Mortierella isabellina IFO 8183에 의한 ${\gamma}$-Linolenic Acid 생산 (Production of ${\gamma}$-Linolenic Acid by Mortierella isabellina IFO 8183)

  • Mortierella isabellina IFO 8183을 이용하여 ${\gamma}-linolenic$ acid를 생산하기 위한 최적 배양조건을 검토한 결과, 최적 탄소원과 최적 질소원은 각각 glucose와 $NH_4NO_3$이었으며, 지질 생산배지내의 최적 C/N율은 40이었다. 또한, 최적 초기 pH와 배양온도는 각각 6.0과 $30^{\circ}C$이었으며, 균체내에 축적된 지질의 지방산 조성은 pH와 온도에 의해 변화되었다. Sodium acetate를 배양 초기에 0.5%(w/v) 첨가하였을 때 대조구에 비해 ${\gamma}-linolenic$ acid 함량과 ${\gamma}-linolenic$ acid 생성량이 각각 15.4%와 37.8% 증가되었다. 최적 조건하에서 8일간 진탕배양하였을 때 균체수율, 지질수율, 지질함량, ${\gamma}-linolenic$ 함량 및 ${\gamma}-linolenic$생성량은 각각 0.347g/g, 0.18g/g, 0.52g/g, 60mg/g 및 347mg/l이었다.

서남해안 검정콩들의 단백질과 지방산 조성변이 (Seed Protein Content and Fatty Acid Composition of Black Seeded Soybeans Collected From Southwestern Islands)

  • 서남해안 부근의 섬에서 재배되고 있는 재래종 검정콩 유전자원 45품종(135점)의 조단백, 지방산 조성을 분석한 결과 조단백 함량 변이는 34.70-44.20%(평균 40.75%),지방산 조성 변이는 palmitic acid에서 9.00-14.40%(평균 11.41%) stearic acid에서 2.90-5.00%(평균 3.93%), oleic acid에서 22.25-26.50%(평균 22.75%), linoleic acid에서 50.30-57.20% (평균 53.55%), linolenic acid에서 6.70-11.20%(평균 8.35%)의 변이를 보였다. 지방산에 대한 유전력은 oleic acid에서는 높았으나 stearic acid와 linolenic acid에서는 낮았으며 palmitic acid, linoleic acid는 조단백의 유전력과 같은 경향으로 중정도 이었다. 형질상호간의 상관 계수에서 조단백은 oleic acid와는 고도의 유의차로 정의 상관을, linoleic acid와는 고도의 유의 차로 부의 상관을 보였고 기타의 지방산간에는 부의 상관을 보였다.

  • In order to study the influence of culture conditions on cell growth and lipid formation by Mucor sp, various carbon and nitrogen sources initial pH and C./N ratio of medium were investigated. Glucose was found to be suitable carbon source in terms of lipid yield and ${\gamma}$-linolenic acid(GLA) content. When NH4Cl and (NH4)2SO4 were used as nitrogen source lipid content was high(19-21%) but GLA content was low(15-17%) On the other hand when NaNO3 and KNO3 were used lipid content was low(about 13%) but GLA content was high(22-23%). The highest production of lipid was obtained at a C/N ratio of 40 using glucose and (NH4)2SO4 as carbon and nitrogen source respectively. it was found that lipid yield was high at pH4.6 Also this fungus did not grow at 35$^{\circ}C$ and lipid yield was highr at 15$^{\circ}C$ than $25^{\circ}C$.

