Changes of the growth plate in children: 3-dimensional magnetic resonance imaging analysis

  • Purpose: This pilot study assessed changes in the growth plate and growth rates in children during a 6-month period. Methods: The study included 31 healthy children (17 boys, 14 girls) under evaluation for growth retardation. Height, weight, bone age, insulin like growth factor-1 (IGF-1), and insulin like growth factor binding protein 3 (IGF-BP3) were measured at baseline and after 6 months. In addition, the diameter, thickness, and volume of the femoral and tibial growth plates were measured using magnetic resonance imaging. Results: The mean bone age in boys and girls was 11.7 and 10.7 years, respectively. In boys, height (z score) (-0.2 vs. 0.0), weight (z score) (0.8 vs. 1.1), body mass index (BMI) (z score) (1.27 vs. 1.5), IGF-1 (ng/mL) (343.6 vs. 501.8), and IGF-BP3 (ng/mL) (5,088.5 vs. 5,620.0) were significantly higher after 6 months. In girls, height (z score) (-1.0 vs. -0.7), weight (z score) (-0.5 vs. 0.1), BMI (z score) (-0.02 vs. 0.3), IGF-1 (ng/mL) (329.3 vs. 524.6), and IGF-BP3 (ng/mL) (4,644.4 vs. 5,593.6) were also significantly higher after 6 months. In both sexes, the mean diameter and volume of the femoral and tibial growth plates were significantly increased 6 months later. Conclusion: No significant correlation was found between changes in the growth plate and clinical parameters in children with growth retardation in this study, other than correlations of change in femoral diameter with weight and BMI. A larger, long-term study is needed to precisely evaluate the correlation between change in the growth plate and growth.

An Herbal Medicine Mixture (HM-10) Induces Longitudinal Bone Growth and Growth Hormone Release in Rats

  • To investigate the growth promoting effects of an herbal medicine formulation (HM-10), Sprague Dawley (SD) male rats (3 weeks old) were divided into 3 groups (8 rats/group). The control group was given a daily oral administration of saline, and the treatment groups, HM-1 and HM-2, were given daily administrations of HM-10 (500 and 1,000 mg/kg BW, respectively). The cumulative tibial bone growth of the HM-1 and HM-2 groups (22.5 and 20.8 mm, respectively), and their cumulative femur bone growth (19.4 and 18.2 mm, respectively), were significantly different compared to the control group (7.5 mm of tibial growth and 7.7 mm of femur growth) (p<0.05). Lastly, the growth hormone levels of the HM-1 and HM-2 groups (1.70 and 1.79 ng/mL, respectively), as well as their insulin like growth factor 1 (IGF-1) levels (165.1 and 171.7 ng/mL, respectively) showed significant differences compared to the control (0.93 ng/mL of growth hormone and 125.6 ng/mL of IGF-1) (p<0.05).

Change of Serum Progesterone and Insulin-like Growth Factor-1 Concentration during the First Oestrus in the Heifers of Hanwoo (한우 초발정 전후 혈중 Progesterone 및 Insulin-like Growth Factor-1 농도의 변화)

  • This study was conducted to determine changes in circulating progesterone and Insulin-like Growth Factor-1(IGF-1) concentrations according to daily gain of 0.5 and 0.7 kg ; from 33 weeks of age until the onset of regular oestrus cycles in the heifer of Hanwoo. In all animals, progesterone concentrations was undetectable until just before first oestrus. A small progesterone elevation of approximately 6 days duration(based upon the mean progesterone profile) preceded the first oestrus in 13 heifers. The mean age at first oestrus of the daily gain 0.7 kg(331.0$\pm$ 15.0 days) group was earlier than those of the daily gain 0.5 kg(358.9$\pm$7.9 days) group, and the mean weight at first oestrus of the daily gain 0.7 kg(236.0$\pm$4.7 kg) group was heavier than those of the daily gain 0.5 kg(224.8$\pm$9.7 kg) group. IGF-1 concentrations from day 3 to 15 of first oestrus were higher than those of the earlier luteal stage(Day 0~1) and the luteal regression stage (Day 18~20). IGF-1 concentrations according to the growth stage (from 33 weeks to 57 weeks) increased gradually. On the basis of these results, we suggest that the onset of first oestrus in Hanwoo heifer is about 345 day of age, and progesterone concentrations are closely related to the IGF-1 concentration.

