Regulation of cementoblast differentiation and mineralization using conditioned media of odontoblast (상아모세포의 조건배지를 이용한 백악모세포의 분화와 석회화 조절)

  • For the regeneration of periodontal tissues, the microenvironment for new attachment of connective tissue fibers should be provided, At this point of view, cementum formation in root surface plays a key role for this new attachment. This study was performed to figure out which factor promotes differentiation of cementoblast Considering anatomical structure of tooth, we selected the cells which may affect the differentiation of cementoblast - Ameloblast, OD11&MDPC23 for odontoblasts, NIH3T3 for fibroblsts and MG63 for osteoblasts. And OCCM30 was selected for cementoblast cell line. Then, the cell lines were cultured respectively and transferred the conditioned media to OCCM30. To evaluate the result, Alizarin red S stain was proceeded for evaluation of mineralization. The subjected mRNA genes are bone sialoprotein(BSP), alkaline phosphate(ALP) , osteocalcin(OC), type I collagen(Col I), osteonectin(SPARC ; secreted protein acidic and rich in cysteine). Expression of the gene were analysed by RT-PCR, The results were as follows: 1. For alizarin red S staining, control OCCM30 didn't show any mineralized red nodules until 14 days. But red nodules started to appear from about 4 days in MDPC-OCCM30 & OD11-OCCM30. 2. For results of RT-PCR, ESP mRNAs of control-OCCM30 and others were expressed from 14 days, but in MDPC23-OCCM30 & OD11-OCCM30 from 4 days. Like this, the gene expression of MDPC23-OCCM30 & OD11-OCCM30 were detected much earlier than others. 3. For confirmation of odontoblast effect on cementoblast, conditioned media of osteoblasts(MG63) which is mineralized by producing matrix vesicles didn't affect on the mineralized nodule formation of cementoblasts(OCCM30). This suggest the possibility that cementoblast mineralization is regulated by specific factor in dentin matrix protein rather than matrix vesicles. Therefore, we proved that the dentin/odontoblast promotes differentiation/mineralization of cementoblasts. This new approach might hole promise as diverse possibilities for the regeneration of tissues after periodontal disease.

The Implementable Functions of the CoreNet of a Multi-Valued Single Neuron Network (단층 코어넷 다단입력 인공신경망회로의 함수에 관한 구현가능 연구)

  • One of the purposes of an artificial neural netowrk(ANNet) is to implement the largest number of functions as possible with the smallest number of nodes and layers. This paper presents a CoreNet which has a multi-leveled input value and a multi-leveled output value with a 2-layered ANNet, which is the basic structure of an ANNet. I have suggested an equation for calculating the capacity of the CoreNet, which has a p-leveled input and a q-leveled output, as $a_{p,q}={\frac{1}{2}}p(p-1)q^2-{\frac{1}{2}}(p-2)(3p-1)q+(p-1)(p-2)$. I've applied this CoreNet into the simulation model 1(5)-1(6), which has 5 levels of an input and 6 levels of an output with no hidden layers. The simulation result of this model gives, the maximum 219 convergences for the number of implementable functions using the cot(${\sqrt{x}}$) input leveling method. I have also shown that, the 27 functions are implementable by the calculation of weight values(w, ${\theta}$) with the multi-threshold lines in the weight space, which are diverged in the simulation results. Therefore the 246 functions are implementable in the 1(5)-1(6) model, and this coincides with the value from the above eqution $a_{5,6}(=246)$. I also show the implementable function numbering method in the weight space.

PKHD1 Gene Silencing May Cause Cell Abnormal Proliferation through Modulation of Intracellular Calcium in Autosomal Recessive Polycystic Kidney Disease

  • Autosomal recessive polycystic kidney disease (ARPKD) is one of the important genetic disorders in pediatric practice. Mutation of the polycystic kidney and hepatic disease gene 1 (PKHD1) was identified as the cause of ARPKD. The gene encodes a 67-exon transcript for a large protein of 4074 amino acids termed fibrocystin, but its function remains unknown. The neoplastic-like in cystic epithelial proliferation and the epidermal growth factor/epidermal growth factor receptor (EGF/EGFR) axis overactivity are known as the most important characteristics of ARPKD. Since the misregulation of $Ca^{2+}$ signaling may lead to aberrant structure and function of the collecting ducts in kidney of rat with ARPKD, present study aimed to investigate the further mechanisms of abnormal proliferation of cystic cells by inhibition of PKHD1 expression. For this, a stable PKHD1-silenced HEK-293T cell line was established. Then cell proliferation rates, intracellular $Ca^{2+}$ concentration and extracellular signal-regulated kinase 1/2 (ERK1/2) activity were assessed after treatment with EGF, a calcium channel blocker and agonist, verapamil and Bay K8644. It was found that PKHD1-silenced HEK-293T cell lines were hyperproliferative to EGF stimulation. Also PKHD1-silencing lowered the intracellular $Ca^{2+}$ and caused EGF-induced ERK1/2 overactivation in the cells. An increase of intracellular $Ca^{2+}$ in PKHD1-silenced cells repressed the EGF-dependent ERK1/2 activation and the hyperproliferative response to EGF stimulation. Thus, inhibition of PKHD1 can cause EGF-induced excessive proliferation through decreasing intracellular $Ca^{2+}$ resulting in EGF-induced ERK1/2 activation. Our results suggest that the loss of fibrocystin may lead to abnormal proliferation in kidney epithelial cells and cyst formation in ARPKD by modulation of intracellular $Ca^{2+}$.

