  • Reactive Oxygen Species (ROS) are produced during normal cellular function. ROS are very transient species due to their high chemical reactivity that leads to lipid peroxidation and oxidation of some enzyme, massive protein oxidation and degradation. Under normal conditions, antioxidant are substances that either directly or indirectly protect cells against adverse effects of ROS. Several biologically important compounds have been reported to have antioxidant functions. These incluce vitamin C, vitamin E, GSH, flavonoids. superoxidee dismutase(SOD), glutathione peroxidase(GPX) and catalase(CAT). (omitted)

  • A study on the qualify changes of fish meat during drying and storage has been carried out with filefish meat. Filefish meat was dried in a forced air dryer at 40 and $55\%$ for 20 hours with an air velocity of 0.4 m/sec under different conditions of relative air humidities in the range of 10 to $50\%$. The dried fish meat was stored at $30^{\circ}C$ in chambers with constant relative humidities controlled by the use of conditioned air stream passing through the saturated salt solutions. The qualify of filefish meat was evaluated with the brown color densities developed by lipid oxidation and Maillard reaction. Changes of viable cell count during drying and storage were also discussed. The predominant reaction for the brown color developed during the study period was the lipid oxidation. The lipid oxidation rate during drying at constant temperature was appreciably affected by water activities at the drying surfaces of filefish meat during the falling drying rate period. The lipid oxidation rate was the slowest under the condition of the relative air humidity of around $30\%$. In samples stored at water activity of 0.33, the lipid oxidation rate was retarded remarkably in comparison with the samples with lower or higher water activities. The addition of $1\%$ table salt, $1.5\%$ D-sorbitol and $6\%$ sucrose slightly lowered the water activity with the slowest lipid oxidation rate. Such additives resulted the increase of the water soluble brown color densities, which seemed due to the increase of mobility of the water soluble substances by the result of the increase of equilibrium water content. Microflora of the samples immediately after drying consisted of ca. $30\%$ of coccus types, ca. $65\%$ of rod types and ca. $5\%$ of molds and yeasts. During the storage of the samples with a water activity of 0.76, the ratio of the coccus types to the total microflora was increased remarkably while that of the Gram negative non-spore rod types was decreased. The ratios of the Gram positive rod types, molds and yeasts during the storage were nearly constant.

  • The effects of antioxidants such as ascorbyl palmitate(AP), tocopherol( Toc), BHA and ascorbyl palmitate+ tocopherol(AP+ Toc) on lipid peroxide formations in lard have been investigated during storage at 50oC after gamma irradiation with the dose of 1~10kGy. Immediately gamma irradiation greatly increased the initiative oxidation of lard as expected. But antioxidants were found to be greatly effective in minimizing the radiation induced peroxidation of lard. The decreasing order of effects on their antioxidative activities was AP>AP+ Toc>BHA> Toc. Oxidation of lard during storage at 50oC after irradiation induced the acceleration of autooxidation. But the additions of an tioxidants inhibited the formation of peroxides. Their antioxidative activities during storage were BHA> AP+ Toc> Toc>AP. Especially AP+ Toc mixture was not significantly different from BHA (p>0.05).

The Influence of Phosvitin on the Inhibition of Iron-, and Copper-catalyzed Oxidation in Egg Oil Model System (철과 구리 이온으로 산화 촉진시킨 난황유 모델시스템에서 Phosvitin의 항산화 효과)

  • Phosvitin, an iron chelating protein in egg yolk, was measured for its ability to inhibit lipid oxidation in egg oil model system. Phosvitin(75$\mu$M) could inhibit both iron(50∼150$\mu$M) and copper(5∼15$\mu$M) catalyzed oxidation of egg oil, and much more effective in the presence of iron than copper. The antioxidant activity of phosvitin in egg oil decreased with increasing temperature up to 121$\^{C}$. But phosvitin was relatively heat stable maintaining 79 and 73% of its antioxidant activity after being heated for 6 min at 100$\^{C}$ and 2 min at 121$\^{C}$, respectively.

