The Contents of Total Lipid and the Component of the Lipid in Porcine Variety Tissue (豚 內臟筋中 總脂質 含量과 構成脂質의 組成)

  • The contents of crude fat were remarkably different according to parts in porcine tissues. The most contents of total lipids in variety tissues were neutral lipid, and the nexts were phospholipid and glycolipid, where these of the most composition in liver were phospholipid and the next neutrallipid and glycolipid. The major component of the nonpolar lipids in fresh was triglyceried(58.09∼74.68%) and phospolipid in the polar lipid were phosphatidyl choline(20.14∼45.17%) and phosphatidyl ethanolamine(18.5∼30.14%) and the main component in glycolipid acylsterol glyceride(26.10∼45.15%), respectively.

Component analysis of the lipid hydroperoxide in the brain and peripheral organs of Senescence-Accelerated Mouse (SAM) model

  • We measured previously the lipid hydroperoxides level in the brain and peripheral organs such as heart, liver, lung and kidney of senescence acceIerated-prone (SAMP8) and -resistant(SAMR1) mice at 3,6 and 9 months of age. It was found that the lipid hydroperoxide leve1s in the brain did not show any age-dependent change, and that they Were significantly higher in SAMP8 than in SAMR1 over the defined periods. In contrast, the lipid hydroperoxide leve1s in the peripheral organs, including liver, Were increased with aging in both substrain, and they were significantly higher in SAMP8 than in SAMR1 at 3 and 6 months of age. In addition, the lipid hydroperoxide levels in the peripheral organs were higher than those in the brain in both substrains. To elucidate the difference of lipid hydroperoxide levels between the brain and the peripheral organs, we further carried out lipid component analysis in the brain and liver, one of the peripheral organs, of SAMP8 and SAMR1 at 6 months of age.

  • The lipid compositions including fatty acid sterol lipid class and the lipid component of the total lipid extracted from the flesh of some fresh water fishes I. e. mandarin cornet cat fish and Korean perch were compared. The levels of total lipid of flesh tissues from the fresh water fishes were high(7.4%) in cat fish but low(1.4-2.2%) in mandarin fish Korean perch and cornet fish. On the contrary the content of unsaponifiable matters found in total lipid was low(2.6%) in cat fish but high(6.0%-6.5%) in mandarin fish Korean perch and cornet fish. Total lipids were mainly composed of triglyceride(74.6-86.5%) as major component in each sample and the other lipid components of total lipid e. g. polar lipid free fatty acids and free sterol were the minor components, The major fatty acids in total lipid of each sample were{{{{ {C }_{16 { }:_{ }0 } }}}}(19.6-29.2%) {{{{ {C }_{16 { }:_{ }1 } }}}}(17.3-30.7%) and {{{{ {C }_{18 { }:_{ }1 } }}}}(16.8-29.2%) and additionally it chiefly consisted of {{{{ {C }_{14 { }:_{ }0 } }}}} and {{{{ {C }_{18 { }:_{ }2 } }}}} Particularly the contents of polyenoic acids in total lipid of cat fish were higher than those of the other fish samples. The level of cholesterol in total lipid was low (8.3mg/g) in cat fish but were high(36.9-59.9mg/g) in mandarin fish Korean perch and cornet fish. The contents of fractionated neutral lipid(NL) were higher than those of polar lipid(PL) in each sample. Particularly phospholipid content in PL was low(6.0%) in cat fish but were high(23.1-36.3%) in mandarin fish Korean perch and cornet fish. Neutral lipids were mainly consisted of triglyceride(84.5-93.4%) as amjor component in each sample and the other lipid components of neutral lipid e, g, free fatty acids and free wterol were exhibited as a minor components, The fatty acid compositon of neutral lipids was very resembled to total lipids. The phospholipid in mandarin cornet and cat-fish were mainly composed of phosphatidyl serine(23.1-49.8%) and phosphatidyl choline(20.8-45.3%) The relatively higher amounts of phosphatidyl serine were observed in mandarine cornet and cat-fist than in Korean perch, But phosphatidyl ethanolamine(42.3%) and phosphatidyl choline (49.9%) were the main phospholipid in Korean perch. The extraordinary high content of phos-phatidyl ethanolamine compared to other fishes was characteristics in phospholipid composition of Korean perch. The major fatty acids in phospholipid of each sample were {{{{ {C }_{16 { }:_{ }0 } }}}} (38.3-46.5%) {{{{ {C}_{18 { }:_{ }0 } }}}}(14.2-21.7) and C16:1(11.6-13.8%) and additionally it chiefly consisted of C18:2, C18:0 and C17:0 The major fatty acids in glycolipid of each sample were C16:0(28.8-40.1%) C18:1(5.4-29.9%) C18:0(5.1-28.9%) and C16:1(8.2-20.1%) and additio-nally it chiefly consisted of C14:0 and C20:1.

