아연과 Phytic Acid 수준별 식이가 흰쥐의 효소활성 및 지질대사에 미치는 영향 (Effects of Dietary Zinc and Phytic Acid Levels on Enzyme Activity and Lipid Metabolism of Rats)

  • In order to investigate the effect of dietary zinc and phytic acid levels on enzyme activity and lipid metabolism in rats, male Sprague-Dawley rats, weighing approximately 60-74g, were fed different diets which contained 0, 0.35 or $1.05\%$ phytic acid each at 3 levels of zinc (0, 30 and 1500ppm zinc) for 28 days. Body weight gain, food consumption, and food efficiency ratio were lower in the rats fed a zinc deficient diet (0ppm zinc) than those consuming 30 or 1500ppm dietary zinc. The activities of GOT, GPT and alkaline phosphatase were lower in the rats consuming 30ppm zinc than those fed 0 or 1500ppm zinc diet. The activity of GOT was increased in rats consuming $0.35\%$ phytic acid, whereas that of alkaline phosphatase was decreased in the rats fed phytic acid-containing diet. The concentration of phospholipid in serum was higher in rats fed $0.35\%$ dietary phytic acid, whereas that of liver phospholipid was higher in zinc deficient groups, and increased by addition of dietary phytic acid. The concentration of triglyceride in serum from rats fed 30ppm zinc was lower than those fed 0 or 1500ppm zinc On the other hand, liver triglyceride was higher in both the rats fed 30ppm zinc and $0.35\%$ phytic acid. The concentration of serum total cholesterol was lower in the rats fed 30ppm zinc diet, and it was increased by addition of dietary phytic acid. But liver total cholesterol was higher in 30ppm zinc group. HDL-cholesterol in serum was the highest in both rats consuming 30ppm zinc and $0.35\%$ dietary phytic acid, and the ratio of HDL-cholesterol to total cholesterol was higher in rats consuming 30ppm zinc diet. In conclusion, we suggest that coronary heart disease or liver disease can be prevented with phytic acid in rats which are fed the high zinc diet.

  • To study the effects of dietary protein and magnesium level on serum lipid contents and enzyme activities, male Sprague-Dawley rats were divided into six groups. Six groups of animals were fed six experimental diets different with protein(8% and 20%) and magnesium levels (Mg-free, 400mg/kg diet and 800mg/kg diet). The serum lipid contents and enzyme activities of rats fed different dietary protein and magnesium levels were examined. Serum triglyceride contents was significantly decreased as the dietary magnesium level was increased. Serum phospholipid contents was significantly higher in magnesium deficient group than those in the other groups. Serum alkaline phosphatase activity was significantly decreased in magnesium dificient group compared with other groups, but serum glutamic oxaloacetic transaminase and glutamic pyruvic transaminase activities was significantly increased in magnesium dificient group than other groups. In conclusion, serum triglyceride contents in low protein level was remarkably decreased as the dietary magnesium level was increased.

대두분 급여가 고지혈증 흰쥐의 지질대사 및 효소 활성도에 미치는 영향 (Effect of Soybean Powder on Lipid Metabolism and Enzyme Activities in Induced Hyperlipidemic Rats)

  • To investigate the effects of yellow soybean powder on serum lipid level and enzyme activities, male Sprague-Dawley(SD) rats were given hyperlipidemic diets for 6 weeks. Rats(n=8) were divided into three groups consisting the normal group(N), hyperlipidemic diets group(C) and hyperlipidemic diets and yellow soybean powder group(YSP). YSP group lowered triglyceride, total lipid, phospholpid, ester-cholesterol and free-cholesterl than control group. YSP group lowered total-cholesterol, VLDL-cholesterol and LDL-cholesterol but increased HDL-cholesterol of the serum compared to the control group. Athergenic Index(AI) and LHR(LDL-cholesterol/HDL-cholesterol ratio) of YSP group was decreased by control group (p<0.05). On the other hand, glutamic oxaloacetate transaminase(GOT), glutamic pyruvate transaminase (GTP), lactate dehydrogenase(LDHase) and alkaline phosphatase(ALP) enzymes activities were decreased by hyperlipidemic diets group (C). These results suggest that yellow soybean powder fed a lipid lowering effect in serum of rats.