  • The root of Astragalus membranaceus Bunge (Leguminosae), which has been used for the treatment of hypertension, chronic hepatitis, duodenal ulcers, chronic nephritis and promotion of immunity in folk remedies. Several lines of evidence indicate that oxidative modification of low-density lipoprotein (Ox-LDL) may play an important role in atherogenesis. Hence, the role of antioxidants in the prevention of LDL oxidation needs to be determined. To investigate the antioxidant activity. we determined the MeOH ex. and fractions of Astragali Radix on the inhibition of LDL oxidation. The CH$_2$C1$_2$ and EtOAc orations inhibited the oxidative modification of LDL by a decrease in the lipid peroxide content and the electrophoretic mobility of LDL. Calycosin-7-0-$\beta$-D -glucoside which was isolated from EtOAc fraction inhibits the oxidative modification of LDL.

Lipid Specificity for Membrane Oxidation Catalyzed by Cytochrome c : An EPR Study

  • Decay of the spin label attached to cytochrome c or to stearic acid has been measured by electron paramagnetic resonance (EPR) spectroscopy to monitor membrane oxidation induced by cytochrome c-membrane interaction. Binding of cytochrome c sequestered the acidic phospholipids and membrane oxidation was efficient in the order linoleic oleic>stearic acid for a fatty acid chain in the acidic phospholipids. The spin label on cyt c was destroyed at pH 7 whereas that on stearic acid embedded in the membrane was destroyed at pH 4, presumably due to different modes of cyt c-membrane interaction depending on pH. Interestingly, cyt c also interacts with phosphatidylethanolamine, an electrically neutral phospholipid, to cause rapid membrane oxidation. Both EPR and fluorescence measurements indicated that electrostatic interaction is at least partially responsible for the process.

  • The purpose of this study was to investigate the effects of onion juice addition on the lipid oxidation of tuna spread. The tuna spread was stored at $4{\pm}1^{\circ}C$ for 49 days. The results of this study were as follows: Lipid oxidation in samples was measured by AV, POV and TBA value. AV and TBA value of the samples treated with 15% and 30% onion juice were lower than those treated with 5% and 45% onion juice. POV of samples treated with onion juice were lower than control group. Lightness of sample treated with onion juice was higher than control group. The score of overall quality in sensory evaluation was the highest in the sample treated with 15% onion juice.

Effects of ${\kappa}-Carrageenan$-Based Film Packaging on Moisture Loss and Lipid Oxidation of Mackerel Mince (${\kappa}-Carrageenan$ 필름을 사용하여 포장한 고등어육의 수분 손실 및 지방 산화)

  • ${\kappa}-Carrageenan-based$ film prepared by mixing 2% ${\kappa}-carrageenan$, 0.1% KCl, 0.75% polyethylene glycol, and 0.75% glycerol was examined to be used as a potential packaging material for mackerel mince for preventing moisture loss and lipid oxidation. Mackerel mince patties were vacuum-packaged with the film and stored at $20^{\circ}C,\;10^{\circ}C,\;0^{\circ}C,\;and\;-15^{\circ}C$; nonpackaged patties were also stored at $0^{\circ}C$. Weight reduction, peroxide value (PV), and thiobarbituric acid (TBA) value were measured during storage. The packaged or nonpackaged samples stored at $20^{\circ}C,\;10^{\circ}C,\;and\;0^{\circ}C$ showed a 60% weight reduction between 2 and 15 days of storage, while the weight reduction of the samples stored at $-15^{\circ}C$ was about 3% after 25 days. The nonpackaged samples stored at $0^{\circ}C$ showed a steady increase in lipid oxidation with the PV reaching 23 mequivalent peroxide (PO)/㎏ on day 20 and with the TBA value at 0.4 mole malonaldehyde (MA)/g on day 5. The PV and TBA values of the samples vacuum-packaged with the carrageenan-based film were below 2 mequivalent PO/㎏ and below 0.1 mole MA/g, respectively, regardless of storage temperature throughout the storage of 28 days.
