The effects of dietary supplementation of the unrefined rice bran oil from 'Suwon 415' pigmented black rice (BRBO) on cholesterol metabolism and cellular antioxidant status were investigated in hypercholesterolemic rats. The significant reduction of total cholesterol (TC) and low density lipoprotein cholesterol (LDL-C) concentrations was observed in the plasma of rats fed BRBO. BRBO also decreased plasma and hepatic oxidative stress as a result of increased levels of hepatic thiobarbituric acid reactive substances (TBARS) levels associated with the elevations of hepatic superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GSH-Px) activities together with increased plasma level of tocopherol. This study indicates that dietary BRBO supplement can leads to the improvement of overall cholesterol metabolism and antioxidant status even more effectively than 'Chuchung' white rice (WRBO). Consumption of BRBO may also protect the liver from oxidative damage caused by lipid peroxidation.
This study was conducted to assess biochemical nutritional status and health status of 53 men and 130 women aged over 65 years and residing in low-income areas in Seoul. Nutritional status and health status were determined by hematological analysis, incidence of disease, and clinical symptoms. Mean serum cholesterol levels were 159.7mg/dl in men and 182.4mg/dl in women and triglyceride levels were 124.6mg/dl in men and 130.6mg/dl in women. The serum lipid levels were higher in women than in men, and 88% of the subjects belonged to normal range and 12% belonged to hyper lipid. The mean hematological indices.; Hb, Hct, RBC, MCH, serum iron belonged to normal ranges. The proportion of anemic state were 16~18% in men and 20~25% in women. Mean serum Ca, Cu and Zn levels were 9.4mg/dl, $84.2\mu\textrm{g}/dl$ and $75.8\mu\textrm{g}/dl$ for male and $9.5\mu\textrm{g}/dl$, $88.3\mu\textrm{g}/dl$ and $75.6\mu\textrm{g}/dl$ for female, respectively. However, proportion of low Ca, Cu and Zn status were 8~20%, 77~84% and 12~34%, respectively. The biochemical indices were not significantly different according to age, BMI, obesity rate. BACKGROUND/OBJECTIVES: Oocyte lipid droplets play a crucial role in meiosis and embryo development. Biotin is associated with fatty acid synthesis and is the coenzyme for acetyl-CoA carboxylase (ACC). The effects of a biotin deficiency on the oocyte lipid metabolism remain unknown. This study examined the effects of a biotin deficiency and its replenishment on murine 1) oocyte lipid droplet levels, 2) ovary lipid metabolism, and 3) oocyte meiosis. MATERIALS/METHODS: Mice were divided into 3 groups: control, biotin deficient (BD), and recovery groups. The control and BD groups were fed a control diet or BD diet (0.004 or 0 g biotin/kg), respectively. The recovery group mice were fed a BD diet until day 21, and were then fed the control diet from days 22 to 64. This study then quantified the oocyte lipid droplet levels, assessed the oocyte mitochondrial function, and examined the ability of oocytes to undergo meiosis. Ovarian phosphorylated ACC (p-ACC), lipogenesis, β-oxidation, and ATP production-related genes were evaluated. RESULTS: The BD group showed a decrease in lipid droplets and mitochondrial membrane potential and increased p-ACC levels. In the recovery group, the hepatic biotin concentration, ovarian p-ACC levels, and mitochondrial membrane potential were restored to the control group levels. On the other hand, the quantity of lipid droplets in the recovery group was not restored to the control levels. Furthermore, the percentage of oocytes with meiotic abnormalities was higher in the recovery group than in the control group. CONCLUSIONS: A biotin deficiency reduced the oocyte lipid droplet levels by downregulating lipogenesis. The decreased lipid droplets and increased oocyte meiosis failure were not fully restored, even though the biotin nutrition status and gene expression of lipid metabolism was resumed. These results suggest that a biotin deficiency remains robust and can be long-lasting. Biotin might play a crucial role in maintaining the oocyte quality.