쥐의 간암화 과정에서 타우린의 공급이 지질과산화물 함량, 생체방어 효소 및 세포막 안정도에 미치는 영향 (Effect of Taurine Supplementation on Lipid Peroxidation, Activities of Defense Enzymes and Membrane Stability During Rat Hepatocarcinogenesis)

  • The purpose of this study was to determine the effects of taurine supplementation on the hepatic lipid peroxidation, activiteis of defense enzymes and membrane stability during rat hepatocarcinogenesis. Hepatocarcinogenesis was induced by Solt & Farber modification. Lipid peroxide contents of carcinogen treated group which was not supplemented with taurine were lower than those of control group. This might be that peroxide is decreased because of the activation of detoxifing enzyme. Glutathione S-transferase(GST) activites of carcinogen treated groups were significantly (p<0.05) increased compared to those of control groups. The GST activities of group supplemented with taurine before treatment of carcinogen and during the all period of experiment were only less increased. In carcinogen treated groups, glutathione peroxidase(GPx) activites of groups supplemented with taurine were higher than those of non supplemented group. By carcinogen treatemtn, glucose 6-phosphatase(G6Pase) activites, index of membrane stability were decreased, but in carcinogen treated groups supplemented with taurine, they were less decreased. These results suggest that taurine supplementation seems to inhibit lipid peroxidation, to change the activities of defense enzymes and to prevent to membrane disintegration during chemically induced hepatocarcinogenesis.

사염화탄소에 의한 랫드의 간손상에 대한 인진호메타놀추출물의 억제효과 (Antihepatotoxic effect of Artemisia Iwayomogi methanol extract on acute hepatic injury by carbon tetrachloride in rat)

  • The purpose of present study was to examine pharmacological effect of Artemisia Iwayomogi methanol extract(AIME) on biochemical parameters(activities of AST, ALT, LDH, and ALP, contents of total bilirubin, total protein, albumin and A/G ratio in serum and levels of hepatic microsomal lipid peroxide and glucose-6-phosphatase activities) against hepatic injury by carbon tetrachloride($CCl_4$) in rats. Increased AST, ALT and LDH activities by $CCl_4$ were decreased in AIMS treatment group at 48 or 72 hours. Together, increased ALP activity by $CCl_4$ almost returned toward normal value in AIME treatment group at 72 hours. Serum total bilirubin contents increased to 87, 79 and 31% compared with normal group by $CCl_4$ which were decreased to 64, 42 and 26% in AIME treatment group at 24, 48 and 72 hours, respectively. Decreased contents of total protein and albumin, and A/G ratio by $CCl_4$ were recovered in AIME treatment group. Hepatic microsomal lipid peroxide levels(nmol malonic dialdehyde/100mg protein) increased to 140, 95 and 78% compared with normal group by $CCl_4$ which were decreased to 107, 74 and 65% in AIME treatment group at 24, 48 and 72 hours, separately. Hepatic microsomal glucose-6-phosphatase activities decreased to 60, 50 and 53% compared with normal group by $CCl_4$ at 24, 48 and 72 hours, respectively, which were increased at 72 hours in AIME treatment group. In conclusion, AIME enhanced the amelioration process from $CCl_4$-induced lipid peroxidation, degeneration of liver cell, and impairment of protein and bilirubin metabolisms.

The Relationship between Serum Ferritin and Bone Mineral Density

  • Several risk factors for osteoporosis are known relatively well. Some nutrients are directly or indirectly needed for metabolic processes related to bone. Recently, an increased prevalence of osteoporosis has been reported in patients with hemochromatosis, an iron overload disease. Thus, the aim of this study was to find out if there was any relationship between serum ferritin and T-score of bone mineral density in healthy women. We recruited 1,101 subjects females aged between 39 and 85 years. We measured serum ferritin, glucose tolerance indices, lipid profiles, inflammatory indices, hormones, calcium, alkaline phosphatase. Also, anthropometric, blood pressure, and bone mineral density measurements were performed. T-score was negatively correlated with age (r=-0.425; P<0.01), systolic (r=-0.109; P<0.01) and diastolic (r=-0.093; P<0.01) pressure, follicular stimulation hormone (r=-0.190; P<0.01), alkaline phosphatase (r=-0.235; P<0.01), and serum ferritin (r=-0.090; P<0.05) and positively with body mass index (r=0.050; P=0.01), HDL-cholesterol (r=0.314; P<0.01), and estradiol (r=0.200; P<0.01). After adjustment for age, alkaline phosphatase, body mass index, HDL-cholesterol, estradiol, and follicular stimulation hormone, serum ferritin was independently inversely correlated with T-score (${\beta}$=-0.001; P<0.05). It is possible that an increase of serum ferritin in females be risk to osteoporosis.