Effect of Dietary Energy Level on Nutrient Utilization, Insulin-like Growth Factor-I and Insulin-like Growth Factor Binding Protein-3 in Plasma, Liver and Longissimus dorsi Muscle in Growing-finishing Pigs Using Soybean Oil as an Energy Source

  • Two experiments were carried out to study the effects of dietary energy level on nutrient digestion, nitrogen (N) utilization, growth performance, insulin-like growth factor-I (IGF-I), and insulin-like growth factor binding protein-3 (IGFBP-3) in plasma, liver and longissimus dorsi muscle in growing-finishing pigs. In experiment 1 (Exp 1), 15 castrated male pigs (Duroc${\times}$Landrace${\times}$Large White) (Body weight, BW, 55.6${\pm}$1.8 kg) were divided into three groups and fed rations containing 13.33, 14.87 and 17.35 MJ digestible energy (DE)/kg as treatments I, II and III, respectively, using soybean oil as an energy source. The experiment lasted 8 days and faecal and urinary samples were collected during the last 3 days. The results showed that the digestibility of dry matter (DM), energy and N was increased from treatments I to III (p<0.01). N-retention and N-retention rate were not influenced by dietary DE level (p>0.05). In experiment 2 (Exp 2), 36 female pigs (Duroc${\times}$Landrace${\times}$Large White) (BW 41.5${\pm}$3.8 kg) were divided into three groups. The pigs were fed with the same three rations used in Exp 1 for 60 days. At the end of Exp 2, eight pigs were selected from each group for blood sampling and 4 pigs for slaughter trial. The results indicated that average daily feed intake (ADFI) and N-intake were significantly decreased (p<0.01), and DE intake (p<0.01) and average daily gain (ADG) (p<0.05) were increased. IGF-I and IGFBP-3 in plasma were increased (p<0.05). No significant differences in IGF-I and IGFBP-3 in liver and longissimus dorsi muscle were found between different treatments. It was concluded that higher dietary DE level improved nutrient digestibility, ADG and feed/gain ratio when soybean oil was used as an energy source in the ration of growing-finishing pigs. No significant differences were found in Nretention and IGF-I and IGFBP-3 in liver and longissimus dorsi muscle between different treatments.

Investigation of the Insulin-like Growth Factor System in Breast Muscle during Embryonic and Postnatal Development in Langshan and Arbor Acres Chickens Subjected to Different Feeding Regimens

  • Nutrient availability may control muscle growth directly and indirectly through its influence on regulatory factors. We analyzed the effects of nutrient availability on the breast muscle insulin-like growth factor system. Real time RT-PCR was used to quantify the level of transcription in breast muscle from Langshan (LS) layer and Arbor Acres (AA) broiler chickens subjected to different feeding regimens during embryonic and postnatal development. The AA chickens were fed AA diet (AA, control group) while the LS chickens were either fed LS diet (LL) or AA diet (LA). According to our results, insulin-like growth factor (IGF)-II (embryonic day 16 (E16) - postnatal day 42 (P42)), IGF-I receptor (IGF-IR, E18-P42), and IGF binding protein (IGFBP)-2 (E18-P42), -5 (E16-P14), -7 (E12-P0), and -3 (E12-P0) were positively correlated with IGF-I, while IGFBP-3 (P0-P28) was negatively correlated with IGF-I. In comparison, IGF-IR (E18-P42), IGFBP-2 (E18-P42), IGFBP-5 (E14-P0), and IGFBP-3 (E16-P0) were positively correlated with IGF-II, while IGF-IR (E10-E16) and IGFBP-3 (P0-P28) were negatively correlated with IGF-II. Moreover, IGFBP-2 (E16-P42), -7 (E10-E16), and -3 (E10-E16) were positively correlated with IGF-IR, while IGFBP-3 (P0-P28) was negatively correlated with IGF-IR. Finally, IGFBP-7 (E12-P0) was positively correlated with IGFBP-3, while IGFBP-2 (P0-P28) and -7 (P0-P42) were negatively correlated with IGFBP-3. Overall, the AA chickens exhibited higher levels of IGF-I, IGF-IR, and IGFBP-2 mRNA expression than the LL chickens, while the opposite was true for IGFBP-7. No strain differences in IGF-I, IGF-IR, and IGFBP-7 mRNA expression were detected between LA and AA chickens; however, a strain difference was observed for IGFBP-2. LA chickens exhibited higher levels of IGFBP-2 than LL chickens, while the opposite was true for IGFBP-7. Our data show the first evidence that certain genes may be correlated during specific developmental periods and that strain differences in the expression of those genes in LS and AA chickens are due to differential responses to the same diet.

