Lysophosphatidylcholine induces azurophil granule translocation via Rho/Rho kinase/F-actin polymerization in human neutrophils

  • Translocation of azurophil granules is pivotal for bactericidal activity of neutrophils, the first-line defense cells against pathogens. Previously, we reported that lysophosphatidylcholine (LPC), an endogenous lipid, enhances bactericidal activity of human neutrophils via increasing translocation of azurophil granules. However, the precise mechanism of LPC-induced azurophil granule translocation was not fully understood. Treatment of neutrophil with LPC significantly increased CD63 (an azurophil granule marker) surface expression. Interestingly, cytochalasin B, an inhibitor of action polymerization, blocked LPC-induced CD63 surface expression. LPC increased F-actin polymerization. LPC-induced CD63 surface expression was inhibited by both a Rho specific inhibitor, Tat-C3 exoenzyme, and a Rho kinase (ROCK) inhibitor, Y27632 which also inhibited LPC-induced F-actin polymerization. LPC induced Rho-GTP activation. NSC23766, a Rac inhibitor, however, did not affect LPC-induced CD63 surface expression. Theses results suggest a novel regulatory mechanism for azurophil granule translocation where LPC induces translocation of azurophil granules via Rho/ROCK/F-actin polymerization pathway.

The Inhibiton Effects of Hypercholesterolemia and Platelet in Fermented and Non-Fermented Preparation of Garlic

  • This Dietary cholesterol augments lipid profile and primes production and activation of platelets, leading to development of atherosclerosis which produce several detrimental effects on cardiovascular health. Ethnomedicine and Mediterranean diet are natural sources and cost effective modes against several ailments including cardiovascular diseases while fermented foods have gained interest due to their increased nutrient profile, enhanced bioavailability and efficacy. Garlic has been known to reduce cholesterol and inhibit platelet activation. We examined whether fermented garlic ameliorates effects of hypercholesterolemia and platelet functions in rats. Methodology: Male SD rats were fed with hypercholesterolemia diet and treated with spirulina, fermented and non-fermented preparations of garlic for one month. Platelet aggregation and granule secretion were assessed to evaluate platelet activation. Liver and kidney weights, lipid and enzymatic profile of serum and whole blood analysis was performed. Expressions of SREBP, ACAT-2 and HMG-CoA were assessed using RT-PCR while liver and adipose tissues were analyzed for histological changes. Both fermented and non-fermented garlic inhibited platelet aggregation and granule secretion while fermented garlic showed greater inhibitor tendency. Fermented garlic significantly reduced liver weight and triglycerides concentrations than non-fermented garlic. Similarly, fermented garlic greatly abrogated the detrimental effects of steatosis on liver and adipose tissues. Fermented garlic significantly improved lipid profile and modulated platelet functions, thereby inhibiting atherosclerosis and platelet related cardiovascular disorders.

Lipofuscin Granule Accumulation Requires Autophagy Activation

  • Lipofuscins are oxidized lipid and protein complexes that accumulate during cellular senescence and tissue aging, regarded as markers for cellular oxidative damage, tissue aging, and certain aging-associated diseases. Therefore, understanding their cellular biological properties is crucial for effective treatment development. Through traditional microscopy, lipofuscins are readily observed as fluorescent granules thought to accumulate in lysosomes. However, lipofuscin granule formation and accumulation in senescent cells are poorly understood. Thus, this study examined lipofuscin accumulation in human fibroblasts exposed to various stressors. Our results substantiate that in glucose-starved or replicative senescence cells, where elevated oxidative stress levels activate autophagy, lipofuscins predominately appear as granules that co-localize with autolysosomes due to lysosomal acidity or impairment. Meanwhile, autophagosome formation is attenuated in cells experiencing oxidative stress induced by a doxorubicin pulse and chase, and lipofuscin fluorescence granules seldom manifest in the cytoplasm. As Torin-1 treatment activates autophagy, granular lipofuscins intensify and dominate, indicating that autophagy activation triggers their accumulation. Our results suggest that high oxidative stress activates autophagy but fails in lipofuscin removal, leaving an abundance of lipofuscin-filled impaired autolysosomes, referred to as residual bodies. Therefore, future endeavors in treating lipofuscin pathology-associated diseases and dysfunctions through autophagy activation demand meticulous consideration.