  • Twelve docked and 12 intact Najdi lambs of equal numbers of males and females were slaughtered at 40 kg shorn shrunk body weight, Lipid in all empty body fat components; namely, subcutaneous, intermuscular, intramuscular, omental, mesenteric, channel, perirenal, pericardial, tail, viscera, bone and hide, were determined. Except for ram lambs, which had a lower percentage of lipid in intermuscular partition in loin cut, sex did not influence the relative proportion of subcutaneous or intermuscular fat in each wholesale cut. The data also showed that docking did not change the distribution of lipid in intermuscular and intramuscular fat partitions in each wholesale cut. Docked lambs tended to accumulate lower proportions of the lipid in subcutaneous fat component in the cuts located along the dorsal line than intact lambs. The total amount of lipid deposited in the empty body of ewe lamb was heavier in weight than that of ram lamb. Docking had no effect on the distribution of total lipid in the empty body, except for subcutaneous fat component, being greater in docked lambs than did intact ones. Generally, the highest proportion of fat was associated with subcutaneous depot followed, in order, by intermuscular, mesenteric, tail, intramuscular and omental fat partitions.

  • Diethylnitrosamine (DEN) is known as a potential hepatic carcinogen by single administration. This study was designed to measure the effects of DEN-induced cell damage on the triglyceride and cholesterol concentration in the liver, excluding dietary effects. Fertilized chicken eggs, 10 days before hatching, were randomly divided into three groups (n=20) and each egg was injected 10 ${mu}ell$ of corn oil (vehicle control), 5 $\mu\textrm{g}$ of DEN/10 ${mu}ell$ of DEN/10 ${mu}ell$ into yolk via air sac. After 48 hr and 96 hr incubation, the damage of the chick-embryo liver cell was investigated by electron microscopy and by measuring the concentration of lipid components (total cholesterol, free cholesterol, phospholipid and triglyceride). For eggs administered 10 $\mu\textrm{g}$ of DEN and incuvated 96 hr, in hepatocyte, the nucleus membrane was roughed, the size of nucleolus was apparently increased and euchromatin was accumulated. Mitochondria were condensed and cristae, located mitochondiral inner membrane, were obscured. Additionally, the leaves of triglyceride and cholesterol classes were significantly increased depend on the amount treated with 10 $\mu\textrm{g}$ DEN at 96 hr, but phospholipids component of cell membrane, were decreased with significance. As a conclusion, carcinogen induced hepatic lesion was correlated with the changes in lipid component of liver.

  • Total lipids extracted from the powdered seeds of Carum roxburghianum were fractionated into hydrocarbons (0.30%), wax esters (0.30%), sterol esters (1.35%), triacylglycerols (72.41%), free fatty acids (6.06%), 1,3-diacylglycerols (4.60%), 1,2- diacylglycerols (0.64%), glycolipids (5.10%), sterols (1.20%), 2-monoacylgylcerols (3.18%), 1-monoacylglycerols (1.46%), phosphatidylethanolamines (1.08%) phosphatidylcholines (0.40%), lysophosphatidylethanolamines (1.48%) and phosphatidylinositols (0.44%) with the help of TLC. The fatty acid composition of all the lipid fractions was determined after converting them into their methyl esters with $BF_3-methanol$ reagent and then analyzing them by GC. Oleic acid was found as a major component in all the lipid classes, whereas palmitic, linoleic and linolenic acids were present in lesser quantities. Arachidic acid was identified as a minor component in only seven out of twelve lipid classes.