A Study on the Form of Modal Music used in K-Pop (K-Pop에서 사용된 선법음악 형태에 관한 연구)

  • According to a recent KOFICE survey, K-Pop's popularity factor is divided into visual and auditory elements, and the importance of hearing elements is found. Among the various types of singers, idol groups led K-Pop and Black Pink, which topped the list of favorite K-Pop singers in 2018, was selected as the study target. In addition, Twice's music, which is similar to Black Pink's period of activity, has been set as a comparative research scope. The common features in the music of the two groups are the chorus and rhythm structure implemented around melody and Harmony rhythm, and the frequency of use of Major and Minor is greatly different. Through this method and modal interchange, K-Pop from 2016 to 2018 is analyzed. Research shows that first, more than one mode is used as a main melody. When more than one mode is used, different lines in the same line will vary depending on the functional form of the song. Second, the rhythm of the melody expressed with vocals and instruments is repeated based on a two-bar pattern. Finally, when using less chord and slowly expressing the igneous rhythms, there is a way to omit or modify the third notes that determine chord characteristics. Through this study, we can see that the usage patterns of the mode and modal interchange are divided according to the major and minor.

Topology Optimization Application for Initial Platform Design of 10 MW Grade Floating Type Wave-wind Hybrid Power Generation System (10MW급 부유식 파력-풍력 복합발전 시스템 플랫폼 초기설계를 위한 위상최적화 응용)

  • This study aims to review a topology optimization based on finite element analysis (FEA) for conceptual design of platform in the 10MW class floating type wave-wind hybrid power generation system (WHPGS). Two topology optimization theories, density method (DM) and homogenization design method (HDM) were used to check which one is more effective for a simplified structural design problem prior to the topology optimization of platform of WHPGS. From the results of the simplified design problem, the HDM was applied to the topology optimization of platform of WHPGS. For the conceptual platform design of WHPGS, FEA model was created and then the structural analysis was performed considering offshore environmental loads at installation site. Hydrodynamics analysis was carried out to calculate pressure on platform and tension forces in mooring lines induced from the offshore environmental loads such as design wave and current. Loading conditions for the structural analysis included the analysis results from the hydrodynamic analysis and the weights of WHPGS. Boundary condition was realized using inertia relief method. The topology optimization of WHPGS platform was performed using the HDM, and then the conceptual arrangement of main structural members was suggested. From the results, it was confirmed that the topology optimization might be a useful tool to design the conceptual arrangement of main structural members for a newly developed offshore structure such as the floating type WHPGS.

Sequencing, Genomic Structure, Chromosomal Mapping and Association Study of the Porcine ADAMTS1 Gene with Litter Size

  • A disintegrin-like and metalloprotease (reprolysin type) with thrombospondin type 1 motif (ADAMTS1) plays a critical role in follicular rupture and represents a major advance in the proteolytic events that control ovulation. In this study, a 9,026-bp DNA sequence containing the full coding region, all 8 introns and part of the 5'and 3' untranslated region of the porcine ADAMTS1 gene was obtained. Analysis of the ADAMTS1 gene using the porcine radiation hybrid panel indicated that pig ADAMTS1 is closely linkage with microsatellite marker S0215, located on SSC13q49. The open reading frame of its cDNA covered 2,844 bp and encoded 947 amino acids. The coding region of porcine ADAMTS1 as determined by sequence alignments shared 85% and 81% identity with human and mouse cDNAs, respectively. The deduced protein contained 947 amino acids showing 85% sequence similarity both to the human and mouse proteins, respectively. Comparative sequencing of three pig breeds revealed one single nucleotide polymorphism (SNP) within exon 7 of which a G-C substitution at position 6006 changes a codon for arginine into a codon for proline. The substitution was situated within a PvuII recognition site and developed as a PCR-RFLP marker for further use in population variation investigations and association analysis with litter size. Allele frequencies of this SNP were investigated in seven pig breeds/lines. An association analysis in a new Qingping female line suggested that different ADAMTS1 genotypes have significant differences in litter size (p<0.01).

Performance Analysis of an Optical CDMA System for multi-user Environment (다중 사용자 환경에서의 광 CDMA 시스템 성능 분석)

  • In this paper, we implement an optical code division multiple access(OCDMA) system and analyze the performance of the implemented system. In the implemented system, a transmitter encodes input data into optical pulses by using laser diode, and spreads the encoded pulses in an encoder which consists of 4 stage delay lines. The decoder which is the same structure as that of encoder delays and combines the spreaded pulses, and thus recovers the original data. At first, we discuss the auto- and cross-correlations of OCDMA signals under both environments of single user and multi-users, and then verify the simulation results with experimental results. We also evaluate the effect of a number of stages of delay line and code length on the system performance through computer simulations. As experimental results we can see that if the decoder have the same configuration as that of encoder, the peak auto-correlation characteristics can be achieved, and thus we can recover the original data from received data. As simulation results we can see that although bit error rate decreases as code length decreases or the number of stage of delay line increases, it is difficult to implement the system because the pulse width becomes narrow. From the results, we can apply CDMA technologies to optical communication networks.