Effects of α-Linolenic, Eicosapentaenoic and Docosahexaenoic Acids on the Content and Fatty Acid Composition of Brain Phospholipid in Rats

  • The effects of dietary n-3 fatty acids, ${\alpha}$-linolenic acid (18:3), eicosapentaenoic acid (EPA, 20:5), and docosahexaenoic acid (DHA, 22:6), on brain phospholipid content and fatty acid composition were compared in rats fed with a diet containing constant ratios of saturated fatty acid/monounsaturated fatty acid/polyunsaturated fatty acid (PUFA) and n-3/n-6. The dietary fat in each diet was added at the level of 10%. In each diet, n-3 PUFA comprised two-thirds of the PUFA and the remaining one-third was linoleic acid (18:2). Dietary fat containing linoleic acid as the sole source of PUFA was also given to the control group. The content of brain phospholipid in the three n-3 PUFA groups was significantly lower than that of the linoleic acid group. This reduction was greater in the EPA and DHA groups than in the ${\alpha}$-linolenic acid group. The decrease in phospholipid content in rats fed n-3 fatty acid-rich diets was largely due to the decrease in the phosphatidylethanolamine fraction. Each dietary n-3 PUFA was found to affect the fatty acid composition of brain phospholipids; the most pronounced alteration was observed in phosphatidylethanolamine fraction. Furthermore, the proportion of DHA in the phosphatidylethanolamine fraction tended to be higher in the DHA group than in other PUFA groups. In conclusion, dietary ${\alpha}$-linolenic acid, EPA and DHA can influence the phospholipid content, phospholipid subclass, and fatty acid composition in rat brain.

  • For a studyon the production of ${\gamma}$-linolenic acid(GLA) by fungi 3 fungal strains were isolated from soil. Their cell growth lipid content and fatty acid composition were compared in shake flask culture. Among these fungi the fungus designated as FA-007 has high lipid content(21.1%) and GLA content(15.6% of total fatty acids) The fungal strain FA-007 was tentatively identified as Mucor sp. on the basis of morphological characteristics, Fungal oil produced by this fungus was composed of 75.2% neutral lipid 5.3% glycolipid and 19.5% phospholipid. Although the GLA content in phospholipid was higher than it in neutral lipid the GLA content in neurtal lipid was high as 15.5%.

  • 120 fungal strains producing Y-linolenic acid (GLA) were isolated from 100 soil samples, and among these, the most suitable one for the production of GLA was identified as Fusarium sp. JK-02. The content of total lipid and dry cell weight was 620mg 1100m1 and 63.5mg 1100m1, respectively. The production of GLA was 10.2% of the total fatty acids.

Fatty Acid Content in Perilla Cultivars and Commercial Oils Determined by GC Analysis

  • The content analysis of fatty acids in Perilla cultivars and commercial oils is conducted through gas chromatography with a flame ionization detector. Results show that Perilla cultivars, such as Deulsaem and Daesil, contain high amounts of ${\alpha}-linolenic$ acid (262.22 and 261.97 mg/g, respectively). Among commercial oils, Perilla oil contains a higher amount of ${\alpha}-linolenic$ acid (515.20 mg/g). Accordingly, ${\alpha}-linolenic$ acid is a major fatty acid of Perilla cultivars and oil. Therefore, Perilla cultivars could be used as a food supplement for nutritional and pharmaceutical purposes.

식이 Linolenic Acid와 Linoleic Acid 함량변화가 흰주의 연령에 따른 지방대사 및 항혈전효과에 미치는 영향 (The Effect of Diet Varing Linolenic Acid and Linoleic Acid Content on Lipid Metabolism and Antithrombosis in Different Aged Rats)