Effects of the Feeding Mixture of Mushrooms and Vegetables Oils on the Lipid Component and Fatty Acid Composition of Liver in Rats (식용버섯과 식물성 유지의 혼합급여가 흰쥐 간장의 지질성분 및 지방산 조성에 미치는 영향)

  • This study was designed to observe the effects of the mixed diets of edible mushrooms and vegetables oils on the lipid component and fatty acid composition in liver of the diet induced hydpercholesterolemic rats. Ten groups of male S.D. rats were fed a basal diet supplemented with 5% of one of three mushrooms(G.I, L.e, A.j) and 10% of one of three vegetable oils (olive ,safflower perilla) for three weeks. In liver, total cholesterol concentration was significantly low in group 3 (olive oil 10 % + L. edodes 5%) and 6 (safflower oil 10 % $_2$L. edodes 5%) , triglyceride concentration was low in groups 8 (perilla oil 10 % + g. lucidum 5%) and 9 (perilla oil 10% + L. edodes 5%) and phospholipid concentration was significantly low in groups 3, 5, (safflower oil 10 % + G.lucidum 5%), 6, 7 (safflower oil 10 % + A .judae 5%) 8, 9 and 10 (perilla oil 10% + a. judae 5%). in the fatty acid composition of total lipid inliver, monounsaturated fatty acid (MUFA) concentration s were high in groups 2 (olive oil 105 + g. lucidum 5%), 3, and 4 (olive oil 10% + A. judae 5%) and all the perilla oil groups, polyunsaturated fatty acid (PUFA) and linoleic aicd concentrations were signifciantly high in all the safflower oil groups. In the fatty acid composition of liver phospholipid , PUFA concentrations were ghih but MUFA concentrations were low. In the triglyceride component, MUFA were some more than saturated fatty acid (SFA) . In the cholesteryl ester component, MUFa concentrations were significantly high. In the fatty acid composition of liver lipid components, linholeic acid was high in the PUFA and so it was major fatty acid. Eicosapentaenoic acid (EPA) of phospholipid component in liver was significantly high.

  • This study was performed to investigate the effects of lipid on cellular senescence, lipid peroxide production, and morphological changes. For this study we used primary skin fibroblasts from neonate rats grown in media various lipid contents. Fibroblasts were cultured until they lost their proliferation potential either in control medium (Dulbecco's modified Eagle's medium supplement with 10% fetal bovine serum) or in media supplemented with various concentrations of lipid-cholesterol rice component from bovine serum. Cumulative population doublings(CPD, as an index of cellular life span), and cellular thiobarbituric acid reactive substances (TBARS, as an index of lipid peroxide) concentrations were measured and morphological changes were observed. CPD were shortened with increasing lipid concentration in media ; 28.12 for cells grown in control medium and 13.42, 11.42, and 6.19 for those grown in 0.1%, 1% and 5% lipid rich components containing media, respectively. Cellular proliferation ratios were those grown in 5% lipid rich components containing media were delayed and they were degenerated soon. TBARS concentrations were increased with increasing concentration of lipid in media. Morphological changes were observed in cells grown in control medium by cellular senescence. Especially lipid droplets were observed in cells grown in 5% lipid rich components containing media. Therefore it seems that lipid contents in media had an effect on cellular proliferation and cellular life span, possibly via lipid peroxide production.