A Study on Cutting Pattern Generation of Membrane Structures Using Spline Curves (스플라인 곡선을 이용한 막구조물의 재단도 작성에 관한 연구)

  • For membrane structure, there are three main steps in design and construction, which are form finding, statistical load analysis, and cutting patterning. Unlike the first two stages, the step of cutting pattern involves the translation of a double-curved surface in 3D space into a 2D plane with minimal error. For economic reasons, the seam lines of generated cutting patterns rely greatly on the geodesic line. Generally, as searching regions of the seam line are plane elements in the step of shape analysis, the seam line is not a smooth curve, but an irregularly divided straight line. So, it is how we make an irregularly divided straight line a smooth curve that defines the quality of the pattern. Accordingly, in this paper, we analyzed interpolation schemes using spline, and apply these methods to cutting pattern generation on the curved surface. To generate the pattern, three types of spline functions were used, i.e., cubic spline function, B-spline, and least-square spline approximation, and simple model and the catenary-shaped membrane was adopted to examine the result of generation. The result of comparing the approximation curves by the number of elements and the number of extracted nodes of simple model revealed that the seam line for less number of extracted nodes with large number of elements were more efficient, and the least-square spline approximation provided smoother seam line than other methods.

Effects of Caffeic Acid Methyl Ester from Mentha arvensis Linne var. piperascens on Function of E6 Oncoprotein of HPV 16 (박하에서 분리한 Caffeic Acid Methyl Ester가 자궁경부암 바이러스 발암단백질 E6의 기능에 미치는 영향)

  • Cervical cancer is one of the leading causes of female death. Viral oncoproteins E6 and E7 are selectively retained and expressed in carcinoma cells infected with HPV (Human pa pilloma virus) type 16 and cooperated in immotalization and transformation of primary keratinocyte. E6 and E7 oncoproteins interfere the functions of tumor suppressor proteins p53 and retinoblasoma protein (pRb), respectively. Among a lots of natural products, Mentha arvensis Linne var.piperascens have inhibitory effects on bindings between E6 oncoprotein and tumor suppressor p53, E3 ubiqutin- protein ligase (E6AP). HPV oncoprotein inhibitors from Mentha piperita L. were isolated by solvent partition and column chromatography (Silica gel, RP-18) and inhibitory compounds were finally purified by HPLC using an ELISA screening system based on binding between E6 and E6AP. The aim of this study is to identify the structure of inhibitory compounds and to investigate whether these compounds have inhibitory effects on functions of E6 oncoprotein. We investigated whether caffeic acid methyl ester (CAM) extracted from Mentha piperita L. could inhibit the function of E6 oncoprotein. CAM inhibited the in vitro binding of E6 and E6AP which are essential for the binding and degradation of the tumor suppressor p53 and also inhibited the proliferation of human cervical cancer cell lines (SiHa and CaSKi) in a dose response manner. These results suggest that CAM inhibited the function of E6 oncoprotein, suggesting that it can be used as a potential drug for the treatment of cervical cancers infected with HPV.

Development of Line Density Index for the Quantification of Oceanic Thermal Fronts (해양의 수온전선 정량화를 위한 선밀도 지수 개발)

  • Line density index(LDI) was developed to quantify a densely isothermal line rate as standard index in the ocean environment. Theoretical background on the LDI development process restricting index range 0 to 100 was described. And validation test was done for the LDI application condition that total line length is not greater than 1/10 of unit area. NOAA SST(Sea Surface Temperature) data were used for the experimental application of LDI in the South Sea of Korea. Using GIS, $0.1^{\circ}C$ isothermal lines were linearized as vector data form SST raster data, and unit area were built as polygon data. For the LDI calculation, spatial overlapping(line in polygon) was implemented. To analyze the effect of unit area size for the LDI distribution, two cases of unit area size were designed and descriptive statistics was calculated including performing normality test. The results showed no change of LDI's essential characteristics such as mean and normality except for the range of value, variance and standard deviation. Accordingly, it was found that complex structure of thermal front and even smaller scale of front width than unit area size could influence on the LDI distribution. Also, correlation analysis performed between LDI and difference of temperature(${\Delta}T^{\circ}C$), and horizontal thermal gradient(${\Delta}T^{\circ}C/km$) on the front was obtained from linear regression model. This obtained value was compared with the results from previous researches. Newly developed LDI can be used to compare the thermal front regions changing spatio-temporally in the ocean environment using absolute index value. It is considered to be significant to analyze the relationship between thermal front and marine environment or front and marine organisms in a quantitative approach described in this study.
