A study on tile processing of liquefied fish protein with a long self life and good solubility has been carried out for the effective utilization of sardine. The whole sadine was chopped, homogenized with same amount of water and then hydrolyzed by the addition of commercial proteolytic enzyme. The hydrolysate was centrifuged and the supernatant was decolorized with active carbon, desodorized by azeotropic distillation with toluene, xylene and cyclohexane. The liquefied sardine protein was then concentrated by rotary vacuum evaporator with the addition of starch. The use of $0.2\%$ commercial proteolytic enzyme to the weight of the whole sardine showed the optimum hydrolysis ratio at $55^{\circ}C$ for 4 hours. The liquefied sardine protein could be decolorized and also desodorized by the treatment with $15\%$ active carl]on at room temperature for 30 minuted. In the view point of lipid concentration and the solubility of the product, the liquefied sardine protein prepared by enzymic hydrolysis from the sardine protein concentrate was better than that prepared by enzymic hydrolysis from the whole sardine and sardine protein concentrate.
Rice bran, treated with heat or ${\gamma}-radiation$, was stored at $-15^{\circ}C,\;4^{\circ}C,\;37^{\circ}C$ and room temperature to determine its lipid stability by monitoring the changes in moisture, free fatty acids (FFA) and peroxide value (POV). Initial moisture content of rice bran was 14% and decreased with time. The higher storage temperature, the more moisture loss. The moisture content became 9% after 80 days of storage at $37^{\circ}C$. The initial FFA of rice bran was 2.5% which increased with time except the samples stored at $-15^{\circ}C$. The higher storage temperature, the more FFA was produced, by 9 times the initial FAA after 80 days of storage at $37^{\circ}C$. POV increased about twice the initial value after 80 days of storage at $-15^{\circ}C,\;4^{\circ}C$, and room temperature, and 5 times at $37^{\circ}C$. Rice bran was treated with heat at $70^{\circ}C,\;90^{\circ}C\;or\;105^{\circ}C$ and stored for 2 weeks at $30^{\circ}C$: The higher the heat treatment temperature and the longer the heat treatment time, the more moisture was lost. The not show any significant changes. Irradiation at $1{\sim}30\;kGy$ and subsequent storage for 4 weeks at $5^{\circ}C\;or\;30^{\circ}C$ caused negligible changes in moisture content. The FFA contents of rice bran irradiated up to 10 kGy were almost similar to these of nonirradiated one when measured just after irradiation. The samples irradiated at 30 kGy were 1.5 times higher in the FFA contents than nonirradiated ones. But there was little influence of irradiation doses on the FFA contents during storage. Irradiation caused the increase in POV of rice bran. resulting in 4 times increase in case of 30 kGy irradiated sample. During the storage, however, the POV of irradiated rice bran decreased significantly.
Burials for the rapid disposal of carcasses have diverse and profound effects on the rural living condition, natural environment, and local economy throughout construction, management and final destruction of burials. In this study, possible residue excavated from standard burials, storage using FRP (Fiberglass Reinforced Plastic) tanks, and microbial-treated burials are characterized as carcasses, contaminated soil by leachate, and wasted plastic film. Treatment technologies for volume reduction of the residue including composting, rendering, and thermal hydrolysis were investigated. If the solid and liquid residues generated during volume reduction treatment are directly transferred to the environmental facilities, it may cause disorder due to high concentrations of organics, antibiotics, and lipid. Benefits and drawbacks of composting as a volume reduction techniques are extensively investigated. We also discussed that proper treatment of excavated soils and the reusing the treated soil as agricultural purpose. For the protection of public health and worker's hygiene, treatment criteria including produced residue qualities, and quality standards for the treated soil as agricultural use are required. In addition, Scientific manual for the proper treatment of residues is required. It is necessary to consider the establishment of a pretreatment facility to the occurrence of large-scale residue treatment.
