Physiological Activities of Rubus coreanus Miq. Extracts Using Different Extraction Methods (추출방법에 따른 복분자 추출물의 생리활성)

  • This study was carried out to evaluate biological activities concerning extracts according to extraction methods from unripened fruit of Rubus coreanus Miq. The extraction methods were HWE (hot water extraction for 4 hr at $100^{\circ}C$), SFE (extraction for 3 hr at $40^{\circ}C$ under 300 bar, 100% of $CO_2$ fluid), USE (ultrasonification extraction for 4 hr at $50^{\circ}C$ with water), USE+HWE (hot water extraction for 2 hr at $100^{\circ}C$ after ultrasonification process for 2 hr), VE (vacuum extraction for 4 hr at $90^{\circ}C$ under 0.9 bar with water). VE extract showed the highest contents of total polyphenol ($178.78{\pm}3.79\;mg/g$) and total flavonoid ($40.93{\pm}0.68\;mg/g$). $IC_{50}$ values of DPPH radical scavenging activity, linoleic acid peroxidation inhibition activity and LDL (low density lipoprotein) oxidation inhibition activity of HWE extract showed the lowest $35.39{\pm}0.25{\mu}g/mL$, $12.61{\pm}0.31{\mu}g/mL$ and $1.31{\pm}0.02{\mu}g/mL$ among other all extracts, respectively. $IC_{50}$ values of ${\alpha}$-glucosidase inhibitory activities of VE and HWE extracts showed lower $14.34{\pm}0.20{\mu}g/mL$ and $15.83{\pm}0.20{\mu}g/mL$ than those of other extracts, respectively. Specifically, HWE and VE extracts have relatively better biological activities than other extracts; these could be potentially used as a bioactive source for health functional foods.

Antioxidant Effect of Histidine Containing Low Molecular Weight Peptide Isolated from Skipjack Boiled Extract (가다랑어 자숙액에서 분리한 히스티딘 함유 저분자 펩타이드의 항산화 효과)

  • This study was carried out to investigate the optimun conditions for the isolation of low molecular weight peptides containing histidine, and to evaluate the antioxidant effects of skipjack boiled extracts(SBE). The results are summarized as follows : L-histidine contents of the ordinary muscle and dark muscle extracts were $ 83.1{\pm}1.75{\mu}M/g\;and\;11.0{\pm}2.39\;{\mu}M/g$, respectively. The L-histidine level of the dark muscle was much lower than that of ordinary muscle in the SBE. The extracts were treated with alcalase and neutrase under different pH levels, temperatures, and times. The optimum hydrolysis conditions of SBE were pH 7.0 and a $60^{\circ}$C temperature for 2 hr in the batch reactor, which hydrolyzed 63% of the SBE. HPLC analysis showed a removing effect of the ultrafiltration permeate (UFP) to high molecular weight impurities in SBE. SBE and pure carnosine participated as inhibiting agents to, which was confirmed through the autoxidation processing of linoleic acid. UFP treatment improved the inhibiting ability of SBE to the autoxidation of linoleic acid. The reducing power of the UFP-treated ordinary muscle extracts were 10-fold higher than the dark muscle extracts, and 0.7-fold higher than 20 mM pure carnosine. The UFP-treated ordinary muscle extracts had greater reducing power activity than pure carnosine. The scavenging activities on DPPH radical of the different treated-SBE and pure carnosine were also investigated. Scavenging activities of the ordinary and dark muscle extracts and the pure carnosine were 90%, 70%, and 45%, respectively. In summary, Skipjack boiled extracts (SBE) demonstrated that low molecular weight peptides containing histidine are capable of inhibiting lipid oxidation. They also possessed effective abilities as free radical scavengers and reducing agents, and these activities may increase with increasing concentrations.

Quality Characteristics and Functional Properties of Ethanol Extract of Propolis (Propolis 에탄올 추출물의 품질특성 및 기능성)

  • Quality characteristics and functional properties of four kinds of propolis collected from 4 different regions were compared. Ethanol extract of propolis (EEP) showed high absorbance of UV at 208 ~ 212 nm and at 292~294 nm. Total flavonoid contents of 4 propolis were at a range of 16.30~21.87 mg/g However, no significant difference in the flavonoid content was observed among four kinds of sample. All EEP showed the high antimicrobial activities, especially against Salmonella typhimurium. Hydrogen donating activities of the EEP were high appear 65.94 to 78.67% in all samples. The EEP from Sunchon showed the highest activity of hydrogen donation among samples. All extracts had the higher nitrite-scavenging activity than 0.1% BHT. Compared to control, a significant decrease in lipid oxidation was found in all 4 extracts.

Changes in Chlorophylls and Carotenoids of Mustard Leaf Kimchi during Fermentation and Their Antioxidative Activities on the Lipid Oxidation (갓김치의 발효과정 중 Chlorophylls 및 Carotenoids의 변화와 동획분의 항산화성)

  • Changes of chlorophylls and carotenoids and antioxidative characteristics of crude chlorophylls and carotenoids(CCC) of mustard leaf kimchi(MLK) were investigated during the fermentation at 15$^{\circ}C$ for 25 days. At the beginning of fermentation chlorophylls were decreased rapidly, however, all chlorophylls and chlorophyllides were converted to pheophytins and pheophorbides at the final stage of fermentation. Total chlorophyll content remained constantly during the fermentation but 43.7% destruction of carotenoids was observed at 25 days of fermentation. Antioxidative activities of CCC from MLK against autoxidation of linoleic acid were significant(p<0.05) and much higher than those of $\alpha$-tocopherol. Fermentation period did not affect antioxidative activities of CCC of MLK. When the CCC of MLK was added to the autoxidation system of linoleic acid, degradation of total chlorophylls were observed to be slower than that of total carotenoids.