Physicochemical Properties of Defatted and Lipid-reintroduced Kidney Bean, Mung Bean and Corn Starches (탈지 및 지질첨가 강남콩, 녹두와 옥수수 전분의 특성)

  • Physicochemical properties of defatted and lipid-reintroduced kidney bean, mung bean, and corn starches were investigated. Granule shapes of kidney bean and mung bean starches were oval/round, but corn starch was polygonal and each shape was not changed after defatting and lipid reintroducing. The granule sizes of starches ranged $12{\sim}45{\mu}m,\;10{\sim}32{\mu}m\;and\;5{\sim}20{\mu}m$ respectively, but those of lipid reintroduced starches increased slightly. All starches showed Ca type in x-ray diffraction but relative crystallinity was decreased by defatting. The amylose contents of starches increased after defatting but decreased after lipid-reintroduction. The swelling power and solubility of untreated starches at each temperature increased by defatting but decreased by lipid-reintroduction except corn starches. The initial pasting temperatures of kidney bean, mung bean and corn starches by amylograph were $87,\;67\;and\;80^{\circ}C$ respectively, and those of them were lowered by defatting. The peak viscosity, height at $50^{\circ}C$, consistency and setback on amylogram of starches decreased by defatting but increased by lipid-reintroduction.

  • Experiments were carried out to investigate the lipid peroxidation (LPO), superoxide dismutase (SOD) activity and histopathological change of hepatic tissue for rockfish, Sebastes schlegeli after feeding sub-chronic dietary Benzo(a)pyrene (BaP) in the concentration of 0 (control), 0.5, 1.0, 1.5, 2.0 mg $kg^{-1}$ dry food of diet for 30 days. In 2.0 mg $kg^{-1}$ dry food group, the significant increase of LPO was observed in all period, and SOD activity was incresed at 30 days significantly in the same concentration. In the histological investigation of liver, there was the swelling of hepatic cells at 10 days over the 1.0 mg $kg^{-1}$ dry food concentration. At 30 days Periodic acid-Schiff (PAS) positive granule was observed in the same group and at 20 days was observed in 2.0 mg $kg^{-1}$ dry food group. And there was necrosis of hepatic cell in some fish of 2.0 mg $kg^{-1}$ dry food group at 30 days.

Property Characterization and Lipid - Compositional Analysis of Lipid Granules Isolated from an Oleaginous Yeast Rhodotorula glutinis

  • Preparative isolation of lipid granules from Fhodotorula glutinis, which has been studied for long time to produce edible lipids, was carried out by flotation method in Ficoll-Linear density gradient. When the isolated lipid granules were suspended in a series of solutions containing varying concentration of osmotic stabilizer (sorbitoal and mannitol) ranging from 0.8M to 0M, the lipid granules appeared to be disrupted at a concentration between 0.8M and 0.7, and again at a concentration below 0.1M, suggesting that lipid granules have a membraneous structure and that at least two types of lipid granules are present. Compositional analysis of lipids from lipid granules revealed that lipids are composed mainly of neutral lipids (87.8% of total lipids), predominantly as triacylglycerols (71.89%). Marked differences were observed inphospholipids between lipids of lipid granules and those of whole cells . The major components of phospholipids in lipid granules and inwhole cells are phosphatidylcholine(38.6%) and phosphatidylserine(42.8%), respectively. In addition, significant differences were also observed in the fatty acid composition of phospholipids. As phospholipids are important structural components of membranes, these differences lead to the suggesting that the membrane of lipid granules may be distinct functionally and structurally from other membranes of yeast cells. The major fatty acid components of neutral lipidss of whole cells and lipid granules are palmitic , oleic and linoleic acid. However , degreeof fatty acid unsaturation of neutal lipids of lipid granules was much lower than that of neutral lipids of whole cells.