Objective: The current study aimed to investigate the impact of high-fat diets composed of different animal and vegetable fat sources on serum metabolic health markers in Japanese quail, as well as the overall lipid content and fatty acid profiles of the edible bird tissues following significantly increased dietary lipid supplementation. Methods: Fifty seven male quail were divided into six groups and fed either a standard diet or a diet enriched with one of five different fats (22% coconut oil, lard, palm oil, soybean oil, or sunflower oil) for 12 weeks. The birds were subjected to an oral glucose tolerance test following the feeding period, after which they were euthanized and blood, liver, breast, and thigh muscle samples collected. Total fat content and fatty acid profiles of the tissue samples, as well as serum uric acid, triglyceride, cholesterol, total protein, albumin, aspartate transaminase, and total bilirubin concentrations were assessed. Results: High-fat diet feeding had no significant effects on the glucose tolerance of the birds. Dietary fatty acid profiles of the added fats were reflected in the lipid profiles of both the liver and breast and thigh muscle tissues, indicating successful transfer of dietary fatty acids to the edible bird tissues. The significantly increased level of lipid inclusion in the diets of the quail used in the present study was unsuccessful in increasing the overall lipid content of the edible bird tissues. Serum metabolic health markers in birds on the high-fat diets were not significantly different from those observed in birds on the standard diet. Conclusion: Thus, despite the various high-fat diets modifying the fatty acid profile of the birds' tissues, unlike in most mammals, the birds maintained a normal health status following consumption of the various high-fat diets.
Secrum lipid and vitamin E levels were determined and smoking, alcohol drinking and exercise habits were asked in 357 healthy male subjects aged 49.4$\pm$6.7 years in Taegu. Average serum levels of total cholesterol, HDL-cholesterol and triglyceride(TG) were 189$\pm$43mg/이, 42$\pm$13mg/dl and 136$\pm$73mg/이, respectively. Serum level of lipid peroxide measured as thiobarbituric acid reactive substances(TBARS) of the subjects was 2.01$\pm$0.73MDA nmoles/ml and that of $\alpha$-tocopherol was 9.53$\pm$3.14ug/ml. The correlation coefficients between $\alpha$-tocopherol and serum lipids were 0.3631 for triglyceride, 0.2993 for cholesterol, and 0.3025 for total lipid. Heavy smokers who smoked more than 20 cigarettes a day had higher levels of TG and TBARS than those who smoke less. Alcohol drinkers had higher levels of TG and TBARS than nondrinkers. Vitamin E level(per ml serum)was significantly higher in the heavy smokers and drinkers, which was reflected by the higher level of serum lipid. When the level of $\alpha$-tocopherol was expressed as $\alpha$-tocopherol/triglyceride, it was negatively correlated with serum TBARS level and was lower in the heavy smokers than in the moderate smokers. It is concluded that vitamin E level as $\alpha$-tocopherol/triglyceride would be better compared to $\alpha$-tocopherol/total cholesterol or $\alpha$-tocopherol/total lipid for the evaluation of vitamin E status in Korean men.