Lycium barbarum Polysaccharide Inhibits Lipid Oxidation and Protein Degradation in Tan Sheep Meatballs during Frozen Storage

  • The aim of the present study was to evaluate the effectiveness of Lycium barbarum polysaccharide (LBP) on lipid oxidation and protein degradation in Tan sheep meatballs during the frozen period. The meatballs were treated with LBP at 0.01%, 0.02%, and 0.03% and stored at -18±1℃ for 0, 3, 6, 9, and 12 weeks. The effects of LBP treatment were investigated using the contents of total volatile basic nitrogen (TVB-N), texture profile (TP), thiobarbituric acid reactive substances (TBARS), colour, and pH values, compared with 0.02% butylated hydroxytoluene treatment and the blank control. The results showed that LBP treatment significantly decreased TBARS content compared with the control, which confirmed LBP to be a highly effective component in preventing lipid oxidation of Tan sheep meatballs during frozen storage, and protein degradation in Tan sheep meatballs had a significant inhibition effect because of TVB-N value reduction. In addition, the colour, TP and pH values of meatballs treated with LBP were improved dramatically. To further determine the quality changes of the blank control and all treated groups during storage, the comprehensive score evaluation equation based on principal component analysis was obtained: Y=0.51632Y1+0.29589Y2 (cumulative contribution rate=81.221%), and the 0.02% LBP-treated group had a higher comprehensive score than the other groups, and the quality of LBP-treated meatballs was better as well. In summary, LBP may reduce or inhibit lipid oxidation and protein degradation, and enhance overall quality and shelf-life in prepared meat products.

Studies on the Lipid of Aquatic Products (Part 4) On the Flesh Lipid Composition of Cephalopods (수산물의 지질에 관한 연구 (제4보) -두족류의 근육지질성분에 대하여-)

  • Differences of lipids, especially total lipid composition, fatty acid and sterol composition of the flesh lipids between three species of cephalopods were investigated, since available researches concerning lipids in flesh tissues of the cephalopod are very limited. Extracted total lipid from the flesh tissues were fractionated by silicic acid column chromatography into three lipid classes of neutral lipids, glycolipids and phospholipids. The lipid compositions of total lipid and neutral lipids were estimated by the method of thin layer chromatography and TLC-scanner. The sterol compositions of unsaponifiable matters from total lipid were determined by using thin layer chromatography and gas-liquid chromatography. The fatty acid composition of each lipid class was also determined by gas-liquid chromatography. Total lipid contents of flesh tissues from three species of the cephalopods were 0.5 in Octopus vulgare, 0.8 in Octopus variabilis and $0.6\%$ in Loligo beka based on wet weight, the contents of total fatty acid in total lipid were 19.3, 47.8 and $38.4\%$, and the contents of unsaponifiable matters were 10.9, 18.8 and $41.1\%$, respectively. Total lipid was mainly composed of sterols and polar lipid-pigments as major components in each sample and the proportion of sterols and polar lipid-pigments to total lipid ranged from 27.0 to $35.5\%$ and 38.3 to $63.4\%$, respectively. The other lipid components of total lipid, e.g. triglycerides, free fatty acids, and carbohydrate-esterified sterols were determined as a minor components. The major component fatty acid in total lipid was palmitic acid and additionaly it chiefly consisted of the other unsaturated acids such as oleic, linoleic, octadecatetraenoic and eicosapentaenoic acid as major components of the acid. The compositions of sterol in three species of cephalopod were found to contain mainly cholesterol for its proportion to total sterols was 82.4 to $89.1\%$. However the other sterols such as 22-dehydrocholesterol and 24-methylenecholesterol were determined in addition to cholesterol as a minor components. The result of fractional composition of lipid class in total lipid was that total lipid had large .amount of polar lipid and small amount of nonpolar lipid i, e, neutral lipid in each sample, and the contents of phospholipid were higher than that of glycolipid in polar lipid. Neutral lipid was mainly composed of free sterol as major components in each sample and its proportion of free sterols to total neutral lipid was 50.0 to $70.5\%$. The other lipid components of neutral lipid showing similar in quantity, esterified sterols, free fatty acids and triglycerides were determined as a minor components. The major components fatty acid in neutral lipid were palmitic, oleic and hexadecadienoic acid. Palmitic acid was the most abundant and additionaly oleic, linoleic, octadecatetraenoic and myristic acid were the major component fatty acid in glycolipid. But, especially, glycolipid of Loligo beka contained a higher amount of arachidonic acid which also consists of major component in addition to those of acids. Palmitic acid was the most abundant and additionaly, oleic, linoleic and octadecatetraenoic acid were the major component fatty acids in phospholipid.
