Cytosolic phospholipase A2, lipoxygenase metabolites, and reactive oxygen species

  • Reactive oxygen species (ROS) are generated in mammalian cells via both enzymatic and non-enzymatic mechanisms. Although certain ROS production pathways are required for the performance of specific physiological functions, excessive ROS generation is harmful, and has been implicated in the pathogenesis of a number of diseases. Among the ROS-producing enzymes, NADPH oxidase is widely distributed among mammalian cells, and is a crucial source of ROS for physiological and pathological processes. Reactive oxygen species are also generated by arachidonic acid (AA) metabolites, which are released from membrane phospholipids via the activity of cytosolic phospholipase $A_2$ ($cPLA_2$). In this study, we describe recent studies concerning the generation of ROS by AA metabolites. In particular, we have focused on the manner in which AA metabolism via lipoxygenase (LOX) and LOX metabolites contributes to ROS generation. By elucidating the signaling mechanisms that link LOX and LOX metabolites to ROS, we hope to shed light on the variety of physiological and pathological mechanisms associated with LOX metabolism.

Lipoxygenase Inhibitory Constituents from Indigofera oblongifolia

  • Indigin, alkylated xanthene (1) and indigoferic acid (2) have been isolated from the chloroform soluble fraction of Indigofera oblongifolia, along with $\beta$-sitosterol (3) and 3-hydroxybenzoic acid (4), which are reported for the first time from this species. Their structures were determined through spectroscopic techniques. Both the new compounds 1 and 2 showed significant activity against enzyme lipoxygenase, while 2 further showed moderate inhibition against BChE.

Changes of physiochemical properties of LOX-3 null rice lines stored at different storage temperatures and periods

  • Due to the lack in storage facility, considerable amount of rice is stocked in the open air, which causes increased stale flavor and deteriorates palatability and merchantable quality. Lipoxygenase-3(LOX-3) is involved in the production of volatile constituents in stored rice, and the development of stale flavor is delayed in LOX-3 null rice. LOX activity in the rice grain is localized in bran fraction and the tropical Japonica cultivar 'Dawdam' was reported that the absence of LOX-3 is inherited as a simple recessive trait. Also, it was reported that the peroxidation of unsaturated fatty acids occurs at lower levels in the 'Dawdam' bran fraction during storage than in rice varieties with LOX-3. This study was conducted to develop LOX-3 null rice lines using 'Dawdam' and investigate changes of physicochemical properties of the lines stored at different storage temperatures and periods. So we analyzed texture, toyo glossiness value, germination rate and lipoxygenase activity of 15 LOX-3 null rice lines on the condition of which rough rice had been stored at different temperatures (high temperature condition at $35^{\circ}C$ and low temperature condition at $15^{\circ}C$ for 4months. Hardness and stickiness of the lines tendered to be increased when it was stored at high temperature and adhesiveness, springiness, cohesiveness and chewiness was not considerably different according to storage temperatures and periods. The germination rate of HR29062-B-98-2-1-B among LOX-3 null rice lines was higher than another lines, 99.3, 94.0% after 4months stored at low temperature and high temperature, respectively. The lipoxygenase activity was 3,304, 1,601unit/mg protein after 4months stored at low temperature and high temperatures, respectively. So, it is thought that this line will be useful to breed rice varieties with high storability after tested on agricultural traits.

Anti-Inflammatory Activity of the Total Flavonoid Fraction from Broussonetia papyrifera in Combination with Lonicera japonica