Partial Purification and Quantification of Insulin-like Growth Factor-I from Red Deer Antler (녹용으로부터 Insulin-like Growth Factor-I의 일부정제 및 정량)

  • Deer antler tissue contains the most rapidly growing bone in the animal kingdom. Thus, it is likely that growing antler tissue is a rich source of local paracrine bone-stimulating factors. Growth factors, at least the insulin-like growth factor (IGF), control the bone-remodelling process. In this study, we tried to isolate and purify IGF-I from fresh antler tissue by the routine isolation and purification of protein. The purification involved ammonium sulfate precipitation, DEAE-Sepharose CL-60 ion-exchange chromatography, CM-Sepharose CL-6B ion-exchange chromatography, and Sephadex G-50 chromatography. Purified fractions from each step were analyzed by high-performance liquid chromatography (HPLC), SDS polyacrylamide gel electrophoresis (SDS-PACE), Dot-blot, and Western-blot methods. Furthermore, the quantification of partially purified IGF-I was calculated by enzyme-linked immunosorbent assays (ELISA) using antibody to human recombinant IGF-1. SDS-PAGE analysis of the final fraction yielded two molecular bands and the signal band was at 12 kDa on the Western-blot film. This purified IGF-I fraction showed a peak at retention time of eight min. The quantity of IGF-I in 20 g deer antler tissue as starting weight was calculated using a standard curve to be 2910 ng/ml, and total IGF-I amount is 0.291 g. The results show that IGF-I, which can be found in deer antler, can be partially purified and quantified by classic protein isolation methods.

Influence of Refeeding of Protein, Carbohydrate and Fat on Hepatic Insulin-Like Growth Factor-I mRNA Level in Fasted Chicks

  • The influence of refeeding either protein, carbohydrate or fat on hepatic insulin-like growth factor-I (IGF-I) mRNA level in chicks which had been fasted for 2 days was examined. The hepatic IGF-I mRNA was measured by ribonuclease protection assay. Fasting reduced hepatic IGF-I mRNA levels to less than half of those in the fed control. When chicks were refed either a control, protein or carbohydrate diet, IGF-I mRNA levels significantly increased to those in the fed control until 2 hours of refeeding. Refeeding of fat did not alter hepatic IGF-I mRNA levels. The significant correlation between liver weight and hepatic IGF-I gene expression suggests that when chicks are refed after 2-d fasting, the acute increase in hepatic IGF-I gene expression brought about after refeeding may be partly regulated by the increase in liver protein metabolism.

  • It is well known that bacterial lipopolysaccharide (LPS) stimulates the prostaglandin (PG) synthesis in various experimental system, but the mechanism and the detailed nature of its action are yet to be understood. Thus, this study was designed to characterize LPS induced PG synthesis in rat alveolar macrophage. Although results were not so much prominent, LPS stimulated PGE2 synthesis in macrophage with short term exposure, and this was thought to be mainly due to the activation of phopholipase A2+ But there was a burst in the PG synthesis 6 hours after the LPS treatment and this was accompanied with the increase of cyclooxygenase activity. This effect was not mediated by tumor necrosis factor (TNF) or platelet activating factor (PAF), and the existence of serum was prerequisite for its action. Growth factors such as epidermal growth factor (EGF) and platelet derived growth factor (PDGF) themselves did not stimulate PG synthesis and the showed stimulatory activities to some extent. Normal rat serum was more effective for the elicitation of the LPS action than growth factors. Thus, considering the amounts of growth fafctors contained in normal serum, it was suggested that another factors like LPS binding protein (LBP) might be involved in the serum effect on LPS action. Conclusively. it was thought that LPS could stimulate PG synthesis through interaction with serum factors such as EGF, PDGF and/or LBP.