The study was to compare the effect of dietary fatty acids on fatty acid profile in tissue and the status of tocopherol and lipid peroxidation, and superoxide dismutase and glutathione peroxidase activities at two fat levels. Male Sprague Dawley rats weighing average 350g(17 weeks) were fed either low fat(LF, 4.3% w/w, 10% kcal) or high fat(HF, 20.8%, w/w, 40% kcal)diet for 6 weeks. The fats used were beef tallow as a source of saturated fatty acid, corn oil for n-6 linoleic acid, perilla oil for n-3 $\alpha$-linolenic acid and fish oil for n-3 eiocosapentatenoic acid(EPA) and n-3 docosahexaenoic acid(DHA). Palsma tocopherol was significantly reduced by fish oil compared to beef tallow at body fat level. However, there was no significant effect on the levels of plasma MDA, RBC MDA and tocopherol, and RBC hempolysis by the type and amount of dietary fat. The peroxidizibility index of fatty acid profile in plasma and liver was increased and liver MDA level was significantly increased by fish oil when dietary fat level was increased. The activities of SOD and GSHPx tended to be increased by perilla oil and fish oil at both fat oil significantly reduced the incorpration of c20:4 and increased the incorporation of c20:5 into liver compared to corn oil. The incorporation of n-3 fatty acids into tissue by perilla oil rich in $\alpha$-linolenic acid was significantly higher tan corn oil and its effect was improved with higher amount of perilla oil in diet by high fat diet. Overall, the lipid peroxidation of tissue could be prevented by tocopherol supplementation when dietary fat level was low in diet. However, at high fat diet, tocopherol supplementation might not be enough to prevent the lipid peroxidation in tissue since the potential for lipid peroxidation was tended to be increased with higher incorporation of higher unsaturated n-3 fatty acids into tissue. Therefore, it could not be recommended to consume large amount of fish oil even with excess amount of tocopherol supplemented to the high fat diet.

This study was conducted to investigate the effect of Platycodi radix Saponin(PRS) on the reduction of lipid status in rats fed on high fat diet for 6 weeks after which lipid contents were measured in serum , liver and feces. The results obtained from this study are as follows : That the levels of total lipid, total cholesterol and triglyceride in serum and liver were significantly lower in the PRS group as compared with the control group. The contents of total lipid and total cholesterol excreted in the feces were tended to be slightly increase in PRS group compared to the control group, which were not significant.
This study was conducted to investigate the effect of the Platycodi radix powder (PRP) and Platycodi radix saponin(PRS) on the reduction of lipid status In rats fed on high fat diet for 6 weeks after which lipid contents were measured in liver. And also by carrying out the histological examination throughout light microscope to observe the effects of fat accumulation reduction. The results obtained from this study are as fellows. In the levels of total lipid in liver, PRS Group significantly decreased compared with Contred Group, but PRP Group was not significantly changed. The content of triglyceride was tended to be slightly decreased in the PRP and PRS groups compared to the control group, which was not significant. It was observed from photomicrographs of hepatic tissue in rats that the PRP and PRS groups inhibits the lipid accumulation induced by high fat diets.
Lipid peroxidation is a free radical oxidation of polyunsaturated fatty acids such as linoleic acid or arachidonic acid. This process has been related with various pathologies and disease status mainly because of the oxidation products formed during the process. The oxidation products include reactive aldehydes such as malondialdehyde and 4-hydroxynonenal. These reactive aldehydes can form adducts with DNAs and proteins, leading to the alterations in their functions to cause various diseases. This review will provide a short summary on the implication of lipid peroxidation on cancer, atherosclerosis, and neurodegeneration as well as chemical and biochemical mechanisms by which these adducts affect the pathological conditions. In addition, select examples will be presented where antioxidants were used to counteract oxidative damage caused by lipid peroxidation. At the end, isoprostanes are discussed as a gold standard for the assessment of oxidative damages.
Autophagy is an intracellular degradation system that breaks down damaged organelles or damaged proteins using intracellular lysosomes. Recent studies have also revealed that various forms of selective autophagy play specific physiological roles under different cellular conditions. Lipid droplets, which are mainly found in adipocytes and hepatocytes, are dynamic organelles that store triglycerides and are critical to health. Lipophagy is a type of selective autophagy that targets lipid droplets and is an essential mechanism for maintaining homeostasis of lipid droplets. However, while processes that regulate lipid droplets such as lipolysis and lipogenesis are relatively well known, the major factors that control lipophagy remain largely unknown. This review introduces the underlying mechanism by which lipophagy is induced and regulated, and the current findings on the major roles of lipophagy in physiological and pathological status. These studies will provide basic insights into the function of lipophagy and may be useful for the development of new therapies for lipophagy dysfunction-related diseases.
