In Vitro Anti-Bacterial and Anti-Inflammatory Effects of Trichosanthes Semen, Gardeniae Fructus, and Angelicae Dahuricae Radix Aqueous Extracts (과루인(瓜蔞仁), 치자(梔子), 백지(白芷)의 시험관내 항균 및 항염 효과)

  • Objectives: The object of this study was to observe the in vitro anti-bacterial and anti-inflammatory effects of three single aqueous herbal extracts(Trichosanthes Semen, Gardeniae Fructus, and Angelicae Dahuricae Radix), traditionally used for treating various gynecological diseases including mastitis in Korea, against Staphylococcus aureus and lipopolysaccharide(LPS)-activated Raw 264.7 cells. Methods: Anti-bacterial activities of three single aqueous herbal extracts against S. aureus were detected using standard agar microdilution methods. In addition, the effects on the cell viability, $PGE_2$, NO, TNF-${\alpha}$, IL-$1{\beta}$ and IL-6 productions of LPS activated Raw 264.7 cells. The anti-bacterial and anti-inflammatory effects were compared with lincomycin and piroxicam, respectively in the present study. Results: MIC of three single aqueous herbal extracts against S. aureus were detected as over 25, $4.063{\pm}2.096$ and $1.641{\pm}0.972$ mg/ml, respectively. MIC of lincomycin was detected as $0.469{\pm}0.297{\mu}g/ml$ at same conditions. In addition, $ED_{50}$ against LPS-induced cell viabilities and cytokine releases of three single aqueous herbal extracts were as follows - cell viability: 7.635, 13.761, 6.986 mg/ml, NO production : 4.808, 22.015, 2.949 mg/ml, $PGE_2$ production : 3.040, 4.312, 0.821 mg/ml, TNF-${\alpha}$ production : 9.563, 54.931, 1.240 mg/ml, IL-$1{\beta}$ production : 1.362, 1.801, 0.534 mg/ml, IL-6 production : 0.371, 0.797, 0.202 mg/ml, respectively. $ED_{50}$ of piroxicam against LPS-induced cell viabilities, NO, $PGE_2$, TNF-${\alpha}$, IL-$1{\beta}$ and IL-6 were detected as 35.179, 6.552, 1.162, 7.273, 7.101 and 5.044 ${\mu}g/ml$, respectively at same conditions. Conclusions: The results of the study showed anti-bacterial effects against S. aureus in the order of Gardeniae Fructus and Angelicae Dahuricae Radix aqueous extracts, except for Trichosanthes Semen. They also showed anti-inflammatory effects against LPS-activated Raw 264.7 cells in the order of Angelicae Dahuricae Radix, Trichosanthes Semen, and Gardeniae Fructus aqueous extracts. These three herbs are expected to be great substitutes to reduce side-effect of lincomycin and piroxicam, if the amount of those three single aqueous herbal extracts is adjusted appropriately.

In vitro antibacterial and synergistic effect of phlorotannins isolated from edible brown seaweed Eisenia bicyclis against acne-related bacteria

  • To develop effective and safe acne vulgaris therapies with a continuing demand for new solutions, we investigated unique efficacy of an antibacterial agent from marine brown alga Eisenia bicyclis in treating acne vulgaris. The methanolic extract of E. bicyclis exhibited potential antibacterial activity against acne-related bacteria. The ethyl acetate fraction showed the strongest antibacterial activity against the bacteria among solvent fractions. Six compounds (1-6), previously isolated from the ethyl acetate fraction of E. bicyclis, were evaluated for antibacterial activity against acne-related bacteria. Among them, compound 2 (fucofuroeckol-A [FF]) exhibited the highest antibacterial activity against acne-related bacteria with a minimum inhibitory concentration (MIC) ranging from 32 to $128{\mu}g\;mL^{-1}$. Furthermore, FF clearly reversed the high-level erythromycin and lincomycin resistance of Propionibacterium acnes. The MIC values of erythromycin against P. acnes were dramatically reduced from 2,048 to $1.0{\mu}g\;mL^{-1}$ in combination with MIC of FF ($64{\mu}g\;mL^{-1}$). The fractional inhibitory concentration indices of erythromycin and lincomycin were measured from 0.500 to 0.751 in combination with 32 or $64{\mu}g\;mL^{-1}$ of FF against all tested P. acnes strains, suggesting that FF-erythromycin and FF-lincomycin combinations exert a weak synergistic effect against P. acnes. The results of this study suggest that the compounds derived from E. bicyclis can be a potential source of natural antibacterial agents and a pharmaceutical component against acnerelated bacteria.