Flour and isolated starch from chickpea (desi type, 328S-8) were evaluated for their in vitro digestibility and physicochemical properties. The protein content, total starch content and apparent amylose content of chickpea flour and isolated starch were 22.2% and 0.6%, 45.8% and 91.5%, and 11.7% and 35.4%, respectively. Chickpea starch granules had an oval to round shape with a smooth surface. The X-ray diffraction pattern of chickpea starch was of the C-type and relative crystallinity was 24.6%. Chickpea starch had only a single endothermic transition (13.3 J/g) in the DSC thermogram, whereas chickpea flour showed two separate endothermic transitions corresponding to starch gelatinization (5.1 J/g) and disruption of the amylose-lipid complex (0.7 J/g). The chickpea flour had a significantly lower pasting viscosity without breakdown due to low starch content and interference of other components. The chickpea starch exhibited significant high setback in the viscogram. The average branch chain length, proportion of short branch chain (DP 6~12), and long branch chains (DP${\geq}$37) of isolated chickpea starch were 20.1, 20.9% and 9.2%, respectively. The rapidly digestible starch (RDS), slowly digestible starch (SDS) and resistant starch (RS) contents of chickpea flour and starch were 9.9% and 21.5%, 28.7% and 57.7%, and 7.1% and 9.3%, respectively. The expected glycemic index (eGI) of chickpea flour (39.5), based on the hydrolysis index, was substantially lower than that of isolated chickpea starch (69.2).
Tectorigenin has an apoptosis-inducing ability and immunosuppressive activity. We investigated the effect of tectorigenin on Phase I and II enzyme activities to elucidate the pharmacological action of the immunosuppressive tectorigenin in the diabetic rat. This compound was obtained from the hydrolysis of tetoridin isolated from the flower of Pueraria thumbergiana (Leguminosae). This crude drug (Puerariae Flos) has been used as a therapeutic for diabetes mellitus in traditional Korean medicine. Tecotrigenin inhibited the formation of malondialdehyde (MDA) and hydroxy radicals in serum and liver but promoted superoxide dismutase (SOD) activity. Low MDA contents and low xanthine oxidase and aldehyde oxidase activities were observed in the tectorigenin-treated rats, suggesting that such Phase I enzyme activities are the major source of lipid peroxidation. However, tectorigenin increased Phase II enzyme activities such as SOD, glutathione peroxidase and catalase, suggesting the activation of free radical-scavenging enzymes. The activities of tectorigenin were comparable to those of glibenclamide, which was employed as a positive control. These results suggest that tectorigenin may share some biological properties with glibenclamide in insulin-dependent-diabetes mellitus (IDDM).
Protein kinase C (PKC) is a family of kinases involved in the transduction of cellular signals that promote lipid hydrolysis. PKC plays a pivotal role in mediating cellular responses to extracellular stimuli involved in proliferation, differentiation and apoptosis. Comparative analysis of the PKC-${\alpha},{\beta},{\varepsilon}$ isozymes of 200 recently sequenced microbial genomes was carried out using variety of bioinformatics tools. Diversity and evolution of PKC was determined by sequence alignment. The ser/thr protein kinases of Streptomyces coelicolor A3 (2), is the only bacteria to show sequence alignment score greater than 30% with all the three PKC isotypes in the sequence alignment. S.coelicolor is the subject of our interest because it is notable for the production of pharmaceutically useful compounds including anti-tumor agents, immunosupressants and over two-thirds of all natural antibiotics currently available. The comparative analysis of three human isotypes of PKC and Serine/threonine protein kinase of S.coelicolor was carried out and possible mechanism of action of PKC was derived. Our analysis indicates that Serine/ threonine protein kinase from S. coelicolor can be a good candidate for potent anti-tumor agent. The presence of three representative isotypes of the PKC super family in this organism helps us to understand the mechanism of PKC from evolutionary perspective.
Quality of rice grain changes during dry storage with internal physiological changes and external injury by organism. Storage rice changes by condition with respiration via variable temperature, hydrolysis enzyme reaction, lipid peroxidation occurs with change of palatability. During dry storage, physiological change with protein variation pattern was examined by image analysis on proteomic technology. Analysis revealed that protein activity had no change store at room temperature and store at $40^{\circ}C$, but decreased store at $60^{\circ}C$. Analysis of variable hydrophobic protein pattern revealed that protein activity of beta-tubulin, protein disulfide isomerase, vacuolar ATPase b subunit, globulin was not significantly decreased all dry and store condition. However, heat shock protein 70, and glutathione transferase was significantly decreased when rice dried at $60^{\circ}C$ compared with room temperature and $40^{\circ}C$ dry condition.