  • This study was done to investigate the effect of age and dietary linolenic acid content and the linolenic acid/linoleic acid(LNA/LA) ratio on the lipid metabolism and formation of PGI2 and TXA2. The male Sprague-Dawley rats were fed 6 different with 0.2, 0.4, 0.6 of LNA/LA ratio within either 8% LNA(high LNA) or 4% LNA(low LNA) of fatty acid content for different feeding period(1, 4, 12 month). The dietary fat used were sesame oil, perilla oil, soybean oil and beef tallow. The concentration of serum total lipid, total cholesterol and HDL-C were increased with aging. Triglyceride concentration was decreased in 0.2 ratio of LNA/LA. The lipid content of liver showed similar tendency to that of serum. The ratio of PGI2/TXA2 was increased in 1 month rats and antithrombotic effect was reduced significantly with increasing age. The PGI2/TXA2 ratio was tended to be higher in diet of 0.2 and 0.4 LNA/LA ratio at high LNA level and in diet of 0.6 LNA/LA ratio at low LNA level. Especially PGI2/TXA2 ratio was increased linearly with rising LNA/LA ratio at low LNA level. It seemed that the LNA content and LNA/LA ratio had interaction to increase the antithrombotic effect bychanging TXA2 synthesis. And the dietary fatty acid related effect lowering the serum and liver lipid content, excepting triglyceride, was increased when dietary n3/n6 ratio was high(0.6) at both high and low n3 level. Therefore, it could not be recommended to consume large amount of n3 fatty acid or high ratio of n3/n6 to prevent cardiovascular diseases. These results suggested that the dietary fatty acid ratio of n3/n6 could be determined based on the n-3 content of dietary fat to reduce the risk of cardiovascular disease.

녹두의 주요 성분함량과 지방산 조성의 품종간 차이 (Varietal Differences of Major Chemical Components and Fatty Acid Composition in Mungbean)

  • 근래에 육성된 한국산 녹두품종에 대하여 녹두 용도별 가공 적성분류 기준이 될 수 있는 주요 일반성분 함량과 지방산 조성을 조사하여 품종 육성의 기초 자료로 제공하고자 수행하였던 바 그 결과를 요약하면 다음과 같다. 1. 평균 조단백질, 조지방 및 조회분 함량은 각각 24.3, 3.6%였는데 수분함량은 약 10% 정도였다. 2. 조단백질과 조탄수화물간, 종자수분과 조지방함량간, 종실중과 조지방함량간 등에서는 각각 고도의 부의 유의조관을 보였다. 3. 지방산조성 중 불포화지방산이 60∼67% 함유되었으며 종류는 oleic acid, linoleic acid, linolenic acid, arachidonic acid 등 4종이었고, 포화지방산이 33∼40%정도 함유되었는데 종류는 stearic acid, palmitic acid, behenic acid, lignoceric acid 등 4종이었다. 4. 지방산 조성 중 linoleic acid가 33.46%로 가장 많았으며, 다음이 linoenic acid와 palmitic acid로 각각 21.87, 21.72%였고, 다음이 stearic acid, oleic acid, arachidonic acid가 각각 5.98, 5.87, 4.87% 였으며 behenic acid와 lignoceric acid는 극히 미량 존재하였다. 5. Palmitic acid와 linoleic acid, oleic acid와 lignoceric, behenic acid간에는 고도의 정상관관계를, palmitic acid와 arachidonic, lignoceric acid간, oleic acid와 linolenic acid, arachidonic acid와 lignoceric acid간에는 고도의 부의 상관관계를 보였다. 6. 종실중과 oleic, arachidonic, behenic acid간에 고도의 정상관관계를 보였다.

Seed Protein Content and Fatty Acid Composition of Soybeans collected from Southwestern Islands in Korea

  • The 129 soybean genotypes were collected in 43 island locations from January to May 2001. Seeds of 129 genotypes collected were analyzed for crude protein and fatty acid composition contents. The crude protein content was averaged to 41.1 % and ranged from 37.4% to 44.4%. The average palmitic acid, stearic acid, oleic acid, linoleic acid and linolenic acid content were 12.0%, 4.0%, 23.2%, 55.5%, and 7.9%, and the ranges of those were 10.0% to 15.0%, 3.0% to 4.8%, 21.7% to 25.5%, 50.2% to 58.3% and 7.0% to 12.0%, respectively. Heritabiliries of palmitic acid, oleic acid, linoleic acid, and crude protein were higher, but that of stearic acid and linolenic acid were relatively lower. The genotypic correlation coefficients between crude protein and oleic acid showed highly positive correlation, but that of linoleic acid showed highly opsitive correlation, but that of linoleic acid showed highly negative correlation and also palmitic acid, stearic acid and linolenic acid showed negative correlation.