  • To establish the anti-inflammatory activity of the total flavonoid fraction of the root barks of Broussonetia papyrifera (EBP) and a new formula, the ethanol extract of the root barks of B. papyrifera was fractionated with ethylacetate, yielding the hydrophobic prenylated flavonoid-enriched fraction. EBP and the ethanol extract of the whole Lonicera japonica (ELJ) plant were then mixed at a ratio of 1:1 (w/w) to give a new preparation (BL) in the hope of obtaining an optimal formula with a higher anti-inflammatory activity. Evaluation of the effects of these preparations on A23187-treated rat basophilic leukemia (RBL-1) cells revealed that EBP potently inhibited 5-lipoxygenase (5-LOX), while ELJ showed weak inhibition. Additionally, the mixture (BL) clearly showed stronger inhibitory effects against 5-LOX than either preparation alone. These preparations also inhibited cyclooxygenase-2-catalyzed $PGE_2$ and inducible nitric oxide (NO) synthase-catalyzed NO production by lipopolysaccharide-treated RAW 264.7 cells. When tested against arachidonic acid-induced mouse ear edema, EBP showed strong inhibitory activity at doses of 5-200 mg/kg when administered orally, but BL had obviously stronger inhibitory effects. When tested against ${\lambda}$-carrageenan-induced paw edema in mice, BL showed a potent and synergistic anti-inflammatory effect. In addition, in the acetic acid-induced writhing test, BL was found to have strong analgesic activity at 50-400 mg/kg. Taken together, these results indicate that each of these preparations exert anti-inflammatory activity in vitro and in vivo. In particular, BL showed stronger anti-inflammatory activity than EBP, and these anti-inflammatory effects were partially related to the inhibition of eicosanoid and NO production. BL may be useful for the treatment of human inflammatory disorders.

Lipoxygenase와 Kunitz Trypsin Inhibitor 단백질 결핍콩으로 제조한 간장의 이화학적 특성 및 항산화 활성 (Physicochemical Characteristics and Antioxidant Activity of Kanjang made from Soybean Cultivars Lacking Lipoxygenase and Kunitz Trypsin Inhibitor Protein)

  • Lipoxygenase(LOX)와 kunitz trypsin inhibitor(KTI) 단백질이 결핍된 non-GM콩(개척#2, 진양콩 및 CJ#1)의 가공적성을 알아보기 위하여 일반콩(태광콩)을 대조로 하여 간장을 제조하여 이화학적 특성 및 항산화 활성을 비교하였다. 원료콩의 일반성분은 시료간에 비슷한 조성을 보였으나, 총 페놀 및 플라보노이드 함량은 태광콩에 비해 LOX와 KTI 단백질 결핍콩에서 유의적으로 높았다. 간장 중 총당 및 환원당 함량은 태광콩 간장이 가장 높았으며, 총 질소 및 아미노태 질소 함량은 진양콩 간장이 가장 높았다. 무기물은 진양콩 및 CJ#1 간장, 아미노산은 3종의 LOX 및 KTI 단백질 결핍콩 간장이 태광콩 간장에 비해 높은 함량이었다. 간장의 갈색도는 진양콩 간장이 가장 높았으며, 황색도는 진양콩 간장이 태광콩 간장과 비슷한 수준이었다. 간장의 종합적인 기호도는 단맛과 구수한 맛이 높았던 진양콩 간장의 점수가 가장 높았다. 간장의 총 페놀 및 플라보노이드 함량은 LOX와 KTI 단백질 결핍콩 간장이 태광콩 간장에 비해 유의적으로 높았다. ABTS 및 DPPH 라디칼 소거활성은 총 페놀 함량에 의존적이었으며, FRAP법에 의한 환원력은 개척#2 및 CJ#1 간장이 태광콩에 비해 유의적으로 높았으며, $Fe^{2+}$ 킬레이트 활성은 태광콩 간장이 여타 간장의 활성에 비해 높았으나, 진양콩 간장과 비슷한 수준이었다. 따라서 LOX와 KTI 단백질 결핍콩으로 제조한 간장은 일반콩 간장에 비해 영양적 성분 및 항산화 활성이 높아 기능성 가공식품 소재로써 활용 가능성이 있는 것으로 판단된다.