The present study was investigated on in vitro anti-obesity effect of 4-hydroxybenzyl alcohol from Cudrania tricuspidata. We isolated various compounds from Cudrania tricuspidata. Among these compounds, anti-obesity effects of 4-hydroxybenzyl alcohol was examined by lipase activity assay, cyclic adenosine monophosphate (cAMP)-specific phosphodiesterase type IV (PDE4) activity assay, and citrate synthase activity assay. 4-hydroxybenzyl alcohol and Cudrania tricuspidata extracts inhibited the enzymatic activities of lipase, PDE4, and citrate synthase. Lipase is known to mediate the hydrolysis of triacylglycerol in adipose tissue and cholesterol esters in other tissue or cells. Also, PDE4 hydrolyses cAMP, a crucial secondary messenger for in metabolic pathways including glucose and lipid metabolism, lipolysis, and thermogenic function. 4-hydroxybenzyl alcohol and Cudrania tricuspidata extracts induced the inhibitory effect against each enzymatic activity on several specific substrates as observed by detection at 405 or 412 nm. These findings might be attributable to the inhibition of adipogenesis, and partial prevention of obesity. In conclusion, these results show that 4-hydroxybenzyl alcohol and Cudrania tricuspidata may be a critical candidate as a natural anti-obesity source.
To produce buckwheat sprouts, buckwheats were germinated at $23{\pm}2^{\circ}C$ up to 8 days in total darkness. In proximate analysis, moisture content increased from 13.87% of buckwheat groats to 93.75% of buckwheat sprouts on the 8th day of germination. On dry weight basis, lipid and ash contents increased like as protein content increased from 13.45% to 21.82% while carbohydrate content decreased due to enzyme hydrolysis. Amino acids were rich in glutamic acid, aspartic acids, and lysine, and also the ratio of the essential amino acids to total amino acids increased from 26.84% to 36.84%. Vitamin C did not exist in buckwheat groats but its content has continued to increase as far as 99.56 mg/100 g buckwheat sprouts. Rutin continued to increase by HPLC analysis from 4.71 mg/100 g buckwheat groats to 1,524 mg/100 g buckwheat sprouts. In analysis of organic acids, oxalic, maleic, and citric acids were commonly found in buckwheat sprouts. Fagopyrin was found almost none in buckwheat sprouts. In conclusion, the conversion of buckwheat seeds into sprouts through germination in darkness results in physically different final product with nutritional changes such as higher content of rutin, generation of vitamin C, abundance of the essential amino acids, and the existence of fagopyrin on very little.
Protein hydrolysate was prepared as a natural flavor stock from the sharp toothed eel (Muraenesox cinereus) mince using com-mercially available proteolytic enzymes, Alcalase, Neutrase, Protamex, and Flavourzyme. A 6 hr hydrolysis of mince, to which water of the equal weight of the mince was added, with $2\%$ (w/w, protein weight) Elavourzyme at $50^{\circ}$ yielded a hydrolysate of the highest acceptability. Removing the access lipid in liquified hydrolysate (not dehydrated) after enzyme hydrolysis, five times repetitive extraction using n-hexane (liquified hydrolysate : n-hexane=4 : 1, v/v) was effective, resulting in less than $1\%$ lipid content of the dehydrated-hydrolysate. The amino acid composition of the hydrolysate (prepared with Flavourzyme) was similar to that of the starting material. Hydrolysis led to an increase in concentration of not only total free amino acid but also free amino acid such as serine, glutamic acid, alanine, and methionine responsible for umami taste, especially up to about 40 times for methionine. Major free amino acids in amount were leucine, phenylalanine, valine, alanine, and isoleucine and they comprised about half of the total free amino acids, Moisture adsorption, fat adsorption, emulsifying capacity, and foaming capacity of the hydrolysate were 870.1 $\pm$$7.9\%$, 352.0$\pm$$5.3\%$, 50.3 $\pm$$1.2\%$, and $87.5\pm$$2.5\%$, respectively, and solubility was 83$\~$$84\%$ at acid pH range of 2$\~$4.