  • Enterotoxigenk E. coli is one of the major causative agents of the infantile diarrhea and traveler's diarrhea. The heat-labile enterotoxin is thought to be a virulence factor in the pathogenesis of the diarrhea and to be a marker for identification of the enterotoxigenic E. coli from non pathogenic E. coli. Therefore knowledge about the heat-labile enterotoxin is essential not only for understanding the pathogenesis but also for the diagnosis of the diarrhea. However the in-vitro heat-labile enterotoxin production is reported to be greatly affected by the cultural condition. In this regards, this study was designed to know the optimal conditions for the production of the heat-labile enterotoxin by assaying the permeability factor in the 18 hours culture supernatant of E. coli 08K25(B2) H9 and of E. coli 015 H11. Results obtained were summerized as follows: 1. Amounts of heat-labile enterotoxin produced were greater at initial pH 8.5 than at 7.0 of CYES-2 broth culture. However, the bacterial growth itself was more abundant at 7.0 than at 8.5. 2. Heat-labile enterotoxin per unit volume of culture supernatant was greater at shaking culture than at standing culture condition, but ratio of the enterotoxin produced over the unit mass of E. coli calculated was greater at standing culture than shaking culture condition, indicating that the greater yields of the toxin produced at shaking culture was due to increase in E. coli cell mass compared to the standing culture condition: 3. The enterotoxin produced in the lincomycin(128 microgram/ml) supplemented media was 5 or 11 times greater on the basis of enterotoxin per unit mass of E. coli, compared to the lincomycin-non-supplemented media, indicating that lincomycin itself increases the enterotoxin production. 4. Treatment of 18 hours culture of E. coli with polymyxin B(0.2 mg/ml) for 1 hour increased the yields of enterotoxin amounting to 2 or 5 times of the non-treated control cultures.

Characteristics of Occurrence of Pharmaceuticals in the Nakdong River (낙동강 중류수계에서 의약물질의 분포특성)

  • This study was suggested as fundamental data to control medical materials remained in Nakdong range gauge. The level of Iopromide detected in Nakdong mainstream was $0.0015{\sim}0.37{\mu}g/L$, Mefenamic acid $0.0087{\sim}0.056{\mu}g/L$, Diclofenac $N.D.{\sim}0.01{\mu}g/L$, Atenolol $N.D.{\sim}0.024{\mu}g/L$, Propranolol $N.D.{\sim}0.0038{\mu}g/L$, Lincomycin $0.0005{\sim}0.038{\mu}g/L$, and Trimethoprim $N.D.{\sim}0.0083{\mu}g/L$. At sewage disposal plant in the region, most of them were detected high levels of density. Especially, the level of Iopromide was found the highest up to $5.38{\mu}g/L$. At livestock wasted water disposal plant, the level of lincomycin was detected the highest figure of $477{\mu}g/L$. As a result, medical materials from Nakdong River mainstream got increasing the concentration due to inflow from sewage disposal plant in Gumi and River Geumho in Daegu, which affects residential and industrial areas significantly. Therefore, to control medical materials remained in Nakdong River efficiently, Geumho River and sewage disposal plants shall be continuously monitored and managed, which is recommendable.

Isolation and characterization of a Bacillus spp. for manufacturing the feed additives in livestock (가축의 보조사료 개발을 위한 Bacillus spp.의 분리 및 특성)

  • The aims of this study were to isolate spore-forming Bacillus strains that exhibit high digestibility and anti-pathogenic bacteria toward feed for calves. Total 136 spore-forming strains were isolated from finished feeds and their ingredients. Among them, 93 strains were identified as Bacillus species when analyzed by 16S rRNA sequencing. For industrial use, three strains named as Bacillus licheniformis SHS14, B. subtilis LCB7, B. amyloliquefaciens LCB10 were selected after evaluating the industrial standards that are related with heat and acid resistance, enzyme activities, and anti-pathogenic activities against Samonella dublin ATCC15480 and E. coli K99. After each culture, 3 selected strains were mixed together at 1:1:1 (v/v/v) ratio and then prepared as the mixed starter culture for feeding. The changes in microbial community were analyzed via 16S rRNA metagenomics. The initial community ratio among three strains was maintained even after manufacturing into final products. Also, in vitro, enzymatic and anti-pathogenic activities were almost same as those when cultured in single culture, and results of anti-pathogenic activities conducted with calves showed 90% activities against lincomycin, which would be indicative of a promising feed starter.