Evaluation of Interfering Substances in Routine Chemistry Tests Using Toshiba TBA-C8000 Chemistry Analyzer

  • In clinical chemistry tests, the interfering substances such as hemoglobin, lipid, bilirubin, and drugs, etc. can cause the changes of test results performed by spectrophotometrical methods. We evaluated the effects of interfering substances on the test results by adding interfering substances on the samples in the 19 kinds of clinical chemistry tests such as aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase, lactate dehydrogenase, gamma-glutamyltransferase, total protein, albumin, glucose, total cholesterol, total bilirubin, triglyceride, uric acid, calcium, inorganic phosphours, high density lipoprotein cholesterol, low density lipoprotein cholesterol, creatinine, blood urea nitrogen, and C-reactive protein using newly implemented automatic chemical analyzer Toshiba TBA-C8000 under the direction of CLSI EP07-A guideline. Hemolytic samples show increased concentration of total protein, aspartate aminotransferase, alanine aminotransferase, and lactate dehydrogenase and reduced concentration of total bilirubin, alkaline phosphatase by interfering effect. Hyperlipemic samples show increased concentration of total protein and alkaline phosphatase and reduced concentration of low density lipoprotein cholesterol. The samples with conjugated bilirubinemia show increased concentration of inorganic phosphours, otherwise the samples with unconjugated bilirubinemia show no interference or allowable range in the test result.

BAG5 regulates PTEN stability in MCF-7 cell line

  • The phosphatase and tensin homolog deleted on chromosome 10 (PTEN) is a tumor-suppressing lipid phosphatase that is frequently absent in breast tumors. Thus, the stability of PTEN is essential for tumor prevention and therapy. The ubiquitin-proteasome pathway has an important role in regulating the functions of PTEN. Specifically, carboxyl terminus Hsp70-interacting protein (CHIP), the E3 ubiquitin ligase of PTEN, can regulate PTEN levels. In this study, we report that BCL-2-associated athanogene 5 (BAG5), a known inhibitor of CHIP activity, reduces the degradation of PTEN and maintains its levels via an ubiquitylation-dependent pathway. BAG5 is identified as an antagonist of cell tumorigenicity.

고콜레스테롤혈증 흰쥐의 당질 및 지질 대사에 볏 붉은잎(Callophyllis japonica)추출액이 미치는 영향 (Effects of Callophyllis japonica Extracts on the Glucide and Lipid Metabolism in Hypercholesterolemic Rats)

  • This study was designed to observe the effects of the feeding Callophyllis japonica extracts on the improvement of the blood glucose, lipid in the serum of dietary hypercholesterolemic rats(S.D. strain, ?) fed the experimental diets for 4 weeks. Concentrations of total cholesterol and LDL-cholesterol in serum were significantly lower in the Callophylis japonica extracts group than in the choloesterol supplemented diet group. Concentrations of HDL-cholesterol in serum were higher in the Callophyllis japonica extracts group than in the cholesterol supplemented diet group. Concentrations of free cholesterol, triglyceride, phospholipid and blood glucose in serum were significantly lower in the Callophyllis japonica extracts group than in the cholesterol supplemented diet group. The activities of aspatate aminotransferae(AST), alanine aminotransferase(ALT), lactate dehydrogenase(LDH) and alkaline phosphatase(ALP) in serum were lower in the CAllophyllis japonica extracts supplemented group than in the cholesterol diet group. From the above research, the physiological activity substances in Callophylis japonica extracts were effective on the improvement of the blood glucose, lipid compositions in serum of dietary hypercholesterolemic rats.

양파즙이 에탄올에 의한 백서의 지질산화물 생성에 미치는 영향 (Effects of Onion Juice on Ethanol-Induced Hepatic Lipid Persoxidation in Rats)

  • The effect of onion juice on ethanol -induced lipid peroxidation were studied were studied in rats. The contents of thiobarbituric acid (TBA) -reactants increased significantly in liver thanol(4ml/kg/day) administered -rats. The activities of serum alanine aminotransferase and alkaline phosphatase increased by ethanol administration compared with control group, but alterations of antioxidant enzymes activities in liver of ethanol administered rats were not significant vs control group. The glutathione contents in liver decreased by ethanol , whereas the glutathione level increased in ethanol and onion juice group compared with ethanol group. The contents of hepatic TBA-reactants and serum aminotrasnferase activity in ethanol group were reduced by onion juice administration. In these results, increased hepatic TBA-reactants of liver in ethanol group might be due to decreased glutathione contents in liver. Reduced glutathione (GSH) plays an important roles in the liver in several detoxification and the reduction of lipid peroxides. So the protective effects of onion juice on ethanol-induced increment of TBA-reactants may be due to the increament of lgutathions content. The glutathione depletion by ethanol was an important factor of ethanol-induced cell damage, and the prevention of onion juice to the glutathione depletion reduced by ethanol may be an important factor on the protection from ethanol-induced lipid perpxidation in rats.
