber can be utilized as loading site in variety of important practical application which could be the basis of new papermaking technologies. One of these includes the manufature of paper containing higher levels of in situ filler precipitated. Hardwood pulp fiber were first impregnated with the solution of sodium carbonate($Na_2CO_3$). The micropores in cell wall of pulp fibers were filled with the liquid salt solution. The second calcium nitrate($Ca(NO_3)_2$) solution formed an insoluble calcium carbonate($CaCO_3$) precipitate within the cell wall micropores by interacting with the first sodium carbonate solution. The effects of chemical concentration and dryness of pulp fibers on the retention of cell wall micropore loaded filler were investigated. The paper properties of cell wall micropore loaded pulp fibers were compared with those of conventionally loaded and lumen loaded pulp fibers. Also the presense of the fillers within the cell wall micropore was observed by SEM. Increasing the chemical concentration to generate the calcium carbonate increased the retention of filler in cell wall micropore loaded pulp fibers. The particle size distribution of precipitated calcium carbonate ranged from $0.1{\mu}m$ to $80{\mu}m$. But, the average particle size of cell wall micropore loaded calcium carbonate was $4{\mu}m$. The paper made from never dried pulp fibers, the cell wall micropores which were filled with calcium carbonate, had better mechanical and optical properties than those of conventionally loaded or lumen loaded pulp fibers.

Removing Lipemia in Serum/Plasma Samples: A Multicenter Study

  • Background: Lipemia, a significant source of analytical errors in clinical laboratory settings, should be removed prior to measuring biochemical parameters. We investigated whether lipemia in serum/plasma samples can be removed using a method that is easier and more practicable than ultracentrifugation, the current reference method. Methods: Seven hospital laboratories in Spain participated in this study. We first compared the effectiveness of ultracentrifugation ($108,200{\times}g$) and high-speed centrifugation ($10,000{\times}g$ for 15 minutes) in removing lipemia. Second, we compared high-speed centrifugation with two liquid-liquid extraction methods-LipoClear (StatSpin, Norwood, USA), and 1,1,2-trichlorotrifluoroethane (Merck, Darmstadt, Germany). We assessed 14 biochemical parameters: serum/plasma concentrations of sodium ion, potassium ion, chloride ion, glucose, total protein, albumin, creatinine, urea, alkaline phosphatase, gamma-glutamyl transferase, alanine aminotransferase, aspartate-aminotransferase, calcium, and bilirubin. We analyzed whether the differences between lipemia removal methods exceeded the limit for clinically significant interference (LCSI). Results: When ultracentrifugation and high-speed centrifugation were compared, no parameter had a difference that exceeded the LCSI. When high-speed centrifugation was compared with the two liquid-liquid extraction methods, we found differences exceeding the LCSI in protein, calcium, and aspartate aminotransferase in the comparison with 1,1,2-trichlorotrifluoroethane, and in protein, albumin, and calcium in the comparison with LipoClear. Differences in other parameters did not exceed the LCSI. Conclusions: High-speed centrifugation ($10,000{\times}g$ for 15 minutes) can be used instead of ultracentrifugation to remove lipemia in serum/plasma samples. LipoClear and 1,1,2-trichlorotrifluoroethane are unsuitable as they interfere with the measurement of certain parameters.

Eletron Microscopic Observation of Calcium-Acetylated Seaweed Alginate Gel

  • Seaweed alginate was acetylated by activated carbon immobilized Pseudomonas syringae in a fluidized bed, up-flow reactor. The acetylation degree of seaweed alginate was about 30%. Calcium-acetylated seaweed alginate gel bead was made and compared to calcium-seaweed alginate gel bead by the scanning electron microscopy (SEM). Structural difference of two gel beads may results from increased viscosity and decreased affinity of acetylated seaweed alginate for calcium ion. On the basis of interior and exterior structure of calcium-acetylated seaweed alginate gels and property of acetylated seaweed alginate, it seems that acetylated seaweed alginate is used for the supporter for electrophoresis and packing materials for liquid chromatography and gel filtration.

고농도 질산성 질소를 함유한 산세폐수의 생물학적 처리에 관한 연구 (A Study on the Biological Treatment of Acid Pickling Wastewater Containing a High Concentration of Nitrate Nitrogen)

  • The purpose of this study is the efficient biological treatment of highly concentrated nitrate nitrogen by calcium ion control present within the pickling wastewater. In laboratory scale's experiments research was performed using a sequencing batch reactor and the evaluation of denitrification reaction in accordance with the injection condition of calcium ions, the surface properties of microorganisms, and the evaluation of sludge precipitability were performed. Results of the study showed that the denitrification reaction was delayed when injecting more than 600 mg/L of the calcium ion within the denitrification process. In addition, we observed the absorption form of calcium ions absorbed on the surface of microorganisms following an increase in the calcium ion dose. It was found that as the calcium ion dose increased the sludge precipitability also increased continuously and it is judged that a smooth denitrification induction is possible when treating the nitrate nitrogen by the calcium ion control of pickling waste water and the shortening of precipitation time enables a liquid operation to increase the reaction time.