  • 레놀레산이 함유된 액상 모델시스템에 있어서 지방산의 산화반응에 미치는 양조간장(분말)의 항산화 특성에 대한 일련의 연구를 행하여 다음과 같은 결과를 얻었다. 양조간장은 산화반응 과정 중 과산화물 생성을 억제하였으며, 반응에 첨가된 양조간장의 함량이 높을수록 항산화 작용이 더 큰 결과를 보였다. 이를 기존 항산화제와 비교하였을 때 알파-토코페롤0.05% 첨가수준의 항산화성은 양조간장 분말 $0.2{\sim}0.5%$ 범위에 있었다. 그리고 10ppm의 염화제일철이 함유된 산화반응에 있어서 양조간장은 미량금속의 산화촉진작용을 저하시켰으며 구연산과 공존할 때 항산화작용의 상승효과도 있었다. 또한, 양조간장은 oxidative free radical과 반응하였을때 수소공여성이 인정되었으며 리놀레산 혼합물의 리폭시제나이제 촉매적 산화반응을 억제하는 효과도 보였다.

Effects of Methyl Gallate on Arachidonic Acid Metabolizing Enzymes: Cyclooxygenase-2 and 5-Lipoxygenase in Mouse Bone Marrow-Derived Mast Cells

  • Methyl gallate (MG) is a medicinal herbal product that is isolated from Paeonia lactiflora that inhibits cyclooxygenase-2 (COX-2) dependent phases of prostaglandin $D_2\;(PGD_2)$ generation in bone marrow-derived mast cells (BMMC) in a concentration-dependent manner with an $IC_{50}$ values of $17.0\;{\mu}M$. This compound also found inhibited the COX-2-dependent conversion of the exogenous arachidonic acid to $PGD_2$ in a dose-dependent manner with an $IC_{50}$ values of $190\;{\mu}M$, using a COX enzyme assay kit. However, at concentrations up to $80\;{\mu}M$, MG did not inhibit COX-2 protein expression in BMMC, indicating that MG inhibits COX-2 activity directly. Furthermore, MG consistently inhibited the production of leukotriene $C_4\;(LTC_4)$ in a dose dependent manner, with an $IC_{50}$ value of $5.3\;{\mu}M$. These results demonstrate that MG has a dual cyclooxygenase-2/5-lipoxygenase inhibitory activity, which might provide the basis for novel anti-inflammatory drugs.

Chemical Constituents of the Root of Dystaenia takeshimana and Their Anti-Inflammatory Activity

  • In our ongoing search for bioactive compounds originating from the endemic species in Korea, we found that the hexane and EtOAc fractions of the MeOH extract from the root of Dystaenia takeshimana (Nakai) Kitagawa (Umbelliferae) showed cyclooxygenase-2 (COX-2) and 5- lipoxygenase (5-LOX) dual inhibitory activity by assessing their effects on the production of prostaglandin $D_2\;(PGD_2)$ and leukotriene $C_4\;(LTC_4)$ in mouse bone marrow-derived mast cells. By activity-guided fractionation, five coumarins, viz. psoralen (2), xanthotoxin (3), scopoletin (4), umbelliferone (5), and (+)-marmesin (6), together with ${\beta}-sitosterol$ (1), were isolated from the hexane fraction, and two phenethyl alcohol derivatives, viz. 2-methoxy-2-(4'-hydroxyphenyl)ethanol (7) and 2-hydroxy-2-(4'-hydroxyphenyl)ethanol (8), three flavonoids, viz. apigenin (9), luteolin (10), and cynaroside (11), as well as daucosterol (12) were isolated from the EtOAc fraction using silica gel column chromatography. In addition, D-mannitol (13) was isolated from the BuOH fraction by recrystallization. Two of the coumarins, scopoletin (4) and (+)- marmesin (6), the two phenethyl alcohol derivatives (7, 8) and the three flavonoids (9-11) were isolated for the first time from this plant. Among the compounds isolated from this plant, the five coumarins as well as the three flavonoids showed COX-2/5-LOX dual inhibitory activity. These results suggest that the anti-inflammatory activity of D. takeshimana might in part occur via the inhibition of the generation of eicosanoids.