  • R. Philippinarum is dioecious and oviparous. In the early vitellogenic oocyte, the Golgi apparatus and mitochondria present in the perinuclear region are involved in the formation of lipid droplets and in lipid granule formation. In the late vitellogenic oocyte, the endoplasmic reticulum, mitochondria in the cytoplasm are involved in the formation of proteid yolk granules. At this time, exogenous lipid granular substance and glycogen particles in the germinal epithelium are passed into the ooplasm of oocyte through the microvilli of the vitelline envelope. Ripe oocytes are about 55-60 $\mu$ m in diameter. The spawning period was once a year between early June and early October, and the main spawning occurred between July and August when seawater temperature was approximately 20 C. The reproductive cycle of this species can be categorized into five successive stages: early active stage (February to March), late active stage (April to May), ripe stage (April to August), partially spawned stage (June to October), and spent/inactive stage (August to March). Gonad developmental phases by histological qualitative analysis showed similar results with those of quantitative image analysis.

The Effects of Taeeumjowi-tang Extract Granule on Metabolic Syndrome Risk Factors with Obesity: A Single Group, Prospective, Multi-Center Trial (태음조위탕 엑스과립이 비만을 포함한 대사증후군 위험인자에 미치는 영향에 대한 연구: 단일군, 전향적, 다기관 임상연구)

  • Objective The purpose of this study is to study the effects of Taeeumjowi-tang extract granule on metabolic syndrome with obesity in Taeeumin. Methods We studied 206 subjects who diagnosed metabolic syndrome with obesity and classified Taeeumin by a Sasang constitution analysis tool. The subjects were administered by Taeeumjowi-tang for 12 weeks. We investigated the changes of body mass index, abdominal circumference, the number of risk factors for metabolic syndrome, lipid, fasting blood sugar, Hemoglobin A1c, free fatty acid, insulin, and blood pressure. Results Body mass index and abdominal circumference, systolic and diastolic blood pressure were significantly reduced after 4 weeks, 8 weeks, and 12 weeks of administration compared to before administration. Triglyceride significantly decreased after 8 weeks and 12 weeks of dosing compared to before taking. Low density lipoprotein cholesterol, fasting blood sugar were significantly increased after 8 weeks of administration compared to before administration. High density lipoprotein cholessterol was decreased significantly after 4 weeks of administration compared to before administration. The number of risk factors for metabolic syndrome, Hemoglobin A1c increased significantly after 12 weeks of administration compared to screening. Conclusions In this study we knew that Taeeumjowi-tang extract granule is very efficient in metabolic syndrome, especially in obesity and hypertension, triglyceride, insulin resistance.

Effect of Freezing of Paste on the Formation of Chou (반죽의 냉동처리가 Chou 형성에 미치는 효과)

  • This study was conducted to know the quality of chou made with flour pastes which were stored at different conditions of quick freezing, slow freezing, cold and room temperature. Also, this study included investigation of the chou properties such as expansion, sensory evaluation, degree of gelatinization, and physical and structural properties of paste were observed. There were not significant differences m diameter, height, volume, appearance, hollow formation, and sensory evaluation between the chou made with the paste stored at freezing condition and chou directly baked after pasting. Quick and slow freezing storages did not significantly affect the properties of chou, and the same results were obtained among the chou made with pastes thawed at room temperature and in microwave ovenrange. The chou of pastes stored at room temperature and in microwave ovenrange. The chou of pastes stored at room temperature and stored in refrigerator showed lowed expansion and value of sensory evaluation than those of frozen pastes. The paste stored at room temperature had the lowest hardness and viscosity compared with the other storage conditions. According to the observation of light microscope. the lipid bodies of the paste of freezing storage smaller those of the room temperature and refrigerator storage. The expantion of chou made with paste stored at room temperature was greatly decreased due to the high coalescence of lipid bodies, and also the paste components such as lipid, starch granule gluten at room temperature had inferior dispersion condition. The general tendency of the degree of gelatinization of chou were low in all treatments of paste. The values were 23.5%~46.0% in freezing, 77.3% in room temperature, 68.7% in directly baked after pasting, and 61.0% in cold storage, respectively. The formation and the taste of chou made with frozen paste were similar to those of chou directly baked pasting.