Analysis of Fatty Acid Compositions and Biodiesel Properties of Seeds of Woody Oil Plants in Korea (국내 목본 유지식물 종자의 지방산 조성 및 바이오디젤 특성 분석)

  • In order to evaluate their potential as sources of biodiesel, oil content and fatty acid composition of seeds and fatty acid methyl ester (FAME) properties from seven woody oil plants in Korea were analysed. The oil content of seed of all woody plant species ranged from 15.1 (Ligustrum lucidum) to 70.3% (Camellia japonica) by dry weight. Fatty acid composition consisted mainly of oleic acid, linoleic acid, linolenic acid, palmitic acid and stearic acid, with oleic acid being the most abundant. The content of unsaturated fatty acids of all species was higher than saturated fatty acids. Oxidation stability of seed oils of all woody plants ranged from 2.25 to 8.62 hours/$110^{\circ}C$. Fatty acid methyl ester of Styrax japonica has been found to have the highest iodine value, indicating that unsaturated fatty acid content is higher than other seed oils. Cold filter plug point(CFPP) was varied over a wide range from $0^{\circ}C$ to $-13^{\circ}C$. The cold fluidity of FAME of Chionanthus retusa were excellent.

  • Oxidations products of ${\alpha}-tocopherol$, ${\gamma}-tocopherol$ and ${\gamma}-tocotrienol$ in lipophilic reaction media were studied. ${\alpha}-Tocopherylquinone$, ${\gamma}-tocopherylquinone$ and ${\gamma}-tocotrienylquinone$ were fractionated using micro column, isolated and identified by HPLC and MS.

  • Polyphenolic compounds widely occurring in the traditional medicine plants have been reported to possess strong antioxidant activity. The antioxidative substances of water soluble extract from leaves, stem bark, root bark and fruit powder of Cudrania tricuspidata were tested in three different in vitro experimental models. In oxidation models using DPPH( , ' diphenyl picrylhydrazyl) method, Fe2+ induced linoleic acid peroxidation, and autooxidation of hepatic microsomal membranes, the antioxidative activities of water soluble extract from stem bark were strong than that from leaves and root bark. Fruits of Cudrania tricuspidata contained the highest amounts of polyphenolic compounds among the parts of this plant. The changes in polyphenolic compound contents of fruit powder caused by heat treatment (20oC, 40oC, and 60oC) were also monitored. After water blanching, contents of phenolic compounds was increased slightly in the following order; 20oC(1454mg), 40oC(1487mg), and 60oC(1511mg). These results supports that water soluble extracts from Cudrania tricuspidata contain antioxidative compounds.

  • Objectives : The verity extract of the Gagam-Hyungbang-Gihwang-tang (GHG) was assessed to determine the mechanisms of its antioxidant activity. Methods : The fellowing effects were measured : GHG exhibited a concentration-treatment; scavenging ${\alpha},\;{\alpha}-diphenyl-\beta-picrylhydrazyl$ (DPPH) radical, linoleic acid oxidation in a thiocyanate assay system, and superoxide anion, hydroxyl radical-induced DNA nicking. We investigated mRNA levels such as superoxide-dismutase. Results : The GHG extract showed dose-dependent free radical scavenging activity, including DPPH radicals, hydroxyl radicals, and superoxide anion, using different systems. The GHG was also found to be effective in protecting plasmid DNA against the strand breakage induced by hydroxyl radicals in Fenton's reaction mixture. Furthermore, SOD-1 mRNA expression levels increased in tat hepatoma H4IIE cells Conclusions : We expect that GHG will to helpful to the development of antioxidant activity treatments.

  • This study was conducted to investigate the effects of environment-friendly materials on garlic cultivation, and the methods for processing garlic at proper ripening stage to increase its antioxidant properties and market value. The functional properties of garlic and processed black garlic were analyzed in vitro and ex vivo. The ethanol extracts of garlic and black garlic were used in the antioxidant, linoleic acid autooxidation, lipid oxidation and RAW264.7 indicator cell using NO induced activity analyses. Also, toxicity tests by RBL-2H3 and RAW264.7 indicator cell using MTT assay were compared. Obesity inhibition test of garlic and black garlic were also studied.

Comparative Meat Qualities of Boston Butt Muscles (M. subscapularis) from Different Pig Breeds Available in Korean Market

  • This study aimed to determine the effects of breed on meat quality characteristics of porcine Boston butt muscles (M. subscapularis) from three different pig breeds: Landrace×Yorkshire×Duroc (LYD), Berkshire, and Ibérico available in Korean market. Ibérico showed significantly higher fat content, yellowness (CIE b⁎), cooking loss, and lower shear force values than LYD and Berkshire. Moreover, the contents of oleic acid (18:1) and palmitic acid (16:0) were significantly higher in Ibérico breed, but stearic acid (18:0) was higher in LYD. As linoleic acid (18:2) and arachidonic acid (20:4) were higher in Berkshire sows as compared to the other breeds, atherogenicity and thrombogenicity indexes were significantly lower in Berkshire sow. Ibérico had lower the ω-6/ω-3 fatty acids ratio, and higher taurine and free amino acids compared with the others. Ibérico also showed significantly greater lipid oxidation, lower antioxidant capacity, and higher hypoxanthine contents, whereas the Berkshire had higher inosine-5'-monophosphate and lower K-index value as compared to the Ibérico. The breed did not impart any significant effect on the size and density of muscle fibers. Thus, quality characteristics of Boston butt varied from breed to breed, and certain consumer preferences for Ibérico can be explained, in part, by the unique quality characteristics imparted by higher contents of intramuscular fat, oleic acid, and free amino acids.