  • It is certainly a difficult work to find out novel-type "practical" antibiotic from natural sources. In fact, chemically modified antibiotics, such as synthetic penicillins, cephalosporins, derivatives of tetracycline, lincomycin, rifamycin and kanamycin are most widely used in medical practice at present. However, as the possibility of success gained by chemical modification is limited, it would be the buty of our applied microbiologists to offer novel antibiotics to chemist and physician without interruption. In this lecture, some practical problems in new antibiotic research will be discussed.(중략)

  • The optimal conditions for protoplast fusion between the cells of kanamycin resistant Lactobacillus bulgaricus IFO 13593 and those of lincomycin resistant Lactobacillus helveticus IAM 12090 were investigated in this study. The highest fusion frequency of 9.1*10-4 was obtained when protoplast fusion was carried out for 0.5 min using 40% PEG 4000 solution as a cell fusion stimulant and subsequent post-PEG-incubation was undergone at 30 for 30 min in the PPI medium.

  • The rational approach to antimicrobial therapy of infected root canals is based on accurate identification of the infecting organism and on the organism's susceptibility to antimicrobial agents as measured by standardized techniques. In establishing criteria for the selection of antibiotics, a susceptibility test should be performed. The purpose of this study was to investigate the susceptibility of 224 aerobic and anaerobic microbial strains isolated from infected root canals to various antibiotics. This was performed by using 7 antibiotic sensi-disc: Penicillin (10 units), Ampicillin (10 mcg), Tetracycline (30 mcg), Streptomycin (10 mcg), Kanamycin (30 mcg), Lincomycin (2 mcg), and Clindamycin (2 mcg). The results were as follows; 1. Strains isolated from infected root canals was shown to be most susceptible to Clindamycin, while Streptomycin exhibited least antibacterial properties. 2. Anaerobes were found to be susceptible to Penicillin, Ampicillin, and Clindamycin. 3. Streptomycin and Kanamycin were shown to be effective against Staphylococcus aureus and Staphylococcus epidermis, however, unidentified G(+) cocci organisms were found to be resistant to these agents. 4. Bifidobacterium sp. was susceptible to Lincomycin while G(+) rods were resistant to it. 5. Staphylococcus aureus, Micrococcus sp., and anaerobes were highly susceptible to Clindamycin. 6. All of the antibiotics tested were shown to be very effective against Eubacterium sp.

  • - Protoplast fusion between lincomycin resistant Lactobacillus casei KCTC 1121 and rifarnpicin resistant Lactobacillus delbrueckii JK-414 was attempted to obtain the improved strains. Protoplasts of L. casei and L. delbrueckii were produced by mutanolysin digestion at $42^{\circ}C$ for 15 min. L. casei cells were converted to protoplasts by treating with 5 $\mu g$ / m l of mutanolysin in 20 mM HEPES buffer (pH 7.0) containing 0.75 M sucrose at the middle logarithmic growth phase. In case of L. delbrueckii 1.0 M sucrose was used osmotic stabilizer. Regeneration of protoplast in both strains was efficiently accomplished on the regeneration medium containing 10% sucrose, 6 mM $MgC1_2, 6 mM CaC1_2$, and 2.5% gelatin. Protoplast fusion between L. casei and L. delbrueckii was carried out in the presence of 40% of PEG 4,000. The frequency of protoplast fusion was found to be about $3.2\times 10^4$. Acid production of L. casei was better than that of L. delbrueckii. Among fusants, F23 and F35 exhibited excellent lactic acid production. F23 and F24 exhibited the improved proteolysis compared to that of the parent strains and they had twice as much as DNA content of the parents.

Production of extracellular protein from Bacillus sp. WY-60 (Bacillus sp. WY-60에 의한 균체외 단백질의 분비조건)

  • A bacteria strain producing extracellular protein was isolated and identified from soil samples, and the optimum conditions of producing protein were investigated. Eleven strains of bacteria were isolated from soil samples. Among which WY-60 strain showed a very strong capability of producing protein and identified as a Bacillus sp. The optimum composition of nutrient medium for the production of the protein by WY-60 was fructose 4.0%, polypeptone 1.0%, $NH_4NO_3$ 0.1%, $K_2HPO_4$ 0.1%, $MgSO_4{\cdot}7H_2O$ 0.005%, $CaCO_3$ 1.0% and the optimum pH and temperature were 8.0 and $30^{\circ}C$, respectively. The penicillin G and lincomycin added to the above medium were effective for the protein production of the WY-60, but other antibiotics were non-effective. The maximum production of protein was obtained after 5 days culture.
